  • The purification of indole from 54.3wt% indole fraction (temperature range of distillate: $250{\sim}255^{\circ}C$) recovered by extraction-distillation combination of coal tar fraction (temperature range of distillate: $240{\sim}265^{\circ}C$) was examined by solute crystallization. The feed consists of eight components such as quinoline, iso-quinoline, indole, quinaldine, 1-methylnaphthalene, 2-methylnaphthalene, biphenyl and phenyl ether. Hexane and an aqueous solution of methanol (50 : 50 vol%) were used as the crystallization solvent and the coolant, respectively. A batch stirred tank of glass material was used as a crystallization apparatus. By increasing the operation temperature and the volume ratio of solvent to feed at initial, the purity of indole increas ed, but yields of indole showed a decreasing tendency. Solute crystallization method using hexane as a solvent was excellent because the purity of 99.3 wt% indole was recovered at the yield of 50% without washing operation.

Effects of 3,3′,4,4′,5-pentachloro biphenyl (PCB126) on the Expression the Tight Junction Genes in Cultured Mouse Neonatal Testis

  • In an effort to uncover the spermatogenic impairment by the polychlorinated biphenyls (PCBs), the expression of tight junctions (TJs) genes important for the formation of the blood testis barrier (BTB) were examined following the 3,3',4,4',5-pentachloro biphenyl (PCB126) treatment in cultured neonatal testis in mice. At 4 days (D4) after 10 and 100 nM PCB126 treatment the expression of claudin-11 was significantly increased when compared with vehicle control. In contrast no difference in occludin and claudin-1 expression was found among the experimental group. On D8, 100 nM PCB126 significantly increased the expression of claudin-11 but not occludin and claudin-1. 1 uM PCB126 treatment significantly decreased expressions of occludin and ciaudin -1, suggesting the general toxic effect on the Sertoli cell. Because PCB126 does not alter the proliferative activity of spermatogenic cells and Sertoli cells in neonatal testis, it is likely that increase in the expression of claudin-11 by low dose of PCB126 may attribute to the alteration of the Sertoli cells differentiation in testis. It also emphasized that PCB126 might have differentially affected the transcription of TJ genes in Sertoli cells. In conclusion, this result suggests that the structure of TJ may be targeted by PCB126 in neonatal testis in mice and that co-PCB is potentially harmful to spermatogenesis by alteration of the development of BTB.

The Luminance Characteristics of Organic ELD Based on Znq2 and TPD (Znq2와 TPD에 기초한 유기 ELD의 발광 특성)

  • The Bis(8-oxyquinolino) zinc lII (Znq2) were synthesized successfully from zinc chloride $(ZnCl_2)$ as a initial material . The organic electroluminescece devices (ELDs) were fabricated with N-N'-diphenyl-N-N'-bis (3-meth-ylphenyl)-1,1'-biphenyl-4,4'-diamine (TPD) which act as a hole transporting layer and the Znq2 act as an EL emitting layer and electron transporting layer. In order to maximize luminance of ELD, TPD/Znq2/Al were deposited onto cleaned indium tin oxide (ITO) by changing thickness of EL emitting layer. The photoluminescence (PL) results show that Znq2 compound emits yellow green from 540nm. electrochemical behavior with V-J and V-L curve of carrier injection was investigated from 6 V. respectively. The maximum luminance were defected about $838 cd/m^2$. From these results, ai synthesized Znq2 material maybe one of the useful material of organic EL display material.

Activity and Selectivity in Low Temperature for Dibenzothiophene Hydrodesulfurization based Zeolite Support (제올라이트 담체상의 디벤조티오펜 수첨탈황반응에서 저온활성 및 선택성)

  • Two types of CoMo/zeolite as well as $NiMo/{\gamma}-Al_2O_3$ were prepared and their activities and selectivities of low-temperature dibenzothiophene(DBT) hydrodesulfurization(HDS) were studied in high pressure fixed bed reactor. The HDS activities of CoMo/zeolites were higher than that of $NiMo/{\gamma}-Al_2O_3$ at temperatures below $225^{\circ}C$ while they were lower than that of $NiMo/{\gamma}-Al_2O_3$ at temperatures higher than $275^{\circ}C$. The main products from $NiMo/{\gamma}-Al_2O_3$ were biphenyl and cyclohexylbenzene. The product distribution of CoMo/zeolite catalysts was different from that of $NiMo/{\gamma}-Al_2O_3$. It is speculated that DBT is converted to alkylcyclohexane over zeolite based catalysts through both alkylation and hydrogenation reactions. The crystal structure of molybdenum was $MoO_3$ in fresh zeolite support while mixtures of $MoO_3$ and $MoS_2$ were observed in the aged catalyst.

Detection of Biodegradative Genes in Oil Contaminated Soil Microbial Community by Oligonucleotide Microarray (Oligonucleotide Microarray를 이용한 유류 오염 토양 미생물 군집내 난분해성 화합물 분해 유전자의 검출)

  • The analysis of functional population and its dynamics on the environment is essential for understanding bioremediation in environment. Here, we report a method for oligonucleotide microarray for the monitoring of aliphatic and aromatic degradative genes. This microarray contained 15 unique and group-specific probes which were based on 100 known genes involved pathways in biodegradation. Hybridization specificity tests with pure cultures, strain Pseudomonas aeruginosa KCTC 1636 indicated that the designed probes on the arrays appeared to be specific to their corresponding target genes. It was found that the presence of 8 genes encoding alkane, naphthalene, biphenyl, pyrene (PAH ring-hydroxylating) degradation pathway could be detected in oil contaminated soil sample. Therefore, the findings of this study strongly suggest that oligonucleotide microarray is an effective diagnostic tool for evaluating biodegradation capability in oil contaminated subsurface environment.

Purification and Characterization of 2,3-Dihydroxybiphenyl 1,2- Dioxygenase from Comamonas sp.

  • A genomic library of biphenyl-degrading strain Comamonas sp. SMN4 was constructed by using the cosmid vector pWE15 and introduced into Escherichia coli. Of 1,000 recombinant clones tested, two clones that expressed 2,3-dihydroxybiphenyl 1,2-dioxygenase activity were found (named pNB 1 and pNB2). From pNB1 clone, subclone pNA210, demonstrated 2,3-dihydroxybiphenyl 1,2-dioxygenase activity, is isolated. 2,3-Dihydroxybiphenyl 1,2-dioxygenase (23DBDO, BphC) is an extradiol-type dioxygenase that involved in third step of biphenyl degradation pathway. The nucleotide sequence of the Comamonas sp. SMN4 gene bphC, which encodes 23DBDO, was cloned into a plasmid pQE30. The His-tagged 23DBDO produced by a recombinant Escherichia coli, SG 13009 (pREP4)(pNPC), and purified with a Ni-nitrilotriacetic acid resin affinity column using the His-bind Qiagen system. The His-tagged 23DBDO construction was active. SDS-PAGE analysis of the purified active 23DBDO gave a single band of 32 kDa; this is in agreement with the size of the bphC coding region. The 23DBDO exhibited maximum activity at pH 9.0. The CD data for the pHs, showed that this enzyme had a typical a-helical folding structures at neutral pHs ranged from pH 4.5 to pH 9.0. This structure maintained up to pH 10.5. However, this high stable folding strucure was converted to unfolded structure in acidic region (pH 2.5) or in high pH (pH 12.0). The result of CD spectra observed with pH effects on 23DBDO activity, suggested that charge transition by pH change have affected change of conformational structure for 23DBDO catalytic reaction. The $K_m$ for 2,3-dihydroxybiphenyl, 3-metylcatechol, 4-methylcatechol and catechol was 11.7 $\mu$M, 24 $\mu$M, 50 mM and 625 $\mu$M.

  • In this paper, the solvent sensing properties of the metallophthalocyanine macrocyclic compounds(ZnPc) have been deposited as thin films by the spin-coated method and evaporated methods onto alumina substrates and quartz substrates. And then the spin-coated materials of Zinc phthalocyanine solutions blended with $N,N'-diphenyl-N,N'-bis(1-naphthyl)-1,\;1-biphenyl-4,4'-diamine\;and/or\; Poly[2-methoxy-5-(2'-ethylhexyloxy)-1,4-phenylene-vinylene]$ solutions. The influences of the blended metallophthalocyanine macrocyclic compounds on the resistance have been measured and analysed in five different vapour organic compounds.

Reductive Dechlorination of Low Concentration Polychlorinated Biphenyls as Affected by a Rhamnolipid Biosurfactant

  • We investigated whether the threshold concentration for polychlorinated biphenyl (PCB) dechlorination may be lower in biosurfactant-amended sediments compared with biosurfactant-free samples. At PCB concentrations of 40, 60, and 120 ppm, the surfactant amendment enhanced the PCB dechlorination rate at all concentrations and the rate was also faster at higher concentrations. On a congener group basis, dechlorination proceeded largely with group A (congeners with low threshold) in both surfactant-free and -amended sediments, accumulating mainly group C (residual products of dechlorination) congeners, and surfactant enhanced the dechlorination rate of group A congeners. Since the PCB threshold concentration for the inoculum in the experiment was lower than 40 ppm, we carried out another experiment using sediments with lower PCB concentrations, 10, 20, and 30 ppm. Sediments with 100 ppm were also performed to measure dechlorination at a PCB saturation concentration. Comparison between the plateaus exhibited that the extent of dechlorination below 40 ppm PCBs was much lower than that at a saturation concentration of 100 ppm. There was no significant difference in the extent of dechlorination between surfactant-free and -amended sediments. Moreover, surfactant did not change the congener specificity or broaden the congener spectrum for dechlorination at PCB concentrations below 40 ppm. Taken together, it seems that at a given PCB concentration, dechlorination characteristics of dechlorinating populations may be determined by not only the congener specificity of the microorganisms but also the affinity of dechlorinating enzyme(s) to individual PCB congeners.
