Arsenic (As) is one of the heavy metals which causes acute bio-toxicity even at low concentration and has disastrous effect on environment. In some countries, As contamination has become alarming and increasing day by day as consequences of unsustainable management practices. Many existing physical, chemical and biological processes for As removal from water system are not feasible due to techno-economic limitations. The present study highlights the scope of biological strategy for As removal through phytoextraction. Arsenic uptake and accumulation in the biomass of three plant species and their As tolerance abilities have been investigated to develop an efficient phytoextraction system in combination of these plant species. Three non-crop plant species, Pteris vittata; Mimosa pudica, and Eichhornia crassipus were treated with 0-200 mg/L As in liquid nutrient solution for 14 days. P. vittata accumulated total 9,082.2 mg (8,223 mg in fronds) As/kg biomass and Eichhornia total 6,969 mg (4,517 mg in fronds)/kg biomass at 200 mg/L As concentration, respectively. Bioaccumulation factor (BF) and translocation factor (TF) were estimated to differentiate between excluders, accumulators and accumulation in above ground biomass. Pteris and Eichhornia have highest BF (67 and 17) and TF (64 and 3), respectively. In contrast, Mimosa accumulated up to 174 mg As/kg plant biomass which is low in comparison with other two plants, and both BF and TF were ${\leq}1$. This study reveals that Pteris and Eichhornia are As hyperaccumulator, and potential candidates for As removal from water system.
Bio-liquid is a liquid by-product of the hydrothermal carbonization (HTC) reaction, converting wet biomass into solid hydrochar, bio-liquid, and bio-gas. Since bio-liquid contains various compounds, it requires efficient sampling method to extract the target compounds from bio-liquid. In this research, fatty acid methyl ester (FAME) in bio-liquid was extracted based on hollow fiber supported liquid phase microextraction (HF-LPME) and determined by Gas Chromatography-Flame Ionization Detector (GC-FID) and Gas Chromatography/Mass Spectrometry (GC/MS). The well-known major components of biodiesel, including methyl myristate, palmitate, methyl palmitoleate, methyl stearate, methyl oleate, and methyl linoleate had been selected as standard materials for FAME analysis using HF-LPME. Physicochemical properties of bio-liquid was measured that the acidity was 3.30 (${\pm}0.01$) and the moisture content was 100.84 (${\pm}3.02$)%. The optimization of HF-LPME method had been investigated by varying the experimental parameters such as extraction solvent, extraction time, stirring speed, and the length of HF at the fixed concentration of NaCl salt. As a result, optimal conditions of HF-LPME for FAMEs were; n-octanol for extraction solvent, 30 min for extraction time, 1200 rpm for stirring speed, 20 mm for the HF length, and 0.5 w/v% for the concentration of NaCl. Validation of HF-LPME was performed with limit of detection (LOD), limit of quantitation (LOQ), dynamic range, reproducibility, and recovery. The results obtained from this study indicated that HF-LPME was suitable for the preconcentration method and the quantitative analysis to characterize FAMEs in bio-liquid generated from food waste via HTC reaction.
BACKGROUND: Tillage systems and fertilization play an important role in crop growth and soil improvement. This study was conducted to determine the effects of tillage and fertilization on the microbial biomass C and dehydrogenase activity of soils in a field under cultivation of soybean. METHODS AND RESULTS: An experimental plot, located in the temperate climate zone, was composed of two main sectors that were no-tillage (NT) and conventional tillage (CT), and they were subdivided into four plots, respectively, in accordance with types of fertilizers (non fertilizer, chemical fertilizer, hairy vetch, and liquid pig manure). Microbial biomass C and dehydrogenase activity were evaluated from May to July in 2016. The microbial biomass C and dehydrogenase activity of NT soils were significantly higher than those of CT in all fertilizer treatments, and they were further increased in hairy vetch treatment than the other fertilizer treatments in both NT and CT. The dehydrogenase activity was closely related to microbial biomass C. CONCLUSION: It is concluded that application of green manure combined with no-tillage can provide viable management practices for enhancing microbial properties of soil.
국내외 석탄화력발전소에서 REC(Renewable energy certificate) 확보를 위해 혼소되고 있는 바이오매스는 기존의 목질계 바이오매스인 우드 펠릿에서부터 최근 들어 초본계 바이오매스로의 확대가 진행되고 있다. 초본계 바이오매스의 경우 목질계 바이오매스에 비해 K, Na 등의 알칼리계 금속 함량이 더 높아 회융점이 더 낮고, 이 때문에 보일러 튜브에의 회 점착을 늘려 보일러의 효율을 떨어뜨리는 원인이 되고 있다. 본 연구에서는 초본계 바이오매스 연소시 회 융점을 높여 회 점착을 감소시키기 위해 많이 사용되고 있는 첨가제인 카올린(kaolin)이 석탄-바이오매스 혼소시 미치는 영향을 열화학 평형 계산을 통해 분석하고자 하였다. 이전 연구에서 수행된 80 kWth 급 미분탄 - 바이오매스 혼소 실험에서 카올린을 사용하는 경우 오히려 회 점착량이 늘어나는 원인을 해석을 통해 분석하였으며, 해석을 수행한 결과 석탄에 많이 포함되어 있는 Al2O3 때문에, Al-Si 계열 첨가제인 카올린 투입시 aluminosilicate 화합물의 생성이 촉진되어 용융점이 낮은 mullite가 많이 형성, 카올린을 더 사용할수록 슬래그 생성이 증가하는 것을 확인하였다. 추가적으로 바이오매스 혼소율을 0~100%까지 10% 간격으로 증가시켜가면서 해석을 수행하였으며, 그 결과 비선형적인 액상 슬래그 생성 특성을 확인하였다. 결과적으로는 바이오매스 혼소율 50~60% 조건일 때 가장 적은 량의 액상 슬래그가 생성됨을 파악하였다. phase diagram을 분석한 결과, 고용융점 화합물(leucite, feldspar)이 해당 조건에서 가장 많이 생성되는 것을 확인하였다.
Rice straw, produced as an agricultural by-product, is usable biomass as fuels if depolymerized to monomer unit, because the chemical structure are similar to high octane materials found in gasoline. In this study, parameters of thermochemical degradation by solvolysis reaction of rice straw such as the effect of reaction temperature, reaction time and type of solvent on conversion yield and degradation products were investigated. It was found that the effectiveness of the solvent on the solvolysis reaction was as follows; acetone>cresol>butanol. When acetone was used as a solvent, the highest rice straw conversion was observed to be 91.5% at $500^{\circ}C$, 40 min. Combustion heating value of liquid products from thermochemical conversion processes was in the range of 7,380 cal/g. The energy yield and mass yield in acetone-solvolysis of rice straw was as high as 69.0% and 38.2 g-oil/100g-raw material after 40 min of reaction at $350^{\circ}C$. Various aliphatic and aromatic compounds were detected in the rice straw solvolysis products. The major components of the solvolysis products, that could be used as fuel, were 4-methyl-2-pentanone, 3,5,5-trimethyl-2-cyclopentan-1-one as ketones.
In this paper, the carbon resources recycling of the overview of coffee waste generation in Vietnam. Since few years, there has been a significant research studies was done in the areas of coffee waste generation areas and also waste water generation from coffee production. The coffee residue (solid) and waste water (liquid) both are caused the underground water contamination and also soil contamination. These residues contain high organic matter and acid content leads to the severe threat to environment. In second stage of coffee production process, the major solid residue was generated. Various solid residues such as spent coffee grounds, defective coffee beans and coffee husks) pose several environmental concerns and specific problems associated with each type of residue. Due to the unlimited usage of coffee, the waste generation is high. At the same time, some researchers have been investigated the spent coffee wastes are the valuable sources for various valuable compounds. Biodiesel or biomass productions from coffee waste residues are the best available utilization method for preventing the landfill problems of coffee waste residues.
The production of adventitious roots derived from root explant of Echinacea angustifolia and its secondary metabolite content were assessed in different types and levels of auxin. The induction of adventitious roots from root explant cultured in Murashige and Skoog solid medium supplemented with 1.0 mg/L indole -3-butyric acid (IBA) attained highest as 20.87 mg fresh weight and 3.07 mg dry weight per culture but root suspension culture at the same concentration of IBA enhanced biomass production as 3.07 g fresh weight and 0.38 g per culture after 4 weeks in culture. 3.0 mg/L ${\alpha}$-naphthalene acetic acid (NAA) treatment had similar effect on root biomass production as 3.07 g fresh weight and 0.38 g per culture with liquid suspension culture, whereas adventitious roots exposed to over 3.0-5.0 mg/L IBA or 5.0 mg/L NAA were less responsive by reducing the number of adventitious roots and/or changing root morphology such as short and thick. The content of secondary metabolites such as phenolic, flavonoids and total caffeic acid in adventitious roots cultured on MS medium supplemented with 1.0 mg/L IBA were attained highest as 27.20, 9.60. 10.67 mg/g dry weight, respectively. Overall, the best production of root biomass and secondary metabolites were given by 1.0 mg/L IBA.
This study was performed to get the basic information concerned to the optimum culture condition of Inonotus obliquus. Several solid media, PDA, MEA and Czapek-Dox, and three liquid media were adopted for the in vitro cultivation. Some main features of the fungal morphological characteristics under cultivation conditions were observed and described. Preliminary results showed that appearance of the mycelial mat, hyphal size and substrate pigmentation differed according to the media. The PDA medium was the most favorable substrate for the growth on solid culture, followed by MEA and Czapek-Dox media. Concerned to the addition of amino acids, 5 amino acids, such as alanine, alginine, isoleucine, leucine and threonine, enhanced to the mycelial growth. Isoleucine was shown the best fungal growth. An important morphological hyphal structure for the fungus, the setae, was found in abundance and diverse its shape and size. In liquid culture, fresh potato broth was the best growth stimulant of the fungus, followed by Malt extract and potato broth. Addition of yeast extract to the liquid media had improved the biomass, but not laccase production.
본 연구에서는 무기염을 첨가한 액-액 추출에 의해 식물세포인 바이오매스로부터 파클리탁셀 회수 방법을 획기적으로 개선하고자 하였다. 다양한 무기염(NaCl, KCl, $K_2HPO_4$, $NaH_2PO_4$, $NaH_2PO_4{\cdot}2H_2O$)을 이용하여 추출효율을 조사한 결과, NaCl에서 가장 낮은 분배계수(0.053)로 가장 높은 파클리탁셀 수율(~96%)을 얻을 수 있었다. NaCl을 이용한 액-액 추출에서 최적의 NaCl/용매 비와 메틸렌 클로라이드/메탄올 비는 각각 1%(w/v)와 26%(v/v)이었다. 또한 최적의 NaCl/용매 비와 메틸렌 클로라이드/메탄올 비에서 파클리탁셀 함량에 따른 영향을 조사한 결과, 순수 파클리탁셀 함량 0.066%(w/v)에서 가장 낮은 분배계수(0.053)로 가장 높은 수율(~96%)을 얻을 수 있었다. 기존 액-액 추출의 경우 총 3회의 추출로 파클리탁셀을 95% 정도 회수 가능한 반면 무기염을 이용한 방법의 경우 단 1회 추출로 대부분의 파클리탁셀을 회수(~96%) 가능하였다.
대기오염물질처리를 위한 생물살수여과법에서 물질전달현상을 이해하기 위한 선행 실험결과를 기초로 각각의 운전조건에서 기체/액체(살수액), 기체/고체(미생물)와 액체/고체에서의 model을 이용하여 물질전달계수를 평가하였다. 생물살수여과법에서 기/액에서는 정상상태물질수지, 그리고 액/고와 기/고에서는 동적물질수지를 이용하여 물질전달 model을 확립하고 그 결과를 고찰하였다. 물질전달 model은 여과탑을 일정크기 구획하여, 각 구획에서 동적 물질수지식을 수치해석 전산코드를 이용해 계산하였다. 동적물질수지식을 이용하여 계산된 결과는 실험결과와 비교하여 생물살수여과법에서 기/액, 기/고, 액/고 각상간의 물질전달계수($K_La$)를 산정하였다. 본 연구에서는 대기오염제어를 위한 생물살수여과법에서 물질전달계수를 결정하기 위한 실험방법개발과 model을 이용하여 물질전달현상을 고찰하였다.
