Isolation and Characterization of the Plant Growth Promoting Rhizobacterium, Arthrobacter scleromae SYE-3 on the Yam Growth (식물성장촉진근권미생물 Arthrobacter scleromae SYE-3의 분리 및 Yam (Dioscorea japonica Thunb.) 성장에 미치는 영향 연구)

  • In this study, Arthrobacter scleromae SYE-3, which was isolated from indigenous plant in a subtropical region, Neigeria, with plant growth promoting activity was evaluated to determine the optimal culture condition. A bacterial strain SYE-3 had the IAA productivity ($89.15{\pm}0.36mg/L$) and ACC deaminase activity ($0.20{\pm}0.06$ at 72 hours). Also, optimal culture conditions such as temperature and pH of strain SYE-3 were $20^{\circ}C$ and 10 in LB medium, respectively. Strain SYE-3 had up to 3% salt tolerance in the LB medium. Plant growth promoting ability of strain SYE-3 using yam (Dioscorea japonica Thunb.) was evaluated. As a result, strain SYE-3 had showed very powerful effect on the increase of the shoot length and root biomass of yam (190.0% and 282.41% increase for 112 days, respectively). These results indicated that Arthrobacter scleromae SYE-3 can serve as a promising microbial resource for the biofertilizers of subtropical crops.

Ethanol Fermentation of Corn Starch by a Recombinant Saccharomyces cerevisiae Having Glucoamylase and $\alpha$-Amylase Activities

  • Starch is an abundant resource in plant biomass, and it should be hydrolyzed enzymatically into fermentable sugars for ethanol fermentation. A genetic recombinant yeast, Saccharomyces cerevisiae GA-7458, was constructed by integrating the structural gene of both $\alpha$-amylase from Bacillus stearothermophilus and the gene (STA1) encoding glucoamylase from S. diastaticus into the chromosome of S. cerevisiae SH7458. The recombinant yeast showed active enzymatic activities of $\alpha$-amylase and glucoamylase. The productivity of ethanol fermentation from the pH-controlled batch culture (pH 5.5) was 2.6 times greater than that of the pH-uncontrolled batch culture. Moreover, in a fed-batch culture, more ethanol was produced (13.2 g/L), and the production yield was 0.38 with 2% of corn starch. Importantly, the integrated plasmids were fully maintained during ethanol fermentation.

Identification of Pseudomonas fluorescens antagonistic to Pseudomonas tolaasii and its cultivation (버섯의 갈변병 유발세균 Pseudomonas tolaasii의 길항세균인 Pseudomonas fluorescens의 분리동정 및 배양조건)

  • A Pseudomonas fluorescens was selected from mushrooms and studied in both batch and fed-batch cultures in order to get maximal biomass concentration. P. fluorescens is an aerobic bacterium and antagonistic to Pseudomonas tolaasii which causes blotch disease on the mushroom cap. P fluarescens and P. tolaasii were identified by Gram staining, gelatin liquefaction, oxidase test, etc. and were characterized by pigment production, temperature sensitivity, salt tolerance and rapid pitting test, etc., Celts of P. fluorescens well in medium containing 30g/L of glucose, whereas the growth was inhibited at the glucose levels at higher than 30g/L. The highest values of specific growth rate and productivity were obtained when using 10g/1 of yeast extract. Optimum concentrations of $NH_4Cl$ and ${(NH_4Cl)}_2SO_4$ for culture were found to be 1.0g/L and 0.1g/L respectively. Optimum concentration of $MgSO_4{\cdot}7H_2O$ used as a sulfursource was 1.0g/L. It was also found that the cell concentrations reached the maximum level when grown on the medium containing 1.0g/L of $KH_2PO_4$ and 0.1g/L of $CaCl_2$. Also, the optimum culture conditions were $30^{\circ}C$ and pH 6.0. Cultivation of P. fluarescens at high dissolved oxygen (DO) concentration led to a decrease of bacterial productivity in batch culture. Maximum productivity was achieved at 40% DO concentration.

Production of Adventitious Ginseng Roots Using Biorectors

  • Panax ginseng is an important medicinal plant that has been used worldwide for geriatric, tonic, stomachic, and aphrodisiac treatments. Ginsenosides contained in the ginseng root are the main substances having active functions for human body. The price of ginseng is very expensive due to a complex process of cultivation, and the yield of ginseng is limited, which cannot meet the demand of the increasing market. Researchers have applied plant biotechnology to solve the problems but there are still things to be determined towards ginsenoside production by large-scale adventitious root culture. In this experiment, 5 to 20 liter bioreactors were employed to determine optimal conditions for adventitious root culture and ginsenoside production of Panax gineng. Callus was induced from the ginseng root on MS agar medium containing 1.0 mg. $L^{-1}$ 2,4-D and 0.1 mg. $L^{-1}$ kinetin. Then the callus was cultured on MS agar medium supplemented with 2.0 mg. $L^{-1}$ IBA, 0.1 mg. $L^{-1}$ kinetin, and 30 g. $L^{-1}$ to induce adventitious roots. The maximum root growth and ginsenoside production were obtained in 1/2 MS medium. 2.0 mg. $L^{-1}$ naphthalene acetic acid resulted in greater root growth than 2.0 mg $L^{-1}$ indole-3-butyric acid. Ginsenoside content increased with 2.0 mg. $L^{-1}$ benzyl adenin or kinetin. High concentrations of benzyl adenin (above 3.0 mg. $L^{-1}$ ) decreased the adventitious root growth and ginsenoside productivity. N $H_{4}$$^{+}$ inhibited the ginsenoside accumulation, while high concentrations of $K^{+}$, $Mg_{2}$$^{+}$, and $Ca_{2}$$^{+}$ increased it. N $H_{4}$$^{+}$ at 0.5 and 1.0 times of the normal amount in 3/4 SH medium resulted in the greatest biomass increase, but the highest ginsenoside productivity was obtained when N $O_{3}$$^{-}$ was used as the sole nitrogen source in the medium. Most microelements at high concentrations in the medium inhibited the root growth, but high concentrations of MnS $O_4$enhanced the root growth. Root dry weight increased with increasing sucrose concentrations up to 50 g. $L^{-1}$ , but decreased from 70 g $L^{-1}$ Ginsenoside productivity was maximized at the range of 20 to 30 g. $L^{-1}$ sucrose. In the experiment on bioreactor types, cone and balloon types were determined to be favorable for both adventitious root growth and ginsenoside production. Jasmonic acid was effective for increasing ginsenoside contents and Rb group ginsenosides mainly increased. These results could be employed in commercial scale bioreactor cultures of Panax ginseng.x ginseng.

  • For the study on the structure and characteristics of the primary production system in the southern waters of the East Sea, chlorophyll, phytoplankton standing stocks. nutrients and hydrographic properties were investigated and analyzed in conjunction with measurement of C-14 based primary productivity. The primary productivity was relatively high in comparison with the previous studies, ranging from 284 to 4,574 mgC$.$m/SUP -2/$.$day /SUP -1/ and averaged to be 2,000 mgC$.$m/SUP 02/$.$day/SUP -1/. The standing stocks within the euphotic zone were fairly high, but ambient inorganic nitrogenous nutrient concentrations were too low to support the high production. This implied that there might be active recycling of nitrogenous nutrients by heterotrophic processes and the upward flux of nutrients by vertical mixing. Subsurface chlorophyll maxima were continuously observed in the lower parts of the euphotic layer and the depth coincided with the nutricline rather than isopycnal surfaces, supporting the view that chlorophyll distributions and primary production were primarily influenced by nutrient supply. Despite low nutrient concentrations, phytoplankton standing stocks and production were fairly high and the fraction of autotrophic nano- and picoplankton production was significant.

Screening of Nutrient Removal Hydrophyte and Distribution Properties of Vegetation in Tributaries of the West Nakdong River (서낙동강 유역 하천의 식생 분포특성과 영양염류 정화 수생식물 탐색)

  • This study was conducted to investigate natural distribution of aquatic plane and to find out natural aquatic plants which highly absorb nutrient N and P. We surveyed vegetation within ${\pm}2m$ from streamside in 12 tributaries of the West Nakdong river watershed during May to October in 2003. Hydrophytes surveyed in tributaries of the West Nakdong river watershed were 27 families, 61 genera, 76 species, 3 varieties. Major dominance species of aquatic plants were Z. latifolia, P. communis, P. thunbergii, P. arundinacea, P. japonica, and P. distichum var. indutum. Aquatic plants having high production ability of biomass were Z. latifolia, P. communis, P. arundinacea, P. japonica, and E. crus-galli var. echinata. In the vertical distribution of hydrophytes within streams, dominant species were P. thunbergii and P. japonica in the upper stream, but dominant species in the downstream were P. communis and Z. latifolia. Species diversity or aquatic, plants was reduced, but their biomass and nutrient (T-N and T-P) content per the natural area unit $(m^2)$ were increased in the downsteaam. Nutrient N and P content of aquatic plants per the natural area unit were high at Joman river, Pyeonggangcheon, Bulam drainage canal, and Hogyecheon. Fifty-seven species of aquatic plants having high biomass were grounped into 4 categories $(I{\sim}IV)$ according to their nutrient content per dry weight unit. I group $(T-N,\;\geqq20gkg^{-1}\;&\;P_2O_5,\;\geqq7gkg^{-1})$ was comprised of 3 submerged plants (H. verticillata, P. crispus, and C. demersum), e emergent plants (O. javanica, P. distichum var. indutum, and R. sceleratus), 1 suspended plant (T. japonica), and 1 riparian plant (A. lobatum). Otherwise, in classification of natural hydrophytes according to their nutrient content per natural area unit, Z. latifolia, P. communis, P. longiseta, P. arundinacea, and P. distichum var. indutum possessing great biomass productivity as emergent plants were included in I group $(T-N,\;\geqq1gm^{-2}\;&\;P_2O_5,\;\geqq0.7gm^{-2})$.

Ecosysteme de I′Etang de Berre (Mediterranee nord-occidentale) : Caracteres Generales Physiques, Chimiques et Biologiques

  • Climatological, hydrological and planktonical research studies, measurements of primary production and photosynthetic efficiency from December 1976 to December 1978 have been carried out in two brackish lakes: Lake Etang de Berre and Lake Etang de Vaine located in the French Mediterranean coast, in the region of Carry-le-Rouet located on the north-west Mediterranean near Marseilles, and in fresh water inflows from 4 Rivers (Touloubre, Durance, Arc, Durancole) to Lake Etang de Berre. Physico-chemical parameters were measured for this study: water temperature, salinity, density, pH, alcalinity, dissolved oxygen (% saturation), phosphate, nitrate, nitrite, silicate etc. Diverse biological parameters were also studied: photosynthetic pigments, phaeopigments, specific composition and biomass of phytoplankton, primary pelagic production etc. Climatical factors were studied: air-temperature, solar-radiation, evaporation, direction (including strength) of winds, precipitation and freshwater volume of the four rivers. The changes in Lake ‘Etang de Berre’ ecosystem depend on the quality of the water in the Durance River, and on the effects of seawater near the entrance of the Caronte Canal. The water quality of the lake varies horizontally and vertically as a result of atmospheric phenomena, maritime currents and tides. The distribution of water temperatures is generally heterogeneous. Southeasterly winds and the Northeasterly Mistral wind are important in the origins of circulated and mixed water masses. These winds are both frequent and strong. They have, as a result, a great effect on the water environment of Lake Etang de Berre. In theory, the annual precipitation in this region is well over eight times the water mass of the lake. The water of the Durance River flows into Lake Etang de Berre through the EDF Canal, amounting to 90% of the precipitation. However, reduction of rainfall in dry seasons has a serious effect on the hydrological characteristics of the lake. The temperature in the winter is partially caused by the low temperature of fresh water, particularly that of the Durance River. The hydrological season of fresh and brackish water is about one month ahead of the hydrological season of sea water in its vicinity. The salinity of Lake Etang de Berre runs approximately 3$\textperthousand$, except at lower levels and near the entrance to the Caronte Canal. However, when the volume of the Durance River water is reduced in the summer and fall, the salinity rises to 15$\textperthousand$. In the lake, the ratio of fresh water to sea water is six to one (6:1). The large quantities of seston conveyed by rivers, particularly the Durance diversion, strongly reduce the transparency in the brackish waters. Although the amount of sunshine is also notable, transparency is slight because of the large amount of seston, carried chiefly by Tripton in the fresh water of the Durance River. Therefore, photosynthesis generally occurs only in the surface layer. The transparency progressively increases from freshwater to open seawater, as mineral particles sink to the bottom (about 1.7kg $m^{-2}a^{-1}$ on the average in brackish lakes). The concentration of dissolved oxygen and the rate of oxygen saturation in seawater (Carry-le-Rouet) ranged from 5.0 to 6.0 $m\ell$ㆍ.$1^{-1}$, and from 95 to 105%, respectively. The amount of dissolved oxygen in Etang de Berre oscillated between 2.9 and 268.3%. The monographs of phosphate, nitrate, nitrite and silicate were published as a part of a study on the ecology of phytoplankton in these environments. Horizontal and vertical distributions of these nutriments were studied in detail. The recent diversion of the Durance River into Lake Etang de Berre has effected a fundamental change in this formerly marine environment, which has had a great impact in its plankton populations. A total of 182 taxa were identified, including 111 Bacillariophyceae, 44 Chlorophyceae, and 15 Cyanophyceae. The most abundant species are small freshwater algae, mainly Chlorophyceae. The average density is about $10^{8}$ cells $1^{-1}$ in Lake Etang de Berre, and about double that amount in Lake Etang de Vaine. Differences in phytoplankton abundance and composition at the various stations or at various depths are slight. Cell biovolume V (equivalent to true biomass), plasma volume VP (‘useful’ biomass) and, simultaneously. the cell surface area S and S/V ratio through the measurement of cell dimensions were computed as the parameters of phytoplankton productivity and metabolism. Pigment concentrations are generally very high on account of phytoplankton blooms by Cyanophyceae, Chlorophyceae and Cryptophyceae. On the other hand, in freshwaters and marine waters, pigment concentrations are comparatively low and stable, showing slight annual variation. The variations of ATP concentration were closely related to those of chlorophyll a and phytoplankton blooms only in marine waters. The carbon uptake rates ranged between 38 and 1091 mg$Cm^{-2}d^{-1}$, with an average surface value of 256 mg; water-column carbon-uptake rates ranged between 240 and 2310 mg$Cm^{-2}d^{-1}$, with an average of 810, representing 290 mg$Cm^{-2}$, per year 45 000 tons per year of photosynthetized carbon for the whole lake. Gross photosynthetic production measured by the method of Ryther was studied over a 2-year period. The values obtained from marine water(Carry-le-Rouet) ranged from 23 to 2 337 mg$Cm^{-2}d^{-1}$, with a weighted average of 319, representing about 110 gCm$^{-2}$ per year. The values in brakish water (Etang de Berre) ranged from 14 to 1778 mg$Cm^{-2}d^{-1}$, with a weighted average of 682, representing 250 mg$Cm^{-2}$ per year and 38 400 tons per year of photosynthesized carbon for the whole lake.

Effects of Experimental Warming on Growth of Quercus variabilis Seedlings (실외 실험적 온난화 처리가 굴참나무 묘목의 생장에 미치는 영향)

  • Climate change affects all biological processes in terrestrial ecosystems including photosynthesis, plant growth and productivity. This study was conducted to investigate the effects of experimental warming on the growth of Quercus variabilis seedlings. One-year-old Q. variabilis seedlings were planted in control and warmed plots in April 2010. The air temperature of warmed plots was increased by $3^{\circ}C$ compared to control plots using the infrared lamp from November 2010. Shoot height and root collar diameter were measured in March 2011 and June 2012, respectively, and aboveground and belowground biomass were also measured in March 2011 and 2012, respectively. Shoot height and root collar diameter were significantly higher in warmed plots than in control plots, except for root collar diameter in March 2011. Increment (mm) of shoot height and root collar diameter were also higher in warmed plots ($529{\pm}30$, $5.6{\pm}0.5$) than in control plots ($464{\pm}28$, $4.5{\pm}0.4$). However, there were no significant differences between warmed and control plots except for root collar diameter. Increment (g/year) of total, aboveground and belowground biomass were higher in warmed plots ($36.88{\pm}6.52$, $11.91{\pm}3.44$, $24.97{\pm}3.73$) than in control plots ($30.59{\pm}5.51$, $8.73{\pm}1.66$, $21.86{\pm}3.88$), however, the differences were not statistically significant. Higher seedling growth and biomass of warmed plots might be related to the enhanced net photosynthetic rates in spring and the extended growing season.

A Study on the Effect of Environmental Pollution on the Biomass Productivity of Paulownia coreana (환경오염(環境汚染)이 오동나무인공림(人工林)의 물질생산(物質生産)에 미치는 영향(影響)에 관한 연구(硏究))

  • To study the comparison of aboveground biomass of paulownia coreana Uyeki of 6-year-old, located in Seongju of non-attacked forest and Ulsan of damaged forest by the air pollution were selected. Ten sample trees in Seongju district and seven trees in Ulsan selected taking account of DBH were measured for 16 trees in total within a $10{\times}10m$ experimental plot. The diagram of oven-dry weight distribution of stem, branch and needle for each 1m segment was constructed. The logarithmic regression equations between dry weight of each component and the two-variables, DBH and tree height, combined term were presented. IF the estimations are extended to a hectare area stand, it contains 47.49 tons of aboveground biomass in Seongju district and 19.05 tons of it in Ulsan. The annual net productions were 11.64 tons of above 2.29 kg/kg/yr in Ulsan and the efficiency of leaf to produce stem was 2.99kg/kg/yr in Seongju and 0.83kg/kg/yr in Ulsan.

  • To assess the ecological changes of Korean fir (Abies koreana E. H. Wilson) under climate change conditions, growth and physiological responses were analyzed over a 5-year period in a control group (outdoors) and in a treatment group where the temperature and CO2 levels were elevated to closely resemble RCP 4.5 conditions. The results showed an increasing trend in annual branch length of A.koreana in the climate change treatment group over time. While climate change conditions did not significantly impact the morphological differences of A.koreana leaves, they did influence the biomass of the leaves, suggesting that as climate change progresses, the productivity of A.koreana leaves may decline. On the other hand, the chlorophyll content in A.koreana under climate change conditions was higher in the climate change treatment group, whereas the photosynthesis rate, transpiration rate, water use efficiency and stomatal conductance was higher in the control group. This suggests that an environment with elevated temperature and CO2 could influence an increase in stomatal density, but having a negative impact on photosynthetic reactions. Further research on stomatal density under each environmental treatment will be required to confirm this hypothesis. Additionally, as this study only observed changes in leaf biomass, further empirical research should be considered to understand the changes in biomass of A.koreana under climate change conditions. In conclusion, the environmental adaptability of A.koreana is expected to weaken in the long term under elevated temperatures and CO2.
