DR (Digital Radiography) 적용을 위한 Biology 초음파 특수용매를 이용한 $PbI_2$ 합성법

  • 최근에 광도전체와 형광체를 기반으로 평판형 디지털 방사선 검출기의 상업적 발전가능성에 많은 관심을 가지고 있다. 본 연구는 기존의 직접변환방식에 널리 사용되었던 비정질 셀레늄 (amorphous selenium) 기반의 디지털 방사선 검출기보다 높은 전기적신호 및 동작특성을 가지는 물질층을 제작하기 위해 High Purity (99.99%)의 상용화된 $PbI_2$를 특수용매에 담가두었다가 약 1시간동안 Biology 초음파 처리한 후 농축기를 사용하여 건조된 $PbI_2$를 3Roll-milling을 사용하여 미세크기의 Powder를 얻어내었다. 합성된 $PbI_2$ Powder를 PIB(Particle-in-Binder)법을 이용하여 전도성을 가진 ITO(Indium-tin-oxide)코팅된 유리판에 제작된 필름의 상부에 Magnetron sputtering system 을 사용하여 전극을 $1cm{\times}1cm$의 크기로 증착하였다. I-V 테스트를 통하여 X선 조사시 $PbI_2$필름의 Sensitivity, Dark current, SNR(signal-to-noise ratio)을 측정하여 필름의 전기적 검출 특성을 정량적으로 평가하였고 SEM(scanning electron microscope)을 통하여 입자의 크기를 관찰하였다.

A pattern of cell death induced by 40 kHz ultrasound in yeast cell model (40 kHz 초음파에 의해 유도된 효모세포 모델에서 세포사멸 패턴)

  • Ultrasound has been widely used for biological and medical applications including induction of cell death, but a precise mechanism of induced cell death by ultrasound is controversial. In this study, an irradiation system with 40 kHz ultrasound was developed for a suitable cell death test of a representative unicellular organism, yeast, and used to study the biological effect of ultrasound on inducing cell death. Potassium Iodide (KI) dosimetry was used to devise an optimal system that successfully delivers 40 kHz ultrasound and produces reactive oxygen species in a 1.5 ml Eppendorf tube. Cell death was observed in an ultrasound transmission time-dependent fashion in this system. Thermal effect during irradiation was not observable in ultrasound induced cell death. Co-treatment of 40 kHz ultrasound and hydrogen peroxide showed a synergistic effect in inducing cell death. This finding suggests that 40 kHz ultrasound is related to reactive oxygen species formation. However, NAC (N-acetyl-L-cysteine) oxygen scavenger slightly inhibited the cell death by 40 kHz ultrasound. It was also found that 40 kHz ultrasound induced cell death was slightly inhibited by inhibitors of necrosis or apoptosis (glycyrrhizin or zVAD-fmk). This study suggests that cell death induced by 40 kHz ultrasound may not be exclusively related to reactive oxygen species formation and thermal effects in irradiated yeast cells.

  • 바이오와 나노 분야의 비파괴검사는 nm 급의 공간분해능을 요구한다. 바이오와 나노 물질의 내부구조의 비파괴 분석을 위한 공간분해능을 nm급으로 향상시키는Soft X-ray현미경 및 초음파현미경에 대한 기술을 소개한다. 세포생물(Biology)의 단백질구조분석, 수문학(Hydrology)연구, 나노입자거동 등의 연구를 위해서 중성자현미경기술개발 및 현황을 소개하고자 한다. 초음파현미경, Soft X-ray현미경 및 중성자현미경에 대하여 Hybrid 이용자 지원 체계를 구축하여 나노 및 Bio 물질 내부구조 분석기술을 확립하여 지원하고자 한다.

Effects of Low Powered Ultrasonic Wave Exposure on Microcystis sp. (Cyanobacteria) (저출력 초음파가 Microcystis sp. (Cyanobacteria)에 미치는 영향)

  • This study aimed to investigate the effects of ultrasonic wave on colonial changes of Microcystis sp. causing water-bloom. The effects of frequency change and exposure time of ultrasonic wave on Microcystis sp. were also perceived by measuring the concentration of chlorophyll-a. The concentrations of chlorophyll-a showed a decreasing tendency in all experimental groups exposed to ultrasonic waves, especially the highest decrease at 70 kHz. Before exposed to ultrasonic wave, single cells of Microcystis sp. were observed in a form of colonies which were densely accumulated onto their mucilage layer. However, after one hour exposure to 70 kHz ultrasonic wave, the bonds of colonies were weakened and single cells were separated from the colonies. Moreover, the single cells of the colony were destroyed with the 5 hour exposure time at 70 kHz. As a result, we proposed that the frequency strength and the exposure time of ultrasonic wave might have effects in decrease of biomass of blue-green algae by reducing the concentration of chlorophyll-a, thereby changing and destroying forms of Microcystis sp. colonies.

Change of Phytoplankton Community by Ultrasonication in Eutrophic Ponds (부영양 연못에서 초음파 작동에 따른 식물플랑크톤의 군집 변화)

  • The effects of ultrasonication on phytoplankton were investigated in two ponds in which physicochemical and biological water quality was similar, one as a treatment and the other as a control. The samples were collected from August 18 to September 30 in 2003. Traditional morphological analysis showed that Bacillariophyceae dominated phytoplankton community in both ponds. The abundance of Cyanophyceae was lower in the phytoplankton community of the sonicated pond than that of control pond. We used DGGE (denaturing gradient gel electrophoresis) to analyze the diversity and change of phytoplankton community in two ponds. The DGGE banding patterns of 16S rRNA gene and sequence analysis demonstrated that Oscillatoria acuminata and CFB (Cytophaga-Flavobacterium-Bacteroides) group bacterium appeared in the treated pond, and the control pond was dominated by Synechococcus sp. and Aphanizomenon flos-aquae. Especially, Pseudanabaena sp. dominated during the ultrasonic cessation in the treated pond. The DGGE profiles of 18S rRNA gene and sequence analysis showed that the treated pond was dominated by Chlamydomonas reinhardtii and the control pond by C. reinhardtii and Pteromonas protracta. In conclusion, the ultrasonication affected the reduced growth of cyanobacteria, particularly Pseudanabaena.

  • Phytoestrogens are one of quasi-estrogens which are not generated within the endocrine system, but consumed by intaking phytoestrogenic plants. Phytoestrogens, also called as "dietary estrogens", are various botanic chemical compound groups naturally occurring nonsteroidal plant compounds of spontaneous generation. Due to their structural similarities to estrogone, they may provide desirable health benefits for reducing the menopausal symptoms. In this paper, various phytoestrogens were extracted from sophora root, pomegranate and kudzu by ultrasonic extraction process. In this study, the amount of extracted phytoestrogens at various ultrasonic power strengths and extraction times were compared, which could be used as basic data in the study of biology and chemistry related to natural products.

A STUDY OF RELATIONSHIP BETWEEN BITE FORCE, MASSETER MUSCLE AND CRANIOFACIAL MORPHOLOGY : CEPHALOMETRIC & ULTRASONOGRAPHIC ANALYSIS (교합력, 교근과 두개안면골 형태의 관계 : 두부규격방사선사진과 초음파진단영상 분석)

  • The aim of this study was to investigate the relationships between bite force, masseter muscle and craniofacial morphology. 141 individuals were included. Bite forces were measured with a bite force transducer(SKT 2004). The facial morphology of the samples was evaluated on the Lateral and PA cephalometric radiographs(Asahi CX-90 SP). Ultrasonographic images were obtained for 36 individuals(Logiq 500). General status of the subjects were also measured for investigating possible relationships. Their relationships were analyzed by means of t-test, Pearson's correlation coefficients and simple regression analysis. In conclusion, bite force of molar was significantly correlated with craniofacial morphology for males and females, but not to masseter muscles. The masseter muscle for males had a significant correlation with craniofacial morphology. The unexpected negative correlations between size of masseter muscle and maxillary width was revealed for males. One possible way to arrive at a plausible explanation for the correlation between masster muscle, bite force and craniofacial morphology of males were the deep layer of masseter muscle had a great influence on males and the superficial layer of masseter muscle had a great influence on females.

Immuno-Regulatory Effect on Centella asiatica L. Urban Extraction Solvent Associated with Ultrasonification Process (병풀의 초음파 추출 시 용매에 따른 면역활성 증진 효과)

  • This study was performed to investigate the increasing of immune activities from Centella asiatica L. Urban by several solvent extraction with ultrasonification process. Ethanol extracts at $60^{\circ}C$ and associated with ultrasofication showed more 15% yield than normal $100^{\circ}C$ water extracts. And ethanol extracts showed higher yield than water extraction at each process. It can be concluded that yield due to polarity of compound in C. asiatica from HPLC peak. All of extracts from C. asiatica revealed lower cytotoxicity than 32%. The ethanol extracts with ultrasofication showed higher promotion on human B and T cell growth, about 10% compared to the control. The secretion of IL-6 and TNF-a was also enhanced as $4.86\;{\times}\;10^{-4}\;pg/cell$ and $5.73\;{\times}\;10^{-4}\;pg/cell$ each, by the addition ethanol extracts with ultrasonification to T cell. NK cell activation was improved up to 10% higher than the control, through adding the samples. It can be concluded that the ethanol extracts from C. asiatica has higher immune activities followed by ultrasonification process, possibly, by higher yield than conventional extraction process.
