Preparation and Drug-releasing Properties of Chitosan-based Thermosensitive Composite Hydrogel

  • The novel chitosan-based thermosensitive hydrogels were prepared as control-releasing drug carriers. N-carboxyethyl chitosan (ACS) was synthesized by microwave heating for 1 h through Michael addition of CS to acrylic acid in a grafting yield of 52.97%, which was proved to be a faster and more efficient way than ordinary methods. 5-Fu was modified with formaldehyde to synthesize N,N'-Bis(hydroxymethyl)-5-fluorouracil (5-Fu-OH). Then an esterification was performed using ACS and 5-Fu-OH to give 5-Fu-ACS. The new thermosensitive hydrogels were prepared by adding sodium glycerophosphate to the solution of compounds under a certain constant temperature. Simultaneously, the hydrogels' swelling rate, in vitro drug release rate and thermosensitive were studied, and found that the 5-Fu-ACS composite hydrogel had more excellent releasing effect, higher drug loading and better thermosensitive.

Expression and Characterization of Truncated Recombinant Human Cytochrome P450 2J2

  • The human cytochrome P450 2J2 catalyzes an epoxygenase reaction to oxidize various fatty acids including arachidonic acid. In this study, three recombinant enzyme constructs of P450 2J2 were heterologously expressed in Escherichia coli and their P450 proteins were successfully purified using a $Ni^{2+}$-NTA affinity column. Deletion of 34 amino acid residues in N-terminus of P450 2J2 enzyme (2J2-D) produced the soluble enzyme located in the cytosolic fraction. The enzymatic analysis of this truncated protein indicated the typical spectral characteristics and functional properties of P450 2J2 enzyme. P450 2J2-D enzymes from soluble fraction catalyzed the oxidation reaction of terfenadine to the hydroxylated product. However, P450 2J2-D enzymes from membrane fraction did not support the P450 oxidation reaction although it displayed the characteristic CO-binding spectrum of P450. Our finding of these features in the N-terminal modified P450 2J2 enzyme could help understand the biological functions and the metabolic roles of P450 2J2 enzyme and make the crystallographic analysis of the P450 2J2 structure feasible for future studies.

Description of ten unrecorded bacterial species isolated from Ostrea denselamellosa and Eriocheir japonica from the Seomjin River

  • Ostrea denselamellosa and Eriocheir japonica samples were collected from the Seomjin River in 2019 as part of the "Research of Host-Associated Bacteria" research program. Almost 200 bacterial strains were isolated from the O. denselamellosa and E. japonica samples and subsequently identified by 16S rRNA gene sequencing. Among the bacterial isolates, ten strains possessed greater than 98.7% sequence similarity with published bacterial species that had not previously been recorded in Korea. These species were phylogenetically diverse, belonging to three phyla, four classes, seven orders, and eight genera. At the genus and class level, the previously unrecorded species belonged to Pseudoalteromonas, Aliivibrio, Rheinheimera, Leucothrix, and Shewanella of the class Gamma-proteobacteria, Olleya of the class Flavobacteriia, Algoriphagus of the class Cytophagia, and Lactococcus of the class Bacilli. The previously unrecorded species were further characterized by examining their Gram staining, colony and cell morphology, biochemical properties, and phylogenetic positions.

Amyloid Polymorphism of α-Synuclein Induced by Active Firefly Luciferase

  • Amyloidogenic proteins often exhibit fibrillar polymorphism through alternative assembly processes, which has been considered to have possible pathological implications. Here, firefly luciferase (LUC) is shown to induce amyloid polymorphism of ${\alpha}$-synuclein, the major constituent of Lewy bodies found in Parkinson's disease, by acting as a novel template. The drastically accelerated fibrillation kinetics of ${\alpha}$-synuclein with LUC required the nucleation center produced by the active enzyme of LUC. Fluorescent dye binding, transmission electron microscopy, and Fourier transformed infrared spectroscopy revealed the morphologically distinctive amyloid fibrils of ${\alpha}$-synuclein prepared in the absence or presence of LUC. As the altered morphological characteristics became inherent to the mature fibrils, those properties were inherited to next-generations via nucleation-dependent fibrillation process. The seed control, therefore, would be an effective means to modify amyloid fibrils with different biochemical characteristics. In addition, the LUC-directed amyloid fibrillar polymorphism also suggests that other cellular biomolecules including enzymes in general are able to diversify amyloid fibrils, which could be self-propagated with diversified biological activities, if any, inside cells.

  • A non-invasive technique to measure absorption and scattering coefficients was investigated The reflected backscattered light from the surface of phantom and biological tissue was obtained by using a time-correlated single photon counting system in pico-second time domain. The absorption and scattering coefficients were acquired by the time of peak and asymptotic behavior of the time-resolved reflectance curve and agreed well the ones that is obtained with deconvolution method It was found that the approximation method was good for biological medium to calculate optical properties due to its convenience and accuracy.

Assessment of Heavy Metal Contamination and Biological Toxicity of Mine Drainages and Sediments from Abandoned Mines (폐광산 배수와 퇴적물의 중금속 오염과 생물독성 평가)

  • Heavy metal contamination and biological toxicity of mine drainages and sediments in abandoned mines were studied. Mine drainages had pH at a range of 2.94~7.86, and contained heavy metals at a toxic level. For coal mines, toxicity of mine drainage to Daphnia magna was attributable to acidic pH. In addition to the low pH, suspended heavy metals such as Zn and Cu contributed to toxicity of mine drainages at abandoned metalliferous mines. All mine sediments studied in this work showed biological toxicity to Chironomus riparius, having mortality at a range of 15~60%. However, its relationship with physicochemical properties including heavy metal content of the sediments was not statistically explained. Exceptionally clay ($< 2{\mu}m$ particle) content was negatively correlated with the biological toxicity for sediment samples collected at the same abandoned mines.

  • A total of 31 bacterial strains were isolated from the Geum River basin in the Republic of Korea during our investigation of indigenous prokaryotic species. The isolated bacterial strains had high 16S rRNA gene sequence similarity (>98.7%) with those of validly published bacterial species, which have not been reported in Republic of Korea. The 31 bacterial strains were phylogenetically diverse and assigned to 4 phyla, 8 classes, 18 orders, 21 families, and 27 genera. At the genus level, the unreported species were affiliated with Kineococcus, Pedococcus, Rhodoluna, Salinibacterium, Rhodoluna, Arthrobacter, Williamsia, Nakamurella, Nocardioides of the class Actinobacteria, Patulibacter of the class Thermoleophilia, Pontibacter, Hymenobacter of the class Cytophagia, Flavobacterium of the class Flavobacteriia, Geomicrobium of the class Bacilli, Brevundimonas, Gellertiella, Rhizobium, Paracoccus, Taonella, Sphingomonas of the class Alphaproteobacteria, Burkholderia, Polaromonas, Hydrogenophaga, Chitinilyticum, Azospira, Zoogloea of the class Betaproteobacteria, and Pseudomonas of the class Gammaproteobacteria. The unreported bacterial species were further characterized by examining their morphological, cultural, physiological, and biochemical properties. The detailed descriptions of the 31 bacterial strains were provided.

Rheological properties and crystallization kinetics of polypropylene block copolymer with repeated extrusion

  • Rheological properties and crystallization kinetics of the polypropylene (PP) block copolymer and recycled PP block copolymer were studied by advanced rheometric expansion system (ARES), differential scanning calorimetry (DSC), and optical microscopy. In the study of the dynamic rheology, it is observed that the storage modulus and loss modulus for the PP block copolymer and recycled PP block copolymer did not change with frequency. In the study of the effect of the repeated extrusion on the crystallization rate, half crystallization time of the PP samples was increased with the number of repeated extrusion in isothermal crystallization temperature ($T_c$). From the isothermal crystallization kinetics study, the crystallization rate was decreased with the increase of the number of repeated extrusion. Also, from the result of Avrami plot, the overall crystallization rate constant (K) was decreased with the increase of the number of the repeated extrusion. From the study of the optical microscopy, the size of the spherulite of the PP samples did not change significantly with the number of repeated extrusion. However, it was clearly observed that the number of the spherulite growth sites was decreased with the number of repeated extrusion. From the results of the crystallization rate, isothermal crystallization kinetics, Avrami plots, and optical microscopy, it is suggested that the crystallization rate of the PP block copolymer is decreased with the increase of the number of repeated extrusion.

The effect of sewage sludge compost amended soils on the growth of Orchardgrass seedlings (하수오니 첨가토양이 Orchardgrass 유식물체의 생육에 미치는 영향)

  • This experiment was carried out to investigate the effect of sewage sludge compost amended soils on the growth and accumulation patterns of heavy metals in plant parts of Orchardgrass seedlings, changes in physical properties and chemical composition, and heavy metal residue in soils. Mixture ratios of sewage sludge compost and soil(loam) were 100:0, 80:20, 60:40, 40:60, 20:80 and 0:100(control), respectively. The results obtained were as follows; 1. The physical properties and chemical compostion of soils were improved by increase in mixture ratios of sewage sludge compost. 2. The biological yield of Orchardgrass seedlings was increased with mixture ratios of sewage sludge compost. 3. The dry weight of shoot(SH) was increased with both of yield components(NT and WT) and biological yield of Orchardgrass seedlings. 4. The total nitrogen concentrations(TN) of plants was increased with quadratically up to the biological yield of 100% mixture ratio of sewage sludge compost. 5. Lead(Pb) concentration of soil in over the 60% mixture ratios of sewage sludge compost were in excess of limiting level(50ppm) of organic fertilizers.

  • In hypersonic aircraft, increase of aerodynamic heat and engine heat leads heat loads in airframe. It could lead structural change of aircraft's component and malfunctioning. Endothermic fuels are liquid hydrocarbon fuels which are able to absorb the heat load by undergoing endothermic reactions. In this study, exo-tetrahydrodicyclopentadiene was selected as a model endothermic fuel and experiments on endothermic properties were investigated with pore structure controlled zeolite catalyst using metal deposition. We secured the catalyst that had better endothermic performance than commercial catalyst. The object of this study is inspect catalyst properties which have effect on heat absorption improvement. Synthetic catalyst could be applied to system that use exo-THDCP as endothermic fuel instead of other commercial catalyst.
