참가사리 분획물의 항산화.항균효과 (The Antioxidative and Antimicrobial Effects of Gloiopeltis Tenax)

  • 참가사리의 항산화효과를 측정하기 위해 ROS와 RNS 제거활성을 측정하였다. ROS 제거활성 측정결과 GTMM층과 GTMB층이 positive control로 사용한 trolox와 유사한 높은 제거능을 보였고, $ONOO^-$ 제거능은 그 효과가 미약하였으나 GTMM 층에서 나타났으며 NO 제거활성 역서 GTMM층과 GTMB층에서 약한 효과가 관찰되었다. 따라서 본 실험의 결과 GTMM 층에서 ROS 제거 활성의 효과가 높았으며 다음으로 GTMB층에서 효과를 보여 GTMM층과 GTMB층에 활성산소 제거효과를 가지는 물질이 있는 것으로 사료되어진다. Paper disc method를 이용한 항균활성 효과 실험결과, Serratia marcescens가 모든 층에서 높은 항균력을 보였으며 Proteus mirabilis에서도 GTMA층을 제외한 모든 층에서 항균효과를 보였다. 사용한 모든 균주에서 각각 항균력의 차이는 있으나 전체적으로 GTMB 층과 GTMM 층에서 높은 항균활성을 관찰할 수 있었다. 이 연구결과를 토대로 참가사리의 항산화 및 항균활성물질에 대한 단계적인 분리 동정이 이루어져 GTMM 층과 GTMB 층의 생리활성물질 개발이 기대되는 바이다.

사상체질별 수면 소증(素證)에 대한 체계적 고찰연구 (Systematic Review on the Sasang Type-specific Pathophysiological Symptoms of Sleep)

  • Objectives: The purpose of this study was to review previous clinical studies on underlying mechanisms of sleep-related type-specific pathophysiological symptoms among the Sasang types. Methods and Procedure: We reviewed seven research databases from December 2003 to August 2015 with the keywords Sasang typology, constitution and sleep. The Sasang type-specific sleep-related symptoms were analyzed based on seven categories, including subjective sleep quality, sleep latency, sleep duration, habitual sleep efficiency, sleep disturbances, use of sleep medications and daytime dysfunction. Results: Total of twelve studies were included in the analysis. The Tae-Yang type showed low subjective sleep quality, long sleep latency and frequent awakening, and the So-Yang type showed long sleep latency. The Tae-Eum type presented high subjective sleep quality, short sleep duration, frequent snore, toss and turn during the sleep, and insomnia and low frequency of dream. The So-Eum type showed long sleep latency in male and high frequency of dream. The Eum type combining Tae-Eum and So-Eum types had higher subjective sleep quality, longer sleep duration and higher frequency of dream than the Yang type combining Tae-Yang and So-Yang types.Conclusions This study reviewed type-specific sleep-related characteristics and discussed possible pathophysiological mechanisms involved. Differences in sleep characteristics among the Sasang types might stem from type-specific temperaments and require further study.

Ginsenoside Re inhibits pacemaker potentials via adenosine triphosphate-sensitive potassium channels and the cyclic guanosine monophosphate/nitric oxide-dependent pathway in cultured interstitial cells of Cajal from mouse small intestine

  • Background: Ginseng belongs to the genus Panax. Its main active ingredients are the ginsenosides. Interstitial cells of Cajal (ICCs) are the pacemaker cells of the gastrointestinal (GI) tract. To understand the effects of ginsenoside Re (GRe) on GI motility, the authors investigated its effects on the pacemaker activity of ICCs of the murine small intestine. Methods: Interstitial cells of Cajal were dissociated from mouse small intestines by enzymatic digestion. The whole-cell patch clamp configuration was used to record pacemaker potentials in cultured ICCs. Changes in cyclic guanosine monophosphate (cGMP) content induced by GRe were investigated. Results: Ginsenoside Re ($20-40{\mu}M$) decreased the amplitude and frequency of ICC pacemaker activity in a concentration-dependent manner. This action was blocked by guanosine 50-[${\beta}-thio$]diphosphate [a guanosine-5'-triphosphate (GTP)-binding protein inhibitor] and by glibenclamide [an adenosine triphosphate (ATP)-sensitive $K^{+}$ channel blocker]. To study the GRe-induced signaling pathway in ICCs, the effects of 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one (a guanylate cyclase inhibitor) and RP-8-CPT-cGMPS (a protein kinase G inhibitor) were examined. Both inhibitors blocked the inhibitory effect of GRe on ICC pacemaker activity. L-NG-nitroarginine methyl ester ($100{\mu}M$), which is a nonselective nitric oxide synthase (NOS) inhibitor, blocked the effects of GRe on ICC pacemaker activity and GRe-stimulated cGMP production in ICCs. Conclusion: In cultured murine ICCs, GRe inhibits the pacemaker activity of ICCs via the ATP-sensitive potassium ($K^{+}$) channel and the cGMP/NO-dependent pathway. Ginsenoside Re may be a basis for developing novel spasmolytic agents to prevent or alleviate GI motility dysfunction.

Association of a Methanol Extract of Rheum undulatum L. Mediated Cell Death in AGS Cells with an Intrinsic Apoptotic Pathway

  • Objectives: Rheum undulatum L. has traditionally been used for the treatment of many diseases in Asia. However, its anti-proliferative activity in cancer has still not been studied. In the present study, we investigated the anti-cancer effects of methanol extract of Rheum undulatum L. (MERL) on human adenocarcinoma gastric cell lines (AGS). Methods: To investigate the anti-cancer effect of MERL on AGS cells, we treated the AGS cells with varying concentrations of MERL and performed 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assays. Cell cycle analyses, measurements of the mitochondrial membrane potential (MMP), caspase activity assays and Western blots were conducted to determine whether AGS cell death occurred by apoptosis. Results: Treatment with MERL significantly inhibited growth of AGS cells in a concentration dependent manner. MERL treatment in AGS cells leaded to increased accumulation of apoptotic sub G1 phase cells in a concentration dependent manner. In control cultures, 5.38% of the cells were in the sub G1 phase. In MERL treated cells, however, this percentage was significantly increased (9.95% at $70{\mu}g/mL$, 15.94% at $140{\mu}g/mL$, 26.56% at $210{\mu}g/mL$ and 38.08% at $280{\mu}g/mL$). MERL treatment induced the decreased expression of pro-caspase-8 and -9 in a concentration dependent manner, whereas the expression of the active form of caspase-3 was increased. A subsequent Western blot analysis revealed increased cleaved levels of poly (ADP-ribose) polymerase (PARP) protein. Also, treatment with MERL increased the activities of caspase-3 and -9 compared with the control. MERL treatment increased the levels of the pro-apoptotic truncated Bid (tBid) and Bcl2 Antagonist X (Bax) proteins and decreased the levels of the anti-apoptotic B-cell lymphoma 2 (Bcl-2) protein, whose is the stabilization of mitochondria. However, inhibitions of p38, extracellular signal regulated kinases (ERKs) and C-Jun N-terminal kinases (JNK) by MERL treatment did not affect cell death. Conclusion: These results suggest that MERL mediated cell death is associated with an intrinsic apoptotic pathway in AGS cells.

위장관 질환의 치료를 위한 한의학 연구 동향 (Latest Research Trends of Korean Medicine for the Treatment of Gastrointestinal Diseases)

  • To research the trends of study about gastrointestinal (GI) disease in Korean medicine, we reviewed and analysed articles published in Korea from 2009 to 2012. From ancient to modern history, traditional plant-based medicines have played an important role in health care, especially GI tract. In spite of the great advances of moderm scientific medicine, traditional medicine is still the primary form of healing methods readily available to the majority of the people in the many countries. In fact many of today's popular drugs have their origins in traditional medicine. Therefore, we suggest that the development and improvement of Korean medicine in GI tract will make good the quality of life in our lifetime.

The Pathophysiologic Roles of TRPM7 Channel

  • Transient receptor potential melastatin 7 (TRPM7) is a member of the melastatin-related subfamily and contains a channel and a kinase domain. TRPM7 is known to be associated with cell proliferation, survival, and development. It is ubiquitously expressed, highly permeable to $Mg^{2+}$ and $Ca^{2+}$, and its channel activity is negatively regulated by free $Mg^{2+}$ and Mg-complexed nucleotides. Recent studies have investigated the relationships between TRPM7 and a number of diseases. TRPM7 regulates cell proliferation in several cancers, and is associated with ischemic cell death and vascular smooth muscle cell (VSMC) function. This review discusses the physiologic and pathophysiologic functions and significance of TRPM7 in several diseases.

혈류역학을 이용한 촌구와 인영의 특성비교 (Comparative study of pulse point using hemodynamics)

  • Objectives : The purpose of this study is to examine the hemodynamic characteristics of pulse point. Methods : The computational analysis algorithms of arterial tree system was derived. In order to investigate the effect of internal organ on the pulse point, the diameter of celiac artery was reduced by half. Results : The sensitivity of flow change at the Inyoung(Renying) is better than that of the Chongu(Cunkou). but the Inyoung was worse than the Chongu in the point of the left and right symmetry. The pressure changes at the Inyoung and the Chongu were in the similar range. Conclusions : It was found from the result that the Chongu shows the more symmetrical hemodynamic characteristics than the Inyoung.
