연부(軟腐)고구마의 생화학적(生化學的) 연구(硏究) (Biochemical Study on Soft-rotted Sweetpotatoes)

  • Rhizopus nigricans에 감염(感染)된 고구마의 생화학적(生化學的) 성질(性質)을 구명(究明)하기 위하여 연부(軟腐)고구마로부터 이상대사산물(異常代謝産物)을 분리(分離)하고 그 성질(性質)에 관(關)하여 실험(實驗)한 결과(結果)를 다음과 같이 요약(要約)한다. 1. 건전(健全)한 고구마에는 없는 이상대사산물(異常代謝産物)로서 19개(個)의 Ehrlich's 시약(試藥)에 대(對)한 양성반응성질(陽性反應性質)과 3개(個)의 형광성물질(螢光性物質)의 존재(存在)를 박층(薄層)크로마토그라프로 확인(確認)하였다. 2. 이상대사산물중(異常代謝産物中)에는 그 생성량(生成量)이 가장 많은 물질(物質)로서 Ipomeamarone과 Umbelliferone을 분리(分離)하고 이들의 정제(精製)와 동정(同定)을 행(行)하였다. 3. Ipomeamarone과 Umbelliferone의 경시적(經時的) 생성량(生成量)을 관찰(觀察)하고 이들의 가능(可能)한 생합성(生合成) 경로(經路)에 관(關)하여 고찰(考察)하였다. 4. 연부(軟腐)고구마의 이상대사산물중(異常代謝産物中) Ipomeamarone과 이의 유사물질(類似物質)은 ${\alpha}-amylase$에 대(對)한 활성(活性)을 저해(抵害)하였으며 그 저해형(抵害型)은 비경쟁적(非競爭的)이었다. 5. 연부(軟腐)고구마로부터 분리한 Ipomeamarone은 Yeast의 호흡(呼吸)을 촉진(促進)시켰으며 산화적(酸化的) 인산화반응(燐酸化反應)의 연합해제제(聯合解除劑)인 2, 4-dinitrophenol과 동일(同一)한 효과(效果)를 나타내었다.

식물세포 배양 및 융합을 통한 유용물질 개발(I) (Development of Useful Secondary Product Through Plant Cell Culture(I))

  • 야생식물인 마디풀과 버드나무의 수용 추출액에는 상치 및 벼종자의 발아를 강하게 억제하는 물질이 존재하여 이들 품질을 GLC로 검정했다. 마디폴에 페놀물질로는 salicylic + vanillic, tannic - gallic, sinapic, p-coumaric, p-cresol 및 catech이 등의 순으로 함유되어 있었고 이들은 강한 활성을 나타냈다. 배양세포에도 유사한 페놀류가 동정되어 2차 대사물질이 세포배양에도 생성됨이 검정되었다. 지방산 가운데는 linolenic산(3.7 mg/g)과 유기산 가운데는 oxalic산(14.288 mg/g)이 가장 많이 함유되어 있었으며 이들도 생리활성을 지닌 물질로 간주된다. 그 밖에 마디풀에는 석유 에테르 추출물도 1.65% 검정되어 생리활성을 발휘하리라 사료되어 계속 연구중이나 알카로이드 함량은 아주 낮았다. 마디풀과 버드나무로부터 MS기본배지에 2,4-D(1.0 mg/l)과 BAP(0.1-0.2 mg/l)조합 처리하면 캘루스의 유기가 잘 되었으며 대량생산을 위한 세포현탁배양에 이용될 수 있는 기초자료가 된다고 사료된다.

버섯 섭취와 장내 미생물 균총의 변화 (Changes in gut microbiota with mushroom consumption)

  • 버섯의 섭취는 버섯의 유용 성분에 의해 면역체계 및 장내미생물균총의 변화를 유발한다. 그중 많이 알려진 β-glucan 구조 기반 물질들은 염증매개물질의 분비는 억제하고 대식세포의 활성은 유발하여 면역력의 증진을 도와인체의 면역기능을 증진시킨다. 직접적인 유용 성분 이외의 다른 물질은 장내 미생물균총에 의해 단쇄지방산으로 변화되며, 변화된 단쇄지방산은 면역 및 다양한 질병의 완화를 유발하게 된다. 버섯이 포함하고 있는 성분은 β-glucan 외에도 장내미생물균총을 변화하게 유도하는 prebiotic로써의 역할을 수행하게 된다. 변화된 장내미생물균총은 외부로부터 들어오는 다양한 감염성 균의 감염을 막으며, 면역체계의 균형 유지를 도우며, 질병을 예방하게 된다. 국내 섭취 빈도가 높은 새송이, 표고, 느타리, 팽이, 목이, 양송이 버섯의 유용 성분의 탐색, 장내미생물균총의 변화 및 섭취 시 인체의 단쇄지방산 변화 연구는 추후 버섯의 유용 성분, 장내미생물 및 질병과의 인과관계를 풀어내는데 기여할 것으로 여겨진다.

Protective Effect of Astaxanthin Produced by Xanthophyllomyces dendrorhous Mutant on Indomethacin-Induced Gastric Mucosal Injury in Rats

  • Nonsteroidal anti-inflammatory drugs such as indomethacin induce severe gastric mucosal damage in humans and rodents. In the present study, the in vivo protective effect of astaxanthin on indomethacin-induced gastric lesions in rats was investigated. The test groups were injected with indomethacin (25 mg/kg) after the oral administration of astaxanthin (25 mg/kg) for 1, 2, and 3 days, while the control group was treated only with indomethacin. Thiobarbituric acid reactive substances in the gastric mucosa, as an index of lipid peroxidation, increased significantly after indomethacin administration and this increase was inhibited by oral administration of astaxanthin. In addition, pretreatment with astaxanthin resulted in a significant increase of the activities of superoxide dismutase (SOD), catalase, and glutathione peroxidase (GSH-px). Histologic examination clearly revealed acute gastric mucosal lesions induced by indomethacin in the stomach of the control group, but were not observed in that of the test group. These results indicate that astaxanthin activates SOD, catalase, and GSH-px, and removes the lipid peroxides and free radicals induced by indomethacin. It is evident that astaxanthin acts as a free radical quencher and antioxidant, and is an effective molecule in the remedy of gastric mucosal lesions.

농작물 해충 및 진균류 방제를 위한 방선균의 분리 및 동정 (Isolation and identification of Actinomycetes for the Control of Agricultual Pests and Fungal Pathogene)

  • Twenty seven out of ca. 5,000 actinomycete strains, which were isolated from soil collected throughout the country, showed antibicrobial effects against fungai, Rhizopus stronifer (ATCC 6227a), Rhizoctonia solani (KCCM 11271) and yeast, Candida albicans (ATCC 10231). From these antifungal microorganisms, we further selected seven strains which seemed to produce insecticidal substances with in vivo test, using silkworm, Bombyx mori and beet armyworm, Spodoptera exigua. Morphological and biochemical experiments revealed that three strains out of seven were Streptomyces. Further investigations on the physical and chemical properties of these antifungal and insecticidal substances are now in progress.

자연치즈의 제조과정 중 숙성의 중요성 (Importance of Ripening during Natural Cheese Making)

  • The ripening of cheese allows for the development of characteristic taste and flavour, nutritional substances, bio-active components and texture, helping to improve quality. Many different microbiological, biochemical and nutritional changes occur during the process depending on the quality of raw milk, added cultures and enzymes, as well as specific processing and ripening conditions. During the ripening lactose is hydrolyzed to lactic, propionic and acetic acid, helping to reduce potential effects of the problem of lactose intolerance. Fat is hydrolyzed to butyric, propionic and conjugated linoleic acid, which function as bio-active substances. Protein is hydrolyzed to different peptides and amino acids which all show various bio-activities. However, errors of cheese ripening can happen and affect the quality of the product. To guarantee good quality cheese the process needs to be managed carefully with the right microbes used and ensuring cleanliness of processing facilities, staff, ventilation and hazard analysis and critical control points (HACCP). Research into and controlling of ripening technology is crucial for producing high quality cheeses.

  • Microalgal bioremediation of CO2, nutrients, endocrine disruptors, hydrocarbons, pesticides, and cyanide compounds have evaluated comprehensively. Microalgal mitigation of nutrients originated from municipal wastewaters, surface waters, and livestock wastewaters has shown great applicability. Algal utilization on secondary and tertiary treatment processes might provide unique and elegant solution on the removing of substances originated from various sources. Microalgae have displayed 3 growth regimes (autotrophic, heterotrophic, and mixotrophic) through which different organic and inorganic substances are being utilized for growth and production of different metabolites. There are still some technology challenges requiring innovative solutions. Strain selection investigation should be directed towards identification of algal that are extremophiles. Understanding and manipulation of metabolic pathways of algae will possible unfold solution to utilization of algae for mitigation of dissolve organic nitrogen in wastewaters.

월동 및 해동기 뽕나무의 생화학적 물질의 변동과 내동성과의 관계 (Studies on the Change of Biochemical Components during Wintering and Thawing Periods and Cold Hardiness of Mulberry(Morus))

  • In relations to cold acclimation, experiment was carried out to understand the seasonal changes in reserve substances of the mulberry. The shoot barks and leaves of three mulberry varieties(Kaeryangppong, Shinilppong and Yongcheonppong) were sampled, after that their reserve substances were analyzed. The cold hardiness of mulberry was investigated by DTA(Differential Thermal Analysis) method. To increase cold hardiness, gibberellin(100 ppm), kinetin(100 ppm) and Jambi 8 were sprayed on the mulberry leaves. After spraying, falling of the leaves of Yongcheonppong occured earier than the other varities. After the first frost, all of treatments except gibberellin were entirely fallen. Growth regulator extended the leaves fallen. After spraying, water of the shoot barks was not showed difference in the content among the treatments, but amino acid, carbohydrate and soluble protein increased from September to October. Starch content of the shoot barks and leaves was maximum in October, but thereafter decreased during wintering stage. In Shinilppong, Jambi 8 spray increased cold hardiness by 1-2$^{\circ}C$ more than no spray. It was concluded that the cold hardiness of the mulberry in midwinter is closely related to the reserve substances with spraying Jambi 8 on the mulberry leaves.

Steinernema glaseri 곤충병원선충으로부터 공생박테리아의 분리 및 배양특성 (Isolation and Culture Characteristics of a Bacterial Symbiont from Entomopathogenic Nematode Steinernema galseri)

  • Asymbiotic bacterium with highly effective toxins was isolated from entomopathogenic nematode Steinernema glaseri which has been widely used against various soil-inhabiting pests. The symbiont of S. glaseri was identified as Xenorhabdus nematophilus sp. by using several biochemical and physiological tests. When this strain was released into the hemolymph of insect larva, it produced highly toxic substances and killed the larva within 2 days. Two colony forms that differed n some biochemical characteristics were observed when cultures in vitro. Phase l colonies were mucid and difficult to be dispersed in liquid. Phase II was not mucoid and was easily dispersed in liquid. It did not adsorb neutral red or bromothymol blue. Rod-shaped cell size was highly variable between two phases, ranging 2-10 ${\mu}{\textrm}{m}$. It was also found that only infective-stage nematodes can carry only primary-phase Xenorhabdus in their intestine.

Application of Biotechnology to Wastewater Treatments in Japan

  • The biotechnology including genetic engineering is expected to be applied to various fields of wastewater treatments in order to promote biological reaction rate, to grade up the effluent quality and to advance the stability of microorganisms against temperature, pH and toxic substances. The current topics in Japan on application of biotechnology to wastewater treatment will be reviewed at the beginning of the presentation. Next, the research of Biochemical Engineering Laboratory is to be presented, especially focused on the following subjects ; (1) Application of genetic engineering to the investiation of heavy metal uptake by the resistant bacteria of Hg Cd or Zn. (2) Relationship between sulfate reducing bacteria and wastewater treatment, offensive odor and corrosion of sewer tranks.
