The Action of Triterpenoidal Glycosides of Dammarane Series and Their Aglycones on $K^{+}$ and $H^{-}$ Fluxes in Erythrocytes, Induced by lonophore $A_{23187}$ and Divalent ions ($A_{23187}$과 2가 이온에 의해 일어나는 $K^{+}$ 이온과 $H^{-}$ 이온의 흐름에 미치는 Triterpenoidal Dammarane Serids의 Glycosides와 그 Aglycones의 영향)

  • Ginsenoside Rb,, at a concentration of 10 $\mu\textrm{g}$/ml and over, initiated the cycle of oscillation of ion flux in erythrocytes after the cells had been treated with a protonophore, carbonyl cyanide p-trifluoro-methoxyphenyl hydrazone (FCCP) and then with a $Ca^{2+}$ ionophore, A23,3,. Its action was similar to the additional portion of $Ca^{2+}$-ionophore or $Ca^{2+}$ ion to the erythrocytes. Effects of $Rg_1$ and Rf were different from that of Rb,. They did not induce the oscillation. They, however, increased the extracellular $K^{+}$ concentration and pH without returning to the initial state in the erythrocytes processed with FCCP and $A_{23187}$. We established that ginsenosides from 20-(5)-panaxatriol family induced the membrane hyperpolarization in erythrocytes, which was attenuated by the pretreatment of $Rb_1$, a major component of 20-(5)-panaxadiol.

  • Recently seaweed polysaccharides have been extensively studied for alternative energy application. Because their producing cost is high and efficiency low, their industrial applications have been limited. The main component of cell wall of red algae represented by Gelidiales and Gracilariales is agar. Red-algae agar or galactan, consisting of D-galactose and 3, 6-anhydro-L-galactose, is suitable for bio-product application if hydrolyzed to monomer unit. For the hydrolysis of algae, chemical or enzymatic treatment can be used. A chemical process using a strong acid is simple and efficient, but it generates together with target sugar and toxic compounds. In an enzymatic hydrolysis process, target sugar without toxic compounds generation. The objective of this review is to summary the recent data of saccharification by chemical and enzymatic means from red seaweed for especially focused on automobile industry.

  • Pretreatment of Glycyrrhizae Radix(GR) to male Sprague-Dawley rats was demonstrated to increase excretion of acetaminophen-glucuronide ocnjugate when bile nad urine were assayed after administration of acetaminophen. In order to study the effect of GR on the glucuronidation in rats, we examined enzymatic activities of hepatic UDP-glucuronosyl-transferases (UDP-GT1 and UDP-GT2) and intracellular concentrations of hepatic UDP-glucuronic acid (UDP-GA), upon the administration of GR (1 g/kg body weight, p.o.) or glycyrrhizin (23 mg/kg body weight, p.o.) a major component of GR, for 6 days. GR and glycyrrhizin caused increases in specific activities of UDP-GT2 111% and 96% respectively. Specific activity of UDP-GT1 was increased 25% by GR treatment whereas it was not significantly increased by glycyrrhizin. Concentrations of UDP-GA were increased 257% by GR and 484% by glycyrrhizin. These data indicate that GR activated glucuronidation and thus suggest the possibility that GR may influence detoxification of xenobiotics in rat liver.

  • The hydrolysis of biomass to fermentable sugar (saccharification) and to oligosaccharide is an essential process in biotechnology including biorefinery and biofood. Various macroalgae are commercially cultivated in several Asian countries as a useful resource for food and agar production. Agar is a major component of the cell walls of red algae that can be hydrolyzed by agarase. Agarases are classified into ${\alpha}$-agarase (E.C. 3.2.1.158) and ${\beta}$-agarase (E.C. 3.2.1.81) according to the cleavage pattern and grouped in the glycoside hydrolase (GH) family (GH-16, GH-58, GH-86, GH-96, and GH-118) based on the amino acid sequences of the proteins. Agarases have been isolated from various bacteria found in seawater and marine sediments. To increase productivity of the enzyme, a research on recombinant enzymes has been done. The application of recombinant agarase can be possible in the various filed such as energy, food, cosmetics, medical and so on. This paper reviews the source, biochemical characteristics and production system of recombinant agarases for further study.

  • To investigate on the biochemical characteristics of muscle fiber, myofibrils and actomyosins were prepared front red muscle and white muscle, and their ATPase activities and SDS-polyacrylamide gel electrophoretic patterns were compared. Also biochemical characteristics of bovine muscle were compared with those of chicken muscle for the detection of species characteristics. SDS-polyacrylamide gel electrophoretic analysis indicated that red muscle contained nlore 30K component of myofibril than white muscle. Differences in KCI concen-tration dependency of actomyosin ATPase activities and ATPase activity-pH cone were observed, when bovine muscle were compared with chicken muscle.

  • The binding characteristics of Anagrapha falcifera nuclear polyhedrosis virus (AtNPV) to Spodoptera frugiperda 21 (Sf21) cells were investigated. The cells displayed an affinity of 4.7×10/sup 10/M/sup -1/ with about 3,300 binding sites per cell. The biochemical nature of the AfNPV-binding sites on the cell surface was also partially identified. Our findings suggest that the binding-site moiety has a glycoprotein component, but that the direct involvement of oligosacccharides containing N-acetylglucosamine or sialic acid residues in binding is unlikely, and that AfNPV entry into Sf21 cells may be via receptor-mediated endocytosis.

Comparison of Biochemical Characteristics of Myofibrillar Protein from Fresh Water Fish and Sea Water Fish (담수어와 해수어의 근원섬유단백질의 특성 비교)

  • Myofibril and actomyosin were prepared from red muscle and white muscle of fresh water fish and sea water fish, and their biochemical characteristics and SDS PAGE patterns of myofibril were compared. SDS PAGE analysis showed that electrophoretic patterns of myofibril were similar be tween white muscle and red muscle, while difference of 30kDa component of myofibril was detected between fresh water fish and sea water fish. When myofibril were treated with trypsin, difference in hydrolysis of heavy chain was observed between white muscle and red muscle. In activities of Ca ATPase, Mg ATPase, EDTA ATPase and ATPase activity pH curve, myofibrillar protein from fresh water fish showed higher specific activity than those from sea water fish.

  • One of the main problems to constitute control method on biologically oriented wastewater purification processes, e.g. activated sludge process, sequential batch reactor, rotating bio-disk contactor, etc. is that it is hard to control the target component directly. For instance, while biochemical oxygen demand, suspended solids, and chemical oxygen demand are the key components to check the process performance, one may not control them directly since they are the results of microbial activities related to numerous physiochemical factors.   • A phylogenetic comparison of homologous protein can often supplement genetic and biochemical analysis by revealing conserved structures that are critical for function(Waugh et al., 1989). I therefore isolated a secY homologue from B. subtilis, a gram positive bacterium evolutionary distant from E. coli. The comparison and interplay between these two bacterial systems should contribute greatly to our understanding of the functions and interactions within systems evolved for protein translocation in both prokaryotic and eukaryotic organisms.
