This study was conducted to estimate the effect of the drought on the changes of chlorophyll, protein and proline content of upper three leaves, and the grain yield components of barley plant (CV. baegdong) subjected to water stress at four stages: late vegetative, boot, anthesis early grain filling. 1. In comparison with leaf posititions in water stress, the first leaf below flag leaf maintained the highest relative turgidity, chlorophyll and protein content and showed the least proline accumulation. And, in terms of growth stages in water stress, chlorophyll was shown to be highest at anthesis stage, protein being highest at boot stage and proline being least at boot stage. 2. In boot stages, culm and spike length, and Number of grains per spike were remarkably decreased. And the weight of 1000 grains was at least level in the early grain filling stage, and also the grain size was comparatively decreased at boot and two following stages. 3. The protein content of grain by water stress, apart from early grain filling stage, was not significantly affected by water stress at different growth stages. 4. The rate of sterility was particularly increased at boot and anthesis stages. 5. It was eventually concluded that the boot stage among four growth stages, and the flag leaf in 3 leaf positions were mostly damaged by water stress at reproductive growth stage.
Purpose. Free radicals are implicated in the pathophysiology of aging, ischemic injury and neurodegenerative disorders. To deform]no whether Hominis Placenta extract prevents $H_2O_2$-induced apoptosis, we have performed morphological and biochemical analyses for the detection of apoptotic phenomena in the pineal tumor cell line $PGT-{\beta}$ We have also peformed cytochemical and immunocytochemical analyses for the detection of changes in nitric oxide synthase (NOS) activity and estimated the expression . of apoptotic genes using reverse transcription-polymerase chain reaction (RT-PCR) Methods. $PGT-{\beta}\;cells$ were pretreated with Hominis Placenta extracts $(0,\;10^{-2}\;{\mu}g/ml)$ for 2 hours and then exposed to $H_2O_2\;(0,\;50\;{\mu}M)$ for 3 hours. Appearance of apoptotic characteristics were monitored using 4, 6-diamidino-2-phenylindole dihydrochloride (DAPI) staining assay, terminal deoxynucleotidyl transferase-mediated dUTP-digoxigenin nick end labeling (TUNEL) assay and flow cytometric analysis. NOS activity was measured by NADPH-diaphorase cytochemistry. Expression of inducible NOS (iNOS) and nuclear factor kappa B (NF k B) was assessed via immunocytochemistry. The expression of apoptotic genes was examined by RT-PCR. Results. After 3 flours of exposure to $H_2O_2$, it was shown that $PGT-{\beta}\;cells$ treated with $H_2O_2(50\;{\mu}M)$ exhibit classical apoptotic features and increases in NOS activity and caspase-3 expression. Treatment with Hominis Placenta extract resulted in a reduced occurrence of apoptotic features. DAPI staining, TUNEL and flow cytometric assays revealed decreases in the occurrence of nuclear fragmentation and in the sub-Gl fraction in the $PGT-{\beta}\;cells$ treated with Hominis Placenta extract. Cells treated with Hominis Placenta extract also showed lower activity of NADPH-diaphorase and immunoreactivities of both iNOS and NF k B than those of $H_2O_2$-treated cells which were not treated with Hominis Placenta extract. By RT-PCR, it was shown that the level of caspase-3 mRNA was derreased In the cells treated with Hominis Placenta . extract. Conclusions. This study shows that Hominis Placenta extract prevents $H_2O_2$-induced apoptosis in $PGT-{\beta}\;cells$; inhibitions of iNOS and caspnse-3 are possible mechanisms of the protection against apoptosis.
The purpose of this work was to investigate the antibacterial activity of Lactobacillus plantarum YK-9 isolated from fermented Kimchi. Morphological and biochemical characteristics of L. plantarum YK-9 were examined. Phylogenetic analysis using 16S rRNA sequencing was performed to identify the strain, and the strain could be assigned to Lactobacillus plantarum, designated as L. plantarum YK-9. The strain was registered in GenBank as [FJ669130]. During the incubation period of L. plantarum YK-9, the changes of bacterial growth and residual organic acids were monitored. HPLC was used to confirm the organic acids produced in the cultures as metabolites. L. plantarum YK-9 produced both lactic acid and acetic acid, which were responsible for the pH decrease during growth. Initial pH 7.0 of the cultures decreased to 3.6 at the incubation after 72 hours, and concentrations of lactic acid and acetic acid increased to approximately 588.7 mM and 255.5 mM, respectively. The antibacterial activities against food-poisoning causing bacteria were examined with 20-fold concentrated culture supernatants from L. plantarum YK-9, and the antibacterial effects were clearly observed against all the bacteria tested in this work.

To evaluate the possible therapeutic effects of growth hormone and vitamin C on multiorgan failure, a rat model was developed for LPS-induced sepsis. Using this model, the effects of growth hormone and vitamin C on tissue damages, catalase and i-NOS activities, and MDA levels were examined in the lung and liver. The level of TNF- in plasm was also examined. Male, Sprague-Dawley rats were injected with LPS intraperitoneally then divided into 3 groups : positive controls injected with LPS only, the ones injected with growth hormone or vitamin C immediately after the LPS injections. The lung and the liver were then isolated, blood samples were collected at 24 or 48 hours after the LPS injection, then examined for histopathological and biochemical changes. The results obtained were as follows. 1. LPS induced sinusoid vasodilation and mild destruction of lobular structure in the liver. In the lung, alveolar structure appeared to be thickened and interstitial edema was observed. The levels of MDA in the liver and the lung was increased by LPS, while the activity of catalase was decreased. The activity of i-NOS of those tissues was also increased, which was more pronounced at 24 hr. The level of TNF- in plasm was increased by LPS 2. In the lung, vitamin C suppressed lymphocyte and neutrophil infiltration, alveolar wall thickening and interstitial edema. In the liver, vitamin C protected against the destruction of the lobular structure. The activity of catalase reduced by LPS was reversed partly by vitamin C. The activity of i-NOS enhanced by LPS was also reversed by vitamin C. The level of TNF- in plasm reduced in some animals by vitamin C, which however was not significant statistically(p<0.05). 3. Growth hormone showed similar protective effects against inflammation and damages in the liver and lung tissues. Growth hormone reversed partly the LPS effects on the level of MDA, the activity of catalase and i-NOS induction in the liver and the lung. Growth hormone reduced plasma level of TNF-${\alpha}$ substantially, which contrasted from vitamin C. Besides this, overall protective effects of growth hormone against LPS-induced experimental sepsis were similar to those of vitamin C. From this results, the mechanism of growth hormone on suppression of LPS-induced tissue damage might be associated with production of antioxidative enzyme and suppression of plasma TNF- level.

In order to investigate the effects of dietary n-3 highly unsaturated fatty acids (n-3HUF A) levels on growth and body composition of the Korean rockfish, six experimental diets with various levels of n-3HUFA, which were adjusted by adding squid liver oil and/or soybean oil at $8\%$ dietary lipid level, were fed to the Korean rockfish (6.2g in mean body weight) for 10 weeks. Daily weight gain, feed efficiency and nutrient retention efficiency were the lowest in the fish fed a diet containing 0% n-3HUFA. These parameters were effectively improved by supplementation with n-3HUFA, and showed linear increase up to $1.2\%$ dietary n-3HUFA level (P<0.01). There was no additional response above this level. A higher concentration of nonpolar lipids in the liver was observed for the fish fed insufficient levels of n-3HUFA in the diets. However the liver glycogen content and hepatosomatic index were slightly deareased. The lipid contents of the whole body and vicera showed significantly higher in the fish fed sufficient levels of n-3HUFA in the diets (P<0.05). The fatty acid compositions of polar lipids in the whole body and liver were affected by dietary fatty acid compositions. The contents of n-3HUFA and 18:1 in the liver increased with increasing the n-3HUFA level in the diets, while the 18:2n-6 and 18: 3n-3 decreased. These results suggest that n-3HUFA plays an essential role for normal growth of the Korean rockfish, and the requirement of n-3HUF A is around $1.2\%$ of the diet.
The distribution of respiratory chain complexes in beef heart and human muscle mitochondria has been explored by immunoeledron microscopy with antibodies made against beef heart mltochondriai proteins in conjundion with protein A cofloidai gold (l2nm particles). The antibodies used were made against NADH-conezyme Q reductase(complex I), ubiquinol-cytochrome-c-oxldoreductase (complex III) and cytochrome-c-oxidase(complex IV). Labeling of bed heart tissue with any of these antihodies gave gold particles randomly distributed along the mitochondrial inner membrane. The labeling of muscle tIssue mitochondria from a patient with a mitochondrial myopathy localized by biochemical analysis to complex III was quantitated and compared with the labeling of human control muscle tissue mitochondria. Four kinds of morphological changes in the mitochondrial fine strudure in the myopathy patient tissue have been found; paracrystalline inclusions consistIng of densely packed multi- lamellar structures, globular crystalline inclusions with high electron density, multilamellar strudure inclusion body(compadly and irregularly arranged concentric whirl shaped cristae)and golbular cyrstalilne inclusions located in the center of the whirl shaped cristae. Compex I and cytochrome-c-oxldase antihodies reacted to the same level in the mitochondria containing the crystalline inclusions and control mitochondria. Antibodies to complex III reacted very poorly to the mitochondria containing the crystalline Inclusions but strongly to control mitchondria. The globular crystalline inclusions in the mitochondria are not reacted antibodies to respiratory chain complexes.

Objectives: The effects of peroxisome proliferator-activated receptor ${\gamma}2\;(PPAR{\gamma}2)$ Pro12Ala (P12A) polymorphism on body mass index (BMI) and type 2 diabetes are well documented; however, until now, only a few studies have evaluated the effects of this polymorphism on body fat distribution. This study was conducted to elucidate the effects of this polymorphism on computed tomography (CT)-measured body fat distribution and other obesity-related parameters in Korean female subjects. Methods & Results: The frequencies of $PPAR{\gamma}2$ genotypes were: PP type, 93.0%; PA type, 6.8%; and AA type, 0.2%. The frequency of the A allele was 0.035. Body weight (P .012), BMI (P .012), and waist-to-hip ratio (WHR) (P .001) were significantly higher in subjects with PA/AA compared with subjects with PP. When body composition was analyzed by bioimpedance analysis, lean body mass and body water content were similar between the 2 groups. However, body fat mass (P .003) and body fat percent (P .025) were significantly higher in subjects with PA/AA compared with subjects with PP. Among overweight subjects with BMI of greater than 25, PA/AA was associated with significantly higher abdominal subcutaneous fat (P .000), abdominal visceral fat (P .031), and subcutaneous upper and lower thigh adipose tissue (P .010 and .013). However, among lean subjects with BMI of less than 25, no significant differences associated with $PPAR{\gamma}2$ genotype were found, suggesting that the fat-accumulating effects of the PA/AA genotype were evident only among overweight subjects, but not among lean subjects. When serum lipid profiles, glucose, and liver function indicators were compared among overweight subjects, no significant difference associated with $PPAR{\gamma}2$ genotype was found. Changes in body weight, BMI, WHR, and body fat mass were measured among overweight subjects who finished a 1-month weight lose program of a hypocaloric diet and exercise; no significant differences associated with $PPAR{\gamma}2$ genotype were found. Conclusions: The results of this study suggest that the $PPAR{\gamma}2$ PA/AA genotype is associated with increased subcutaneous and visceral fat areas in overweight Korean female subjects, but does not significantly affect serum biochemical parameters and outcomes of weight loss programs.
To study the follicular atretic mechanism in mammalian ovary, the medium-sized (3.0-6.0mm) follicles of porcine ovary were morphologically classified as nonnal and atretic. Steroid concentrations in the follicular fluid were analyzed by radioimmunoassay, and the proteins in that fluid were electrophoretically separated. Concentrations of progesterone in the atretic follicular fluid of follicular phases were higher than those of normal ones (p < 0.05). Concentrations of testosterone were high in normal luteal and atretic follicular-phases follicles. The concentrations of estradiol remained significanily lower in atretic follicular-phases follicles than normal. After sodium dodecyl sulfate-polyacrylamide gel electrophoresis of follicular fluid proteins, four kinds of proteins (20K, 32K, 33K, 38K) were detected, and those proteins were not present in sera. According to the ovarian cycle, proteins of MW of 112K and 141K were identified. In atretic follicular fludies, MW of 23K and 24K were specifically detected. From the above results, we can conclude that, as ovarian cycle changes, steroid contents and protein composition in atretic follicular fluid are different from the normal developing follicular fluid. To further understand the physiologic roles of the proteins present in the atretic follicular fluids, these proteins need to be characterized and identified.
A study has been made to correlate morphological and biochemical differentiation in the oocytes of a tubiculous polychaete, Schizobranchia insignis. The pressent paper is concerned with an examination of the cytological changes during oogenesis and annual size distribution of oocytes, The oocytes are released from the ovary into the coelomic sac at the end of the oogonial division and grow to a maximum size (180 $\\mu$ diameter). Oogensis takes place continuously throughout a year, although the breeding season is the period between January and March. When the oocytes reach the largest size class, they remain constant in size thereafter and accumulate in the coelomic sac. The nucleolus, which first appears in the oocytes 5-10$\\mu$ diameter, grows in the early stages of oogenesis, becomes maximum in the oocytes 100-120$\\mu$ diameter, and is constant throughout the rest of the pertiod. The nuclelus initially has a single comartment but becomes bipartite prior to vitello genesis. Three types of yolk including lipid droplets, proteid granules and oval granules oof unknown composition form at different times of oogenesis. The lipid droplets and oval granules appear in the early stage, but mainly in the oocytes larger than 80 $\\mu$ diameter. Proteid yolk and cortical granules appear only in the oocytes larger than 80 $\\mu$ diameter Microvilli are abundant in the oocyte 80 $\\mu$ diameter and embedded in the vitelline membranc. In the oocytes 180 $\\mu$ diameter they have retracted from the vitelline membrane.
Despite the risk of high altitude disease (HAD), raising cattle at higher altitudes does have advantages such as beef quality due to minimal chance of infection and stress. The ideal situation is to determine the optimum altitude suitable for raising cattle while at the same time minimizing the risk of HAD. Therefore, in this study, we documented the health status of three groups of steers, raised at three different altitudes (200m, 400m and 800m) in Gangwon province, using routine hematological, biochemical and hormonal tests. The red blood cell counts and hemoglobin concentration were highest in the 800m group, and lowest in the 200m group, possibly due to hypoxia induced myeloid and erythroid stimulation. The mean concentration of AST, BUN and serum cortisol were lowest in the 800m. These findings suggest the stress factor will contribute the general health status of animal and indicate a difference in the Hanwoo groups raised at 800m compared to 200m, where the cattle raised at the higher altitude exhibit better health status compared to the lower altitude, possibly due to less stressful environment in the higher altitude.
