저온 처리가 포도주스의 품질에 미치는 영향 (Effects of Cold Stabilization on the Quality of Grape Juice)

  • '캠벨' 포도주스의 이화학적 및 관능적 품질특성에 저온 안정화 처리기간이 미치는 영향을 조사하기 위하여, 포도 과실로 압착 주스를 제조하여 $-5^{\circ}C$에서 30일간 보관하면서 주스의 각종 품질특성 변화를 조사하였다. 주스의 당/산비와 pH는 증가하였으나 적정산도와 가용성 고형분 함량은 경시적으로 감소함을 보였다. 주스의 명도($L^*$ 값)와 황색도($b^*$ 값)는 증가하는 경향을 보였으며 적색도($a^*$ 값)는 감소하는 경향을 각각 나타내었다. 주스의 총페놀, 안토시아닌, 플라보노이드 함량과 유리기 소거능 및 관능적 기호도는 안정화처리 기간이 길어질수록 점차적으로 감소하였다. 이로써 저온 안정화 처리기간은 포도주스의 품질 변화에 영향을 미치며 장기간 처리는 품질을 오히려 저하시키는 것으로 확인되었다.

The I/LWEQ Domain in RapGAP3 Required for Posterior Localization in Migrating Cells

  • Cell migration requires a defined cell polarity which is formed by diverse cytoskeletal components differentially localized to the poles of cells to extracellular signals. Rap-GAP3 transiently and rapidly translocates to the cell cortex in response to chemoattractant stimulation and localizes to the leading edge of migrating cells. Here, we examined localization of truncated RapGAP3 proteins and found that the I/LWEQ domain in the central region of RapGAP3 was sufficient for posterior localization in migrating cells, as opposed to leading-edge localization of full-length Rap-GAP3. All truncated proteins accumulated at the leading edge of migrating cells exhibited clear translocation to the cell cortex in response to stimulation, whereas proteins localized to the posterior in migrating cells displayed no translocation to the cortex. The I/LWEQ domain appears to passively accumulate at the posterior region in migrating cells due to exclusion from the extended front region in response to chemoattractant stimulation rather than actively being localized to the back of cells. Our results suggest that posterior localization of the I/LWEQ domain of RapGAP3 is likely related to F-actin, which has probably different properties compared to newly formed F-actin at the leading edge of migrating cells, at the lateral and posterior regions of the cell.

  • Cow's milk and dairy products are elements of the human diet that could play an important role in improving human health. The macronutrients and micronutrients found in milk could supply the nutrients required to maintain human health. Among them, milk-derived bioactive peptides have been identified as potential ingredients found in health promoting functional foods. These bioactive peptides target diet-related chronic diseases, particularly non-communicable ones such as cardiovascular disease, diabetes and obesity. Additionally probiotics such as lactic acid bacteria (LAB) are can be considered live microorganisms that confer health benefits for the host-, when administered in adequate amounts. Further, the calcium, vitamin D, and protein content of milk and dairy products could play a role in proving bone health. The effect of milk and calcium on bone mineral density could prevent against fracture, osteoporosis and rickets. Furthermore, milk and dairy products also contain which factors that, which protect against dental caries (anti-cariogenic properties). This paper reviews the various nutritional functions of milk and dairy products in improving human health.

Antioxidant and Antifungal Activity of Endophytic Fungi Associated with Agarwood Trees

  • Several species of Aquilaria and Gyrinops are native to Indonesia and well known as agarwood-producing trees with a high economic value. Their bioactive compounds have a wide spectrum of uses, such as in medicine and cosmetics. These genera have undergone extensive search for novel bioactive compounds. The purpose of this study was to isolate, identify, and characterize the endophytic fungi community associated with Aquilaria malaccensis, A. microcarpa, Gyrinops versteegii, and A. crassna trees and investigate their bioactive properties as antioxidant agents and antagonists. A total of 50 fungi were successfully isolated from different tissues of the four species of agarwood-producing trees. Two isolates exhibited strong antioxidant activity, namely, Apodus oryzae (R2MC3A, $IC_{50}$ 60.92 mg/mL) and Diaporthe sp. (P1DS1[C], $IC_{50}$ 76.65 mg/mL). Two isolates, Pestalotiopsis theae (P3BS3[B]) and Curvularia sp. (P2CD3A), showed >75% antifungal activity against pathogenic Fusarium solani. The results revealed that endophytic fungi associated with the studied agarwood-producing trees had potential antioxidant and antifungal activities for further applications in biotechnology.

1H-NMR-Based Metabolic Profiling of Cordyceps militaris to Correlate the Development Process and Anti-Cancer Effect

  • The study of metabolomics in natural products using the diverse analytical instruments including GC-MS, LC-MS, and NMR is useful for the exploration of physiological and biological effects and the investigation of drug discovery and health functional foods. Cordyceps militaris has been very attractive to natural medicine as a traditional Chinese medicine, due to its various bioactive properties including anti-cancer and anti-oxidant effects. In this study, we analyzed the metabolite profile in 50% ethanol extracts of C. militaris fruit bodies from three development periods (growth period, matured period, and aging period) using $^1H-NMR$, and identified 44 metabolites, which are classified as 16 amino acids, 10 organic acids, 5 carbohydrates, 3 nucleotide derivatives, and 10 other compounds. Among the three development periods of the C. militaris fruit body, the aging period showed significantly higher levels of metabolites including cordycepin, mannitol (cordycepic acid), and ${\beta}-glucan$. Interestingly, these bioactive metabolites are positively correlated with antitumor growth effect; the extract of the aging period showed significant inhibition of HepG2 hepatic cancer cell proliferation. These results showed that the aging period during the development of C. militaris fruit bodies was more highly enriched with bioactive metabolites that are associated with cancer cell growth inhibition.

  • Field of enantioseparations represents one of the actual topics of chemistry. Significant, in some cases dramatic differences in a desired activity as well as in toxic properties of enantiomers of chiral drugs, food additives, agrochemicals, cosmetic products, etc. represents a driving force in this field. The enantiomers of all chiral bioactive compounds have to be isolated and to be tested. Besides the ethical or environmental reasons for developing single enantiomers, it may be a real therapeutic benefit and in some cases, it has been used as a strategy for extending patent life.   • The branch-like, feather-like $BaCrO_4$ nano-superstructures were synthesized with bioactive eggshell membrane as directing and assembly template. Studies on the two products revealed that they formed through the self-assembly of nanoparticles, and that the optical properties of the products were different from $BaCrO_4$ bulk materials.

  • Quercetin is one of flavonoids widely distributed in the plants and well known to have antioxidants, antiinflammatory, antimicrobial properties. In this study, alginate sponge containing quercetin-encapsulated nanocapsules was prepared by miniemulsion polymerization, dyring/crosslinking method and their bioactive characteristics were investigated. Alginate sponge containing quercetin-encapsulated nanocapsules were evaluated using a field emission scanning electron microscope(FE-SEM), a high performance liquid chromatography, cell viability, DPPH radical scavenging activity and antibacterial activity. The study indicates that alginate sponge containing quercetin-encapsulated nanocapsules had significant antioxidant, antiinflammatory and antibacterial activities. This study suggested that alginate sponge containing quercetin-encapsulated nanocapsules can be a potential candidate for medical materials.

  • Polyamine dendrimers functionalized with electrochemiluminescent (ECL) polypyridyl Ru(II) complexes, dend-$[CO-(CH_2)_3-mbpy{\cdot}Ru(L)_2]_3(PF_6)_6$ (dend: N$(CH_2CH_2NH)_3$-, L: bpy, o-phen, phen-Cl, DTDP), were synthesized through the complexation of dendritic polypyridyl ligands to Ru(II) complexes. Their electrochemical redox potentials, photoluminescence (PL), and relative ECL intensities were studied. The ECL emissions produced by the reaction between the electro-oxidized $Ru^{3+}$ species of polyamine dendrimers and tripropylamine as a coreactant were measured in a static system with potential cycles between 0.8 and 1.3 V or through flow injection analysis with a potential of +1.3 V, and were compared to that of $[Ru(o-phen)_3](PF_6)_2{\cdot}Dend-[CO-(CH_2)_3-mbpy{\cdot}Ru(bpy)_2]_3(PF_6)_6$ showed an ECL intensity that was two-fold greater than that of the reference complex $[Ru(o-phen)_3](PF_6)_2$.

Adventitious root culture for secondary metabolite production in medicinal plants: A Review

  • Medicinal plants are high-value natural resources that have been used as precautionary drugs by many people globally. The increasing global demand for bioactive compounds from medicinal plants has led to the overexploitation of many valuable species. One widely used approach to overcome this problem is the use of adventitious root cultures as a propagation strategy. This review examines the scientific research published globally on the application of adventitious root cultures for many medicinal plants. Adventitious roots generated under aseptic environments in suitable phytohormone-augmented medium exhibit high growth rates and production of important secondary metabolites. Parameters such as medium properties and composition, growth hormone type, and elicitation strategies for in vitro grown adventitious roots of medicinal plants, are the main topics discussed in this review. We also examine current developments in bioreactor system cultivation for plant bioactive compounds using adventitious root cultures, a technology with possible commercial applications, via several studies on adventitious root culture of medicinal plants in which bioreactor systems play a role. In conclusion, the development of adventitious root cultures for medicinal plants is highly useful because of their capability for vegetative propagation and germplasm preservation.
