Comparison of Bioactive Compounds and Quality Traits of Breast Meat from Korean Native Ducks and Commercial Ducks

  • The aim of this research was to compare the bioactive compound content and quality traits of breast meat from male and female Korean native ducks (KND) and commercial ducks (CD, Cherry Valley). Meat from three 6-wk old birds of each sex from KND and CD were evaluated for carcass and breast weights, pH, color, cooking loss, shear force, and bioactive compound (creatine, carnosine, anserine, betaine, and L-carnitine) content. KND showed significantly higher carcass weights than CD whereas no such difference (p>0.05) was found between male and female ducks. The breed and sex had no significant effects on the breast weight, pH value, and shear force. However, KND had significantly lower cooking loss values than did CD. Creatine, anserine, and L-carnitine contents were significantly higher in KND than in CD and were predominant in female ducks compared to males. The results of this study provide rare information regarding the amounts and the determinants of several bioactive compounds in duck meat, which can be useful for selection and breeding programs, and for popularizing indigenous duck meat.

Effect of Nitrogen and Silicon Nutrition on Bioactive Gibberellin and Growth of Rice under Field Conditions

  • Gibberellins are growth hormones that play a pivotal role in the growth and development of plants. Present investigations were carried to check the effect of nitrogen(N) and silicon(Si) on bioactive $GA_1$ and its immediate precursor $GA_{20}$ at different growth stages of two rice cultivars with different maturity traits. It was observed that the endogenous bioactive $GA_1$ level gradually increased during vegetative stage and anthesis stage of both Junghwabyeo(early flowering cultivar) and Daesanbyeo(late flowering cultivar). However, the $GA_1$ and $GA_{20}$ content start decreasing during the seed filling stage in both rice cultivars, which indicated a possible relationship of bioactive $GA_1$ and floral development. Our results also confirmed that early 13-hydroxylation pathway was operated at all developmental stages of rice plant. Variation in the levels of the endogenous gibberellins in rice shoots were measured by GCMS-SIM using $^2H_2$-labeled gibberellins as internal standards. Combined application of N and Si enhanced growth parameters and reduced lodging index of both rice cultivars. It was thus concluded that the level of physiologically active $GA_1$ increased during vegetative and early reproductive stage, but starts declining at seed filling stage.

Histological Observations on Bone Healing with Bioactive Glass in Horizontal Ridge Augmentation;A Report of Four Cases (수평적 치조제증대술에 사용된 Bioactive glass의 골재생에 관한 조직학적 관찰;증례보고)

  • 임프란트 식립을 필요로 하는 환자의 수평적 치조제 결손의 증대를 위해 골유도재생술과 병용한 bioactive glass (BG) $(Biogran^{(R)})$ 이식의 골재생 양상을 각기 다른 치유기간을 부여한 4명의 환자에서 평가하였다. 6, 8, 10, 18개월의 치유기간 후 임프란트 식립부위에서 조직절편을 채득하여 골재생을 조직계측학적으로 평가하였다. 임프란트 식립을 위한 surgical reentry시 모든 이식부위는 임상적으로 명확한 수평적 치조제 폭경의 증가를 관찰할 수 있었다. 하지만 조직학적 분석결과 BG는 불량한 골전도성을 나타내었다. 6, 8개월의 치유기간후, 이식부위에서 신생골이 거의 관찰되지 않았으며(2.5%이하), 이식부와 기존 골의 경계부위에서 BG particle에 대한 신생골 성장과 결합양상 또는 관찰할 수 없었다. 10개월의 치유기간후 기존 골조직으로부터 성장한 신생골의 BG particle과의 직접적인 접촉양상을 일부 관찰할 수 있었다. 이식부는 13.2%의 광물화된 신생골조직을 보였고, 대부분의 BG particle은 결체조직으로 둘러싸여 있었다. 18개월의 치유기간이 부여된 환자의 조직절편에서 신생골은 이식부의 10.7%를 차지하여 비교적 낮은 신생골 형성양을 나타내었고, 이식부에 존재하는 잔존BG particle은 대부분은 결체조직으로, 일부분에서 광물화된 골조직으로 둘러싸여 있었다. 6, 8, 10, 18개월에서 잔존 BG particle양은 전체 이식부 면적에 대해서 각기 22.3%, 26.5%, 30.7%, 18.7%로 나타났다. 본 증례보고는 비록 한정적인 4명의 환자에서의 조직계측학적 평가결과이지만, 수평적 치조제 결손의 증대를 위해 골유도재생술과 병용한 bioactive glass이식은 불량한 골전도성으로 인해 효과적인 골재생을 위한 이식재로서는 적절하지 않을 수 있음을 나타낸다.

Amperometric Glucose Biosensor Based on Sol-Gel-Derived Zirconia/Nafion Composite Film as Encapsulation Matrix

  • An amperometric glucose biosensor has been developed based on the use of the nanoporous composite film of sol-gel-derived zirconia and perfluorosulfonated ionomer, Nafion, for the encapsulation of glucose oxidase (GOx) on a platinized glassy carbon electrode. Zirconium isopropoxide (ZrOPr) was used as a sol-gel precursor for the preparation of zirconia/Nafion composite film and the performance of the resulting glucose biosensor was tuned by controlling the water content in the acid-catalyzed hydrolysis of sol-gel stock solution. The presence of Nafion polymer in the sol-gel-derived zirconia in the biosensor resulted in faster response time and higher sensitivity compared to those obtained at the pure zirconia- and pure Nafion-based biosensors. Because of the nanoporous nature of the composite film, the glucose biosensor based on the zirconia/Nafion composite film can reach 95% of steady-state current less than 5 s. In addition, the biosensor responds to glucose linearly in the range of 0.03-15.08 mM with a sensitivity of 3.40 $\mu$A/mM and the detection limit of 0.037 mM (S/N = 3). Moreover, the biosensor exhibited good sensor-to-sensor reproducibility (~5%) and long-term stability (90% of its original activity retained after 4 weeks) when stored in 50 mM phosphate buffer at pH 7 at 4 ${^{\circ}C}$.

  • Initial screening assay for depigmenting agents includes in vitro mushroom tyrosinase assay and antioxidant assay. Based on this screening result, melanin synthesis in melanocyte, in screened samples, is further measured. Measuring cellular melanin needs time, human resource, and skills. Therefore initial screening method should be reliable. We examined, 34 Chinese herbs, correlated the screening assay methods with cellular melanin. No reliable relationship was observed between factors, indicating the limitation in the use of these assays, probably due to the complexicity of melanogenesis.

  • Two species, P. pyrifolia and P. ussuriensis, of the genus Pyrus native to Korea, are valuable genetic resources that can be used for food, dietary supplement, pharmaceutical, and cosmetics. Bioactive compounds of the plant leaves are the main components that are used for the products. Farmers had cultivated a few individuals from the wild to produce fruits or leaves for traditional remedy or tea; however, bioactive components of their leaves are not tested. We selected some trees from the natural stand that have distinct traits for the improvement program. We investigated the bioassay on the extracts' bioactive compounds and antioxidant capacity from the selected accessions and other accessions, including newly developed cultivars. The contents of the phenolic compounds and flavonoids from the leaf extracts of the selected accessions were higher than the commonly cultivated trees in both species but lower than 'Sanhyang' in P. ussuriensis. The antioxidant capacity was measured using two assay methods, DPPH and ABST. The selected cultivars also had higher inhibitory activity than common trees. The selected accession 'Cultivar 3' in P. pyrifolia had the highest radical scavenging activity than others. Although leaves of the accessions were used only in this study, all three selected individuals have the potential for cultivar in containing high bioactive compounds and antioxidant capacity.

Effect of Feeding Alfalfa and Concentrate on Meat Quality and Bioactive Compounds in Korean Native Black Goat Loin during Storage at 4℃

  • The primary aim of this study was to evaluate the effect of feeding alfalfa: Concentrate at different ratios (8:2 or 2:8) to Korean native black goats (KNBG) for 90 days on meat quality and bioactive compound content. Feeding KNBG alfalfa and concentrate at different ratios did not impact meat pH, color, microorganism composition, volatile basic nitrogen levels, or lipid oxidation. The low alfalfa (KLA) group exhibited increased oleic acid and monosaturated fatty acid levels, both of which impact the palatability traits of meat. The abundance of bioactive compounds increased in the loin meat of the KLA group, leading to an increase in antioxidant activities. Our results suggest that feeding alfalfa and concentrate at a 2:8 ratio to KNBG can increase taste-related fatty acids and bioactive compounds in loin meat, relative to that achieved by feeding at an 8:2 ratio. Further investigation is required to evaluate the quality and the metabolites of bioactive compounds in KNBG meat and the effect of the different dietary ratios of forage and concentrate.

Evaluation of Dermal Bioactive Properties of the Gastrodiae Rhizoma Extract by Steaming Times (증숙 횟수에 따른 천마 추출물의 피부 생리 활성 평가)

  • Objectives : Gastrodiae Rhizoma extract (GE) is possess the various bioactive compounds such as gastrodin, vanilyl alcool and p-hydroxybenzyl alcohol. Various processing methods such as steaming have been widely applied to ease ingestion and enhance the therapeutic effects of plant materials including GE in East-Asia area. The aim of this study was to evaluate the dermal bioactive properties of GE. Methods : First, total phenol, total flavonoid, gastrodin and ergothionein contents of GE were measured. In order to evaluate the dermal bioactive properties of steamed GE compared with not-steamed GE, tyrosinase, collagenase and elastase inhibitory activity were tested. Furthermore, the anti-oxidant activity of GE assessed based on DPPH and ABTS radical scavenging assay. Results : In results, total phenol and total flavonoid contents were increased when 9 times steamed compared to not-steamed GE. Also, GE increased gastrodin contents, in proportion to the number of steaming times and ergothioneine content was abolished in the steaming state. The DPPH radical scavenging activity of GE increased by steaming, but the ABTS radical scavenging activity was not related to the steaming process. In addition, the tyrosinase inhibitory activity was increased as the number of steaming times of GE increased. Collagenase was most inhibited by 4 times steamed GE, and elastase was inhibited by 8 times steamed GE. Conclusion : In conclusion, these results suggest that steamed GE extract has the potential as a cosmetic material which possess anti-oxidant and whitening activities than not steamed GE.

  • A photo-labile compound which is bioinactive but, upon irradiation with light, yields bioactive species is called as "caged compound". Photolysis of caged compounds generating bioactive species, has become a general method to produce a desired amounts of bioactive species in the specific time interval at the desired place or area of the target biological systems. For this purpose, we designed and synthesized caged hydroxyl radical., "Photo-Fenton Reagent" NP-IIl. NP-IIl has a strong absorption maximum at 377 nm and yields hydroxyl radicals upon UV light irradiation. The antioxidant activity of the ${\alpha}$ -lipoic acid and other naturally occurring compounds has been examined by using NP-IIl as a molecular probe. For example, upon photoirradiation of NP-lII with BSA or apolipoprotein of human low density (LDL), the significant oxidative modifications were observed in both cases. The oxidation was completely suppressed in the presence of ${\alpha}$-lipoic acid, which clearly demonstrates the strong hydroxyl radical scavenging activity of ${\alpha}$-lipoic acid.   • The average life span is over 80 years of age, and various biomaterials have being studied. Many research institutes and companies around the world have been commercializing bioactive glass through R&D, however, there is not much research in Korea. Most bioactive glass is applied to bone regeneration in powder form due to its excellent bio-compatibility. Recently, new applications such as scaffolds for tissue engineering and nerve regeneration have been found in composite form. The global market size is not as large as US $ 556 million in 2019, but the growth rate is very high at a CAGR of 14.35 %. This field is waiting for the challenge of new researchers.
