This study was conducted to investigate changes of vegetation and soil characteristics to tidal land of Saemangeum reclaimed land from 2010 to 2012. Soil salinity was 0.16-22.3 dS $m^{-1}$ in the first survey, while the three years later, it was decreased to 0.12-4.22 dS $m^{-1}$. Vegetations were classified as 6 families and 26 species but it was increased to 8 families and 31 species after three years. Numbers of average species in survey site were increased from 7.1 species to 10.6 species. Numbers of vegetation were increased at each survey sites except for survey site 7 : there was decreased halophyte according to decrease in the soil salinity. Biomass production was increased at low production site, and showed highest production in area of dominant vegetation as Phragmites communis. Simpson's dominance ratio(SDR) of main vegetation as Phragmites communis, Calamagrostis epigeios were increased but Suaeda maritima, Salicornia europaea, Puccinellia nipponica and Zoysia sinica were decreased.

The objective of this study was to determine the effect of replacing perlite (PL) with pellets processed with poultry feather fiber as an inert material to prepare growing medium. The growth and flowering of Salvia splendens 'Vista Red' grown in individual growing medium $Biosangto^{(R)}$, peat moss (PM), PL, and two pellets (P45-1 and P45-2) were evaluated. Peat moss was mixed with PL, P45-1, or P45-2 at various ratios (1:0 to 1:3 or 3:1 by volume) to investigate the feasibility of replacing PL with pellets. Nutrient composition of the growing medium and leaf tissues was analyzed. The number of florets, inflorescence length, plant height, and fresh weight of plants grown in media containing P45-1 or P45-2 were reduced compared to those grown in individual growing medium PM or PL. As the mixing ratio of P45-1 or P45-2 to PM was higher, the growth of salvia, such as inflorescence length, plant height, number of leaves, and fresh weight was inhibited. Our results indicate that mixing three parts PM with one part of P45-1 (PM/P45-1/3:1) or P45-2 (PM/P45-2/3:1) accelerated flowering and increased the number of florets and leaves compared to other mixing ratios of PM and pellets media. The concentrations of phosphorus (P), calcium (Ca), boron (B), iron (Fe), and copper (Cu) in individual growing medium PL, P45-1, and P45-2 were significantly lower than those in PM. The concentration of N was the highest in leaves of plants grown in P45-1 or P45-2 amended media, and the concentrations of P, Ca, and zinc (Zn) in leaves were lower in individual growing medium P45-1 or P45-2 than in PM and PL. The pH of PM/P45-1/3:1 or PM/P45-2/3:1 media was maintained at optimal level (5.8-5.9) and the concentrations of macro- and micro-elements in the media and leaves were considered to be optimal levels. Therefore, mixing three parts PM with one part P45-1(PM:P45-1/3:1) or P45-2 (PM:P45-2/3:1) is recommended for improved growth and flowering in salvia. This suggests that P45-1 or P45-2 can replace PL as an inert material to prepare growing medium.
A wastewater purifying system using phototrophic bacterium, Rhodopseudomonas capsulata, is currently in operation in several countries, One of them, is a continuously aerated bioreactor(CABR) system, which treats concentrated swin wasterwater using small amounts of phototrophic bacteria as additive bacterial seeding. Using this plant, total biochemical oxygen demand was decreased to 13%, and most of volatile fatty acids were removed. About 40% of the wastewater(Influx) was evaporated during aerobic digestion for 24h, and 60% of that erupted in a decodorized foam(Efflux). The efflux had enough nutrients, N, P and K kor growing plant, as well as organic matters. When the efflux was applied to Italian ryegrass with high dose, fresh shoot and root weights were significantly greater, and $NO_3-N$ contents of the dried shoot were lower than those of control plant (CDU). These results indicate that the CABR plant is useful for reduction and deodorization of swine wastewater and the efflux from CABR can be used for crop production as an organic fertilizer.
An efficient method of plant regeneration from Acanthopanax chiisanensis somatic embryos was developed. Cotyledonary somatic embryos were obtained in liquid Murashige and Skoog (MS) medium from embryogenic cell suspension cultures. They were desiccated for 0 to 72 hr and then cultured on MS medium containing NAA, BA, GA$_3$, (0-0.5mg/L). The highest multiple shoots formation (100%) was obtained from 72 hr desiccated somatic embryos on ifs medium with 0.5mg/L NAA+0.5mg/L BA or 0.5 mg/L NAA+0.5mg/L BA+0.5mg/L GA$_3$ after 6 weeks culture. Plant conversion from multiple shoots was not high. The highest plant conversion from multiple shoots was obtained on 1/3MS medium with 1.0mg/L GA$_3$. Converted plantlets were transferred to ex vitro condition and the highest survival rate (70%) of the plantlets was obtained on plastic pots containing vermiculite and sand. These results indicate that micropropagation procedure can be applied for an efficient mass propagation of Acanthopanax chiisanensis.
Inhibitor of DNA binding protein or inhibitor of differentiation(Id) is largely considered as positive and/or negative regulators of proliferation, differentiation, angiogeneisis, and apoptosis. The four Id genes(Id1, Id2, Id3, and Id4) were known in mammals. However, little is known about the expression and function of these genes in reproductive physiology. Among them, this study was conducted to analyze the expression pattern of Id3 mRNA on folliculogenesis in rat ovary. After PMSG administration, the ovaries were obtained at 3, 6, 12, 24, 36, and 48hrs, fixed, dehydrated, and paraffin embedded. For in situ hybridization, anti-sense and sense Id3 cRNA probes were prepared and applied to the ovarian section. The ovarian sections were coated with NTB-2 emulsion. After that, the slides were developed and counterstained with hematoxylin and eosin staining. The hybridization signal was estimated on a scale of 1+ to 4+. In oocyte, the intensity of Id3 mRNA in primordial and primary follicles was scored at ${\geq}2+$, but the intensity was less than 1+ in secondary, dominant, and preovulatory follicles. In granulosa cells, the Id3 mRNA was strongly expressed(3+ or 4+) in dominant and preovulatory follicles. Taken together, Id3 mRNA was expressed specifically at follicle stages and follicular tissue and might be closely related with follicle development.

This study was conducted to investigate the effect of microbial inoculation as additive consisting of novel lactic acid bacteria on quality and fermentation characterization of Miscanthus sinnensis silages. The contents of crude protein, acid detergent fiber, neutral detergent fiber in treatments of additive of lactic acid bacteria (ALAB) inoculation had similar to control. pH of Miscanthus sinnensis (MS) silage in treatments of ALAB inoculation significantly decreased as compared to control (p<0.05). The content of lactic acid in in treatments of ALAB inoculation significantly increased (p<0.05), but the content of acetic acid in treatments of ALAB inoculation decreased. In addition, number of lactic acid bacteria in treatments of ALAB inoculation significantly increased as compared to control (p<0.05). Therefor, we suggest that MS silage improved by inoculation of additive consisting of novel lactic acid bacteria.

This study was conducted to investigate the effect of addition of lactic acid bacteria inoculants on quality of rye silage harvested at early heading stage. The nutritive values in lactic acid bacteria (LAB) inoculated group showed similar results to control. The pH of rye silage in LAB inoculation significantly decreased as compared to control (p<0.05). In addition, the content of lactic acid in LAB inoculation significantly increased (p<0.05), but the content of acetic acid in LAB treatments decreased. In addition, lactic acid bacterial counts in LAB inoculation significantly increased as compared to control (p<0.05). Therefore, we suggest that rye silage could be improved by LAB inoculation.
To verify the sex steroids which are involved in oocyte maturation of the blacktip grouper, $Epinephelus$$fasciatus$, we incubated vitellogenic oocytes (0.41 and 0.50 mm in average diameter) in the presence of exogenous steroid precursor ($[^3H]17{\alpha}$-hydroxyprogesterone). Steroids were extracted, separated and identified by thin layer chromatography. The major metabolites produced were androstenedione, estradiol-$17{\beta}$, estrone and progestogens. Progestogen metabolites in the oocytes of 0.50 mm were more abundant than those of 0.41 mm. Also, we investigated the $in$$vitro$ effects of human chorionic gonadotropin (HCG; 5, 50 and 500 $IU/m{\ell}$), $17{\alpha},20{\beta}$-dihydroxy-4-pregnen-3-one ($17{\alpha}20{\beta}P$) and $17{\alpha},20{\beta}$-trihydroxy-4-pregnen-3-one ($17{\alpha}20{\beta}21P$; 5, 50 and 500 $ng/m{\ell}$, respectively) on oocyte maturation. In the oocytes of 0.41 mm, treatment with 50 IU HCG stimulated GVBD ($55.30{\pm}1.20%$) compared with controls ($32.41{\pm}3.13%$, $p$<0.05). In the oocytes of 0.50 mm, treatment of $17{\alpha}20{\beta}P$ (50 and 500 $ng/m{\ell}$) stimulated GVBD ($50.13{\pm}2.52$ and $51.77{\pm}5.91%$, respectively) compared with controls ($36.81{\pm}2.89%$, $p$<0.05). Treatment with 500 IU HCG also stimulated GVBD ($49.59{\pm}5.15%$) compared with controls ($p$<0.05). Taken together, these results suggested that both HCG and $17{\alpha}20{\beta}P$ were effective on in vitro oocyte maturation and $17{\alpha}20{\beta}P$ may act as a maturation inducing hormone in blacktip grouper.

The approach presented in this article refers to the bioanalytical method validation for the detection and quantitative determination of arsenic species including arsenite (As(III)), arsenate (As(V)), dimethylarsinic acid (DMA) and monomethylarsonic acid (MMA) in dog plasma by high-performance liquid chromatography inductively coupled plasma mass spectrometry (HPLC-ICP/MS). The arsenic species were separated using an agilent As speciation column by a mobile phase of 2 mM sodium phosphate monobasic, 0.2 mM ethylenediaminetetraacetic acid disodium salt dehydrate, 10 mM sodium acetate, 3 mM sodium nitrate and 1 % ethyl alcohol at pH 11 (adjusted with 1M NaOH). The method validation experiment was obtained selectivity, linearity, accuracy, precision, matrix effect, recovery, system suitability, dilution integrity and various stabilities. All calibration curves showed good linearity (R2>0.999) within test ranges. The lower limit of quantitation (LLOQ) was 5 ng/mL for As(III), As(V) and DMA, and 20 ng/mL for MMA. The system suitability and dilution values were within 6.5 % and 7.7 %. Subsequently, the developed and validated HPLC-ICP/MS method was also successfully applied to determine the arsenic speciation in dog plasma samples, and the recoveries for the spiked samples were in the range of 91.5–102.2 %. Therefore, this method could be applied to the evaluation of arsenic exposure, health effect assessment and other bio-monitoring studies in biological samples.

Aromatic hydrocarbons are toxic environmental pollutants that are detrimental to the ecosystem and human health. Among them, chlorotoluene and nitrotoluene are toxic to hydrobios and irritate the skin, eyes, and respiratory organs of humans. We herein report the development of recombinant microbial biosensors for cheap and rapid monitoring of chlorotoluene and nitrotoluene compounds. Plasmids were constructed by inserting the xylR regulatory gene for BTEX (benzene, toluene, ethylbenzene, and xylene) degradation into upstream of Po' (the DmpR activator promoter Po with the deletion of its own upstream activating sequences) or Pu (the cognate promoter of XylR)::lacZ (the ${\beta}$-galactosidase gene) and transformed into Escherichia coli $DH5{\alpha}$. In the presence of inducers, the biosensor cells immobilized in agarose developed a red color in 1-2 h due to the hydrolysis of chlorophenol red ${\beta}$-D-galactopyranoside (CPRG), a substrate of ${\beta}$-galactosidase that was expressed by the inducers. Among BTEX, high responses were specifically observed with o-, m-, p-chlorotoluene ($0.1{\mu}M-100 mM$) and o-, m-, p-nitrotoluene (0.1 mM-100 mM). Po' demonstrated higher responses than those with Pu. The biosensors immobilized in agarose showed good stability after 21 days' storage at $4^{\circ}C$, and responses in untreated wastewater spiked with chlorotoluene and nitrotoluene, suggesting they can be used to detect compounds in wastewater.
