New Isolation Technique and Culture System for Clinical Applications of Human Amniotic Epithelial Stem Cells (인간태반양막유래 상피줄기세포의 임상적용을 위한 새로운 세포분리 및 배양 기술)

  • Human placenta is abundant source of adult stem cells. Especially, amniotic epithelial cells have stem cell characteristics, expressing surface markers normally present on embryonic stem cells and germ cells. However, culturing and expanding amniotic epithelial cells in vitro without feeder cells are difficult due to endogenous characteristics of epithelial cells. In the present study, amniotic epithelial cells are isolated and proliferated in several passages by applying dithiothreitol and a Rho-associated kinase inhibitor in culture media. The cultured amniotic epithelial cells showed the epithelial and stem cell characteristics. In conclusion, human placenta-derived amniotic epithelial stem cells can be a major source of stem cells for medical treatment of various diseases without any controversial issues.

  • 자갈축열 태양열온실은 주간에 일사로 데워진 온실 내부공기를 온실하부에 설치된 자갈축열층사이로 강제순환시켜 자갈에 에너지를 축열한다. 이러한 축열과 방열과정을 통하여 겨울철 야간에 난방시스템으로 이용하고, 여름철에는 냉방효과를 꾀하게 된다. 온실내 공기가 축열층을 통과하는 동안의 열전달은 강제대류열전달이며, 이 경우 축열층내의 열이동은 축열층내 공기와 자갈표면온도가 초기에는 열적으로 평형상태로 존재하다가 순환공기의 온도상승에 따라 열전달이 일어나게 된다. (중략)

Variations of Size and Density of Sea Cucumber (Stichopus japonicus) Released to the Habitat Conditions (서식 환경에 따른 방류 돌기해삼(Stichopus japonicus)의 크기 및 서식밀도 변화)

  • We investigated the effects of environmental variations on the growth and survival rate of Stichopus japonicus to determine the optimum environmental conditions for its growth. Literature studies and a 12 month-long diver survey were carried out to understand the habitat, ecology and size of the surveyed area. Based on the collected data, we suggested optimum habitat conditions for releasing S. japonicus. Experiments on releasing S. japonicus were conducted in the breakwater of the Hwagye fishing cooperative in Hwagey-ri, Namhae-un, Gyeongsangnam-do, Korea. To implement the experiments, we divided the surveyed area into 4 sub-areas with different characteristics: (1) sand and silt zone; (2) artificial sea cucumber bank zone; (3) artificial rock bank zone; and (4) marine algae zone. The experiment lasted for 12 months. We released 32,000 sea cucumbers over $120m^2$ of each of the sand and silt zone, artificial rock bank zone and marine algae zone and released 6,000 sea cucumbers over $120m^2$ of the artificial sea cucumber bank zone. The average density of the released sea cucumbers from day 30 to day 360 after the releasing was conducted was the highest in the artificial sea cucumber bank zone ($23.7animal/m^2$), which was followed by artificial rock bank zone ($2.0animal/m^2$), marine algae zone ($1.9animal/m^2$) and sand and silt zone ($0.8animal/m^2$). The analysis on growth showed that the initial average weight of 2.3 g increased on day 360 after the releasing to 12.5 g in the artificial sea cucumber rank zone, 20.2 g in the sand and silt zone, 23.3 g in the artificial rock bank zone and 22.9 g in the marine algae zone. Results from the experiment along with the literature analysis suggest the following optimum habitat conditions: $10-15^{\circ}C$ water temperature; 28-34 psu salinity; 5-10 m water depth; 0.2-0.5 m/s velocity; rock, stone and muddy sand as substrate; and less than 20% mud in the substrate.

  • We constructed a rock bream (Oplegnathus fasciatus) gill cDNA library and a total of 1450 expressed sequence tag (EST) clones were generated. Gene annotation procedures and homology searches of the sequenced ESTs were locally done by BLASTX for amino acid similarity comparisons. Of the 1450 EST clones, 1022 EST clones showed significant homology to previously described genes while 428 ESTs were unidentified, and 259 clones were hypothetical, or unnamed proteins. Encoding 313 different sequences were identified as putative bio-defense genes or genes associated with immune response.

Recurrent odontogenic fibroma in a Formosan rock macaque (Macaca cyclopis)

  • The animal in this case report was a 10-year-old male Taiwanese monkey (Macaca cyclopis) kept at a zoo of South Korea. Over the last three years, a cauliflower-shaped masses have been noted on the gingiva near the incisor and molar teeth on right maxilla. Consequently, this monkey have undergone surgical removal of the mass annually. Grossly masses showed pinkish color. Histopathological findings, typical spindle cell tumor composed of collagen fibers. Infiltration by plasma cells and lymphocytes is found unrelated to ulceration of the surface epithelium. This is the first report of peripheral odontogenic fibroma in a Formosan rock macaque.

Optimal culture conditions for mass production of rock polypody (Polypodium vulgare L.)

  • This study aimed to develop a suitable method for inducing the proliferation of prothallus and producing sporophytes of rock polypody (Polypodium vulgare L.). The prothalli used in all experiments were obtained from spore germination and sub-cultured for 8-week intervals. The most appropriate media for prothallus propagation were investigated by culturing 300 mg of prothallus in MS ($1/4{\times}$, $1/2{\times}$, $1{\times}$, and $2{\times}$ strength) medium and in Knop medium for 8 weeks. Cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $30{\pm}1.0{\mu}mol-m-2{\cdot}s-1$, and a photoperiod of 16/8 h (light/dark). Fresh weight of prothalli was 4.8 g on $1{\times}$ MS, 4.5 g on $1/2{\times}$ MS and 4.3 g on 1/4 MS medium. To select a suitable soil combination for sporophyte formation, 1.0 g of prothallus was ground with distilled water, spread in five combinations onto different soil substrates (decomposed granite, horticultural substrates, peat moss, and perlite), and then cultivated for 13 weeks. The sporophyte cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $43{\pm}2.0{\mu}mol-m-2{\cdot}s-1$, humidity of $84{\pm}1.4%$, and a photoperiod of 16/8 h (light/dark). The results showed that a mixture containing a 2:1 (v:v) ratio of horticultural substrate and perlite, increased sporophyte formation to 462.5 sporophytes per pot (7.5 cm2). The other soil substrates produced from 314.5 to 405.3 sporophytes per pot. Therefore, our results will provide conditions suitable for mass production of Polypodium vulgare L.
