Ichthyotoxic Cochlodinium polykrikoides red tides offshore in the South Sea, Korea in 2014: III. Metazooplankton and their grazing impacts on red-tide organisms and heterotrophic protists

  • Cochlodinium polykrikoides red tides have caused great economic losses in the aquaculture industry in many countries. To investigate the roles of metazooplankton in red tide dynamics of C. polykrikoides in the South Sea of Korea, the abundance of metazooplankton was measured at 60 stations over 1- or 2-week intervals from May to November 2014. In addition, the grazing impacts of dominant metazooplankton on red tide species and their potential heterotrophic protistan grazers were estimated by combining field data on the abundance of red tide species, heterotrophic protist grazers, and dominant metazooplankton with data obtained from the literature concerning ingestion rates of the grazers on red tide species and heterotrophic protists. The mean abundance of total metazooplankton at each sampling time during the study was 297-1,119 individuals $m^{-3}$. The abundance of total metazooplankton was significantly positively correlated with that of phototrophic dinoflagellates (p < 0.01), but it was not significantly correlated with water temperature, salinity, and the abundance of diatoms, euglenophytes, cryptophytes, heterotrophic dinoflagellates, tintinnid ciliates, and naked ciliates (p > 0.1). Thus, dinoflagellate red tides may support high abundance of total metazooplankton. Copepods dominated metazooplankton assemblages at all sampling times except from Jul 11 to Aug 6 when cladocerans and hydrozoans dominated. The calculated maximum grazing coefficients attributable to calanoid copepods on C. polykrikoides and Prorocentrum spp. were 0.018 and $0.029d^{-1}$, respectively. Therefore, calanoid copepods may not control populations of C. polykrikoides or Prorocentrum spp. Furthermore, the maximum grazing coefficients attributable to calanoid copepods on the heterotrophic dinoflagellates Polykrikos spp. and Gyrodinium spp., which were grazers on C. polykrikoides and Prorocentrum spp., respectively, were 0.008 and $0.047d^{-1}$, respectively. Therefore, calanoid copepods may not reduce grazing impact by these heterotrophic dinoflagellate grazers on populations of the red tide dinoflagellates.

Structual Observation of Fruit Skin and Influence of Rainfall Inducing Fruit Cracking in 'Sato Nishiki' Sweet Cherry (체리 '좌등금'의 과실표면 구조 관찰과 강우가 열과발생에 미치는 영향)

  • Fruit cracking of sweet cherry fruit due to rain near harvest is a major source of crop loss in the cherry industry. The objection of this study was to understand the sweet cherry fruit cracking. Depending on the year, the disorder is characterized by a cracking of the outside layer of the cherry skin, as called cuticle. The crackings were appeared around the stem end, where water could accumulate, but was also seen fruit side or apical end. The cracking was observed mostly in the fruit apical end in 2004 and around the stem end in the other years. It had more rainfall at early season of fruit growth, early May, in 2004 and 2006 compared to other years. Those years showed higher rates of the fruit cracking occurrence. The hypodermal layer was examined on cracking susceptible cultivar, 'Sato Nishiki' from young to mature fruit cuticle. The hypodermal layers of the stem end area were decreased by one to two layers and fruit apical end area was cracked as fruit development. When fruits were immersed in distilled water, the larger fruit (> 6 g) had more cracking ratio than smaller fruits (< 4.5 g).

  • In days to calyx-flex stage test, flower development affected by light intensity, light duration and the prevailing temperature. As, 'Jacaranda' variety shorter in under 46 days of flower bud formation and 13 days of calyx-flex stage than the other variety with 'Queen Elizabeth' control variety. About cut flower only 'Grand Vicki Brown' (as within 46 days of flower formation, 13 days of calyx stage and 'Jacaranda', both with shorter plants than the other varieties produced cut flower classified as short and medium-stemmed cut flowers. 'Jacaranda', 'Vicki Brown', 'Golden Times', 'Champagne', 'Osiana', and 'Queen Elizabeth (control variety)' flowered earlier from 46.7 to 50.8 days from pruning. Flower development was likewise faster in all the above mentioned cultivars; while 'Tineke' had significantly delayed flower development reaching calyx-flex stage from 0.5 cm bud size after 51.0 days. The top ten high yielding varieties were 'American Orange', with a net income of P3,081,676.00; 'Yellow Island' with P1,572,874.00; 'Frosty Pink' with P1,085,183.00; 'Champagne' with P980,620.00; 'Coral Pink' with P959,669.00; 'Lara Pink' with P918,444.00; 'Lara Orange' with P714,252.00; 'Grand Gala' with P705,787.00; 'Golden Times' with P806,489.00; and 'Lady X' with P778,060.00 net returns.

Effect of the Extract of Hydrangea Dulcis Folium on Alcohol-induced Psychiatric Deficits (수국 추출물이 알코올로 유도한 기억 장애 및 long-term potentiation 억제에 미치는 영향)

  • Consumption of high doses of ethanol can lead to amnesia, which often manifests as a blackout. This incoordination of blackout may be a major cause in various social problems in alcohol consumption. However, there is still no treatment for preventing these alcohol-induced problems. Hydrangeae dulcis folium is a drug or a tea which is made from the fermented and dried leaves of Hydrangea serrata Seringe. The present study, we tested the ethanol extract of the Hydrangeae dulcis folium (EHDF) on ethanol-induced psychological deficits. To test behavioral deficits, an object recognition test was conducted using a mouse model. To evaluate synaptic deficits, N-methyl-D-aspartate (NMDA) receptor-mediated excitatory postsynaptic potential EPSP and long-term potentiation (LTP) in the mouse hippocampal slices were tested, as they are known to be vulnerable to ethanol and are associated with ethanol-induced amnesia. In the tests, ethanol (1 g/kg, i.p.) impaired object recognition memory, but EHDF (10 or 30 mg/kg) prevented this impairment in object recognition test. Interestingly, EHDF ($30{\mu}g/ml$) significantly ameliorated ethanol-induced LTP and NMDA receptor-mediated synaptic transmission in the hippocampal slices. EHDF prevented ethanol-induced object recognition memory deficits induced by ethanol. Interestingly, EHDF significantly ameliorated ethanol-induced LTP and NMDA receptor- mediated synaptic transmission in the hippocampal slices.

Biological Activities of Ethanol Extracts from Monascus-Fermented Chinese Yam (발효 홍국마 에탄올 추출물의 생리활성 효과)

  • This study was conducted to investigate antioxidative and physiological activities of ethanol extracts concentration from Monascus-Fermented Chinese Yam (MFCY). The ethanol extracts from MFCY were measured to examine pigment, monacolin K content, total polyphenol content, DPPH radical scavenging activity and angiotensin converting enzyme inhibitory effects. In this study, the results show that Monascus sp. MK2, with Dioscorea batatas Dence. as the substrate, can produce pigments (red, orange, and yellow), monacolin K content, total polyphenol content and DPPH radical scavenging activity at 13.48 (red), 11.66 (orange), 12.77 (yellow), 462.78 (mg/kg), 658.8 (mg/kg) and 92.8% in EtOH extract, respectively. In addition, ACE inhibitory activity was shown to be 74.55% in EtOH extract. Therefore, it can be concluded that Dioscorea batatas Dence is the best fermentative substrate for Monascus species to produce secondary metabolites as biomedicinal substances.

Wound Healing Effects of Lespedeza cuneata Extract (야관문 추출물의 창상치유 효과)

  • In this study, the anti-inflammatory effects of Lespedeza cuneata extract on macrophages and wound-healing in wound-induced animal experiments were investigated. In an anti-inflammatory test, 0.1 mg/mL of Lespedeza cuneata extract did not affect growth of RAW 264.7 cells, and Lespedeza cuneata extract suppressed nitric oxide (NO) generation from inflammation-induced macrophages in a concentration-dependent manner. Wounds on the skin of rats were treated with vehicle containing Lespedeza cuneata extract (SSP), vehicle (SCO), and commercial ointment (CCO). The wound and scar sizes in the SSP group were significantly reduced in comparison to the SCO and CCO groups (P<0.05). The epidermis and dermis of the SSP group also recovered faster than the SCO group based on Masson's trichrome staining. The gene expression levels of vascular endothelial growth factor (VEGF) decreased and transforming growth factor-beta 1 (TGF-${\beta}1$) increased in wound tissue from the SSP group compared to that from the SCO group. These results show that Lespedeza cuneata extract accelerates wound-healing through anti-inflammatory activity and induction of collagen regeneration as well as reduces the scar area surrounding wounds. Accordingly, Lespedeza cuneata extract could be useful as a cosmeceutical in the cosmetic industry.

Biological Activities of Fermented Dioscorea batatas Dence by two Stage Fermentation (2단 발효에 의한 발효마의 생리활성)

  • This study was conducted to investigate antioxidative and physiological activities of two stage fermented Chinese Yam (TSFCY) by Monascus sp. MK2 and Lactobacillus brevis HLJ59. The extracts from TSFCY were measured to examine pigments, monacolin K contents, total polyphenol and flavonoid contents, DPPH radical scavenging activity, reducing power, ACE inhibitory activity and GABA contents. In this study, the results show that Monascus sp. MK2 and Lactobacillus brevis HLJ59, with Dioscorea batatas Dence. As the substrate can produce pigments (yellow, orange and red), monacolin K contents, total polyphenol and flavonoid contents, DPPH radical scavenging activity, reducing power, ACE inhibitory activity, and GABA contents at 14.03 (yellow), 13.88 (orange), 15.71 (red), 487.9 (MK, mg/kg), 723.8 (TP, mg/kg), 326.4 (TF, mg/kg), 81.7 (DPPH, %), and 1.5 (RP, O.D), respectively. In addition, the showed ACE inhibitory activity and GABA contents was 86.9% and 977.4 mg/kg in EtOH extract, respectively.

Phenolic Contents and Antioxidant Activities of Six Edible Seaweeds (식용해조류 6종의 페놀성화합물 함량 및 항산화 활성)

  • Phenolic contents and antioxidant activities were determined in the water and ethanol extracts from six species of edible seaweeds, Phaeophyceae (Laminaria japonica, Hizikia fusiformis and Undaria pinnatifida), Rhodophyceae (Porphyra tenera and Gracilaria verrucosa) and Chlorophyceae (Ulva lactuca). The highest extraction yield was observed in water extract (pH 8.0) of G. verrucosa (44.23 %) and phenolic content was the highest in ethanolic (75 %) extract of H. fusiformis (52.82 ${\mu}g/mg$). 25 % ethanolic extract from U. lactuca was found to have the highest DPPH radicals scavenging activity(19.29 %) and superoxide anion scavenging activitiy was higher in water extract (pH 8.0) from H. fusiformis (81.20 %) than that other seaweeds. Phenolic contents were strongly correlated with antioxidant activity in the six edible seaweeds extracts ($R^2$=0.852). Taken together, these results indicate that H fusiformis may be a excellent source for development of natural antioxidants.

Enhancement effects of sea mustard and starfish mixtures on skin activity (미역 및 불가사리 추출 혼합물의 피부활성 증진 효능)

  • The enhancement effects of sea mustard extract (SME), starfish collagen peptide (SCP) and a mixture of the two (MIX) on skin activity were evaluated to investigate the possibility of using marine-derived extracts as cosmetic additives. The anti-inflammatory activity, whitening activity and skin elasticity activity of the extracts were analyzed to evaluate their skin-activating effects. Inhibiting the generation of nitric oxide (NO) and the tyrosinase and elastase inhibitory activities were assessed as the bio-markers for evaluating skin activity. SME, SCP and MIX did not show cytotoxicity within the concentration range of 1.0-50 ㎍/mL. In addition, SME, SCP and MIX all increased NO production and the tyrosinase and elastase inhibitory activities in a concentration-dependent manner. The activity of MIX was significantly increased compared to that with using SME or SCP alone. Taken together, when natural extracts are applied as cosmetic additives, the results demonstrate that using a mixture of SME and SCP may have a greater synergistic effect than that when using only a single extract. Therefore, this study contributes to the knowledge about the kinds and composition of several natural extracts when they are used as cosmetic additives.

Sleep-Induction Effects of GABA Coated Rice from Fermentation of Mono Sodium Glutamate (Mono Sodium Glutamate 발효로 얻은 GABA 코팅 쌀에 의한 수면유도 효과)

  • Gamma-aminobutyric acid (GABA) is a non-protein amino acid widely present in organisms, which has shown several important physiological functions such as neurotransmission, hypotension induction, as well as diuretic and tranquilizer effects. They have also been extensively used in food industry. GABA contents in the grain and brown rice are about 1~4 mg/100 g and 4~8 mg/100 g, respectively but it is difficult to expect physiological activity from such low amounts of natural food intake. We investigate the effects of GABA-coated rice on the secretion of melatonin and serotonin which both have been used as sleep inductive compounds. As a result, the secretion of melatonin and serotonin from mice were found to be $3.578{\pm}0.158pg/mL$, $5.918{\pm}0.169ng/mL$ respectively. The melatonin and serotonin in mice increased significantly up to the 8.7 and 22.8 times respectively, when compared to that of the rice, but there was no cumulative effects. Due to the continuous intake of GABA-coated rice, which was developed as a functional food nutrient, inductive effects of melatonin and serotonin from general rice could be achieved and also the similar effect as taking up directly 25 mg/mL of GABA could be obtained.
