Growth, Flowering, and Nutrient Composition of Salvia Grown in Peat moss Media Containing Pellets Processed with Poultry Feather Fibers at Different Mixing Ratios

  • The objective of this study was to determine the effect of replacing perlite (PL) with pellets processed with poultry feather fiber as an inert material to prepare growing medium. The growth and flowering of Salvia splendens 'Vista Red' grown in individual growing medium $Biosangto^{(R)}$, peat moss (PM), PL, and two pellets (P45-1 and P45-2) were evaluated. Peat moss was mixed with PL, P45-1, or P45-2 at various ratios (1:0 to 1:3 or 3:1 by volume) to investigate the feasibility of replacing PL with pellets. Nutrient composition of the growing medium and leaf tissues was analyzed. The number of florets, inflorescence length, plant height, and fresh weight of plants grown in media containing P45-1 or P45-2 were reduced compared to those grown in individual growing medium PM or PL. As the mixing ratio of P45-1 or P45-2 to PM was higher, the growth of salvia, such as inflorescence length, plant height, number of leaves, and fresh weight was inhibited. Our results indicate that mixing three parts PM with one part of P45-1 (PM/P45-1/3:1) or P45-2 (PM/P45-2/3:1) accelerated flowering and increased the number of florets and leaves compared to other mixing ratios of PM and pellets media. The concentrations of phosphorus (P), calcium (Ca), boron (B), iron (Fe), and copper (Cu) in individual growing medium PL, P45-1, and P45-2 were significantly lower than those in PM. The concentration of N was the highest in leaves of plants grown in P45-1 or P45-2 amended media, and the concentrations of P, Ca, and zinc (Zn) in leaves were lower in individual growing medium P45-1 or P45-2 than in PM and PL. The pH of PM/P45-1/3:1 or PM/P45-2/3:1 media was maintained at optimal level (5.8-5.9) and the concentrations of macro- and micro-elements in the media and leaves were considered to be optimal levels. Therefore, mixing three parts PM with one part P45-1(PM:P45-1/3:1) or P45-2 (PM:P45-2/3:1) is recommended for improved growth and flowering in salvia. This suggests that P45-1 or P45-2 can replace PL as an inert material to prepare growing medium.

  • This study was carried out to investigate the antioxidative and antimicrobial activities of Celastrus orbiculatus (CO). CO was extracted with methanol and then further fractionated with n-hexane (COMH), butanol (COMB), methanol (COMM) or aqueous (COMA) to get active fractions. The antioxidative activity of fractions from CO was investigated by measuring the scavenging activities of CO against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen speicies (ROS). Among the various solvent fractions, the COMB showed a marked scavenging effect against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen species(ROS). The antimicrobial activity was increased in proportion to its concentration by the paper disk method. Among the various solvent fractions, the COMM and COMB fractions of CO showed strong antimicrobial activities. The results suggest that The CO may be suitable for development as a food preservative and alternative antioxidant.

  • Two-dimensional gel electrophoresis (2-DE) is an essential tool of proteomics to analyse the entire set of proteins of an organism and its variation between organisms. Helicobacter pylori was tried to identify differences between strains. As the first step, whole H. pylori was lysed using high concentration urea contained lysis buffer [9.5 M Urea, 4% CHAPS, 35 mM Tris, 65 mM DTT, 0.01% SDS and 0.5% Ampholite (Bio-Rad, pH 3-10)]. The extract ($10\;{\mu}g$) was rehydrated to commercially available immobilised pH gradient (IPG) strips, then the proteins were separated according to their charges as the first dimensional separation. The IPG strips were placed on Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis (SDS-PAGE) to separate according to molecular mass of the proteins as the second dimension. The separated protein spots were visualised by silver staining in order to compare different expression of proteins between strains. Approximately 120 spots were identified in each mini-protein electrophoresised gel, furthermore about 65 to 75 spots were regarded as identical proteins in terms of pI value and molecular weight between strains used. In addition, distinct differences were found between strains, such as 219-1, Y7 and Y14, CH150. Two representative strains were examined using strips which had pH range from 4 to 7. This strips showed a number of isoforms which were considered large spots on pH range 3-10. Furthermore, the rest of spots on pH 4-7 IPG strips appeared very distinctive compared to broad range IPG strips. 2-DE seems to be an excellent tool for analysing and identifying variations between H. pylori strains.

Effects of Persimmon Leaf Tea Extract, Green Tea Extract and Oolong Tea Extract on the Frequencies of Mutagen-Induced Sister Chromatid Exchange in Chinese Hamster Ovary Cells (감잎차, 녹차, 우롱차 추출물이 돌연변이 물질로 유발된 Sister Chromatid Exchanges 빈도에 미치는 영향)

  • The suppressing effects of crude extracts of three Korean teas, persimmon leaf tea extract (PLTE), green tea extract (GTE) and oolong tea extract (OTE), were studied on the induction of sister chromatid exchange (SCE) in cultured Chinese hamster ovary cells. When cells were treated with tea extract after mitomycin C (MMC) treatment, the frequency of MMC-induced SCEs were decreased at the high concentration $(1000\;{\mu}g/mL)$ of PLTE in the presence of S9 mix and at low concentrations $(20{\sim}80\;{\mu}g/mL)$ of PLTE in the absence of S9 mix, Whereas GTE and OTE showed suppressing effects on the MMC-induced SCEs at low concentrations $(10{\sim}20\;{\mu}g/mL)$ for OTE and $160\;{\mu}g/mL$ for GTE only in the presence of S9 mix. MMC-induced SCEs were decreased by post-treatment with each tea extracts with S9 mix in the G1 phase of the cell cycle. These results suggest that PLTE, GTE and OTE could have bio antimutagenic activities, and also suggest that PLTE might have unknown antimutagenic components which would be responsible for the inhibitory effect against direct acting mutagenicity.

Estimation of Future Design Flood Under Non-Stationarity for Wonpyeongcheon Watershed (비정상성을 고려한 원평천 유역의 미래 설계홍수량 산정)

  • Along with climate change, it is reported that the scale and frequency of extreme climate events show unstable tendency of increase. Thus, to comprehend the change characteristics of precipitation data, it is needed to consider non-stationary. The main objectives of this study were to estimate future design floods for Wonpyeongcheon watershed based on RCP (Representative Concentration Pathways) scenario. Wonpyeongcheon located in the Keum River watershed was selected as the study area. Historical precipitation data of the past 35 years (1976~2010) were collected from the Jeonju meteorological station. Future precipitation data based on RCP4.5 were also obtained for the period of 2011~2100. Systematic bias between observed and simulated data were corrected using the quantile mapping (QM) method. The parameters for the bias-correction were estimated by non-parametric method. A non-stationary frequency analysis was conducted with moving average method which derives change characteristics of generalized extreme value (GEV) distribution parameters. Design floods for different durations and frequencies were estimated using rational formula. As the result, the GEV parameters (location and scale) showed an upward tendency indicating the increase of quantity and fluctuation of an extreme precipitation in the future. The probable rainfall and design flood based on non-stationarity showed higher values than those of stationarity assumption by 1.2%~54.9% and 3.6%~54.9%, respectively, thus empathizing the necessity of non-stationary frequency analysis. The study findings are expected to be used as a basis to analyze the impacts of climate change and to reconsider the future design criteria of Wonpyeongcheon watershed.

Herbicidal Characteristics of Soil Bacteria Actinomycetes G-0299 to Southern Crabgrass (토양 방선균 Actinomycetes G-0299의 바랭이에 대한 선택적 살초특성)

  • An actinomycetes isolate G-0299 obtained from a forest soil showed strong phytotoxic activity to Digitaria ciliaris. For the foliar application study, the culture filtrate of the isolate G-0299 showed strong herbicidal activity only to D. ciliaris among the 12 monocot and 5 dicot weed species. And herbicidal activity at a concentration of 500, 250, 125 and $62.5{\mu}gmL^{-1}$ of culture filtrate was 100%, 98%, 70% and 40%, respectively. Phytotoxic symptoms of the culture filtrate by foliar application were desiccation and burn-down or bleaching of leaves and finally plant death. And then the herbicidal activity was exhibited only under the light condition. Also, chlorophyll loss of D. ciliaris leaf tissues in the light condition was much higher than in the dark condition and then chlorophyll content decreased 82%, 5%, respectively. In conclusion, our results suggest that soil bacteria, isolate G-0299 could be a good candidate for new bio-herbicide and provide a new lead molecule for a more unique herbicide.

Effects of Dietary Bio Ethanol By-product and Complex Enzyme on Meat Quality of Pork Loin (바이오 에탄올 부산물 DDGS와 복합 효소제 첨가급여가 돈육의 품질 특성에 미치는 영향)

  • This study was conducted to evaluate the effects of dietary complex enzyme (${\beta}$-mannanase 800 IU/kg and xylanase 700 IU/kg) in a diet containing corn distiller's dried grain with soluble (DDGS) on meat quality and pork fatty acid composition. Ninety-six pigs ([(Landrace${\times}$Yorkshire)${\times}$Duroc], with an average body weight of 68.77 kg were used in the 8 wk growth assay. Dietary treatments included 1) corn-soybean meal diet, 2) corn-soybean meal diet + 0.05% enzyme complex, 3) cornsoybean meal diet with DDGS and 4) corn-soybean meal diet with DDGS + 0.05% enzyme complex. The pigs were allotted randomly into pens (n=4 per pen) with six replicate pens per treatment by a completely randomized design. Pigs were slaughtered at the end of the experiment and the loin muscle was obtained for meat quality. Meat pH (p<0.01), firmness (p<0.01) and redness (p<0.05) were higher in DDGS-supplemented diet than in the corn-soy bean meal diet. However, color, marbling, lightness, yellowness, thiobarbituric acid reactive substances, water holing capacity, driploss, cooking loss and loin muscle area were not significantly different among the diets. The pigs fed the DGGS-supplemented diet had higher total unsaturated fatty acids (UFA) and total UFA/saturated fatty acid (SFA) ratio of loin and backfat. The results indicate that a diet containing DDGS can influence pH, firmness, redness and total UFA concentration and total UFA/SFA ratio of meat and backfat, but that enzyme addition has no affect on meat quality.

Analysis of Air Circulation in Oyster Mushroom Farm

  • Oyster mushroom farm which could not meet optimum temperature range yields non-uniform sized, low quality products. Thus, this study, utilizing STAR CCM+, one of the computational fluid dynamics (CFD) programs, analyzed the impact of air circulation and temperature distribution. Methods: After we visited numerous mushroom farms, we measured the temperature at the discharge ports of heaters, fan capacity, and the locations of the air circulators in the farms. According to the data, most mushroom growers installed the heaters near the entrance and discharge ports of the heaters at the third growing bed on the same height as the heaters in the entrance. The temperature at the discharge port of heater was $1,26^{\circ}C$, and the fan capacity was 4,500 $m^3$/hr. The air circulator was placed in the center of the mushroom farm 50cm above the ground, and its capacity of inlet port was 1,100 $m^3$/hr and discharge port 1,600 $m^3$/hr. The mushroom farm was insulated. Results: According to the analysis of the temperature distribution in the vertical plane of the entrance side, no air circulation causes the high temperature zone of 296~299K at the discharge port of the heater to take up 34% of area while the operation of air circulators causes it to occupy only 9%. This means that not using air circulators leads to a concentration of high temperature at the discharge port near the entrance. In addition, with the results of the analysis of the temperature distribution in the vertical planes of the center, no air circulation causes the temperature zone of 295~298K at the discharge port of the heater to take up 48% of area while the operation of air circulators causes it to occupy 80%. This shows that the high outlet port temperature disseminated to the center. Conclusions: After ninety minute operation of both heater and air circulator, the interior temperature became stabilized in the mushroom farm. Air circulation made the high temperature at the discharge port disseminate to the center and exit in the farm and equalize the temperature distribution.

Effects of Medium Components and Composition on Mass Propagation of Arachniodes aristata (G. Forst.) Tindale (가는쇠고사리의 대량번식에 미치는 배지구성물질과 배양토의 영향)

  • This study was conducted to investigate in vitro mass propagation methods suitable for each growth stage of A. aristata (G. Forst.) Tindale, from spore germination to sporophyte formation. Among spores germinated in $1/8-1{\times}MS$ medium and Knop medium, Knop medium yielded the highest germination percentage (87.1%). We cultured prothalli obtained from germinating spores for 8 weeks on media with different concentrations of sucrose and active carbon, as well as different concentrations and ratios of nitrogen, to select a suitable growth medium. A. aristata (G. Forst.) Tindale prothalli grew most actively in MS medium with 3% sucrose and 20 : 40 mM of $NH_4Cl$ and $KNO_3$ (total concentration of 60 mM). We investigated sporophyte formation according to soil type, finding that bedding soil mixed with perlite at a 2 : 1(v / v) ratio yielded the highest number of sporophytes per pot ($73.8/7.5{\times}7.5cm\;pot$). By contrast, when peat moss was used alone or mixed with other substrates, prothallus development and sporophyte formation were suppressed. Therefore, the most effective propagation method for A. aristata (G. Forst.) Tindale is to grow prothalli in MS medium and to induce sporophyte formation in a mixture of bedding soil and perlite (v / v = 2 : 1).

Characterization of a Fibrinolytic Enzyme from Pickled Anchovy (멸치젓갈유래의 혈전용해호소에 대한 특성)

  • In the previous study, we isolated a myulchikinase (MK), which has fibrinolytic activity and cytotoxicity to the tumor cell line, from myl- chi-jeot-gal. In this study, the effect of NaCl concentration, metallic ions, pH, temperature, and plasminogen on the activity of MK was analysed. The MK activity was maintained at least $80\%$ activity up to $30\%$ NaCl, which indicates that the enzyme may be halotolerant. The optimal pH and temperature were 8 and $40^{\circ}C$, respectively. The fibrinolytic activity of MK was completely inhibited with 0.5 mM $Hg^{2+}$ and inhibited to $50^{\circ}C$ with 1 mM $Cu^{2+}\;and\;Zn^{2+}$. The MK showed strong activity in plasminogen- rich fibrin plate but not in plasminogen-free fibrin plate. The result indicates that the MK may be a plasminogen activator type fibrinolytic enzyme.
