Monitoring on Alcohol Fermentations Properties for Aronia Juice for Aronia(Aronia melanocarpa) Vinegar

  • This study is to establish optimizing condition of alcohol fermentation in vinegar production with aronia, to confirm whether this can be industrially used, and to compare and analyze a change in anti-oxidative activity and quality characteristic according to alcohol fermentation of aronia. The optimized conditions for alcohol fermentation were as follows: Saccharomyces cerevisiae 5645 of yeast strain, a 5% inoculum size, aronia juice with a brix value of 14, and a glucose content of 7%. As a result to conduct scale up with optimizing conditions of alcohol fermentation of aronia, 8 days (192 hrs) of total alcohol fermentation time and 7.4% of the final alcohol content. The harvest volume accounted for approximately 90.2% with a loss of about 2.8%. As a result of antioxidant test, anti-oxidative activity of alcohol fermented liquor is lower than anti-oxidative activity of aronia extract, because of the decrease of antioxidant by oxidation of the fermentation process. However, the decrease of tannin by the fermentation process reduces acerbity of aronia, so increases overall preference

Involvement of Hepatic Innate Immunity in Alcoholic Liver Disease

  • Excessive alcohol consumption is one of the critical causative factors leading to alcoholic liver disease (ALD). ALD is characterized by a wide spectrum of liver damage, ranging from simple uncomplicated liver steatosis (fatty liver) to steatohepatitis and liver fibrosis/cirrhosis. It has been believed that the obvious underlying cause for ALD is due to hepatocyte death induced by alcohol itself. However, recent sparkling studies have shown that diverse immune responses contribute to ALD because liver is enriched with numerous immune cells. Especially, a line of evidence has suggested that innate immune cells such as Kupffer cells and natural killer (NK)/NKT cells are significantly involved in the pathogenesis of ALD via production of pro-inflammatory cytokines and other mediators. Indeed, more interestingly, hepatic stellate cells (HSCs), known as a major cell inducing liver steatosis and fibrosis, can be killed by liver NK cells, which could be suppressed by chronic alcohol consumption. In this review, with the view of liver as predominant innate immune organ, we describe the pathogenesis of ALD in which what roles of innate immune cells are and how they are interacting with HSCs.

Effects of Propylene Glycol on the Physical Properties of Poly(vinyl alcohol) Solutions and Films

  • To trace the plasticizing effects of propylene glycol (PG) on poly(vinyl alcohol) (PVA), the rheological properties of PVA solutions in dimethyl sulfoxide (DMSO) and the physical properties of PVA films were discussed in terms of PG content. Both properties were closely related to the hydrogen bond breaking effects of PG The 6 and 12 wt% PVA solutions containing PG exhibited Bingham behavior, which was more noticeable at lower plasticizer content and higher polymer concentration. The 6 wt% PVA solutions containing more than 30 wt% PG showed a sudden decrease of viscosity over the frequency range of 0.08 and 0.2 rad/s. However, the 12 wt% PYA solutions showed no viscosity reduction even at a PG content up to 40 wt%. The glass transition temperature of the PVA/PG films was almost linearly decreased with increasing PG content but an abrupt reduction was observed at a plasticizer content 30 wt%, suggesting that the hydrogen bond breaking effects of PG on PVA became dominant between 20 and 30 wt%. This effect was further supported by the similar tendency of the tensile properties.

Effect of probiotics Lactobacillus paracasei GKS6, L. plantarum GKM3, and L. rhamnosus GKLC1 on alleviating alcohol-induced alcoholic liver disease in a mouse model

  • BACKGROUND/OBJECTIVES: Heavy alcohol consumption causes the development of alcoholic liver disease (ALD), a neglected but important public health problem. Many studies have pointed out that probiotics could improve gut health, which is also considered to be a cause of ALD. Therefore, this study screened the probiotics, Lactobacillus casei GKC1 (GKC1), L. fermentum GKF3 (GKF3), Bifidobacterium lactis GKK2 (GKK2), L. rhamnosus GKLC1 (GKLC1), L. paracasei GKS6 (GKS6), and L. plantarum GKM3 (GKM3), for their potential benefits in alleviating ALD for applications to disease prevention. SUBJECTS/METHODS: C57BL/6N mice were divided into 8 groups (n = 6 in each): normal control, positive control (alcohol-diet fed), and treatments of feeding probiotics GKC1, GKF3, GKK2, GKLC1, GKS6, and GKM3 under an oral dose 0.82 g/kg B.W. per day by oral gavage. The experiment was conducted for 8 weeks, and the concentrations of alanine aminotransferase (ALT), aspartate aminotransferase, triglyceride (TG), and total cholesterol (TC) in mice were measured. The glutathione (GSH), catalase (CAT), and histology were analyzed after sacrifice. RESULTS: The results showed a decrease in the serum ALT, liver TG, and liver TC levels in the GKS6, GKM3, and GKLC1 groups compared to the positive control. In addition, the decreasing GSH and CAT levels were inhibited in the GKS6 and GKM3 groups. The histopathological results showed that all probiotics could reduce the accumulation of liver fat. Furthermore, there was a significant difference in GKLC1 with lower stomach damage compared to the alcohol-fed mice without any addition of probiotics. CONCLUSIONS: GKLC1, GKS6, and GKM3 can be used as supplements for alleviating the development of ALD.

Kiwi-persimmon wine produced using wild Saccharomyces cerevisiae strains with sugar, acid, and alcohol tolerance

  • 100 different yeast colonies were isolated from spontaneously fermented kiwis, persimmons, apples, pears, watermelons, grapes, grape fruits, peachs, and plums, and selected yeast strains were used to produce kiwi-persimmon mixed wine (KPMW). Among the isolates, five representative strains exhibited tolerance to sucrose, alcohol, pH, and potassium metabisulfite when compared with the control yeast strain (Saccharomyces cerevisiae KCCM 12615). All five yeast strains (Y4, Y10, Y28, Y78, and Y81) exhibited 99% 26S rDNA sequence similarity to S. cerevisiae. The pH, acidity, Brix, reducing sugar, alcohol, and organic acid contents were consistent in KPMW prepared from the S. cerevisiae KCCM 12615 and Y28 strains. KPMW made from the Y4, Y10, and Y28 strains exhibited lower quantities of free sugars than those of the KPMW made from the other yeast strains. The level of ethyl esters in KPMW prepared from the Y28 was higher than that in the other KPMWs. All strains, except for Y28, produced lower concentrations of sulfur and ketone compounds. Furthermore, the KPMW produced by the Y28 strains had total phenolic contents with 1.1 g/L, with DPPH and ABTS radical scavenging activities of 57.06% and 55.62%, respectively, and a FRAP assay value of 0.72. Our results suggest that Y28 is a promising yeast strain for producing high-quality wines.

Bulnesia sarmienti 를 함유한 숙취해소 음료가 알코올대사 및 항산화 효소활성에 미치는 영향 (Effect of Alcohol Detoxification Beverage that Contained Bulnesia sarmienti on Alcohol-metabolizing Enzymes and Antioxidant Enzyme Activities)

  • 허브의 일종인 B. sarmienti를 이용하여 숙취 해소 음료를 개발하고 그에 따른 알콜 대사 및 항산화 효소에 미치는 영향을 알아보고자 하였다. 알코올을 섭취한 쥐에 B. sarmienti 음료군(BSP)을 투여 한 경우 타사 제품군인 PC 그리고 B. sarmienti만을 제외한 음료군인 NBS보다 더 뛰어난 알코올 분해능 및 ADH, ALDH에서도 뛰어난 활성을 보였다. 또한 생화학적 검사를 통해 안전성을 확인하였으며, ALP, ALT 그리고 AST에서도 PC군에 비해 유의적인 감소 효과를 보였다. 항산화 활성 및 효소계에 있어서도 BSP군에서는 GSH에서는 CO군에 비해 효소의 함량은 높으나, 다른 군에 비해 유의적인 차이는 보이지 않았으며, TBARS에는 다른 군에 비해 함량이 감소됨을 알 수 있었으나 뚜렷한 차이는 보이지 않았다. 그러나 GST, CAT 그리고 SOD 효소에 있어서는 CO군에 비해 유의적인 효소 활성이 감소되었음을 알 수 있었다. 이상과 같은 결과를 토대로 B. sarmienti 추출물이 첨가된 음료는 뛰어난 숙취 해소 및 간세포의 손상을 보호하는 음료로서 이용 가능하리라 기대 된다.

Swelling Behavior and Drug Release of Poly(vinyl alcohol) Hydrogel Cross-Linked with Poly(acrylic acid)

  • Thermal cross-linking method of poly(vinyl alcohol) (PVA) using poly(acrylic acid) (PAA) was carried out on PVA/PAA hydrogels. The level of gelation was measured in the PVA/PAA hydrogels with various PAA contents. The swelling behavior at various pHs showed that the swelling kinetics and water contents of the PVA/PAA hydrogels reached equilibrium after 30 h, and the time to reach the equilibrium state decreased with increasing PAA content in the hydrogel. The water content increased with increasing pH of the buffer solution. The permeation and release of the drug were tested using indomethacin as a model drug. The permeated and released amounts of the drug increased with decreasing the PAA content because of the low free volume in the hydrogel due to the higher cross-linking density. The kinetic profile of drug release at various pHs showed that all samples reached the equilibrium state within the 5 h.

Pyrolysis of Lignin Obtained from Cinnamyl Alcohol Dehydrogenase (CAD) Downregulated Arabidopsis Thaliana

  • Despite its potential as a renewable source for fuels and chemicals, lignin valorization still faces technical challenges in many aspects. Overcoming such challenges associated with the chemical recalcitrance of lignin can provide many opportunities to innovate existing and emerging biorefineries. In this work, we leveraged a biomass genetic engineering technology to produce phenolic aldehyde-rich lignin structure via downregulation of cinnamyl alcohol dehydrogenase (CAD). The structurally altered lignin obtained from the Arabidopsis thaliana CAD mutant was pyrolyzed to understand the effect of structural alteration on thermal behavior of lignin. The pyrolysis was conducted at 400 and $500^{\circ}C$ using an analytical pyrolyzer connected with GC/MS and the products were systematically analyzed. The results indicate that aldehyde-rich lignin undergoes fragmentation reaction during pyrolysis forming a considerable amount of C6 units. Also, it was speculated that highly reactive phenolic aldehydes facilitate secondary repolymerization reaction as described by the lower yield of overall phenolic compounds compared to wild type (WT) lignin. Quantum mechanical calculation clearly shows the higher electrophilicity of transgenic lignin than that of WT, which could promote both fragmentation and recondensation reactions. This work provides mechanistic insights toward biomass genetic engineering and its application to the pyrolysis allowing to establish sustainable biorefinery in the future.

액상환원법과 알코올환원법으로 제조한 은나노입자특성 비교에 관한 연구 (Comparison Study of the Synthesized Silver Nano-particles using Liquid Phase Reduction Method and Alcohol Reduction Process)

  • Silver nano-particles have been synthesized by liquid phase reduction method and alcohol reduction process. Silver nano-particles of the size 30 ~ 40 nm were formed successfully by alcohol reduction process. The formation, structure, morphology and size of silver nano-particles have been studied using FE-SEM, TEM, XRD, UV-visible spectroscopy. In particular high dispersion stability of the synthesized silver nano-particles could be obtained by PVP binding. Antibacterial activity of Ag/PET master batch sample made from its nano-silver particles showed excellent antibacterial activity against S. aureus and E. coli.
