Optimized purification and characterization of expressed hMC4R-TM2

  • Human melanocortin-4 receptor (hMC4R) among MC-Rs, expressed in the brain, is in charge of the control on energy homeostasis and food intake. The structure and function of human MC4R have been studied to understand their essential function and roles. To investigate the structure and function, it is necessary to prepare sufficient amounts of proteins. However, their expression and purification is demanding and time-consuming due to their innate insoluble and toxic properties. The heterozygous mutations of hMC4R, exchange of Asp 90 to Asn located in second transmembrane, cause severe obesity in human. To obtain purified hMC4R wt-TM2 for structural studies, it was first over-expressed and purified by fast protein liquid chromatography (FPLC) and then solution NMR studies were performed to get high-resolution spectra. In here, we established optimized purification scheme to get more purified target peptide.

Rehabilitation of a crossed-occlusion patient using implant-assisted removable partial denture with Locator® attachment: A case report (엇갈린 교합 및 교합평면 부조화를 가진 부분무치악 환자에서 임플란트를 이용한 가철성 국소의치 증례)

  • In crossed occlusion, displacement of removable partial denture is likely to occur during function due to different characteristics of abutment and supporting tissue. It increases discomfort to the patient. In addition, adverse effect on the residual ridge and abutment can induce an unfavorable prognosis of the denture. In this case, a small number of implants can be placed in strategic locations. Attachment can be added for additional support and retention of removable partial denture assisted by implants. This article describes the rehabilitation of a crossed occlusion patient using implant-assisted removable partial denture with Locator$^{(R)}$ attachment. After 24 months, the patient was satisfied with the aesthetic and function of the prosthesis that is maintained stable.

  • The skin protects the body from excessive water loss and the invasion of harmful substances, such as chemicals and microbes. The stratum corneum, is recognized as a very important physical barrier. However, in recent years evidence emerged that tight junctions (TJ) might also play a crucial role in barrier function of the skin. In the present study, TJ proteins including transmembrane proteins and plaque proteins, skin permeability barrier function and skin diseases of TJ were reviewed.

  • This network system for home care realized communication by the bio-physical signal, to convey physical rhythm. Four function of displacement had point of a Vision, Somatosensory, Vestibular and CNS. Bio-physical signal was decided to design a maximum points and minimum points with 0.01unit in reference level. Bio-physical signal was checked to compound physical condition of body posture for sensory organ. There detected a measurement of Vision, Somatosensory, Vestibular, CNS and BMI. The service of network system of home can be used to support a health care system for health assistant in health care center. It will expect to manage a physical parameter for network communication.

In vitro Anti-inflammation Effect of Adventitious Shoots of Toona sinesis in Propionibacterium acnes-induced Skin Dermatitis

  • Toona sinensis (TS) leaf is known to antinociceptive, antioxidative stress and skin moisturizing effects. Acnes vulgaris is a chronic skin disease with various symptoms including itchiness, pain and interruption of normal skin function. Propionibacterium acnes (P. acnes) is a major factor in the occurrence of inflammatory acnes. This study evaluated the antioxidant and anti-inflammation effects by TS extract from adventitious shoots. TS extract showed anti-inflammatory activities by suppression of pro-inflammation mediators (iNOS and COX-2) in LPS-stimulated RAW264.7 cells. TS extract also has anti-inflammatory activities by inhibiting the secretion of pro-inflammatory cytokines on P. acnes-stimulated HaCaT cells. These effects were regulated by MAPK signaling pathway. Therefore, we suggest that TS extract from adventitious shoots might have applications as a medicine for treating P. acnes-induced skin diseases.

A Mutation of cdc-25.1 Causes Defects in Germ Cells But Not in Somatic Tissues in C. elegans

  • By screening C. elegans mutants for severe defects in germline proliferation, we isolated a new loss-of-function allele of cdc-25.1, bn115. bn115 and another previously identified loss-of-function allele nr2036 do not exhibit noticeable cell division defects in the somatic tissues but have reduced numbers of germ cells and are sterile, indicating that cdc-25.1 functions predominantly in the germ line during postembryonic development, and that cdc-25.1 activity is probably not required in somatic lineages during larval development. We analyzed cell division of germ cells and somatic tissues in bn115 homozygotes with germline-specific anti-PGL-1 immunofluorescence and GFP transgenes that express in intestinal cells, in distal tip cells, and in gonadal sheath cells, respectively. We also analyzed the expression pattern of cdc-25.1 with conventional and quantitative RT-PCR. In the presence of three other family members of cdc-25.1 in C. elegans, defects are observed only in the germ line but not in the somatic tissues in cdc-25.1 single mutants, and cdc-25.1 is expressed predominantly, if not exclusively, in the germ line during postembryonic stages. Our findings indicate that the function of cdc-25.1 is unique in the germ line but likely redundant with other members in the soma.

  • Self-authentication technology using electrocardiogram (ECG) signals is drawing attention as a self-authentication technology that can replace existing bio-authentication. A device that recognizes a digital electronic key can be mounted on a vehicle to wirelessly exchange data with a car, and a function that can lock or unlock a car door or start a car by using a smartphone can be controlled through a smartphone. However, smart keys are vulnerable to security, so smart keys applied with bio-authentication technology were studied to solve this problem and provide driver convenience. A personal authentication algorithm using electrocardiogram was mounted on a watch-type wearable device to authenticate bio, and when personal authentication was completed, it could function as a smart key of a car. The certification rate was 95 per cent achieved. Drivers do not need to have a smart key, and they propose a smart key as an alternative that can safely protect it from loss and hacking. Smart keys using personal authentication technology using electrocardiogram can be applied to various fields through personal authentication and will study methods that can be applied to identification devices using electrocardiogram in the future.

Double Processing Obturator for the Hemi-maxillectomy Treated Patient (Hemi-Maxillectomy 환자에서 이중온성법을 이용한 폐색장치의 제작)

  • In the case of congenital malformation, acquired trauma, or maxillectomy by neoplasm, the defect in maxilla is combined with nasal cavity, maxillary sinus and oral cavity. This results in abnormal function in pronunciation, mastication, and swallowing. In this situation, the purposes of prosthodontics are also applied. The purposes of obturator for acquired maxillary deficiency patients are restoring pronunciation, mastication, and swallowing, and make patients comfortable and esthetic satisfaction. Reducing weight of the obturator makes edentulous ridge heathy and functional. It also improves patients' comfort. Hollow obturator also reduces unnecessary stress because improves physiologic function, as it weights less than 6.55-33.06% compare to usual one. This case presents the double-processing method for maxillary obturator in the patient treated hemi-maxillectomy due to malignancy neoplasm. We report this case because the patient acquired functional and esthetic satisfaction from double-processing obturator.

The Change of Electroencephalogram According to Bio-Feedback Training in Dementia (치매노인들의 바이오피드백 훈련에 따른 뇌파 변화)

  • Purpose : This study was to evaluate the effects of cognitive rehabilitation training on the cognitive decline of dementia patients. Therefore the purpose of this study was to examine the influence of brain activation according to bio-feedback training in dementia. Methods : Ten dementia patients were recruited this study. Experiment was performed for 30min per session, five times a week through 4 week and two measurements before and after bio-feedback training. Brain activity was measured by Korea Electroencephalogram(EEG) system. Statistical analysis was used Wilcoxon signed rank test to know difference of EEG between pre and post-test in each group and Mann-Whitney U test was to know difference between experimental and control group. Results : Significant improvement of slow-alpha wave was observed following bio-feedback in experiment group. There was no significant change in experiment and control group. Conclusion : In this study, the bio-feedback training was effective in improving slow-alpha wave in dementia patients. It is suggested that bio-feedback training with dementia patients can be useful to ameliorate the cognitive decline. And it will be effective for prevention of cognitive function decline.

The Study to Diagnose the Road-Driver Compatibility I: Comparison of Methods for Bio-Signal Analysis (운전자 주행 적합성 진단을 위한 연구 I: 생체신호 분석방법 비교)

  • The aim of this study is to compare the methods in analyzing bio-signals representing measure driver's psychophysiological staus. This study has considered three approaches: first, the deterministic approach calculating the mean and standard deviation of bio-signal, second, probabilistic approach converting driver's bio-signal values to probability density function and identifying individual state relative to overall distribution, and third, diagnostic approach identifying the pattern change of signal over certain period of time. For evaluation of analysis methods, driver's bio-signal was collected under various road conditions, and three analysis approaches were applied respectively. In result, the deterministic approach was found to be simple to use, but generated a large variability of bio-signal. The probabilistic approach provide a relative status of individual driver among overall population, but too much affected by temporal variability of individual driver. The diagnostic approach seemed to reasonably find driver's psychophysiological change over certain period of time, but still needs to develop quantification method of the bio-signal.
