Pesticide and Heavy Metal Residue Monitoring in 13 Types of Agroforestry Products in 2019 (2019년 유통 임산물 중 산나물류와 약초류, 과실류 13종의 농약 및 중금속 잔류 실태)

  • In accordance with the implementation of the Positive List System (PLS), the proper usage of pesticides is now being enforced. It is assumed that unregistered pesticides are being used on agroforestry products due to the low number of registered pesticides in the agricultural industry. In this study, pesticide and heavy metal residues were investigated in 13 types of products to determine the status of usage. The levels (%) of pesticides detected in Pimpinella beachscape, Platycodon grandiflorum, Codonopsis lancekolate, Artemisia dubia, Angelica gigas, Pyrus pyrifolia, and Punica granatum were 40.0, 20.0, 26.7, 13.3, 56.3, 57.1, 33.3, 26.7, 66.7, and 46.7%, respectively, while, those in Petridium aquilinum, Disoscorea batata and Senna tora were zero. Heavy metals (Pb, Cd) were detected only from P. grandisflorum and A. dubia. The pesticide usage and registration data by agroforestry product obtained in the study will be useful in the future for ensuring the safety of domestic agroforestry products.

Antioxidative Activity and Chemical Characteristics of Leaves, Roots, Stems and Fruits Extracts from Acanthopanax senticosus (가시오가피 잎·뿌리·추출물의 이화학적 특성 및 항산화 효과)

  • The contents of bioactive and antioxidative activities (DPPH (${\alpha},{\alpha}'$-diphenyl-${\beta}$-picrylhydrazyl), free radical scavenging activity, peroxidation of linoleic acid and rat hepatocyte microsome, and Fe/Cu reducing power, tyrosinase inhibition activity) were tested by in vitro experimental models using water, hot water, ethanol and methanol extracts of leaves (ASL), roots (ASR), stems (ASS) and fruits (ASF) from Acanthopanax senticosus. Hot water extract from ASL showed the highest extraction yield (16.04%) as well as highest contents of phenolic compounds (2.67%) and flavonoids (1.43%). Major minerals were K, Ca and Mg. In oxidation in vitro models using DPPH free radical scavenging activity, Fe/Cu reducing power, $Fe^{2+}$/ascorbate-induced linoleic acid peroxidation by ferric thiocyanate and thiobarbituric acid (TBA) methods, tyrosinase inhibition activity and autooxidation of rat hepatic microsomes membrane, and antioxidative activities were strong in Acanthopanax senticosus. From these results, ASL extracts were shown to have the most potent antioxidative properties and contain the highest amounts of antioxidative compounds such as phenolic compounds and flavonoids. These results may provide the basic data to understand the biological activities of bio-active materials derived from leaves of Acanthopanax senticosus.

Growth Inhibitory Effect of Extracts of Propolis on Epithelial Ovarian Cancer Cells (상피성 난소암 세포에서 프로폴리스 추출물의 세포 증식 저해 효과)

  • Propolis is a natural product collected from plants by honey bees product used extensively in traditional medicine for its antioxidant, anti-inflammatory, immunomodulatory and anti-cancer effects. Propolis exhibits a broad spectrum of biological activities because it is a complex mixture of natural substances. Ovarian cancer is the second most common newly diagnosed cancer from all cancers among women in Korea and the leading cause of death from gynecological malignancies. While most ovarian cancer patients initially respond to surgical debulking and chemotherapy, patients later succumb to the disease. Thus, there is an urgent need to test novel therapeutic agents to counteract the high mortality rate associated with ovarian cancer. In this study, we investigated the anti-cancer properties and the active mechanism of Australian propolis in human epithelial ovarian cancer A2780 cells. Our data revealed that propolis showed a cytotoxic activity in a dose-dependent manner. Flow cytometric analysis for cell cycle arrest and apoptosis using propidium iodide staning and annexin V-FITC indicated that propolis could induce cycle arrest in the G0/G1 phase and apoptosis in a dose-dependent manner on human epithelial ovarian cancer cells. These results suggest that the Australian propolis is potential alternative agent on ovarian cancer prevention and treatment.

Degradation of Alginate Solution by Using ${\gamma}-Irradiation$ and Organic Acid (감마선과 유기산을 이용한 알긴산 용액의 저분자화에 대한 연구)

  • Alginates were irradiated in an aqueous solution with $Co^{60}$ gamma rays in the dose ranges from 0 to 100 kGy, and investigated the relationship between the intrinsic viscosity $([{\eta}])$ and the molecular weight $(M_w)$ of alginates. The molecular weight of alginate was measured by gel permeation chromatography and the ranges from 1,894 to 135,174 Da were obtained. The molecular weight of alginate decreased markedly with increasing the degree of ${\gamma}-ray$ dose rate. The intrinsic viscosity of alginate solution after ${\gamma}-irradiation$ showed the ranges from 9.83 (g/g) to 602.69 (g/g), depending upon the ${\gamma}-irradiation$ dose. The molecular weight of alginate dependence of the intrinsic viscosity of the alginate solution would be expressed by Mark-Houwink equation. With a linearization of molecular weight and the intrinsic viscosity of the alginate solution, Mark-Houwink equation could be expressed with constant variables and the real data fitted to the equation of $[{\eta}]=2.2{\times}10^{-6}\;{M_w}^{1.656}\;(R^2=0.998)$.

Effects of Mandibular Midline Shift and Difference of Mandibular Height on the Masticatory Muscle Activity (하악 정중선의 편위와 하악골의 높이차가 저작근 활성에 미치는 영향)

  • This study was performed to investigate the effect of mandibular midline shift and difference of mandibular height between both sides on the electromyo- graphic(EMG) activity of the masticatory muscles on clenching or gum chewing movement. For this study, 105 patients with temporomandibular disorders(TMD) were selected and panoramic radiograph were taken. Amount and side of the midline shift and height of the mandible from antegonial notch to the top of the condylar head were measured on panoramic view. $BioEMG^{(R)}$ (Bioresearch Inc., Milwaukee, USA) was used for recording of EMG activity(${\mu}V$) of the anterior temporalis and the superficial masseter on clenching or gum chewing movement. EMG activity on clenching during 533msec period were measured for activity of the starting point and the one second-after activity as the early EMG and the maximum EMG, respectively. EMG activity on gum chewing movement were measured for activity of the first and the second chewing stroke. The data collected were analysed by SPSS windows program, and the results of this study were as follows : 1. Height of the mandible was 8.06cm on right side and 8.03cm on left side, and showed no difference by age, but significantly differed by sex with higher in male subjects. 2. Mean value of the midline shift was 0.1mm with range of 0~5mm on both sides. The amount and side of the midline shift did not related with height difference of the mandible and/or the EMG activity of the masticatory muscles on clenching. 3. Prevalence of higher right side and higher left side of the mandible were almost same, and the EMG activity of higher side was not higher than that of the other side. 4. In the subjects with height difference of more than 5mm between both sides of the mandible, the early EMG activity on clenching were differed for the anterior temporalis, but the maximum activity were differed for the superficial masseter. 5. In the subjects with height difference of more than 5mm between both sides of the mandible, EMG activity of the anterior temporalis of the gum chewing side was not higher than that of the other side when chewing on the side of lower height, but in the subjects with height difference of less than 5mm, the EMG activity was higher than that of the other side.

Monitoring and risk assessment of pesticide in school foodservice products in seoul, Korea (서울지역 학교급식 식재료의 잔류농약 위해성 평가)

  • We tested for residual pesticide levels in school foodservice agricultural products in Seoul, Korea from 2010 to 2012. A total of 316 samples of 23 different types of agricultural products were analyzed via gas chromatography-nitrogen phosphate detector (GC-NPD), an electron capture detector (GC-${\mu}ECD$), a mass spectrometry detector (GC-MSD), and a high performance liquid chromatography-ultraviolet detector (HPLC-UV). We used multi-analysis methods to analyze 185 different pesticide types. Among the selected agricultural products, residual pesticides were detected in 26 samples (8.2%), of which 6 samples (1.9%) exceeded the Korea Maximum Residue limits (MRLs). We detected pesticide residue in more than 65% of the Chwinamul, while 6 among 9 analyzed samples contained pesticide residue, and 1 sample exceeded the Korea MRLs. Among the 185 kinds of pesticides that we have tested, 18 were detected, while 7 of them were detected more than twice. Data obtained were then used for estimating the potential health risks associated with the exposures to these pesticides. The most critical commodity is carbofuran in the perilla leaves, which has contributed 3.8% to the hazard index (HI). These results showed that the detected pesticides could not be considered as a serious public health problem. Nevertheless, constant supervision is recommended.

Biological Activities and Chemical Characteristics of Monascus-Fermented Korean Red Ginseng (홍국발효홍삼의 이화학적 특성 및 생리활성)

  • This study investigated the biological activities and chemical characteristics of Monascus-fermented Korean red ginseng (MFRG). The comparative activities of water, ethanol, and methanol extracts from MFRGE and Korean red ginseng (RG) were tested in vitro of anti-oxidative models of linoleic acid peroxidation by thiocyanate and thiobarbituric acid (TBA) methods and DPPH ($\alpha,\alpha'$-diphenyl-$\beta$-picrylhydrazyl)radical scavenging activities. In addition, measurements of their bioactive total phenolic compounds and minerals, and extract yield, were obtained. The extract yield of each solvent extracted from MFRG and RG was aqueous by 6.58% and 5.83%, ethanol by 0.62% and 0.98%, and methanol by 1.27% and 3.04%, respectively. Total phenolic compounds were higher in all solvents extracted from MFRG than those from RG. Major mineral contents (ppm) of MFRG and RG were K by 16,936 and 22,386, Ca by 2,310 and 3,693, Mg by 2,703 and 2,647, respectively. The DPPH radical scavenging activities were higher in all solvents extracted from MFRG than those from RG, however, all these extracts exhibited a relatively low level of radical-scavenging activity compared to the butylated hydroxytoluene (BHT). In antioxidative activities determined by TBA method using linoleic acid peroxidation, 70% methanol extract from MFRG and RG showed the highest antioxidative activity at a concentration of 0.1%. These results may provide the basic data to understand the biological activities of bio-active materials derived from MFRG.

Phenolic Content, DPPH Radical Scavenging, and Tyrosinase Inhibitory Activities of Ecklonia cava Extracted with the Ultrasonic Wave Method (초음파 추출법에 의한 감태 추출물의 페놀성 화합물 함량, DPPH 라디칼 소거 활성 및 tyrosinase 저해 활성 분석)

  • The applicability of the ultrasonic wave method to the extraction of useful components from seaweeds was investigated. Extracts from freeze-dried Ecklonia cava powder were prepared with hot water ($65^{\circ}C$), water ($24^{\circ}C$), 50% ethanol, and 100% ethanol, and ultrasonic extraction was also performed. The content of phenolic compounds and the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and tyrosinase inhibitory activity of the extracts were analyzed, and differences in the data obtained by the ultrasonic extraction and the traditional extraction methods were compared. The phenolic content in the E. cava extract by ultrasonic extraction (142.80 mg/g) was approximately 14 times higher than the phenolic content in the hot water extract (10.03 mg/g). The DPPH radical scavenging and the tyrosinase inhibitory activities of the ultrasonic extract were approximately 4 times and 14 times higher than the hot water extracts, respectively. The correlation between the phenolic content and the DPPH radical scavenging activity ($R^2$=99.47) and between the phenolic content and the tyrosinase inhibitory activity ($R^2$=99.99) was very high. These results indicate that ultrasonic extraction is more suitable than traditional extraction for the extraction of useful components from E. cava.

Induction of Mitotic Arrest and Apoptosis by Diallyl Trisulfide in U937 Human Leukemia Cells (U937 인체혈구암세포에서 diallyl trisulfide에 의한 mitotic arrest와 apoptosis 유발)

  • Diallyl trisulfide (DATS), one of the major organosulfur components of garlic (Allium sativum), has various biological effects such as anti-microbial and anti-cancer activities. However, the molecular mechanisms of growth inhibition related to cell cycle arrest are poorly understood. In this study, we investigated the effects of DATS on cell cycle progression in U937 human leukemia cells. Treatment with DATS in U937 cells resulted in inhibition of cell viability through G2/M arrest and apoptosis. DATS-induced G2/M arrest was associated with up-regulation of cyclin B1 and cyclin-dependent kinase 1 (CDK1). DATS also significantly increased levels of phospho-histone H3, which is a mitosis-specific marker, indicating that DATS induced mitotic arrest but not G2 arrest in U937 cells. DATS treatment also generated the reactive oxygen species (ROS) in U937 cells; however, pretreatment with N-acetyl-l-cysteine (NAC), a ROS scavenger, significantly attenuated DATS-induced mitotic arrest and apoptosis. Taken together, our data indicate that DATS exhibits anti-cancer effects through mitotic arrest and apoptosis in a ROS-dependent manner.

The Analysis of Function and Factors for the Value Assessment of Ecosystem Service at Rice Paddy Wetland (논습지의 생태계서비스 가치평가를 위한 기능 및 요인분석)

  • Recently, wetland and paddy wetland are being highlighted as the international environmental issues. However, research system which can assess paddy wetland is lacking. Thus, the purpose of this study is to assess the value of paddy wetland by applying RAM to paddy wetland. In addition, we would like to use this study as the data to establish assessment method for paddy wetland. 16 sites were selected as research targets based on altitude and soil. As the results of wetland assessment, 16 research target sites had total score of 212~227 and the average of 2.17~2.50. It was similar to those of Lacustrine Wetlands, Palustrine Wetland and Riverine Wetland which were sites in previous studies. The value could be recognized as the wetland. However, there was no difference in assessment results based on altitude and soil. It was found that all conservation values were the same. The factor the most closely affecting conservation value was the area. However, there was limitation to apply existing wetland assessment system to paddy wetland. In order to assess paddy wetland, factors such as rice farming methods, topography, vegetation, growth environment and biodiversity should be added. It was thought to supplement wetland assessment system through various further studies.
