Extracellular Proteome Profiling of Bacillus pumilus SCU11 Producing Alkaline Protease for Dehairing

  • Bacillus pumilus is one of the most characterized microorganisms that are used for high-level production of select industrial enzymes. A novel B. pumilus SCU11 strain possessing high alkaline protease activity was obtained in our previous work. The culture supernatant of this strain showed efficient dehairing capability with minimal collagen damage, indicating promising potential applications in the leather industry. In this study, the strain's extracellular proteome was identified by LC-MS/MS-based shotgun proteomic analysis, and their related secretory pathways were characterized by BLAST searches. A total of 513 proteins, including 100 actual secreted and 413 intracellular proteins, were detected in the extracellular proteome. The functions of these secreted proteins were elucidated and four complete secretory systems (Sec, Tat, Com, and ABC transporter) were proposed for B. pumilus. These data provide B. pumilus a comprehensive extracellular proteome profile, which is a valuable theoretical and applicative basis for future genetic modifications and development of industrial enzymes.

  • The chlorinated dioxins and furans have attracted considerable scientific and public concern because of their environmental persistence and super-toxicity through the foodchain. Recent dioxin scandals in several military bases have also contributed to a higher awareness on the side of food consumers as well as foodwaste combustion. However, there is continuing uncertainty over the relative importance of different sources of dioxins and furans to the soil environment. In difference to those awareness there is a main influence of potential soil contamination on the dioxin contents in groundwater. It is, therefore, important to provide a sound scientific framework and basis by which to evaluate the significance of the presence of dioxin in soils. Consequently, we have to identify the characteristics and nature of dioxin released into the soil environment, especially in agricultural aspect.

System Design for Developing the Remote Controlled Sprayer of Pear Trees (배나무 무인 방제기의 개발을 위한 살포 시스템 설계)

  • A remote controlled sprayer has designed, manufactured and experimented to spray well on pear trees with pesticides. This study was executed to automate pest management of pear trees. Types of spray nozzle, which was used on the system, were analyzed experimentally to find an optimal spray equipment configuration with several nozzles. Attributions of ultrasonic sensors were analyzed to adjust spraying distance of an unmanned sprayer system. This paper investigated shapes of pear trees and cultivating environment of pear orchard. In order to select optimal spray environment, liquid distribution was measured while angle of nozzle was changed. Additionally, liquid distribution by distance and sprayed liquid capacity by side distance were measured. According to information of shapes of pear trees and cultivating environment of pear orchard, sprayer frames of an unmanned sprayer system were manufactured and sprayer frames were suitable for interval of pear trees. The sprayer system could adjust width of sprayer frames to 2.5 m and height of sprayer frames to 1.7 m. Optimal angle of nozzle, and optimal distance between objects and nozzle were $15^{\circ}$ and 0.8 m. When side distance was placed from 1.2 m to 1.8 m, sprayed capacity reached to the highest amount.

The Norwegian Model of Fisheries Bio-Resources Management (노르웨이 해역 수산생명자원 관리모델)

  • The Norwegian coastal area is the most efficient region for fishery production in the world's oceans, the Norway is the world's top 10 fisheries countries through efficient fishing and fishing aquaculture technology and its scientific management of fisheries bio-resources, with Norwegian salmon having attained the world's highest level. In the late 1980s, fisheries resources were depleted due to overfishing and fish diseases, resulting in a crisis in the fishing industry that lasted until the early 1990s. Since the national fishery emergency, people involved in the fishing industry, including fishermen, research scientists, and government officers, have tried to overcome the challenges facing the industry and identify an appropriate management model for fisheries bio-resources in the Norwegian coastal area. First, research vessels were used to monitor water and sediment conditions and fishery species, with the long-term aim of predicting fishery resources in real time and collecting information on species diversity, abundance, and distribution. Second, a "Healthy Fish Project" was promoted to counter natural disasters and fish disease problems with the development of vaccines against viruses and bacteria, eventually allowing for a decrease in the use of antibiotics and the production of notably healthier fish in the 2000s. Third, a systematic management model was developed to help with preparations for decreases in the total number of fishermen and increases in the proportion of elderly fishermen in the fishery industry using the development of automatic fishing aquaculture systems and short-chain systems. We could learn from the Norwegian model of fisheries bio-resources, management and could adopt it for the preparation of fishery bio-resources management policy for South Korean coastal areas in the near future.
