Characteristics and Action Pattern of Alikaline Lipase from Serratia liquefaciens AL-11 (Serratia liquefaciens AL-11이 생산하는 Alkaline Lipase의 특성 및 작용양상)

  • The optimum temperature and pH for the enzyme activity were 45$^{\circ}C$ and 10.0, respectively. The enzyme was stable in a pH range of 5 to 10, and 62% of its activity was lost on heat treatment of 60$^{\circ}C$ for 20 min. The activity of the purified enzyme was inhibited by $Fe^{2+},\;Zn^{2+}\;and\;Pb^{2+}$, and slightly activated by $Mn^{2+}\;and\;Ca^{2+}$. ${\gamma}$-Chloromercuribenzoic acid, 2,4-dinitrophenol and $H_{2}O_{2}$ did not show inhibitroy effect on the lipolytic activity of the alkaline lipase but ethylenediaminetetraacetic acid inhibited the enzyem activity. This suggested that the enzyme have metal group in its active site. Sodium salts of bile acids stimulated the enzyme activity. Analysis of hydrolyzates of olive oil after the reaction revealed that Serratia liquefaciens AL-11 produced non-specific lipolytic enzyme.

Hypocholesterolemic Response to Karaya Saponin and Rhodobacter capsulatus in Broiler Chickens

  • Dietary karaya saponin and Rhodobacter capsulatus (R. capsulatus) are known to have hypocholesterolemic actions, as reported in our previous studies. This study examined possible synergistic hypocholesterolemic effects of karaya saponin and R. capsulatus in broilers. A total of 150 broilers were allocated into 10 treatments: control, saponin 25 mg, saponin 50 mg, saponin 75 mg, saponin 25 mg+R. capsulatus 0.2 g, saponin 25 mg+R. capsulatus 0.4 g, saponin 50 mg+R. capsulatus 0.2 g, saponin 50 mg+R. capsulatus 0.4 g, saponin 75 mg+R. capsulatus 0.2 g and saponin 75 mg+R. capsulatus 0.4 g. Feed intake and feed efficiency were improved when karaya saponin and R. capsulatus were synergistically supplemented in the diet. Combinations of karaya saponin, especially supplementation of karaya saponin 50 mg+R. capsulatus 0.4 g were shown to have potential hypolipidemic actions in breast and thigh muscle cholesterol and triglycerides, serum cholesterol, low density lipoprotein-cholesterol and triglycerides, as well as improved high density lipoprotein (HDL)-cholesterol (p<0.05). Compared to the control, almost all the treatments significantly increased serum, liver and fecal concentrations of bile acids (p<0.05). Supplementation of both karaya saponin (75 mg) and saponin 50 mg+R. capsulatus 0.4 g reduced palmitic acid (C16:0) and stearic acid (C18:0) in a similar fashion (p<0.05). The ratios of PUFA:SFA or PUFA+MUFA:SFA in the thigh and breast muscle of broilers were greater in karaya saponin and R. capsulatus supplemented groups than in the control group. Thus, our study concluded that supplementation of karaya saponin synergistically with R. capsulatus in the diet of broilers is an effective way to obtain low-cholesterol, low-triglyceride and high HDL-cholesterol enriched poultry meat with a unique fatty acid balance.

Effect of Lecithin Intake on Lipid Metabolism and Antioxidative Capacity in Rats Fed High Fat Diet (레시틴섭취가 고지방 식이를 섭취한 흰쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate the effect of lecithin on lipid metabolism and antixidative capacity in 9-week-old rats. Forty-five male Sprague-Dawley rats weighing 249.8 g were blocked into three groups according to their body weight and raised for 8 weeks with experimental diets containing 1% (LM) or 5% lecithin (LH) and control (C) diet. Plasma and liver total lipids, triglyceride, total cholesterol and plasma HDL-cholesterol concenterations, and fecal total lipids, triglyceride, total cholesterol and bile acid excretions were measured. Malondialdehyde (MDA) levels in plasma, liver, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities in red blood cell and liver, xanthine oxidase (XO) activities in plasma and liver, and total antioxidant status (TAS) in plasma were also measured. Effect of lecithin intake on antioxidative capacity was not significantly different among all the groups. Plasma total lipids, triglyceride and total cholesterol levels were lower in lecithin groups compared to control group, and these three lipid levels of lecithin groups were lowered dose-dependently as dietary lecithin level increased. But liver total lipids, triglyceride and total cholesterol levels were not different among all the groups. Also fecal total lipids, triglyceride and total cholesterol excretions were highest in high lecithin groups compared to two other groups. Thus it is plausible that lecithin intake decreases plasma lipid levels through increasing fecal lipid excretions, and may be beneficial for treatment and prevention of hyperlipidemia, but has no effect on antioxidative capacity.

  • To select potent probiotics from lactic acid bacteria in Yulmoo Kimchi, an in vitro and stimulated human intestinal model system (SHIMS) test were performed. One Leuconostoc mesenteroides strain from five strains of Yulmoo Kimchi and one Lactobacillus plantarum from 12 strains of KCTC and KCCM were selected according to survival in acidic and bile salts conditions. Between the two species, Leu. mesentroides displayed higher survival activity in a SHIMS test. The strain was identified as Leu. mesentroides by 16S rRNA sequencing and was designated as Leu. mesentroides K01.

Effect of Alfalfa Saponin on the Serum Cholesterol Level in Rats (알팔파 사포닌이 흰쥐 혈청 콜레스테롤 농도에 미치는 영향)

  • Alfalfa saponin was isolated from the roots of alfalfa(Medicage sativa L.) and incoporated at the level of 0.2% and 0.4% cholesterol-free diets and cholesterol enriched-diets. Wistar male rats weighing approximately 80g were fed with for 21 days. The serum lipids and fecal excretion lipids of the rats were determined and the result obtained are as follows. 1. Alfalfa saponin contained 36.4% medicagenic acid. 2. The feeding of alfalfa saponin increased cholesterol excretion but not bile acid excretion. 3. Alfalfa saponin showed hypocholesterolemic activity at 0.4% level in both the diets with and without cholesterol but the effect was not observed at 0.2% level in a diet. 4. It was indicated that the hypocholesterolemic effect of alfalfa saponin was associated with the decreased absorption of cholesterol from alimentary canal.

Effect of Garlic Diets on the Changes of Serum Cholesterol Glucose Level and Coagulation Time in Rats (마늘첨가식이(添加食餌)가 흰쥐의 혈청(血淸) 콜레스테롤, 글루코오스의 함량(含量) 및 혈액응고시간(血液凝固時間)에 미치는 영향(影響))

  • Effects of garlic on hypocholesterolemia, anticoagulation and hypoglycemia were studied in the present experiments using male rats. The results were summarized as follows. 1. The supplementation of $2{\sim}4%$ garlic to 2% cholesterol diets did not affect food intake and weight gain in male rats. 2. Rat's groups fed the diets supplemented with $2{\sim}3%$ garlic (C.D.E.F) to 2% cholesterol diet (B) decreased serum total cholesterol levels by $16{\sim}32%$, triglyceride levels by $18.6{\sim}26.8%$ and ${\beta}/{\alpha}-lipoportein$ rations by $42{\sim}58%$, but increased HDL-cholesterol levels by $29{\sim}65%$ as compared to B group, and so the author assumes that garlic supplementation exerts hypocholesterolemic effect on cholesterol - fed rats because of the increase of HDL and HDL - cholesterol levels. 3. Rat's groups fed the diets supplemented with $2{\sim}4%$ garlic (C.D.E.F) to 2% cholesterol plus 0.25% bile salt diet (B) increased whole blood coagulation times, prothrombin times and fibrinolytic activities, but decreased plasma fibrinogen levels as compared to B group, and so the author assumes that garlic supplementation exerts anticoagulative effect because of the inhibition of fibrinogen synthesis in male rat's liver. 4. Rat's groups fed the diets supplemented with $2{\sim}4%$ garlic (B.C.D.E) to standard diet (A) decreased serum glucose levels by $1{\sim}24%$, but increased serum insulin concentrations by $0.5{\sim}3.0$ times as compared to A group, and so the author assumes that garlic supplementation exerts hypoglycemic effect because of the increase of serum insulin levels by stimulation pancreatic secretion of insulin from ${\beta}-cells$ in the islets of Langerhans.

Effect of Garlic Diets on Blood Coagulation of Cholesterol-fed Male Rats (마늘첨가식이(添加食餌)가 콜레스테롤 투여(投與)흰쥐의 혈액응고(血液凝固)에 미치는 영향(影響))

  • The purpose of this study was to investigate the effects of garlic diet on blood coagulation in cholesterol-fed rats. 36 male rats were divided into six groups and fed experimental diet for three weeks. Two-levels (2% and 4%) of fresh and boiled garlic were supplemented to the stock diet with 2% cholesterol and 0.25% bile salt for the experimental diet. After the rats fed with experimental diets during 3 weeks, the whole blood coagulation time, the content of plasma fibrinogen, the fibrinolytic activity and the prothrombin time were measured. The results are summarized as follows; 1. Garlic diet have no influence on body weight and food consumption in rats. 2. In the case of cholesterol-fed group, the whole blood coagulation time was decreased 24.3% compared with the normal group(A). But garlic diet group(C) was similar to the normal group(A) and groupD-F was increased 4.9-11.5%. 3. The content of plasma fibrinogen of cholesterol-fed group was higher 114.5% than that of normal gropu(A). In the garlic diets(C-F) the contents of plasma fibrinogen were increased 33.1-63.8%. 4. The fibrinolytic activity of cholesterol diet was decreased 16.2% compared with the normal group(A) but was increased 10.9-44.5% compared to the garlic diets(C-F). 5. The prothrombin time of cholesterol-fed group was decreased 8.9% compared with normal group(A) but was increased 8.0-46.3% in garlic diets(C-F). As mentioned above, the garlic diets increased whole blood coagulation time, prothrombin time and fibrinolytic activity and decreased the content of fibrinogen. For that reason it is thought that the garlic diets have a anticoagulation effect regulating the fibrinogen synthesis in liver.

Difference of Toxicities among Tissues in the Pufferfish Fugu xanthopterus('Ggachibog') (까치복, Fugu xanthopterus의 조직별 독성차이)

  • Twenty-four specimens of the pufferfish Fugu xantheptens, Korean name, 'Ggachibog', collected at a fish market of Pusan were examined for anatomical distribution of toxicity by the mouse bioassay method. Frequency of toxic specimens was 88, 75, 54, 13, 71, 80 and $71\%$, in terms of liver, intestine, skin, muscle, testis, ovary and bile, respectively. Their the highest toxicity scores were 417, 387, 112, 17, 39, 403 and 178 MU/g, respectively; and average toxicity values were $110\pm25.0(mean \pm S.E.)$, $73\pm20.3,\;17.8\pm 5.1,\;2.7\pm1.1,\;15.6\pm5.4,\;115\pm33.0\;and\;34\pm9.3 MU/g,$ respectively. A significant correlation between the toxicities of liver and intestine(r=0.93), between those of liver and skin (r=0.79) and between those of liver and ovary(r=0.83) was observed.

Proteomic Analysis of Proteins Increased or Reduced by Ethanol of Lactobacillus plantarum ST4 Isolated from Makgeolli, Traditional Korean Rice Wine

  • LAB were isolated from makgeolli locally produced around Jinju, Gyeongnam, S. Korea during spring of 2011. Randomly selected 11 isolates from MRS agar plates were identified first by API CHL 50 kits and then 16S rRNA gene sequencing. All 11 isolates were identified as Lactobacillus plantarum. Among them, ST4 grew in MRS broth with ethanol up to 10%, showing the highest alcohol resistance. L. plantarum ST4 was moderately resistant against acid and bile salts. When cellular proteins of L. plantarum ST4 under ethanol stress were analyzed by two-dimensional gel electrophoresis (2DE), the intensities of 6 spots increased, whereas 22 spots decreased at least 2-fold. Those 28 spots were identified by peptide mass fingerprinting (PMF). FusA2 (elongation factor G) increased 18.8-fold (6% ethanol) compared with control. Other proteins were AtpD (ATP synthase subunit beta), DnaK, GroEL, Tuf (elongation factor Tu), and Npr2 (NADH peroxidase), respectively. Among the 22 proteins decreased in intensities, lactate dehydrogenases (LdhD and LdhL1) were included.

Changes of Liver Function In Korean Black Goats Dosed wit Carbon Tetrachloride and 1-naphthylisothiocyanate (한국흑염소에 있어서 사염화탄소와 1-naphthylisothiocyanate 투여시의 간기능 변화)

  • In order to study the effects of administration of carbon tetrachloride(CCI$_4$) and 1-naphthylisothiocyanate(ANIT) on the liver of Korean black goats, some liver function tests and liver biopsy were done on 4 Korean black goats dosed with CCI$_4$(0.4m1/kg of body weight) in-traruminally and 4 Korean black goats dosed with ANIT(400mg/kg of body weight) by stomach tube. BSP Tl/2 and serum total bilirubin concentration in goats dosed with CCI$_4$ were increased gradually, reached to maximum value on 2nd and 1st day, respectively, and then began to decrease in normal range, gradually. In goats dosed with ANIT, BSP Tl/2 and serum total bilirubin concentration were increased rapidly, reached to maximum value on 0.5 and 1st day, respectively, and then returned to normal ragne, rapidly. Serum SDH, AST and GGT activities in goats dosed with CCI$_4$ were increased rapidly and reached to maximum value on 3rd, 1st and 2nd day, respectively. Thereafter, the serum enzyme activities began to decrease in normal range gradually. In goats dosed with ANIT, however, serum SDH, AST and GGT activities were not changed. The histopathologic changes in goats dosed with CCI$_4$ were lipidosis and centrilobular nee-rosis of the hepatic parenchyma. In goats dosed with ANIT, hyperplasia of bile duct epithelium was noticeable, but pathologic changes in liver parenchyma were not noticed. Conclusively, in Korean black goats dosed with CCI$_4$, main finding was necrosis of hepatic parenchyma. In Korean black goats dosed with ANIT, main finding was cholestasis.
