  • Bicarbonate anion ($HCO_3{^-}$) takes the role of major buffer systems in our body by maintaining the pH at 7.4. Epithelial $HCO_3{^-}$ secretion also hydrolyzes the mucus which protects body from noxious infections. It has been widely known that such infections are closely related to $HCO_3{^-}$ permeability through membrane and, thus, increasing the $HCO_3{^-}$ permeability is essential. To evaluate the $HCO_3{^-}$ permeability through ion channels, the free energy changes relevant to ion pumping are calculated with the Integral Equation Formalism-PCM (IEF-PCM) theory. Molecular structures of various anions including $HCO_3{^-}$ were optimized with the density functional theory at the level of B3LYP/6-311++G(d,p) in gas and solution phase. In addition, the anion permeability is significantly influenced by the relative size of the anion and pore. We introduce a shifted volume factor model that describes the pore size effect when the charged solutes transfer through ion channels. We found excellent agreement between experimental and calculated permeability when our novel model of the size effect was taken into account to.

수분산성 아크릴계 점착제 중합 조건에 관한 연구 (Study on Polymerization Condition of Water-based Acrylic Adhesion)

  • 본 연구는 주단량체로 2-에틸헥실아크릴레이트(2-EHA), n-부틸아크릴레이트(n-BA), 메틸메타크릴레이트(MMA) 그리고 기능성 단량체로 아크릴산(AAc)을 사용한 유화중합에서 아크릴계 점착제를 합성하기 위한 최적의 합성 조건을 구하고자 한다. 이를 위하여 계면활성제 농도의 영향, 계면활성제 Hydrophilic lipophilic Balance(HLB) 값, 중합개시제의 농도와 완충제가 아크릴계 점착제의 물성에 미치는 영향을 연구하였다. 그 결과 계면활성제는 단량체 사용량에 대하여 3~5 wt%를 사용하였을 때, HLB 값이 12.3일 때 가장 안정한 점착체를 얻을 수 있었으며, 중합 개시제의 농도가 1 wt% 이상 사용하였을 때는 중합 속도는 증가하였으나 안정성이 저하되었다. 완충제로 sodium bicarbonate를 사용할 경우 0.4 wt% 이상 사용하여야 충분한 완충 효과가 있었으며, 단량체의 종류에 따라 약간의 차이는 있으나 완충제의 농도가 0.6~0.8 wt%가 적당하여 4성분계 유화중합에서 최적의 중합 조건을 제시할 수 있었다.

Effects of Monensin and Fish Oil on Conjugated Linoleic Acid Production by Rumen Microbes in Holstein Cows Fed Diets Supplemented with Soybean Oil and Sodium Bicarbonate

  • The present study was conducted with four ruminally canulated Holstein cows to observe the effects of monensin or fish oil on diet fermentation and production of conjugated linoleic acids (CLAs) in the rumen when fed diets supplemented with soybean oil and sodium bicarbonate. Cows of the control treatment were fed a basal diet (CON) consisting of 60% commercial concentrate and 40% chopped rye grass hay. Cows of other treatments were fed the same diet as CON, but the concentrate was supplemented with 7% of soybean oil and 0.5% of sodium bicarbonate (SO-B), SO-B supplemented with monensin (30 ppm, SO-BM) or concentrate supplemented with 6.3% of soybean oil, 0.5% of sodium-bicarbonate, 30 ppm of monensin and 0.7% of fish oil (SO-BMF). Dry matter (DM) intake of the cows was significantly (p<0.011) reduced by feeding the SO-BMF diet compared to the other diets which did not differ in DM intake. Whole tract digestibility of major dietary components was significantly (p<0.004-0.027) higher for SO-BMF than the other supplement-containing diets. Dietary supplements did not clearly affect rumen pH and ammonia concentrations compared to the CON diet. Significantly reduced (p<0.05) total VFA concentration was obtained by the addition of fish oil to the diet (SO-BMF) compared to other diets. No differences, however, were obtained in major VFA proportions as well as in total VFA between the supplemented diets. The SO-BM diet increased (p<0.01-0.05) the concentrations of trans-11 $C_{18:1}$ and linoleic acid in rumen fluid. Total CLA concentration was also increased by the feeding of SO-B and SO-BM diets during early fermentation times (up to 3 h) post-feeding. Cis-9, trans-11 CLA concentration in rumen fluid was highest (p<0.05) for SO-B up to 1 h while the highest (p<0.01) value for SO-BM occurred at 3 h post-feeding. An increased trans-10, cis-12 CLA concentration was obtained from the SO-B and SO-BM diets at 1 and 3 h post feeding compared to the other diets. Supplementation of oils with monensin and sodium bicarbonate increased (p<0.05) the proportions of $C_{18:1}$ and CLA in the plasma of cows, but the effect of monensin and/or fish oil was limited to trans-10, cis-12 CLA.

적출간 관류법을 이용한 카드뮴과 니켈의 간독성 비교 (Comparative Hepatotoxicity Assessment of Cadmium and Nickel with Isolated Perfused Rat Liver(IPRL))

  • 목적 : 이 연구의 목적은 적출간 관류법을 통하여 염화니켈과 염화카드뮴의 간 독성을 비교하는 데에 두었다. 방법 : AST(aspartate aminotransferase ferase), ALT(alanine aminotransferase), LDH(lactate dehydrogenase) 등의 간기능을 나타내는 생화학적 지표와 관류속도를 간독성 지표로 이용하였으며 산소 소모율은 생존력을 나타내는 지표로 이용하였다. $300({\pm}50)g$ 정도의 흰쥐를 대조군과 각 노출군($50{\mu}M$$200{\mu}M$의 염화니켈과 염화카드뮴)에 5마리씩 총 25마리를 무작위로 할당하였다. Krebs-Ringer bicarbonate buffer solution을 관류액 용도로 제조하여 간문맥에 투입하고 간세포를 통과하여 vena cava로 배출되는 관류액을 일정한 시간 간격을 두고 계속적으로 채취하였으며 채취한 관류액은 간독성 지표를 측정하는 데에 이용하였다. 결과 : AST, ALT, LDH 모두 염하니켈 보다는 염화카드뮴 투여군에서 시간의 경과에 따라 큰 폭으로 증가하였으며 반복 측정된 2요인 분산분석을 실시한 결과, 통계적으로 유의한 차이를 나타냈다. 생존력은 두 물질 모두에서 시간의 경과에 따라 감소하는 경향을 나타냈다. 결론 : 결론적으로, 이 실험상에서는 염화니켈 보다 염화카드뮴의 간독성이 더욱 강한 것으로 드러났다. 적출간 더욱 강한 것으로 드러났다. 적출간 관류법의 특성상 급성 간독성 평가에서 적절한 기법으로 보이며 국내 연구에서도 이 방법이 널리 이용되기를 기대한다.

흰쥐 관류간 모델에서 저산소 및 산소재도입시 vitamin C가 간장기능에 미치는 영향 (The Effect of Vitamin C on Hypoxia/reoxygenation Induced Hepatic Injury in Isolated Perfused Rat Liver)

  • This study was done to investigate the effect of vitamin C on hypoxia/reoxygenation-induced hepatic injury ul isolated perfused rat liver. Isolated livers from rats fasted 18 hours were subjected to 45 min of hypoxia followed by reoxygenation for 45 min. The perfusion medium used was Krebs-Henseleit bicarbonate buffer (pH 7.4) and 0.5 mmol/L of vitamin C was added to the perfusate. Alanine aminotransferase (ALI) and lactate dehydrogenase (LDH) levels were significantly increased by hypoxia/reoxygenation. These increases were augmented by vitamin C. Glucose output and bile flow were markedly decreased by hypoxia/reoxygenation. Vitamin C aggavated the decrease of glucose output but had little effect on bile flow. Our findings suggest that hypoxia/reoxygenation diminishes hepatic metabolic and secretory functions, and vitamin C significantly aggravates these changes.

  • Lead and cadmium are potent environmental toxicants that affect populations living in Europe, Americas, and Asia. Identifying transporters for lead and cadmium could potentially 1 help us better understand possible risk factors. The iron transporter divalent metal transporter 1(DMT1) mediates intestinal transport of cadmium, and lead in yeast and fibroblasts overexpressing DMT1. In human intestinal cells knocking down expression of DMT1 attenuated uptake of cadmium and iron but not lead. A possible explanation is the expression of a second transporter for lead in intestine. In astrocytes, however, DMT1 appears to transport lead in an extracellular buffer at pH value. At neutral pH, transport was not mediated by DMT1 but rather by a transporter that is stimulated by bicarbonate and inhibited by 4,4'-diisothiocyanatodihydrostilbene-2,2'-disulfonic acid. The identity of this lead transporter is under study.

Optimization of Automated Suspension Trapping Digestion in Bottom-Up Proteomics via Mass Spectrometry

  • The Suspension Trapping (S-Trap) method has been a prominent sample preparation technique since its introduction in 2014. Its capacity to induce protein aggregation using organic solvents has significantly improved protein purification and facilitated peptide identification. However, its full potential for automation has been limited by the lack of a suitable liquid handling system until recently. In this study, we aimed to enhance the automation of S-Trap sample preparation by optimizing the S-Trap digestion process, incorporating triethylammonium bicarbonate (TEAB) and CaCl2. The utilization of TEAB buffer conditions in this innovative process led to a noteworthy 12% improvement in protein identification. Additionally, through careful observation of various incubation conditions, we streamlined the entire sample preparation workflow into a concise 4 hours timeline, covering reduction, alkylation, and trypsin incubation stages. This refined and expedited automated S-Trap digestion process not only showcased exceptional time efficiency but also improved trypsin digestion, resulting in increased protein identification.

인삼 Saponin이 백서부고환지방조직에서의 유리지방산동원에 미치는 효과 (Effect of Ginseng Saponin on the Mobilization of Free Fatty Acid from Epididymal Fat Pad of Rat)

  • Krebs-Ringer bicarbonate 완충용액(pH 7.4, 4%(w/v) bovine serum albumin 함유)에 백서부고환지방조직을 incubation 하면서 FFA 유리에 대한 인삼 saponin의 작용을 관찰한 saponin 농도 1mg/ml에서 FFA의 유리가 현저히 증가되었으며 0.01mg/ml에서는 오히려 감소하였다. Epinephrine으로 전처치하여 hormone-sensitive lipase system을 활성화시킨 백서부고환지방조직 homogenate에서는 $0.1{\sim}1mg/ml$의 인삼 saponin에 의하여 FFA 동원이 현저히 증가하였으며 0.01mg/ml에서는 역시 감소하였다. 유효농도에서의 인삼 saponin은 epinephrine에 의한 지방조직으로 부터의 FFA 동원에 아마도 협동적으로 작용할 것으로 사료된다.

염소의 산·염기 균형 특성에 관한 연구 (A Study on Characteristics of Acid-Base Balance in Goats)

  • The changes of acid-base status in vitro of the venous blood for 24 hours in ten Korean native goat were investigated. The acid-base parameters were measured within ten minutes after collection of the blood, and every hour during the first six hours and finally after twenty four hours of storage. Blood samples were stored at two different temperatures ($0-4^{\circ}C$ and $21-24^{\circ}C$). Twelve goats were induced acute acid-base disturbances by intravenous infusion of either hydrochloric acid or sodium bicarbonate and inhalated with $CO_2$ gas mixture (20% $CO_2$, 80% $O_2$) or hyperventilation were performed by means of respirator. The results were as follows; 1. Blood samples could be stored during the first two hours in ice water ($0-4^{\circ}C$) and one hour at room temperature without significant changes in pH. The magnitudes of changes were similar to those of cow, and lower than those of men and dogs. 2. The mean values of acid-base parameters in normal goat were arterial pH, 7.40; $P_{CO_2}$, 35.4mmHg; $HCO_3{^-}$, 21.8mEq/L. 3. Both the base excess and the bicarbonate showed high correlation (r=0.99) during the metabolic disturbance and were represented as $B.E.=1.38\;HCO^-{_3}-29.7$. 4. The slope of blood buffer curve obtained from the in vivo experiment was 16.3mEq/L/pH. 5. The magnitudes of changes in hydrogen ion concentration per unit change of $P_{CO_2}$ were 0.8nM/mmHg in hypercapnia and 1.0nM/mmHg in hypocapnia. 6. The ranges of acid-base parameters in normal goat urine were pH, 6.0-8.1; $P_{CO_2}$, 42-61mmHg; $HCO_3{^-}$, 2-110mEq/L. The concentration of potassium was higher (60-200mEq/L), and that of sodium was lower (8-70mEq/L) than those of human urine.

A Study of a Sodium Bicarbonate Cotransporter Functionally Expressed in Xenopus laevis Oocytes

  • Purpose: $HCO_3{^-}$ is the most important ion to buffer the acidity of saliva. The transport of $HCO_3{^-}$ is mediated by electrogenic $Na^+/HCO_3{^-}$ cotransporter 1 (NBCe1), which expressed in various tissues including salivary glands, kidney and pancreas, etc. This experiment was performed to investigate regulatory site of NBCe1involved in the pH regulation using various mutants of NBCe1. Materials and Methods: Human parotid gland NBCe1 (hpNBCe1) and mutants by deletion of 1~285 bp and 1~1,035 bp were prepared. After microinjection of each cRNA to oocytes of Xenopus laevis, they were incubated for 2~3 days. The function of each protein was tested by electrophysiological method. Results: When oocytes were exposed to the $HCO_3{^-}$ buffered solution, 1~285 bp deleted mutant hpNBCe1 evoked a marked hyperpolarization ranging from -90 mV to -160 mV (average: -134 mV; n=12) compared to the full length of hpNBCe1. Although 1~1,035 bp deleted mutant hpNBCe1 was also expressed in the plasma membrane, but it did not show any changes of membrane potentials. Conclusion: Our deletion mutant study demonstrated that 1~285 bp of the NBCe1 is the major domain to determine $HCO_3{^-}$ transport ratio.
