Hydrodynamic and Chloride Ion Effects on Corrosion of Cobalt in Bicarbonate Buffer Solution (Bicarbonate 완충용액에서 코발트의 부식에 대한 대류와 염화이온의 영향)

  • Bicarbonate 완충용액에서 Co-RDE를 이용하여 RDE 회전속도와 완충용액 속의 염화이온이 Co의 부식과 부동화에 미치는 영향을 연구하였다. Co-RDE의 회전속도가 부식에 미치는 영향은 Levich 식과 일치하였으며 부동화 막을 파괴하는데 염화이온의 효과가 큼을 알 수 있었다. 혼합 전위 이론을 사용하여 대류확산 조건에서 회전속도의 증가에 따라 부식전위가 양의 방향으로 증가하는 모형을 발견하였다. Tafel 영역에서 Co의 용해반응과 수소가 발생하는 환원반응은 전하이동과 물질이동을 이용하여 설명할 수 있었다.

Effect of Bicarbonate and Phosphate Buffer Treatments on the Structure and Thermal Stability of Spent Layer Meat (중 탄산 및 인산염 완층액 처리가 노계육의 조직구조 및 열안정성에 미치는 영향)

  • Spent layer breast meat and leg meat samples washed with 0.05 M sodium bicarbonate solution and 0.04 M phosphate buffer(pH 8.3) showed decreases in heat denaturation temperature indicating the destabilization of myofibrillar proteins. The destabilization was attributed to the solubilization of 95 Kdalton and 55 kdalton proteins from the myofibrils observed in gel-electrophoretograms. Transmission electron microscopy further indicated the breakage of Z-lines.

  • The effect of ginseng saponin on the release of free fatty acid from adipose tissue was studied in vitro. 1. The mobilization of free fatty acid from intact epididymal fat pad of rat, incubated in Krebs-Ringer bicarbonate buffer (pH 7.4) contained bovine serum albumin (4 w/v%), was increased by addition of ginseng sponin (1 mg/ml). 2. The mobilization of free fatty acid from homogenate of the epididymal fat pad of rat, of which hormone-sensitive lipese system was activated by pre-incubation of intact fat pad in the presence of epinephrine, was markedly increased by addition of ginseng saponin $(0.1{\sim}1mg/ml)$. 3. It is considered that ginseng saponin potentiate the actions of epinephrine on hormone-sensitive lipase system of the adipose tissue of rat.

Effects of Washing and Desinewing Treatments on the Composition and Quality Characteristics of Spent Layer Meat (세척 및 결체조직 제거 처리가 노계육의 성분 및 품질 특성에 미치는 영향)

  • The effects of washing and desinewing on the composition, fuctional properties, storage stability and texture of spent layer meat were investigated. Spent layer meat subjected to treatments involving water washing, bicarbonate washing and desinewing showed increases in moisture content and decreases in protein content, hydroxyproline content and heme pigment content. Increases in salt extractable protein content and water holding capacity and decreases in buffering capacity and emulsifying capacity were also observed. The 2-thiobarbituric acid values of washed and desinewed samples increased slowly during storage indicated the increase in storage stability compared to the control. Sample rolls prepared from bicarbonate washed and desinewed thigh and drumstick meats were scored by trained sensory panelists as less tough than the products made of ground thigh and drumstick meats.

  • To elucidate the effects of calcium carbonate applied under upland and flooded conditions on the changes of soil pH, an incubation study of a soil was carried out. The experiment was conducted under the conditions with and without ground rice straw application. The results of the experiment are summarized as follows: 1. The incubation under the upland moisture condition after the application of calcium carbonate raised soil pH as high as calcium hydroxide did, whereas incubation with calos treatment under the flooded condition showed a low pH. 2. Reduction of a soil brought about by the application of ground rice straw has a significant effect on the rise of soil pH. The high pH of a soil thus brought about or raised by the use of lime is gradually reduced as the accumulation of bicarbonate ions and carbon dioxide increases in the soil solution. It may, however, be possible to keep the high pH of a flooded soil by practicing intermittent drainage or cultivation which reduces the content of bicarbonate and $CO_2$ pressure in soil solution. 3. Carbonate and bicarbonate salts which may be produced in flooded and reduced soil increases the buffering capacity of the soil.

  • Optimization and standardization of solid phase enzyme immunoassay were done for the diagnosis of edwardsiellosis in fish. The analyzed degree of immobilized antibody on surface of solid phase with peroxidase saturation method showed the optimized result by using partially purified $50{\mu}g/ml$ of rabbit anti-E. tarda Edk-2 antibody in sodium bicarbonate buffer for overnight incubation to cover the surface of polystyrene beads. Optimized immunoreaction was observed in the treatment of $50{\mu}g/ml$ of biotin conjugated antibody followed extravidin-peroxidase diluted 1 : 2,000 in PBS. The detectable concentrations of the this method were $1{\times}10^5$ cells/ml and $1{\times}10^5$ cells/ml expressed as the source of antigen amount for EDTA extraction and heat extraction, respectively. High cross-reaction of solid phase ELISA with the prepared rabbit and-E. tarda Edk-2 was observed against E. tarda strains isolated from flounder suffering from edwardsiellosis in aquatic farms of Korea. It suggested that the potential of this solid phase of ELISA technique is very powerful for the application to different strains of E. tarda isolated in farms of many different areas.

Enzymatic Synthesis of Flame Retardant Phenolic Polymers Catalyzed by Horseradish Peroxidase (Horseradish Peroxidase 효소촉매에 의한 난연성 페놀고분자의 합성)

  • The optimum synthetic conditions of poly(p-phenylphenol) by horseradish peroxidase in dioxane:water (80:20 v/v) mixtures were studied. The stability against thermal degradation and structural properties of the synthesized phenolic resins were investigated by thermogravimetric analysis (TGA) and differential scanning calorimetry (DSC), respectively. The synthetic yield of poly(p-phenylphenol) increased upon the increase of the amount of enzyme up to 0.25 mg HRP/mL, then leveled off for further increase of the enzyme usage. When sodium acetate (100 mM, pH 4~6) and sodium phosphate (100 mM, pH 7~9) were used as the buffering salts for the aqueous component (20% v/v), the synthetic yield of the resin increased at higher pH of the aqueous buffer. But when the pHs of the aqueous buffer were 6 and 9, the synthetic yield strongly depended on the types of the buffering salts; if sodium phosphate was used instead of sodium acetate at pH 6, the yield decreased by about 15% and if sodium bicarbonate was used instead of sodium phosphate, the yield decreased by almost 20%. When the pH range of the aqueous buffer was from 4 to 7, the addition of a radical mediator, 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonate) (ABTS), up to 2 mM improved the synthetic yield of the resin by about 10%. TGA experiments revealed that the thermal stability of the resin synthesized in dioxane:water (100 mM sodium phosphate, pH 9) (80:20 v/v) was high having the char yield of 47% upon the heating at $800^{\circ}C$. DCS results showed that the structures of the polymers synthesized in acidic aqueous buffers were different from those of the polymers synthesized in the basic aqueous buffers. However, all the synthesized resins were found to have the property of the thermosetting resins.

  • As for analytes which did not represent the differences of the selectivity on the stationary and mobile phase, secondary chemical equilibrium theory was applied to study pH effects on the separation of alkylphenols. Mobile phase was consisted of an aqueous sodium carbonate-bicarbonate buffer and acetonitrile. The maximum selectivity for adjacent peak pairs was predicted from those values of $k^{\prime}_{HA}/k^{\prime}_{A-}$ and ${\Delta}pK$. The optimum pH determined by this method was 11.18 pH and solvent selectivity were considered at the same time to invoke the full range of selectivity effects possible for separations. Quaternary mixture composed of methanol, acetonitrile, tetrahydrofuran and water was adjusted to optimum pH 11.18. As the statistical simplex technique of an overlapping resolution map (ORM) was used to predict the optimized solvent system. The optimum solvent, which gives complete separation of alkylphenols, was determined as follws MeOH : ACN : THF = 14.4 : 81.8 : 2.8.
