Diversity of Nigrospora (Xylariales, Apiosporaceae) Species Identified in Korean Macroalgae Including Five Unrecorded Species

  • Nigrospora (Xylariales, Apiosporaceae) consists of species of terrestrial plant endophytes and pathogens. Nigrospora has also been reported in marine environments such as mangroves, sea fans, and macroalgae. However, limited research has been conducted on Nigrospora associated with macroalgae. Here, we isolated Nigrospora species from three types of algae (brown, green, and red algae) from Korean islands (Chuja, Jeju, and Ulleung) based on phylogenetic analyses of multigenetic markers: the internal transcribed spacers (ITS), beta-tubulin (BenA), and translation elongation factor 1 (TEF1-α). A total of 17 Nigrospora strains were isolated from macroalgae and identified as nine distinct species. The majority of Nigrospora species (seven) were found on brown algae, followed by red algae (three), and then green algae (two). To our understanding, this study represents the first account of N. cooperae, N. covidalis, N. guilinensis, N. lacticolonia, N. osmanthi, N. pyriformis, and N. rubi occurring in marine environments. Additionally, this study provides the first report of the occurrence of N. cooperae, N. covidalis, N. guilinensis, N. lacticolonia, and N. osmanthi in South Korea. This study will provide valuable insights for future research exploring the functions of fungi in macroalgal communities.

Dietary supplementation of solubles from shredded, steam-exploded pine particles modulates cecal microbiome composition in broiler chickens

  • This study evaluated the effects of supplementing solubles from shredded, steam-exploded pine particles (SSPP) on growth performances, plasma biochemicals, and microbial composition in broilers. The birds were reared for 28 days and fed basal diets with or without the inclusion of SSPP from 8 days old. There were a total of three dietary treatments supplemented with 0% (0% SSPP), 0.1% (0.1% SSPP) and 0.4% (0.4% SSPP) SSPP in basal diets. Supplementation of SSPP did not significantly affect growth or plasma biochemicals, but there was a clear indication of diet-induced microbial shifts. Beta-diversity analysis revealed SSPP supplementation-related clustering (ANOSIM: r = 0.31, p < 0.01), with an overall lower (PERMDISP: p < 0.05) individual dispersion in comparison to the control group. In addition, the proportions of the Bacteroides were increased, and the relative abundances of the families Vallitaleaceae, Defluviitaleaceae, Clostridiaceae, and the genera Butyricicoccus and Anaerofilum (p < 0.05) were significantly higher in the 0.4% SSPP group than in the control group. Furthermore, the linear discriminant analysis effect size (LEfSe) also showed that beneficial bacteria such as Ruminococcus albus and Butyricicoccus pullicaecorum were identified as microbial biomarkers of dietary SSPP inclusion (p < 0.05; | LDA effect size | > 2.0). Finally, network analysis showed that strong positive correlations were established among microbial species belonging to the class Clostridia, whereas Erysipelotrichia and Bacteroidia were mostly negatively correlated with Clostridia. Taken together, the results suggested that SSPP supplementation modulates the cecal microbial composition of broilers toward a "healthier" profile.

Identification of Host-Resistant and Susceptible Varieties of Korean Grapes to Plasmopara viticola, a Pathogen Causing Grapevine Downy Mildew

  • Grapevine downy mildew, caused by Plasmopara viticola, significantly damages vineyards and is one of the most devastating diseases affecting cultivated grapes worldwide. In this study, we characterized the phenotypic and molecular traits of 11 P. viticola isolates from four grape-growing regions in South Korea. Additionally, we investigated the diversity of pathogenicity among these isolates and conducted an assay to evaluate the response of grape cultivars to P. viticola infection. Lemon-shaped sporangia were identified in the collected isolates, which released zoospores into the suspension at room temperature. Within a few hours of inoculation, the zoospores developed germ tubes. We tested 11 P. viticola isolates for pathogenicity in 845 grape cultivars to screen for grape host resistance to downy mildew infection. Among the tested isolates, JN-9 showed the highest virulence. Grape cultivars displayed varying phenotypic reactions to P. viticola infection: approximately 7% were highly susceptible, 41% were susceptible, 20% were moderately susceptible, 8% were resistant, and 24% exhibited extreme resistance. Phylogenetic analysis based on four genomic regions (internal transcribed spacer 1 [ITS1], actin, beta-tubulin, and cytochrome c oxidase II) revealed a close evolutionary relationship among all the Korean isolates, forming a single monophyletic lineage. Notably, these isolates showed greater similarity to European isolates than to American isolates. This comprehensive study contributes to a deeper understanding of the identity and behavior of P. viticola, which is crucial for developing effective resistance strategies against this pathogen in grape cultivars cultivated in South Korea.

Alterations in immunized antigens of Anisakis pegreffii by ampicillin-induced gut microbiome changes in mice

  • The gut microbiome plays an essential role in host immune responses, including allergic reactions. However, commensal gut microbiota is extremely sensitive to antibiotics and excessive usage can cause microbial dysbiosis. Herein, we investigated how changes in the gut microbiome induced by ampicillin affected the production of IgG1 and IgG2a antibodies in mice subsequently exposed to Anisakis pegreffii antigens. Ampicillin treatment caused a notable change in the gut microbiome as shown by changes in both alpha and beta diversity indexes. In a 1-dimensional immunoblot using Anisakis-specific anti-mouse IgG1, a 56-kDa band corresponding to an unnamed Anisakis protein was detected using mass spectrometry analysis only in ampicillin-treated mice. In the Anisakis-specific anti-mouse IgG2a-probed immunoblot, a 70-kDa band corresponding to heat shock protein 70 (HSP70) was only detected in ampicillin-treated and Anisakis-immunized mice. A 2-dimensional immunoblot against Anisakis extract with immunized mouse sera demonstrated altered spot patterns in both groups. Our results showed that ampicillin treatment altered the gut microbiome composition in mice, changing the immunization response to antigens from A. pegreffii. This research could serve as a basis for developing vaccines or allergy immunotherapies against parasitic infections.

복합미생물 생물증강법을 이용한 인공해수하천의 친환경 효율적 현장 수질정화 (Eco-friendly and efficient in situ restoration of the constructed sea stream by bioaugmentation of a microbial consortium)

  • 부산시 영도구의 혁신지구의 인공해수천은 높아진 하상과 조류의 특성으로 인해 물이 순환되지 않고 더구나 주위의 오수가 유입되고 있어서 수질이 나빠지고 악취를 발생하고 있다. 이 문제를 해결하기 위한 방안으로 가장 오염되고 조류이동을 감안한 하천의 지점에 생물증강법을 적용하여 친환경적, 효율적으로 하천을 정화하고자 하였다. 현장에서 활성화된 복합미생물을 가장 오염된 지점(Site 2)에 7~10일 간격으로 투입하여, 수질의 pH, 온도, DO, ORP, SS, COD, T-N, 및 T-P를 측정하였고 또한 하상퇴적토의 COD 및 미생물군집을 분석하였다. 13개월 후 Site 2의 수질 SS, COD, T-N 및 COD (퇴적토)의 처리효율은 각각 51%, 58%, 27% 및 35%으로 나타났으나 T-P는 오히려 47% 증가를 보였다. 대부분의 측정지점에서 황산염환원세균(sulfate reducing bacteria)그룹 (Desulfobacteraceae_uc_s, Desulfobacterales_uc_s, Desulfuromonadaceae_uc_s, Desulfuromonas_g1_uc and Desulfobacter postgatei)과 Anaerolinaeles의 밀도는 대체적으로 생물증강에 의한 정화가 진행될수록 감소하였으며, 반면에 Gamma-proteobacteria (NOR5-6B_s and NOR5-6A_s), Bacteroidales_uc_s, 및 Flavobacteriales_uc_s의 밀도는 증가하는 경향이었다. 상대적으로 COD가 낮고 DO가 높은 청정지점인 St. 4에서는 호기성미생물인 Flavobacteriaceae_uc_s가 우점하였다. 이러한 미생물군은 하천의 정화과정을 추적할 수 있는 지표미생물로 활용될 수 있을 것으로 판단되었다. 생물증강 시행 후의 대표적 시점 퇴적토시료의 미생물군집 alpha diversity 지수(OTUs, Chao1 및 Shannon 지수)는 시행 전에 비해 감소하는 경향을 보였으며, 또한 beta diversity 분석기법(fast unifrac 분석)으로 분석한 결과 정화 전이나 초기에 비해서 정화가 진행될수록 전반적으로 시료에 무관하게 미생물군집의 유사성을 보여 생물증강이 현장 토착 미생물의 군집구조를 변화시키고 있음을 확인하였다. 이러한 사실로 보아 본 복합미생물에 의한 현장 생물증강법은 brine water 및 담수로 이루어진 오염된 하천을 환경친화적, 경제적으로 정화할 수 있는 대안으로 판단이 되었다.

토양효소활성을 이용한 미생물제제 처리 고추경작지의 토양미생물군집 분석 (Soil Microbial Community Analysis using Soil Enzyme Activities in Red Pepper Field Treated Microbial Agents)

  • 친환경 농법이 대두되면서 식물 병원균에 대한 길항능과 식물생장 촉진능을 동시에 가지는 plant growth promoting rhizobacterium (PGRP)을 이용해 식물병을 방제하는 미생물 농법이 선호되고 있다. 본 연구에서는 토양비옥도 지표효소로 알려진 5종의 토양효소활성 측정을 이용해 plant growth promoting rhizobacterium 균주인 Bacillus subtilis AH18, Bacillus licheniformis K11, Pseudomonas fluorescens 2112를 조합한 복합미생물제제와 시판중인 미생물농약, 화학농약을 처리한 고추경작지에서 토양미생물상을 분석하고 고추의 생장 및 수확량을 측정하여 메타지노믹스를 이용한 토양미생물 다양성 연구의 기초자료로 사용하고자 하였다. 토양효소활성의 측정에서 복합미생물제제 처리구가 dehydrogenase 3.5584 ${\mu}g$ TPF $g^{-1}h^{-1}$, urease 15.8689 ${\mu}g$ $NH_4{^-}N$ $g^{-1}h^{-1}$, phosphatase 0.5692 ${\mu}g$ PNP $g^{-1}h^{-1}$, ${\beta}$-glucosidase 2.4785 ${\mu}g$ PNP $g^{-1}h^{-1}$, cellulase 86.1597 ${\mu}g$ glucose $g^{-1}h^{-1}$의 수치를 나타내 타처리구보다 높은 활성을 보여 토양미생물상의 다양성이 증대됨을 확인하였다. 또한, 고추의 생장촉진도측정에서 복합미생물제제가 타처리구에 비해 주경장에서 최대 6.1%, 경경에서 최대 8.1%의 생장촉진능을 보여 복합미생물제제의 생장촉진능을 확인하였다. 생고추의 수확량 측정에서는 복합미생물제제가 무처리구를 기준으로 했을 때 14%의 수확량 증대효과를 나타내었고, 화학농약 처리구보다도 7.3%의 증대효과를 나타내어 복합미생물제제에 의한 수확량 증대효과도 확인하였다. 따라서 본 연구에 사용된 복합 미생물컨소시움제제가 고추경작지 토양의 미생물상 다양성 증가와 고추의 생장촉진 및 수확량 향상 모두에 기여함을 알 수 있었다.

Allele Frequencies of the Single Nucleotide Polymorphisms Related to the Body Burden of Heavy Metals in the Korean Population and Their Ethnic Differences

  • This study was performed to select single nucleotide polymorphisms (SNPs) related to the body burden of heavy metals in Koreans, to provide Korean allele frequencies of selected SNPs, and to assess the difference in allele frequencies with other ethnicities. The candidate-gene approach method and genome-wide association screening were used to select SNPs related to the body burden of heavy metals. Genotyping analysis of the final 192 SNPs selected was performed on 1,483 subjects using the VeraCode Goldengate assay. Allele frequencies differences and genetic differentiations between the Korean population and Chinese (CHB), Japanese (JPT), Caucasian (CEU), and African (YIR) populations were tested by Fisher's exact test and fixation index ($F_{ST}$), respectively. The Korean population was genetically similar to the CHB and JPT populations ($F_{ST}$ < 0.05, for all SNPs in both populations). However, a significant difference in the allele frequencies between the Korean and CEU and YIR populations were observed in 99 SNPs (60.7%) and 120 SNPs (73.6%), respectively. Ten (6.1%) and 26 (16.0%) SNPs had genetic differentiation ($F_{ST}$ > 0.05) among the Korean-CEU and Korean-YIR comparisons, respectively. The SNP with the largest $F_{ST}$ value between the Korean and African populations was cystathionine-${\beta}$-synthase rs234709 ($F_{ST}$: KOR-YIR, 0.309; KOR-CEU, 0.064). Our study suggests that interethnic differences exist in SNPs associated with heavy metals of Koreans, and it should be considered in future studies that address ethnic differences in heavy-metal concentrations in the body and genetic susceptibility to the body burden of heavy metals.

Nutritional aspects of night eating and its association with weight status among Korean adolescents

  • BACKGROUND/OBJECTIVES: A growing body of research has indicated that night eating could be associated with poor diet quality and negative health outcomes. This study examined the nutritional aspects of night eating, its related factors, and the association between night eating and body weight among Korean adolescents. SUBJECTS/METHODS: This study analysed the data from a one day 24-hour dietary recall as well as a demographic survey of 1,738 Korean adolescents aged 12 to 18-years-old obtained from the 2010-2012 Korea National Health and Nutrition Examination Survey. 'Night eating' was defined as consuming 25% or more of one's daily energy intake between 21:00 and 06:00. Subjects complying with the preceding condition were classified as 'night eaters', whereas the rest were considered 'non-night eaters'. Logistic regression analysis examined factors related to night eating. Multiple linear regression analyses were used to examine the relationship between night eating and BMI z-scores, whereas multinomial logistic regression analysis was used to examine the relationship between night eating and weight status. RESULTS: About 21% of Korean adolescents appeared to be night eaters. Night eaters showed increased breakfast skipping (P = 0.001), higher energy intake from snacks (P < 0.001), greater proportion of energy intake from fat (P = 0.029), and lower Dietary Diversity Scores (P = 0.008) than non-night eaters. Male adolescents presented 1.9 times higher odds of being night eaters than females. Adolescents whose both parents were night eaters were 4.4 times as likely to be night eaters as those whose neither parents were. Female adolescents showed a significant relationship between night eating and BMI z-scores (${\beta}=0.28$, P = 0.004). However, night eating did not increase odds of being overweight or obese in adolescents. CONCLUSIONS: Night eating in Korean adolescents was related to undesirable dietary behaviours and low diet quality in general as well as higher BMI z-scores in females. Male gender and parental night eating appeared to be the factors that significantly increased odds of night eating. These results suggest that night eating should be considered when designing nutrition education or intervention programs targeting adolescents.

작약 연작재배지의 토양 이화학성 및 미생물 분포특성 (Soil Physico-Chemical Properties and Characteristics of Microbial Distribution in the Continuous Cropped Field with Paeonia lactiflora)

  • 작약 연작장해 경감대책을 마련하기 위하여 연작재배지의 토양 이화학성과 미생물상 변화 및 토양 미생물 군락의 기질 이용성을 조사하였다. 작약 초작지 토양에 비해 연작지 토양에서 표토 깊이가 얕고 표토와 심토의 경도차이가 심하였다. 토양화학성에서는 초작 2년에 비해 연작 4년생 토양이 pH 7.6, 유기물함량 $44.1g\;kg^{-1}$, 치환성 Ca, Mg함량이 높았다. 작약 재배연수에 따른 미생물의 변화는 세균과 방선균 밀도는 초작 3년까지 증가하다가 그 이후 감소하였고 사상균 밀도는 재배년수에 따라 크게 증가하였다. Biolog EcoPlate에 의한 토양 미생물 군락의 탄소기질 이용성 차이는 제 3주성분까지 57.5%의 자료를 해석할 수 있었고, 주성분에 영향을 미치는 탄소기질은 D-cellobiose, ${\beta}$-methyl-D-glucoside 등이었으며, 작약 재배기간이 길어짐에 따라 뚜렷하게 구분되었다.

Analysis of 16S rRNA gene sequencing data for the taxonomic characterization of the vaginal and the fecal microbial communities in Hanwoo

  • Objective: The study of Hanwoo (Korean native cattle) has mainly been focused on meat quality and productivity. Recently the field of microbiome research has increased dramatically. However, the information on the microbiome in Hanwoo is still insufficient, especially relationship between vagina and feces. Therefore, the purpose of this study is to examine the microbial community characteristics by analyzing the 16S rRNA sequencing data of Hanwoo vagina and feces, as well as to confirm the difference and correlation between vaginal and fecal microorganisms. As a result, the goal is to investigate if fecal microbiome can be used to predict vaginal microbiome. Methods: A total of 31 clinically healthy Hanwoo that delivered healthy calves more than once in Cheongju, South Korea were enrolled in this study. During the breeding season, we collected vaginal and fecal samples and sequenced the microbial 16S rRNA genes V3-V4 hypervariable regions from microbial DNA of samples. Results: The results revealed that the phylum-level microorganisms with the largest relative distribution were Firmicutes, Actinobacteria, Bacteroidetes, and Proteobacteria in the vagina, and Firmicutes, Bacteroidetes, and Spirochaetes in the feces, respectively. In the analysis of alpha, beta diversity, and effect size measurements (LefSe), the results showed significant differences between the vaginal and fecal samples. We also identified the function of these differentially abundant microorganisms by functional annotation analyses. But there is no significant correlation between vaginal and fecal microbiome. Conclusion: There is a significant difference between vaginal and fecal microbiome, but no significant correlation. Therefore, it is difficult to interrelate vaginal microbiome as fecal microbiome in Hanwoo. In a further study, it will be necessary to identify the genetic relationship of the entire microorganism between vagina and feces through the whole metagenome sequencing analysis and meta-transcriptome analysis to figure out their relationship.
