Enhancement of Antioxidant and Anti-aging Activities of Spirulina Extracts by Fermentation (스피루리나 발효에 의한 항산화력 증진 및 항노화 효과)

  • It is known that Spirulina extracts have strong antioxidant activities since it contains diverse antioxidants such as phycocyanian, ${\beta}$-carotene, vitamin E and other carotenoids. In order to enhance antioxidant activity of Spirulina, Spirulina extracts were fermented by Lactobacillus plantarum P23 and Bacillus subtilis TP6. The resulting fermented supernatants were analyzed for their antioxidant activities by DPPH (1,1-diphenyl-2-picrylhydiazyl) method. The results indicated that fermentation process significantly enhanced total antioxidant activities. Increased levels of UV-induced TNF-${\alpha}$ and IL-6 were reduced back to normal level even by treatment of all three of the Spirulina extracts. The result suggested that the fermentation process enhanced the anti-inflammatory activities at least ten times higher than the simple extract. Zymography is used to determine the expression of UV-induced MMP. Spirulina extracts fermented by Bacillus subtilis TP6 were found to suppressed the expression of MMPs. Also treatment with the fermented Spirulina extracts resulted in an increase of collagen synthesis in vitro. In conclusion, the fermented Spirulina extracts are expected to be used as anti-aging cosmeceuticals.

Characteristics of agronomy to vitamin A strengthening rice at large scale GMO field (대규모 GMO 포장에서 비타민 A 강화 벼의 농업특성 검정)

  • This study was carried out to develop of macro-protocol and the biosafety guide Golden Rice, modified vitamin A at large scale GMO field. Commercialization of genetically modified (GM) plants will be required the assessment of risks associated with the release of GM plants that should include a detailed risk assessment of their impacts in the environment and human health. Prior to GM plant release, applicants should provide the information on GM crops for approval. A total of $4,700m^2$ GM field, each of Golden Rice and 'Nakdong' were used for further analysis on agronomic traits and pathogenesis. Generally plant growth and panicle number in the transgenic lines were lower than in the wild type under field condition. The major agronomic traits such as plant growth, grain quality in Golden Rice were similar than in 'Nakdong'. The yield difference was due to reduced ripened rate and the 1,000 grain weight. The grain shape was similar donor plant, however, color of brown rice was different from segregation. Also the difference of pathogenesis reaction was not significant in the leaf blast and sheath blight between Golden Rice and 'Nakdong'. These results provided the agronomic data and pathogenesis for risk assessment analysis of Golden Rice and suggested that the macro-protocol could be useful to detect GM plants.

Studies on Insect Diversity Related to Genetically Engineered Vitamin A Rice under Large Scale Production (비타민 A 강화 벼의 대규모 GMO 포장에서 곤충다양성 분석)

  • This study was carried out to develop of macro-protocol and the biosafety guide for 'Nakdong', and 'Golden Rice' (genetically modified vitamin A rice) by large scale field trial of GM crops. Typically, when a new GM crop is created, breeders should conduct field test to make sure the GM crop is safe, and provide some information on GM crops for approval. A total of isolated 4,700 $m^2$ field for trial of GM crops were prepared, and 'Nakdong' and 'Golden rice' were cultivated by standard method of RDA (Rural Development Administration, Korea). Field studies indicated that the population densities of insect pests and natural enemies have no difference between two varieties. While insect pest density on 'Nakdong' was slightly higher than on Golden Rice, but natural enemy density on Golden Rice was a little higher. These results provided the insect diversity for risk assessment analysis of Golden Rice and suggested that the macro-protocol could be useful to detect GM plants.

A study on the eating behaviors and food intake of diabetic patients in Daegu·Gyeongbuk area (대구·경북지역 당뇨 환자의 식행동 및 식품 섭취에 관한 조사 연구)

  • Rapid economic development has changed the dietary habits and patterns. Especially, western style diet has increased the risk of type 2 diabetes (T2DM) in Korea. To provide more specific and appropriate diet guideline for the prevention and for the treatment of T2DM, the investigation on the characteristics of diebetic patients related to the diet needs to be done. In this study, we therefore analyzed eating behaviors, dietary attitudes, and intake of food and nutrients of normal subjects (control, n=26) and diabetic patients (case, n=18) diagnosed T2DM within one year in Daegu Gyeongbuk area. Body mass index of the patients were significantly higher than the control (p<0.05). Overeating, high fried food and low whole grain intake were revealed as risk factors for T2DM. From the food frequency questionnaire analysis, salty foods such as fermented soybean paste (Doenjang) and watery Kimch intake were associated with T2DM. Intake of vegetable lipid, ${\beta}$-carotene, calcium, copper, and vitamin K were also shown to be associated with T2DM. Taken together, these findings suggest that maintaining ideal body weight and intake the foods with low salt, fat, and refined grain in appropriate amount may help to prevent and to manage T2DM.

Quality Changes in Tomato Fruits Caused by Genotype and Environment Interactions (재배환경과 유전형의 상호작용에 따른 토마토 과실 품질 변화)

  • Bred and grown around the world, tomato (Solanum spp.) has highly valuable fruits containings various anti-oxidants such as lycopene, flavonoids, glutamine, and ${\beta}-carotene$. Several studies have explored, way in which to enhance the growth, management and quality of tomato, we focus on the management of growth for yield rather than quality. The expression of superior agronomic traits depends on where cultivars are grown. We evaluated 10 cultivars grown in three environment for their lycopene. HTL3137 ($70.48mg{\cdot}kg^{-1}$), which was grown in Yoeju in spring/summer, contained the highest lycopene content, while HTL10256 ($20.9mg{\cdot}kg^{-1}$), which was grown in Suwon in spring/summer, contain the least lycopene.Correlations between color components and lycopene content varied according to growing location and season. In spring/summer-grown tomatoes from Suwon, no significant correlation was observed between any color component (redness [R], greenness [G], blueness [B], luminosity, $L^*$, $a^*$, $b^*$, hue and chroma) and lycopene content. A correlation was observed between B and lycopene content in tomatoes grown in Yeoju during the same season. In tomatoes grown in Yeoju in fall/winter, significant correlations were found between lycopene content and G, luminosity, $L^*$, and hue. Variance in interactions between genotype, environment, and genotype ${\times}$ environment (G ${\times}$ E) using Minimum Norm Quadratic Unbiased Estimate (MINQUE) analysis indicated that lycopene content depends on genotype (51.33%), environment (49.13%), and G ${\times}$ E (21.43%). However, when the Additive Main Effects and Multiplicative Interaction (AMMI) was used, the G ${\times}$ E value was highest.

Studies on Organic Acid, Vitamin and Free Sugar Contents of Commercial Dried Lavers (Porphyra yezoensis) Cultivated in Korea (시판 건조김의 유기산, 비타민 및 유리당 함량)

  • The purpose of this study was to provide basic date for organic acid, vitamin and free sugar contents of commercial dried lavers cultivated in Korea (Seocheon, Wando, Goheung and Busan). The contents of organic acid per 100 g of dried lavers were in the range of 1,238.67~1,437.00 mg of malic acid, 1,021.67~1,309.67 mg of oxalic acid, 1,035.67~1,247.33 mg of citric acid, 1,001.00~1,146.00 mg of succinic acid, 902.00~1,107.00 mg of tartaric acid, 862.67~1,070.67 mg of lactic acid and 370.33~421.67 mg of acetic acid. The concentration of vitamin A and ${\beta}$-carotene were the highest (0.286 mg and 22.19 mg/100 g) in dried lavers cultivated from Seocheon and Wando. Vitamin $B_1$, $B_3$ and $B_{12}$ concentrations were the highest (9.79 mg, 19.15 mg and 1.77 mg/100 g) in dried lavers cultivated from Goheung, and vitamin $B_2$, $B_6$ and $B_9$ concentrations were the highest (4.55 mg, 2.36 mg and 2.17 mg/100 g) in dried lavers cultivated from Wando. The concentration of vitamin C and E contents were the highest (219.33 mg and 1.75 mg/100 g) in dried lavers cultivated from Wando. The contents of free sugar per 100 g of dried lavers were in the range of 214.27~347.72 mg of galactose, 59.10~71.69 mg of glucose, 37.86~54.34 mg of fucose and 6.26~14.65 mg of mannose.

  • Statistical models of shape variability based on active shape models (ASMs) have been successfully utilized to perform segmentation and recognition tasks in two-dimensional (2D) images. Three-dimensional (3D) model-based approaches are more promising than 2D approaches since they can bring in more realistic shape constraints for recognizing and delineating the object boundary. For 3D model-based approaches, however, building the 3D shape model from a training set of segmented instances of an object is a major challenge and currently it remains an open problem in building the 3D shape model, one essential step is to generate a point distribution model (PDM). Corresponding landmarks must be selected in all1 training shapes for generating PDM, and manual determination of landmark correspondences is very time-consuming, tedious, and error-prone. In this paper, we propose a novel automatic method for generating 3D statistical shape models. Given a set of training 3D shapes, we generate a 3D model by 1) building the mean shape fro]n the distance transform of the training shapes, 2) utilizing a tetrahedron method for automatically selecting landmarks on the mean shape, and 3) subsequently propagating these landmarks to each training shape via a distance labeling method. In this paper, we investigate the accuracy and compactness of the 3D model for the human liver built from 50 segmented individual CT data sets. The proposed method is very general without such assumptions and can be applied to other data sets.

Effect of Wine Yeast, Temperature and Moisture Contents on Characteristics of Jeung-Pyun Batter (Wine Yeast와 온도 및 수분함량이 증편 반죽의 특성에 미치는 영향)

  • In this study, the characteristics of Jeung-pyun hatter were investigated by wine yeast. The processing conditions were optimized by physicochemical characterization including pH, volume, reduced sugar. The effect of yeast concentration, moisture content on the fermentation time and temperature were investigated in view of improving productivity. It was found that the volume was increased at maximum state when the fermentation was carried out at 35 $^{\circ}C$ with 0.1% yeast concentration 60% of moisture. The quality of Jeung-Pyun was most preferable in the condition of 0.1% wine yeast(Pasteur Red) for 8 hrs at 35$^{\circ}C$.

Comparison of Phytochemical and Antioxidant Activities in Different Color Stages and Varieties of Paprika Harvested in Korea (국내산 파프리카의 색상별 phytochemical 및 항산화능 비교)

  • This study qualitatively and quantitatively analyzed various carotenoids, L-ascorbic acid, and tocopherol and their anti-oxidant properties from four varieties (Special, Chelsea, Cupra, and Fiesta) and three different color stages [green (GP), yellow (YP) and red (RP)] of paprika harvested in Korea. Seven carotenoids were identified, mainly lutein in GP (Special: 4.65${\pm}$0.84 mg/kg fresh weight (fw)) and YP (Fiesta: 5.19${\pm}$0.05 mg/kg fw), and capsorubin (3.16${\pm}$0.35 mg/kg fw) and capsanthin (53.70${\pm}$6.23 mg/kg fw) in Special of RP. RP was the highest in total carotenoids, L-ascorbic acid, and ${\alpha}$-tocopherol contents, while GP was the highest in ${\gamma}$-tocopherol content. RP showed the strongest antioxidant activity ($IC_{50}$=62.40${\pm}$0.03 ${\mu}g/mL$ in an ABTS assay and 182.77${\pm}$31.74 ${\mu}g/mL$ in a DPPH assay). Paprika in different color stages has many phytochemicals even though they have different kinds of carotenoids. Therefore, dietary intake of paprika may be helpful for improving human health.

Effects of Gardenia jasminoides Ellis Peel Extract in Namhae Korea on the Bioactivity Compounds and Lipid Peroxidation Inhibition Activity (남해산 치자(Gardenia jasminoides Ellis fructus) 껍질의 생리활성 및 지질과산화 저해 활성에 미치는 영향)

  • The object of this study was to measure the bioactivity and lipid peroxidation inhibition activity of peel from Gardenia jasminoides Ellis fructus (GJE) in Namhae Korea. The amount of phytic acid was also determined. Extraction was performed using three solvents, CM (choloform:methanol, 2:1, v/v), n-butanol and 70% ethanol. To investigate by the solvent extract of total phenol content and value as a functional food ingredient of GJE peel through nitrogen oxide scavenging activity, antioxidant activity, reducing power and lipid peroxidation inhibition were performed. The bioactivities of the extract solvents increased significantly with increasing concentrations (0.2, 0.4, 0.6 mg/mL, p<0.05). The total phenol contents of GJE peel extracts were highest in CM ($39.74{\pm}0.15mg\;CAE/g$) extract. The order of total phenol contents, antioxidant activity and reducing power of the solvents in the GJE peel were the same, in the analysis of nitrogen oxides scavenging activity and lipid peroxidation inhibition, it was confirmed the results were inconsistent. As a result, the GJE peel showed excellent bioactivities. Considering the extraction yield and various physiological activities, it is considered that efficiency is better when extracted from CM and 70% ethanol extracts.
