사방오리(Alnus firma) 추출물의 in vitro 항산화 활성 (In Vitro Antioxidant Activity of Alnus firma Extracts)

  • 본 연구는 사방오리 추출 분획물의 항산화 활성을 검증하기 위하여 시행되었다. 사방오리는 메탄올(MeOH) 분획에 가장 높은 총 페놀화합물($452.80{\pm}7.01{\mu}g$ gallic acid equivalents/mg)을 그리고 에틸에세테이트(EA) 분획 에 가장 높은 총 플라보노이드 함량($112.29{\pm}11.14{\mu}g$ rutin equivalents/mg)과 가장 높은 총 항산화 활성($936.23{\pm}0.07{\mu}g$ ${\alpha}$-tocopherol equivalents/mg)을 가지고 있었다. 사방오리의 용매 추출분획물에 대한 항산화 활성은 아스코빅산 등 표준 물질과 비교하여 1,1-diphenyl-2-picrylhydrazyl (DPPH) 라디칼 소거능 측정, 슈퍼옥사이드 음이온 라디칼 소거능 측정, ${\beta}$-carotene-linoleate 에세이, 환원력 측정, ferric thiocyanate법으로 지질과산화 억제력 측정 그리고 금속 킬레이팅능 측정 등 다양한 항산화 활성을 측정하였다. $250{\mu}g/ml$ 농도의 사방오리 EA와 MeOH 분획은 DPPH 라디칼 소거능 측정에서 각각 92.43와 89.20% 항산화 활성을 가지고 있었다. $50{\mu}g/ml$의 EA분획은 72.49% 슈퍼옥사이드 음이온 라디칼 소거능을 보였으며 이는 gallic acid의 소거능(60.88%)보다 조금 높았다. 또한 ${\beta}$-carotene-linoleic acid 시스템에 의한 과산화 억제능 조사에서 0.5과 1 mg/ml 농도의 EA 분획이 각각 73.45와 73.29% 억제능을 보였다. 항산화 활성은 EA 분획 > BuOH 분획 > DCM분획 > HX분획 순 이였다. 이상의 본 연구 결과에서 사방오리는 천연 항산화제제 개발을 위한 용이하게 접근 가능한 잠재적인 자원으로 판단된다.

β-Carotene Production from Dunaliella salina Cultivated with Bicarbonate as Carbon Source

  • Bicarbonate has been considered as a better approach for supplying CO2 to microalgae cells microenvironments than gas bubbling owing to cost-effectiveness and easy operation. However, the β-carotene production was too low in Dunaliella salina cultivated with bicarbonate in previous studies. Also, the difference in photosynthetic efficiency between these two carbon sources (bicarbonate and CO2) has seldom been discussed. In this study, the culture conditions, including NaHCO3, Ca2+, Mg2+ and microelement concentrations, were optimized when bicarbonate was used as carbon source. Under optimized condition, a maximum biomass concentration of 0.71 g/l and corresponding β-carotene content of 4.76% were obtained, with β-carotene yield of 32.0 mg/l, much higher than previous studies with NaHCO3. Finally, these optimized conditions with bicarbonate were compared with CO2 bubbling by online monitoring. There was a notable difference in Fv/Fm value between cultivations with bicarbonate and CO2, but there was no difference in the Fv/Fm periodic changing patterns. This indicates that the high concentration of NaHCO3 used in this study served as a stress factor for β-carotene accumulation, although high productivity of biomass was still obtained.

Comparison of Forage Quality, Productivity and β-carotene Content according to Maturity of Forage Rye (Secale cereale L.)

  • These experiments were to investigate the variations of rye on forage quality, productivity and β-carotene concentration affected by maturity in Pyeongchang region. Limited information are available about how forage quality and β-carotene content are affected by various factors. Samples were collected from rye harvested every 5 days, from April 25 to May 31 (April 25, April 30, May 4, May 9, May 15, May 21, May 25 and May 31). Dry matter (DM) content, plant height, DM yield and total digestible nutrient (TDN) yield increased continuously with the progressed maturity. However, crude protein (CP) content, in vitro dry matter digestibility (IVDMD) and relative feed value (RFV) decreased markedly with the delay of harvesting, while TDN content decreased from April 25 till May 15, then followed by a stable fluctuation. Conversely, acid detergent fiber (ADF) and neutral detergent fiber (NDF) value increased and then fluctuated slightly after blooming stage. For quality of plant parts, stem contained the lowest CP content and RFV value, and the highest ADF and NDF contents compared with other parts, while the grain showed the higher CP, IVDMD, RFV and lower fiber contents than others. With the plant matured, leaf proportion decreased while stem and grain proportion increased, and feed value of all the three parts decreased till blooming stage and followed by a stable phase. β-carotene concentration showed its highest on jointing stage, and then fell down sharply on the sequential stages. In conclusion, harvest around May 15 (blooming) is proper for forage rye if directly consumed by livestock as green chop in Pyeongchang under the consideration of both nutritive yield and forage quality.

Effect of harvest dates on β-carotene content and forage quality of rye (Secale cereale L.) silage and hay

  • Limited data about the effects of various factors on forage quality and β-carotene content of rye produced in Korea are available, so this study investigated the effects of two preservation methods. Samples were collected from rye harvested every 5 days between April 25 and May 31, and comparisons were done among rye silage wilted for different periods of time and hay of three growth stages of rye. For the silage, dry matter (DM), acid detergent fiber (ADF), and neutral detergent fiber (NDF) contents increased with advanced maturity of rye, whereas crude protein, in vitro dry matter digestibility (IVDMD), total digestible nutrients (TDN), relative feed value (RFV), and DM loss decreased (p < 0.0001). Wilting increased the DM content and pH value significantly (p < 0.0001). Silage harvested at the heading stage had the lowest pH value (4.45), propionic acid (0.83 g/kg DM), butyric acid (0 g/kg DM), and fungi and yeast populations (3.70 Log CFU/g of fresh matter [FM]); conversely, it had the highest lactic acid (9.7 g/kg DM), lactic acid bacteria (LAB) (6.87 Log CFU/g of FM), total microorganisms (TM) (7.33 Log CFU/g of FM), and Flieg's score (70) (p < 0.0001). Wilting elevated LAB and TM populations, but it had no consistent effect on other fermentation products. Both delayed harvest and prolonged wilting decreased β-carotene content. Rye silage harvested around May 9 (heading stage) with 24 h of wilting was preferred for highland, Pyeongchang. For rye hay, advanced maturity decreased DM loss, IVDMD, TDN, and RFV, but it increased DM, ADF, and NDF significantly (p < 0.05). β-carotene was decreased by delay of hay-making. Consequently, to attain lower DM loss and higher hay quality, the harvest date of May 9 (heading stage) is recommended.

Identification and characterization of Dunaliella salina OH214 strain newly isolated from a saltpan in Korea

  • Carotenoids are effective antioxidants that are found in various photosynthetic organisms. Marine microalgae are an advantageous bioresource for carotenoid production because they do not compete with other crops for freshwater and arable land. This study reports a newly isolated Dunaliella strain from the Geumhong Saltpan on Yeongjong Island, West Sea, Korea. The new strain was isolated and classified as Dunaliella salina through phylogenetic analysis and was named the OH214 strain (Deposit ID: KCTC14434BP). The newly isolated strain can survive in a wide range of NaCl concentrations (0.3-5.0 M NaCl), but grows well in 0.6 to 1.5 M NaCl culture medium. Under high-light conditions (500 ± 10 μmol photons m-2 s-1), the cells accumulated three times more β-carotene than under low-light conditions (50 ± 5 μmol photons m-2 s-1). The cells accumulated 2.5-fold more β-carotene under nitrogen-deficient (1 mM KNO3) conditions (3.24 ± 0.36 ㎍ 106 cells-1) than in nitrogen-sufficient conditions (>5 mM KNO3). The lutein content under nitrogen-deficient conditions (1.73 ± 0.09 ㎍ 106 cells-1) was more than 24% higher than that under nitrogen-sufficient conditions. Under the optimized culture condition for carotenoid induction using natural seawater, D. salina OH214 strain produced 7.97 ± 0.09 mg g DCW-1 of β-carotene and 4.65 ± 0.18 mg g DCW-1 of lutein, respectively. We propose that this new microalga is a promising strain for the simultaneous production of β-carotene and lutein.

유전자변형 베타-카로틴 강화 쌀의 주요 영양성분 분석 (Analyses of Nutrient Composition in Genetically Modified β-Carotene Biofortified Rice)

  • 본 연구는 국내에서 개발된 베타-카로틴 강화 유전자변형 벼(GM 벼)와 그 모종 벼인 낙동벼(non-GM 벼)로부터 쌀(현미, 백미)의 일반성분, 베타-카로틴, 지방산, 아미노산, 무기질 함량을 분석하여 유전자변형 쌀에서 주요 영양성분 조성에 차이가 있는지를 비교하기 위해 수행되었다. GM 쌀은 non-GM 모종 쌀과 일반성분 함량이 유사하였고 GM 쌀 100 g당 약 0.2 mg의 베타-카로틴을 함유하는 것으로 분석되었다. GM 쌀의 지방산 조성은 oleic acid, linoleic acid, palmitic acid가 지방산 전체의 94% 이상으로 거의 대부분을 차지하고 있었으며 non-GM 쌀과 전체적인 지방산 조성이 유사한 것으로 나타났다. GM 쌀의 아미노산은 Asp>Arg>Leu>Ala>Ser>Val 순으로 그 함량이 높았고 무기질 함량은 P>K>Mg>Na>Ca>Zn>Fe의 순이었으며, non-GM 쌀에 비해 대부분의 아미노산과 무기질 함량이 약간 높은 수치로 측정되었지만 유의적인 차이는 없는 것으로 평가되었다. 베타-카로틴 강화 유전자변형 쌀은 의도한 바의 베타-카로틴이 쌀의 종실에 생합성 됨과 함께 쌀의 일반성분, 지방산, 아미노산 및 무기질 함량에는 별 다른 영향을 미치지 않는 것으로 나타났다.

Effects of the P/S Ratio of Dietary Lipids and Antioxidant Vitamin Supplements on the Level of Serum Lipids and Liver. Lipid Peroxidation in Rats Treated with DMBA

  • This study an analyzes the effects of the P/S ratio of dietary lipids and antioxidant vitamin supplements on serum lipids level and fatty acid profile, the degree of lipid peroxidation, and the antioxidant enzyme activities in the liver of rats treated with 7,12-dimethylbenz($\alpha$) anthracene(DMBA). P/S ratio of dietary lipids was made into 0.5, 1 and 2 by mixing palm oil, soybean oil, sesame oil and perilla oil at 10%(w/w) fat level and n-6/n-3 ratio was fixed to 4. Antioxidant vitamin of $\alpha$-tocopherol or $\beta$-carotene was supplemented in addition to vitamin mixture which was given at 1 % of the standard diet. female Sprague-Dawley strain rats, about 60 days old, were divided into three groups(LP : low P/S ratio(0.5), MP : medium P/S ratio (1.0), HP , high P/S ratio(2.0)) and each group was sub-divided into three groups(S ; standard, T ; tocopherol supplemented, C : carotene supplemented): Two weeks after feeding experimental diets, all groups were treated with a single dose of DMBA(2mg/100g BW) by gastric intubation and fed experimental diet for 9 week. The results were as follows ; 1) Serum total cholesterol(TC) level was not significantly influenced by diet but tended to be lower in HP groups compared to LP and MP groups. Triglyceride level was the highest in LP groups and the lowest in $\alpha$-tocopherol supplemented groups. 2) Thiobarbituric acid reactive substance(TBARS) level, representing lipid peroxidation in hepatic microsome, tended to be increased as the unsaturation of dietary lipids increases. $\alpha$-Tocopherol supplement significantly decreased TBARS level. 3) The activities of superoxide dismutase(SOD) and glutathione peroxidase(GSHPx) in hepatic cytosol showed the tendency to be high with increasing P/S ratio of dietary lipids. SOD activity was not significantly influenced by antioxidant vitamin, but GSHPx activity was significantly increased in $\alpha$-tocopherol supplemented groups. In summary, high polyunsaturated fat diet was effective on reducing the serum level of total cholesterol and triglyceride, while it increased unsaturation and peroxidizability of serum fatty acid. With increasing P/S ratio of dietary lipids, lipid peroxidation was increased in the liver and antioxidant enzyme system was induced to inhibit lipid peroxidation against oxidative damage. $\alpha$-Tocopherol supplement was effective in lowering lipid peoxidation, but $\beta$-carotene supplement did not exhibit antioxidant effect. Blanching 조건이 당근쥬스의 품질에 미치는 영향 (Effect of Blanching Condition on the Quality of Carrot Juice)

  • 당근 고유의 색상, 맛, 향을 가지는 당근쥬스를 제조하기 위하여 데치기용액의 종류와 시간을 달리하여 제조한 당근쥬스의 수율, pH, 가용성 고형분, 적정산도, 색도, 저장 중 탁도 변화, $\alpha,$ $\beta-carotenes$ 함량, PE 활성도 및 관능검사를 실시하여 당근의 데치기 조건을 최적화하였다. 당근쥬스의 수율은 생착즙액이 65.9%로 가장 높았으며, 물로 데치기한 경우는 51.7~55.3% 였고 초산용액으로 데치기한 경우는 44.6%로 가장 낮았다. 당근쥬스의 pH는 생착즙액인 경우 6.20으로 물로 데치기 한 경우인 6.12~6.37과 거의 유사하였으나, 초산용액으로 데치기한 경우는 4.85로 가장 낮았다. 가용성 고형분(。Brix)은 생착즙액인 경우 9.5로 가장 높았으며, 물로 데치기한 경우는 7.1~8.1%로 데치기 시간의 증가에 따라 감소하였으며 초산용액으로 데치기한 경우는 6.8로 가장 낮았다. 물로 1분 데치기한 경우와 0.05N 초산용액으로 5분 데치기한 후 착즙한 쥬스는 3일 저장 후에도 초기 탁도의 94% 이상이 유지되어 시판중인 당근쥬스 보다도 색상침전이 적었다. 생당근 중의 u-carotene이 약 20%, p-carotene이 약 44~46%가 물과 초산용액으로 데치기한 후 착즙한 당근쥬스 중에 추출되었다. 관능검사 결과 종합적인 선호도에 있어서 물 데치기 >초산용액 데치기=시판쥬스 순으로 생당근을 물에서 1분 데치기한 후 착즙여과한 쥬스가 가장 우수하였다.
