식물플랑크론 5종에 대한 benzo[a]Pyene의 독성에 관한 연구

  • 본 연구는 해양 식물플랑크톤에 지속성 유기오염물질의 영향을 이해하기 위한 목적으로, 연안역에서 우점하는 식물플랑크톤 주요 5종 즉, 규조류 Skeletonema costatum, 침편모조류 Heterosigma akashiwo, 와편모류 prorecentrum dentatum, Prorocentrum minimum, Akashiwo sanguinea를 이용하여을 benzo[a]pyrene (PAHs)에 72시간동안 노출시킨 다음 각 종의 성장 및 저해, 회복능력 등을 조사하였고, benzo[a]Pyrene의 농도 0.1, 1, 5, 10 $\mu\textrm{g}$/l에 Heterosigma akashiwo(Raphidophyceae)를 노출시켜 시간에 따른 광합성률의 변화를 측정하였다. benzo[a]Pyrene에 72시간 노출시킨 후 S. costatum, P. minimum, P. dentatum, akashiwo의 세포수는 1-10 $\mu\textrm{g}$/l의 농도범위에서 극적인 감소를 보인 반면, A. sanguinea는 0.1-1 $\mu\textrm{g}$/l의 낮은 농도범위에서 지수함수적인 감소를 나타냈다. 성장저해 농도 ($IC_{50}$/)는 A. sanguinea가 0.43 $\mu\textrm{g}$/l로 가장 낮았고, H akashiwo(1.17 $\mu\textrm{g}$/l) S. costatum (3.34 $\mu\textrm{g}$/l), p dentatum (3.97 $\mu\textrm{g}$/l), p minimum (7.24 $\mu\textrm{g}$/l)의 순서로 증가하였다. BenEo[a]pyrene의 낮은 농도(1 $\mu\textrm{g}$/l)에 노출되었던 세포들은 5종 모두 시간이 경과함에 따라 회복하는 경향을 나타냈으나 고농도(10, 100 $\mu\textrm{g}$/l)에 노출시에는 P. minimum을 제외하고는 회복되지 않았다. 이러한 결과들은 조사된 5종 중에서 유각 와편모류 P. minimum이 benzo[a]pyrene에 가장 내성이 강하며, 무각 와편모류 A. sangulinea가 가장 약함을 의미한다. benzo[a]pyrene의 여러 농도수준에 노출시켰을 때 H. akashiwo의 광합성 능력은 낮은 농도들에서는 대조구와 유사하였으나, 5 $\mu\textrm{g}$/l의 높은 농도에서는 초기에 매우 낮은 광합성 능력을 보이다가 시간이 경과하면서 대조군보다 더 높은 경향을 나타냈다. 이러한 결과는 식물플랑크톤이 benso[a]pyrene의 낮은 농도에서 노출될 때는 이 물질을 탄소원으로 사용할 가능성이 있음을 시사한다. 본 연구의 결과들은 연안해역에 benso[a]pyrene과 같은 지속성 유기오염물질이 유입되었을 때 내정여부에 따라 식물플랑크톤 군집내 종 천이와 일차생산력에 크게 영향을 미칠 수 있음을 시사한다.

훈연공정이 훈연액 중의 다환방향족 탄화수소의 함량에 미치는 영향 (Effect of Smoking Process on the Contents of Polycyclic Aromatic Hydrocarbons in Smoke Flavouring)

  • The contents of holocellulose, one of the main components of the wood, were 83.9% in oak wood and 76.9% in apple wood, respectively. Those of hemicellulose were 16.41 and 20.33%, and in lignin 23.0 and 19.7%, respectively. Six species of domestic oak wood and apple wood were considered to be suitable for smoking materials due to the low content of lignin. Benzo(a)pyrene contents in smoke flavoring prepared with oak wood at 150, 400 and 500$^{\circ}C$ were 0. 4, 3. 7 and 5.6$\mu\textrm{g}$,/kg, respectively. The amounts of phenanthrene were 112.7, 131.4 and 255.9$\mu\textrm{g}$/kg, respectively, in each temperature. The amounts of polycyclic aromatic hydrocarbons(PAH) in smoke flavory were in the order of phenanthrene>anthracene>pyrene>benzo(a)anthracene>chrysene>benzo(b)fluoranthens>benzo(a)pyrene Benzo(a)pyrene contents in smoking extracts prepared with apple wood at 150, 400 and 500$^{\circ}C$ were 0.4, 3.3 and 5.5$\mu\textrm{g}$/kg, respectively. Phenanthrene contents in those samples were 72.7, 100.2 and 220.5$\mu\textrm{g}$/kg, respectively. Contents of each PAH showed the same order as in oak wood.

오미자 메탄올 추출액이 흰쥐에 있어서 Benzo(a)pyrene에 이해 유도된 간장해에 미치는 영향 (Effect of Omija(Schizandra chinensis Baillon) Methanal Extract on Benzo(a)pyrene induced Hepatotoxicity in Rats)

  • The protective effect of omija methanol extract on benzo(a)pyrene induce liver injury was studied in rats in vitro and in vivo. In vitro experiment, primary cultured hepatocytes(5${\times}$105cells/$m\ell$) were cultured for 20∼24 hours after adding omija methanol extract(5.1$\mu\textrm{g}$/$m\ell$) and B(a)P(50$\mu\textrm{m}$) in culture medium. In vivo experiment, omija methanol extract(0.1g/kg/day, per os) was administered for 7days and B(a)P(0.1mg/kg body weight, intraperitoneally) was given to the rats after the last administration of extract. Omija methanol extract significantly recovered serum enzyme activities(AST, ALT and LDH) and lipid contents(total cholesterol, triglyceride and HDL-cholesterol) changed by benzo(a)pyrene (B(a)P) to normal levels in vivo. In vitro experiment, as a result of 3-(4, 5-dimethlythiazol-2-yl)-2, 5-diphenyl tetrazolium bromide(MTT) assay, omija methanol extract showed a little hepatotoxicity compared with group I (normal) but significantly recovered enzyme activities(AST, ALT and LDH) changed by B(a)P in comparison to group IIadministered B(a)P only. It was suggested that omija methanol extract has a protective effect on liver injury induced by B(a)P.

벤조피렌의 급성독성시험 연구 (An Experimental Study on Acute Toxicity of Benzo[a]pyrene)

  • Objectives: This study was performed to observe the toxicological changes caused by a single exposure to benzo[a]pyrene. Methods: Based on the results of a preliminary study, 300 mg/kg was set as the middle dose. A highest dose of 2,000 mg/kg and a lowest dose of 50 mg/kg were selected based on GHS guidelines. Benzo[a]pyrene was orally administered once to female and male SD rats at dose levels of 50, 300, and 2,000 mg/kg(body weight). All animals were monitored daily for clinical signs and mortality over 14 days. Hematological and biochemical values were examined as well. Results: There were neither dead animals nor significant changes in body weights during the experimental period. In addition, no differences were found between the control and treated groups in clinical sign, hematology, serum biochemical, and histopathological analysis. Conclusion: Compared with the control group, we could not detect any toxic alteration in all treated groups. These studies indicate that the acute toxicity of benzo[a]pyrene is relatively low.

Benzo(a)pyrene에 의해 유도된 간기능장해에 미치는 자근 추출액의 영향 (The Effect of Lithospermi Radix on Benzo(a)pyrene-Induced Hepatotoxicity)

  • 민간에서 주로 사용되는 약용식물들 중 간독성의 발현 저해 및 간기능 보호 효과가 우수한 약물을 찾아 임상적인 약효를 밝혀보고자 하는 실험과정으로 탄소화합물의 불완전 연소 및 열분해에 의해 생성되는 간장해 물질인 benzo(a)pyrene으로 유도한 rats의 간독성 발현에 미치는 자근 수침액의 예방 및 치료 효과를 실험한 결과 자근 수침액의 투여는 B(a)P 투여로 현저하게 증가된 혈청 및 간장의 AST, ALT, LDH, ALP 활성을 유의성 있게 감소시켰고 B(a)P 투여로 증가한 혈청 total cholesterol 및 phospholipid 함량 역시 감소시킬 수 있었으나 그 효과는 현저하지 않았다. 즉, 자근 수침액의 투여는 B(a)P에 의한 간독성 발현을 유의성 있게 감소시켰으며 그 효과는 전처리가 후처리에 비해 우수하였다.

Effect of Chinese Yam on Benzo[a]pyrene Hydroxylase Activities in Rats Fed Dietary Benzo[a]pyrene

  • To investigate the effect of yam on the activity of benzo[a]pyrene hydroxylase(BPH), the key enzyme associated with polycyclic aromatic hydrocarbons(PAHs) metabolism, rats were fed a fiber free diet for 7 days, whereupon they were switched to experimental diets for another 7 days. Diets contained benzo[a] pyrene(BP, 400 mg/kg diet) and 25% or 50% yam powder (freezer dried and hot air dried ). Diets containing pectin and cellulose were compared with diets containing yam. BPH activities were assessed in the liver, lung, kidney, stomach, small intestine and large intestine of rats. BP induced BPH activities in various tissues ; 8 fold in liver, 28 in lung and stomach , and 32 in large intestine. The addition of yam significantly lowered BPH activity in liver, lung and stomach and hot air dried yam was nmor eeffectivie than freeze dried yams. These data suggested that yam containing diet may influence carcinogen metabolism in liver and extrahepatic target tissues by altering activities of BPH and may reduce exposure of these tissues to dietary carcinogens.

대추 메탄올 추출액이 Benzo(a)pyrene에 의해 유도된 간기능 장해에 미치는 영향 (Effect of Jujube Methanol Extract on Benzo(a)pyrene Induced Hepatotoxicity)

  • The protective effect of jujube methanol extract on benzo(a)pyrene(B(a)P)-induced liver injury was studied in rats in vitro and in vivo. Jujube methanol extract significantly recovered the enzyme activities(GOT, GPT, LDH and ALP) and lipid contents(total cholesterol, triglyceride and HDL-cholesterol) changed by B(a)P to normal levels in vivo. in viro experiment jujube methanol extract didn't stimulate hepatocyte proliferation but significantly recovered the enzyme activities(GOT, GPT and LDH) in comparison to group Ⅱ administered B(a)P only. It was suggested that jujube methanol extract have a protective effect on liver injury by B(a)P.

Structural Damage of DNA by 6-Sulfooxymethyl Benzo(a)pyrene

  • The effect of 6-sulfooxymethyl benzo(a)pyrene (SMBP) on conformational changes of calf thymus DNA was investigated. As SMBP is a strong electrophile, the covalent binding of SMBP to DNA should distort three dimensional conformation of DNA at the binding sites. A formaldehyde-unwinding methods were used to determine the rate of DNA denaturation. The increase in absorbance at 251nm was detected by addition of formaldehyde following treatment with SMBP. SMBP changed supercoiled DNA to relaxed and linear DNA as determined by electrophoresis, which was similar to the change in DNA due to in vitro treatment with benzo(a) pyrene diol epoxide. Treatment with SMBP completely denatured DNA under alkaline conditions. However, DNA was nicked or partially denatured under neutral condition. The absorption band of DNA was increased by the treatment with SMBP in V79 cells, which may be explained by the formation of stabilized SMBP-DNA adduct.

GC-MS/Ms Analysis of Benzo(a)pyrene by Ion Trap Tandem Mass Spectrometry

  • The mass spectrometry using an ion trap tandem mass spectrometer has been investigated to find optimum conditions for the analysis of benzo(a)pyrene (3,4-benzpyrene). The applicability to a real soil sample was also investigated to verify the usef ulness of the MS/MS (or collision induced dissociation, CID) analysis. The optimum CID condition was 1.5 and 0.45 for the RF excitation voltage and the q value, respectively. For comparison, CID and EI were applied to the analysis of a soil sample. CID analysis was more sensitive than EI analysis of the soil sample. The limit of detection (LOD) of benzo(a)pyrene was 3.18 ng mL-1 and 0.85 ng mL,-1 for EI and MS/MS analysis, respectively. The precision at the soil sample for EI and CID showed relative standard deviations of 6.1% and 4.1%, respectively, and the concentrations were 168 ㎍ kg-1 and 162 ㎍ kg-1 , respectively.

오매(烏梅)와 증숙 오매(蒸熟 烏梅)의 생리활성 연구 (Biological Activity Review of Mume Fructus and Processed Mume Fructus)

  • Objectives: Presently Mume Fructus (MF) undergoes fumigation, which produces benzo[a]pyrene. As a primary analysis with the aims to minimize the production of benzo[a]pyrene and to suggest standards for processing the MF, the steaming method was chosen among the various processing methods, and reviewed through a series of experiments.Methods: Methods:Pitted and un-pitted MF were steamed and processed into samples. After testing level of benzo[a]pyrene, the samples were analyzed for amount of polyphenol and flavonoids. Scavenging activities of the samples for the DPPH and ABTS radicals were tested. In order to measure anti-inflammatory effects of the samples, cell survival rate was investigated using CCK-8 Assay. Also, water extracts of dried and steamed MF were administered to the RAW 264.7 cells to compare expressions of NO, PGE2, IL-1β, and TNF-α. In addition, anti-diarrhea effects of the herbal medicine were tested on animal models with diarrhea induced by MgSO4 and Castor oil.Results: Regardless of pitting, processed MF contained no benzo[a]pyrene. Anti-oxidation effect increased in relation to the frequency of steaming process. However, extracts of dried and steamed MF suppressed different kinds of inflammation factors, and extract of dried MF showed superior anti-diarrhea effect than extract of steamed MF.Conclusions: It is suggested that steaming method of MF is recommended for processing the herbal medicine without the production of benzo[a]pyrene. But regarding that dried and steamed MF showed differences in their anti-oxidative, anti-inflammatory, and anti-diarrhea effects, it is recommended to perform further researches on different efficacies of MF according to their processing methods.
