  • The immunopotentiating effect of ethanol extract, butanol fraction and petroleum ether extract of Panax ginseng on the immunotoxicity of benzo(a)pyrene were investigated in mice. A single administration of benzo(a)pyrene induced an apparent but relatively transient reduction in HY titer, Arthus reaction, delayed type hypersensitivity, rosette forming cell and natural killer cell activity Ethanol extract very significantly restored HY titer, Arthus reaction. RFC and natural killer cell activity. Butanol fraction have no effect. But petroleum ether extract very significantly restored humoral and cellular immune response and especially natural killer cell activity.

Screening of Ecotoxicant Responsive Genes and Expression Analysis of Benzo[a]pyrene-exposed Rockfish (Sebastes schlgeli)

  • Benzo[a]pyrene is a representative ecotoxicant in marine environment and a model compound of polycyclic aromatic hydrocarbons, which has an ability to bioaccumulate in aquatic organisms. This study aimed to identify molecular biomarkers suitable for assessing environmental pollution using a microarray technique. We examined the effects of benzo[a]pyrene on gene expressions in the rockfish, Sebastes schlegeli. We constructed the subtractive cDNA library with hepatic RNA from benzo[a]pyrene-exposed and non-exposed control fish. From the library 10,000 candidate clones were selected randomly and cDNA microarray was constructed. We determined benzo[a]pyrene-responsive genes using a high-density microarray. Statistical analysis showed that approximately 400 genes are significantly induced or reduced by benzo[a]pyrene treatment ($2\;{\mu}m$). Especially gene expression changes of 4 candidate clones among the up- or down-regulated genes were investigated in 6, 12 and 24 hr BaP-exposed fish groups. Many methods have been developed to monitor marine environmental status, which depend on quantifying the levels of the toxic components in polluted seawater or on ecological accessing, such as species diversity or richness. However, those methods could not provide information on physiological or genetic changes induced by such environmental stresses. Comparing with the conventional methods, these data will propose that benzo[a]pyrene-responsive genes can be useful for biological risk assessment of polycyclic aromatic hydrocarbons on marine organism at molecular level.

유통 한약재 초과(草果) 중 벤조피렌 오염실태 조사 (A Survey on Benzo(a)pyrene Contamination in Amomum Tsao-ko Fruit of Medicinal Herbs)

  • Amomum Tsao-ko used as a traditional oriental herbal medicine, is indigenous to several Asia countries. This study was carried out to investigate the contamination by Benzo(a)pyrene in Amomum Tsao-ko Fruit of Medicinal Herbs. 20 samples of Amomum Tsao-ko Fruit were evaluated for the Benzo(a)pyrene contamination. They were analyzed for Benzo(a)pyrene using high-performance liquid chromatogrphy(HPLC)-fluorescence detection and the positive samples were confirmed using gas chromatography tandem mass spectrometry. The levels of Benzo(a)pyrene were from 9.2 to 95.5 ㎍/kg and the average was 40.6 ㎍/kg. There are no Benzo(a)pyrene standards for Amomum Tsao-ko Fruit of Medicinal Herbs. These data will be used as a basic data for the future legislation on the regulation and control of benzo(a)pyrene of Amomum Tsao-ko Fruit of Medicinal Herbs.

가공조건에 따른 참기름의 benzo(a)pyrene 변화 (The Changes of Benzo(a)pyrene in Sesame Oil Affected by Processing Conditions)

  • Sesame oil is a simple pressed oil as unrefined oil. During manufacturing process of roasting-expression, benzo(a)pyrene[B(a)P] formed as a strong carcinogenic substance is cause a social problem. In manufacturing process of sesame oil, it had following the forming pathway of benzo(a)pyrene[B(a)P] as well as minimizing plan of B(a)P formation. Suitable roasting condition by roaster was during 15~20min at $220^{\circ}C$, B(a)P content in sesame oil was $1.35{\sim}1.57{\mu}g/kg$. Between roasting temperature and/or roastingtime and forming amount of B(a)P was showed a linear correlation. As a point of view the turbidity and yield of final product, roasting process of the more regular level was required.

유통 한약재 중 벤조피렌 함량 모니터링 및 안전성 평가 (Monitoring and Risk Assessment of Benzo(a)pyrene Content in Medicinal Herbs)

  • In this study, to investigate of safety for Benzo(a)pyrene in medicinal herb, 8 kinds of selected commercial herbal medicines (Rehmanniae Radix, Rehmanniae Radix Preparata, Amomi Tsao-Ko Fructus, Cimicifugae Rhizoma, Cyperi Rhizoma, Magnoliae Cortex, Scutellariae Radix, Scrophulariae Radix) were analysed using the high performance liquid chromatography with fluorescence detector and assessed the health risk. The levels of benzo(a)pyrene were from non-detection to $28.1{\mu}g/kg$, and the average was $3.6{\mu}g/kg$. Based on a nationwide survey of the consumption of medicinal herb by the Korean population, we estimated the potential risk from the ingestion of benzo(a)pyrene. The estimated daily intake of benzo(a)pyrene was 1.6 ng/kg b.w./day for group only know the daily average intake of medicinal herb. The MOE (margin of exposure) of benzo(a)pyrene for estimate of health risk was $1.93{\times}10^5$. Therefore, health risk from benzo(a)pyrene through intake of herbal medicine was considered negligible.

국내 유통 식용유지 및 식용유지 종류별 제조 공정에 따른 벤조피렌 함량 (Benzo(a)pyrene Contents in Commercial Vegetable Oils and Changes during Processing of Vegetable Oils)

  • 국내에서 유통되는 식용유지의 벤조피렌 함량과 정제유와 압착유의 제조 공정별 벤조피렌 함량변화를 살펴보았다. 측정된 모든 식용유지의 벤조피렌 함량은 허용한계 이하로 검출되긴 하였으나, 동일한 식용유지라 하더라도 벤조피렌 함량에 큰 차이를 나타내어 원료 및 제조 공정에 따라 최종제품의 벤조피렌을 저감화시킬 수 있음을 확인할 수 있었다. 정제유 및 압착유의 제조 공정별 벤조피렌 함량을 분석한 결과, 정제 공정을 거치면서 원유 및 착유 원유에 함유되어있던 벤조피렌의 함량이 꾸준히 감소되는 것을 확인할 수 있었다. 또한 옥수수유의 경우는 옥수수 배아를 압착 착유하는 과정에서 벤조피렌의 발생이 증가되었으며, 참기름의 경우는 참깨의 볶음 과정에서 벤조피렌이 급격히 발생하는 것을 확인할 수 있었다. 즉 식용유지의 제조 공정 중고온 처리과정에서 벤조피렌의 발생량이 급증하였으며, 이후 정제 공정에서 꾸준히 감소되는 현상을 나타내었다. 이러한 결과는 이후 벤조피렌의 저감화를 위한 공정개선의 기초자료로 활용할 수 있을 것이다.

동물플랑크톤에 대한 지속성 유기오염물질 PAHs와 TBT의 독성 및 생존능력에 미치는 영향 (Toxicity of Persistent Organic Pollutants, PAHs and TBT, in Zooplankton and Influence on Their Viability)

  • 인간이나 환경에 해로운 영향을 주는 지속성 유기오염물질의 독성을 평가하기 위해서 염분과 온도에 대해 내성이 강한 Acartia 종들과 Artemia을 대상으로 실험하였다. 지속성 유기오염물질인 PAHs와 TBT에 대한 요각류의 독성을 평가하기 위해 3가지의 실험을 실시하였다. 1) 광양만에서 주로 나타나는 5가지 PAHs(anthracene, benzo〔a〕pyrene, fluoranthene, phenanthrene, pyrene)에 대한 A. omorii의 48h-LC$_{50}$ 을 구하였다. 2) Artemia를 이용하여 온도에 따른 benzo〔a〕pyrene과 TBT의 독성의 변화를 측정하였다. 3) PhMs중에서 독성이 강한 benzo〔a〕pyrene에 노출된 먹이를 섭취한 A. erythruea와 A. omorii의 난 생산, 부화율, 고형화된 배설물 양의 변화를 측정하였다. A. omorii에 대한 5가지 PAHs 중에서 fluoranthene (48 h-LC$_{50}$ 19.20 $\mu\textrm{g}$ L$_{-1}$)과 benzo〔a〕pyrene (48h-LC$_{50}$ 29.89$\mu\textrm{g}$ L$_{-1}$)의 독성이 강하게 나타났다. 온도실험에서는 동일한 유해물질을 가지고 실험을 하더라도 온도의 변화에 따라 급격한 독성의 차이가 나타날 수 있고, 유해물질 간에도 온도에 따라서 나타나는 독성에 대한 특성이 다르게 나타났다. 특히 15$^{\circ}C$에서는 TBT (9.982 $\mu\textrm{g}$ L$^{-1}$)의 독성이 benzo〔a〕pyrene (1,173 $\mu\textrm{g}$ L$^{-1}$)의 독성보다 약 100배 정도 더 독성이 강하게 나타났다. Benzo〔a〕pyrene에 노출된 먹이는 요각류의 난 생산, 부화율, 고형화된 배설물 양의 변화에 영향을 주었다. 특히 benzo〔a〕pyrene의 농도 증가는 요각류의 생산력 변동에 영향을 미칠 수 있는 것으로 사료된다. 본 연구는 요각류를 이용하여 해양생태계에 유입되는 지속성 유기오염물질에 대한 위험성을 알리는 지표생물로서 사용할 수 있을 것으로 판단된다.

녹각추출물이 Benzo(a)pyrene에 의한 간손상에 미치는 영향 (Effect of Old Antler Extracts on the Benzo(a)pyrene-Induced Hepatotoxicity in Rats)

  • 녹각이 동양의학에서 보혈강장제라는 사실에 근거하여 생체내 물질대사에 중요한 역할을 하는 간장의 장해시 효소활성 변동에 미치는 녹각의 효능을 구명할 목적의 일환으로 간독성 물질인 benzo(a)pyrene으로 간장해를 유도한 후 각각의 녹각추출물을 급여함으로써 간해독 과정에 관여하는 효소의 활성을 생화학적 측면에서 비교하였다. 각각의 녹각추출물 급여로 인한 체중증가량, 식이섭취량 및 식이효율은 녹각추출물 군간의 유의성은 나타나지 않았으나, Control군에 비하여 유의적인 증가를 보였고 B(a)P 투여군에서도 Cont-B군에 비하여 녹각추출물 급여군에서 유의적인 증가를 나타내었다. 체중 100g당 장기무게는 간장의 경우 benzo (a)pyrene 투여로 증가하는 경향을 보였으나 비장과 신장 및 심장에는 별다른 차이가 없었다. 녹각추출물 급여로 4주간 사육한 흰쥐에서 간해독계 효소중 cytochrome P-450의 함량은 Control 군에 비하여 ben-zo(a)pyrene 투여로 유의적인 감소를 보였으며, 녹각추출물과 benzo(a)pyrene을 병행 투여한 실험군 중 Water-B군이 정상군에 가깝게 증가되었다. 간조직 중의 glutathione함량은 Water군과 Neutral군에서 증가를 보였으나 benzo(a)pyrene 투여로 감소하였다. 또한 간조직 중의 glutathione peroxidase 활성은 녹각추출물 급여에 따른 영향은 없었으며, benzo(a)pyrene 투여로 감소된 활성이 water-B군에서 정상군에 가까운 증가를 나타내었다. 간조직 중의 과산화지질 함량은 Water-B군에서 그 감소 정도가 가장 현저하였다. 이상의 결과를 종합해 볼 때 녹각추출물 중의 water-ext.와 neutral-ext.에 함유되어 있는 유효성분이 간장의 해독기구 효소활성을 유도하고, benzo(a)pyrene의 대사를 촉진시키므로 간손상을 억제할 수 있을 것으로 생각된다.

식품가열에 따른 Benzo[a]pyrene 생성 및 한국인의 발암 위해성 평가 (Pyrolytic Formation of Benzo[a]pyrene in Foods During Heating and Cancer Risk Assessment in Koreans)

  • The pyrolytic formation of benzo[a]pyrene during the cooking procedure was analysed in beef, pork, pacific saury, rice, and soybean by HPLC. In raw foods, benzo[a]pyrene (B[a]P) was not detected or negligible, but it was increasingly formed when foods were boiled (0.010~0.037 ppb) and more dramatically during broiling (0.302~0.851 ppb) in a time dependent manner. Human daily intake of B[a]P in Koreans and cancer risk assessment were estimated based on food consumption per capita and carcinogenic potency of B[a]P. When cooked foods were consumed for entire life time, cancer risk was estimated to bo 1.77$\times$10-6>1.65$\times$10-7>1.32$\times$10-8 by the order of broiled, boiled, and raw foods consumption. These data suggest that broiled foods produce more benzo[a]pyrene than water boiled foods. Thus cooking procedure is an important factor for the formation of carcinogens and needs to bo modified to reduce cancer risk for man.

Dietary Fiber Reduces Benzo[a]pyrene Hydroxylase Induced by Dietary Benzo[a]pyrene

  • This study was conducted to determine if dietary fiber would reduce exposure of the tissues to dietary benzo[a]pyrene(BP) , a well-known carcinogenic polycyclic aromatic hydrocarbon, as evaluated by benzo[a]pyrene hydroxylase (BPH) activity. The effects of three different sources of dietary fiber(pectin, polydextrose, and clellulose) on BPH activity were studied using Sprague-Dawley rats. In this study, male rats were fed a fiber-free purified diet for 7 days, whereupon they were switched to experimentla diets for 48h. After 48h, their liver, stomach , small intestinal mucosa and large intestinal mucosa were assayed for BPH activity. Thissues exposed to benzo[a]pyrene(400mg/kg diet, fiber-free) showed significant increse in the activity of BPH ; 27 times in liver, 7 times in stomach, 18 times in small intestinal mucosa and 3 times in large intestine. The inhibition in BP -induced BPH activity by dietary fiber in liver, stomach and small intestinal mucosa was observed in the decreasing order : 10 % perctin > 10% polydextrose >5 % polydextrose > 10% cellulose. Decreased BPH induction indicates that soluble dietry fibers, especially pectin and polydextrose in this study, protect the tissues of digestive system from exposure to BP.
