Isolation of Marine Bacterium Decomposing Sea tangle (Laminaria japonica) to Single Cell Detritus (다시마(Laminaria japonica)를 single cell detritus로 분해하는 해양세균의 분리)

  • Seventy-one marine bacteria decomposing sea tangle (Laminaria japonica) into single cell detritus (SCD) were isolated from sea water, sea tangle, sea mustard (Undaria pinnatifida), sea urchin (Anthocidaris crassispina), star fish (Acanthaster planci), and turban cell (Batillus cornutus), among which 14 strains decreased cutting strength of sea tangle and had alginate-degrading activity. Marine bacterium No. 34 isolated from turban cell showed lowest cutting strength of sea tangle, strongest alginate-degrading activity, and produced high content of $5-10\;{\mu}m$ SCD from sea tangle. This strain was identified as Vibrio sp. based on morphological, physiological, and biochemical characteristics and named as Vibrio sp. YKW-34.

Mantle Ultrastructure of the Spiny Top Shell, Batillus cornutus (Gastropoda: Turbinidae) (소라(Batillus cornutus) 외투막의 미세구조)

  • The histochemical characteristics and ultrastructure of the mantle in the spiny top shell, Batillus cornutus were described using light and electron microscopy. The simple epidermal layer wrapped on the top and bottom of the centrally located connective tissue. And then the epidermal layer were divided into the outer epidermal layer near a shell and the inner epidermal layer closed to the visceral mass. The connective tissue layer was composed of the collagen fiber muscularfiber bundle and hemolymph sinus. Mucous cells in the apical mantle contained acid and neutral mucopolysaccaride, and acidic carboxylated mucopolysaccaride in the mid and marginal mantle. The mantle thickness, epidermal layer thickness and hemolymph sinus area displayed a trend of reduction from the marginal zone to the apical zone. From TEM observation, it was possible to distinguish epithelium, ciliated cell, absorptive cell and secretory cell in the epidermal layer. The epithelia were columnar and the nucleus was elliptical. The free surface were covered with microvilli. The lateral membranes of epithelium was con nected with neighboring cells by the zonular occludens, zonular adherens and membrane interdigitation. Ciliated cell on free surface had cilia and microvilli, and numerous mitochondria in the apical cytoplasm. In the epidermal layer, it observed 2 type cells having absorptive function. The absorptive cells were columnar in shape, and contained microvilli, pinocytotic vesicles, mitochondria and lysosomes of various electron density. Secretory cells can be divided into four types (A, B, C, D) depending on the cell shape and characteristics of secretory granules. These cells were unicellular glands and had similar characteristics to previously reported on the mantle of the gastropod and bivalves.

Quality Characteristics of Sea Tangle Single Cell Detritus (SCD) Manufactured by Vibrio sp. Isolated from Batillus cornutus (소라에서 분리한 Vibrio균으로 제조한 다시마 Single Cell Detritus(SCD)의 품질특성)

  • Obtaining powder form of seaweed is essential for the use of seaweed as a food additive. The deterioration of seaweed caused by high temperatures during homogenization and powder processing is a serious problem and limits the use of seaweed as a food or pharmaceutical ingredient. Furthermore, many powder particles are not fluidized very well because of the interaction between particles. In order to solve this problem, sea tangle was hydrolyzed to a level of single cell detritus (SCD) by Vibrio sp., isolated from Batillus cornutus. with strong hydrolytic activity. The crude protein and amino acid contents of sea tangle SCD were higher than those of the powder, whereas the reverse was true for ash content. Sea tangle powder contained more mineral than its SCD, whereas total amino acid content was 5 times more in SCD than in power. The anticancer activities of sea tangle SCD and powder were 31.20 and 29.07%, respectively, with no significant difference (p<0.05), but about 15% higher than that of the control. The ACE inhibitory activity of the sea tangle powder, 39.31%, was higher than the 26.07% of the SCD. The antithrombin activity of the sea tangle powder, 55.3 seconds, was higher than the 34.5 seconds of the SCD. Moreover, there was no antioxidative and ischemic activities in both tile sea tangle powder and SCD.

  • Samples of Turbo (Batillus) cornutus Lightfoot, 1786 in Jeju Island were collected from September 2009 to May 2010. Population ecological parameters and stock assessment of the turban shell were determined, based on the length and age composition data from 2000 to 2009 and ecological parameters. Instantaneous coefficient of total mortality (Z) of turban shell was estimated to be 2.2062/year. The estimated instantaneous coefficient of natural mortality (M) was 0.874/year. The age of turban shell at its first capture ($t_c$) was 2.636 year. Yield-per-recruit were estimated under harvest strategies that based on $F_{max}$, $F_{0.1}$, $F_{35%}$, and $F_{40%}$ was 10.44 g, 1.87 g, 6.53 g and 7.46 g.

Nuclear DNA content determinations in 15 seawater shellfish species in Korea (한국 해산 패류 15종의 DNA 함량)

  • The object of this study was to obtain nuclear DNA content data for representatives of the 15 shellfish species that inhabit the coast of Korea. In the gastropoda group, the DNA content (pg DNA nucleus-1) was 3.3±0.08 in Haliotis discus hannai and 2.4±0.18 in Batillus cornutus. In the bivalvia group, the DNA content(pg DNA nucleus-1) was 2.0±0.15 in Scapharca broughtonii, 3.0±0.12 in Mytilus galloprovincialis, 2.9±0.05 in Meretrix lusoria, 2.2±0.03 in Meretrix lamarkii, 2.6±0.05 in Fulvia mutica, 1.8±0.18 in Tegillarca granosa, 3.3±0.01 in Solen corneus, 2.2±0.04 in Barnea manilensis, in 2.5±0.32 in Crassostrea gigas, 3.9±0.24 in Atrina pectinate, 3.5±0.15 in Patinopecten yessoensis, 1.9±0.16 in Amygdala philippinarum, and 2.3±0.14 in Pseudocardium sachalinensis. The results of this study provide new information for a better understanding of the genomic evolution process of the shellfish species investigated in this experiment.

Residue of Organophosphorus Pesticides in the Coastal Environment on the Cheju Island (제주도 연안 해양환경 중 유기린계 농약의 잔류)

  • To study the residue of organophosphorus pesticides in the ocean environment on the Cheju island, EPN[O-ethyl-O-4-nitrophenyl phenylphosphonothioate] and monocrotophos [Dimethyl-1-methyl-2-methyl carba-moylvinylphosphate] of organophosphorus pesticides are surveyed on coastal environment of Cheju city and Wimi in May and October 1996. The qualified limit detection of EPN and monocrotophos are 0.005ng/mL and 0.006ng/mL in the water by GC-FPD, respectively. The qualified limit detection of EPN and monocrotophos are 0.010ng/g and 0.012ng/g in the solid such as sediment and sea organism by GC-FPD, respectively. EPN and monocrotophos are not detected in seawater and sedimnet. Moreover EPN and monocrotophos are neither detected in seaweed cava (Ecklonia cava), Agar (Gelidium amansii), turban sell(Batillus cornutus) and sea urchin(Anthocidaris Crassispina). EPN, monocrotophos used in the farm on the Cheju island are not residued in the coastal environment in the Cheju island.

  • Some enzymatic properties of Cynthia roretzi v. (Drasche Korean : U-Rung-Shei) was studied by author and obtained the following results; 1. The optimum pH of the digestive gland amylase was 6.8-7.0 2. Activity of metallic ion on the amylase showed the following order; 10$^{-3}$ M M $n^{++}$>10$^{-3}$ M $Co^{++}$>10$^{-4}$ M $Mg^{++}$>10$^{-4}$ M $Ca^{++}$>10$^{-2}$ M Z $n^{++}$>10$^{-2}$ M P $b^{++}$ 3. The digestive gland enzyme inactivated at 70.deg. C. 4. When the enzyme concentration increase 2 times, the enzymatic activity also increase, but not propertionally. 5. The digestine gland amylase showed remarkably higher enzymatic activity than the intestinal amylase. 6. The digestive gland amylase from the ascidian showed remarkably higher enzymatic activity than the heptancreatic amylase from shell fish (Turbo (Batillus) Cornutus Solander).nder).nder).

Species Appearance and Seasonal Variation of Macrobenthic Invertebrate in the Coastal Water of Chagwi-do, Jeju-Island (제주 차귀도 연안역 저서 무척추동물의 출현과 계절별 변동)

  • Macrobenthos were collected in each season by SCUBA diving to investigate the benthic faunal assemblages in the coastal water of Chagwido from September 2004 to August 2005. A total of 201 macrobenthos identified, 74 species (37.2%) of Mollusca were found; 43 species of Arthropoda (19.2%); 34 species of Cnidaria (16.7%) and others including 18 species of Porifera (9.8%). Mean density and biomass were estimated to be 455 individual/$m^2$ and 15,565.0 $g/m^2$, respectively. Whereas annelids was predominant in biomass (1,558.8 g), gastropods were the most dominant faunal group in terms of abundance (5,391 individuals) and the number of species. The dominant species were Trochus sacellus, Batillus cornutus, Pagurus gracilipes, and Cantharus cecillei. The seasonal variation of the number of species and individual Mollusca and Arthropoda was observed. There was a slow increase in spring, the peak in summer, and a slow decrease in autumn and winter. The dominance index was the highest in Gosan and lowest in Yongdang. The biodiversity indice (H') were 1.760-3.497 in each station. Mean biodiversity index was 2.858 (H'). The diversity index, the eveness index (E'), and the dominance index (R) were 3.218-3.743, 0.648-0.720, 17.690-22.826, respectively, in each station.

Ecological Studies on the Culture Bed and Production of Young Top Shell, Batillus cornutus in Cheju Island (제주도산 소라의 치패생산 및 서식생태에 관한 연구)

  • In order to improve top shell seed production techniques spawning and larvae rearing were done in rearing tanks. Growth of young top shell in the nursing ground were also investigated. For induced spawning, top shells were maintained in still water during night time. Then they were treated with ultra violet iradiated sea water after dried up in air for 60 minutes. Spawning rate were 10 to $39.77\%$. It was found that young top shells moved in the growing grounds from nursing grounds when they reached approximately 30-40mm in shell heignt. Among main food algae for top shell in the natural growing grounds, sea mustard were melted away during June. Therefore, presence of another food algae such as Ecklonia cava or Sargassum spp. seems to be the main limiting factor for survival of top shell during summer. The tolerance of top shells ranging from 30mm to 60mm to low density of seawater for were tested at the temperature between 29.5 and $31.4^{\circ}C$. Hundred percent mortality occoured in 20, 55 and 90 hours after first stocking at the specific gravity of 1.010, 1.015, and 1.020, respectively.

Vertical Distribution of Mega-invertebrate and Calculation to the Stock Assessment of Commercial Species Inhibiting Shallow Hard-bottom in Dokdo, Korea (독도 연안 암반에 서식하는 초대형 저서동물의 수직분포와 산업종의 현존량 추정)

  • Distribution and stock assessment of mega-benthos living in the shallow hard bottom around Dokdo were studied in July,2000. Depth and topographical conditions have affected to the densities and biomass of benthic animals. In shallow area, less than 10 m depth, turbo shell Batillus cornutus, mussel, Mytilus corusucs were dominated and showed distinct patterns in vertical distribution. On the other hand, the area over 10m depth, it showed diverse pattern depending on topography. Turbo shell, mussel and sea cucumber Stichopus japonicus were dominated in terms of fishery resources, but abalones were rarely sampled. Stock assessment were estimated to be 6.54 M/T, 3.89 M/T and 8.92 M/T, respectively. Some parts of coastal hard bottom around Dokdo, such as the area between Dongdo and Seodo, seemed to play an Important role as nursery ground. Therefore, it is necessary to the environmental monitoring for coastal fishery managements aspects.
