Ethanol Production by Synchronous Saccharification and Fermentation of Foodwastes

  • In the previous research about ethanol production, we confirmed that SFW(saccharified foodwastes) medium(0.56g-ethanol/g-glucose) is mere efficient than YM medium(0.538g-ethanol/g-glucose). Ethanol production using SFW needs large enzyme cost due to the enzymatic hydrolysis of foodwastes, although the enzymes was obtained from our economical enzyme production methods, using the intact whole culture broth of Trichoderma harzianum FJ1. Therefore, in this research we used synchronous saccharification and fermentationmethod to produce ethanol using foodwastes. Ethanol production yield was 0.45g-ethanol/g-reducing sugar in synchronous saccharification and for-mentation by a fed-batch mode.

Haloarcular sp. EH-1으로부터 생분해성 Poly-3-Hydroxyalkanoate의 생산 (Production of Poly-3-Hydroxyalkanhoate by Haloarcular sp. EH-1)

  • 천일염에서 분리된 고호염성균인 Haloarual sp. EH-1으로부터 PHA를 생산하여 PHA의 구조 및 성질 등을 검토하였다. 그 결과는 다음과 같다. PHA를 GC chromatogram, IR 및 300MHz H-NMR로 분석한 결과 Haloarcnly sp. EH-1으로부터 추출된 PHA는 3-hydroxybutyric acid와 3-hydroxyvaleric acid 와 3-hydroxyvaleric acid의 중합체인 PBB/HV로 추정되었으며, thermal property는 $152.46^{\circ}C.$ 점도는 1.25dl/g 그리고 분사량은 $1.44 X 10^5$이었다.

Production of Antibacterial Violet Pigment by Psychrotropic Bacterium RT102 Strain

  • The antibacterial action of violet pigment, a mixture of violacein and deoxyviolacein, isolated from phychrotrophic bacterium RT102 strain was examined, and the operational conditions for the effective production of violet pigment were studied. The antibacterial activity of the violet pigment was confirmed for several bacteria such as Bacillus licheniformis, Bacillus subtilis, Bacillus megaterium, Staphylococcus aureus, and Pseudomonas aeruginosa, and the high concentration of violet pigment, above about 15mg/L, caused not only growth inhibition but also death of cells. The growth properties of RT102 strain were clarified under various incubation conditions such as pH, temperature, and dissolved oxygen concentration. The maximum violet pigment concentration, i.e. 3.7 g/L, and the maximum productivity of violet pigment, i.e. 0.12 g .L$\^$-1/H$\^$-1/, were obtained in a batch culture of pH 6, 20$^{\circ}C$, and 1 mg/L of dissolved oxygen concentration.

Rapid Purification of Recombinant Human Lipocortin-I Secreted from Saccharomyces cerevisiae

  • Human lipocortin-I was expressed as a secretory product by Saccharomyces cerevisiae harboring an expression system consisting of GAL10 promoter, inulinase signal sequence and lipocortin-I terminator. Fed-batch fermentation was carried out to overproduce recombinant human lipocortin-I. The culture medium was desalted and concentrated by ultrafiltration, and then subjected to hydroxyapatite column chromatography. The lipocortin-I was purified to >98% purity by single-step hydroxyapatite column chromato-graphy. However, it was found that the purified lipocortin-I was a proteolytically-cleaved form which was cleaved immediately after the basic amino acid Lys26.

Production of Poly(3-hydroxybutyrate) [P(3HB)] with High P(3HB) Content by Recombinant Escherichia coli Harboring the Alcaligenes latus P(3HB) Biosynthesis Genes and the E. coli ftsZ Gene

  • Filamentation-suppressed recombinant Escherichia coli strain harboring the Alcaligenes latus polyhydroxyalkanoate (PHA) biosynthesis genes and the E. coli ftsZ gene was constructed and cultivated for the production of poly(3-hydroxybutyrate) [P(3HB)] with high concentration and high content. By the pH-stat fed-batch culture of this recombinant E. coli strain XL1-Blue(pJC5), the final cell concentration and P(3HB) concentration obtained in 44.25h were 172.2g cell dry weight/l and 141.9g P(3HB)/l, respectively, resulting in productivity of 3.21g P(3HB)/l-h. More importantly, the P(3HB) content obtained was 82.4 wt %, which was significantly higher than that obtained with the recombinant E. coli harboring only the PHA biosynthesis genes.

  • Sophorolipid was produced by Candida bombicola ATCC 22214 from soybean dark oil, a byproduct of soybean oil processing. With a fed-batch culture of C. bombicola for 7 days, 90 g/l of sophorolipid was obtained. The CMC (critical micelle concentration) and minimum surface tension of the sophorolipid in aqueous solution were found to be 150 mg/l and 48 mN/m, respectively. The dispersion capability of sophorolipid was higher than that of the chemical surfactants such as SDS and Brij30. The molar solubility ratio (MSR) of 4-methylnaphthalene was 0.2. Linoleic and oleic acids were the main constituents of the fatty acid composition of the sophorolipid. The sophorolipid showed antimicrobial activity against Propionibacterium acne and Bacillus subtilis.

  • Fermentative hydrogen production by Citrobacter sp. Y 19 was investigated in batch culture. Optimal hydrogen production activity was observed at pH 6 - 7 and temperature of $36^{\circ}C$, and hydrogen yield and maximal hydrogen production rate were 1.12 mmol/mmol glucose and 32.3 mmol/g cell${\cdot}$h, respectively. With glucose as a substrate, the bacterium produced ethanol, acetate, and carbon dioxide as major glucose fermentation by-products. Y19 could utilize various sugars such as galactose, fructose, lactose, sucrose, and starch for cell growth and hydrogen production.

Optimal conditions for biological hydrogen production from food waste

  • Biohydrogen production from food waste via dark fermentation was conducted by using mixed culture under various environmental conditions (initial pH, initial F/M ratio, initial ferrous iron ($Fe^{2+}$), and temperature condition) in batch reactor. The results revealed that the maximum hydrogen yield of $46.19mL\;H_2/g\;COD_{add}$ was achieved at the optimal conditions (initial pH 8.0, initial F/M ratio 4.0, initial iron concentration 100 mg $FeSO_4/L$ and thermophilic condition ($55{\pm}1^{\circ}C$)). Furthermore, major volatile fatty acid (VFA) productions of butyrate (765.66 mg/L) and acetate (324.69 mg/L) were detected and COD removal efficiency was detected at 66.00%. Therefore, these optimal conditions could be recommended to operate a system.

Hansenular capsulata S-13의 변이주에 의한 Killer Toxin의생산 (Production of Killer Toxin from a Mutant of Hansenular capsulata S-13)

  • Killer yeast, Hansenular capsulata S-13 were treated with heat, ethylmethane sulfonate and N-methyl-n'-nitro-n-nitrosoguanidine and a mutant(S13-E1), showing 2-fold higher killer toxin activity than that of parent strain to killer sensitive strain, Saccharomyces cerevisiae ATCC 38026 was obtained. Hansenular capsulata S13-E1 showed strong killer toxin activity to Saccharmyces mellis and Saccharomyces sal년 and four strains of gas-producing yeasts from traditional Doenjang and Kochujang. The culture condition for killer toxin production by Hansenular capsulata S13-E1 was optimized to be 1.0% potato extract, each 0.5% of peptone and glucose, and 0.025% MgSO4 with initial pH 4.5 at 3$0^{\circ}C$ and 36 hr of batch cultivation.

연속 발효조를 이용한 soluble glucan 생산 공정 개발 (Development of Soluble Glucan Production Process with Continuous Stirred Tank Fermentor)

  • Continuous fermentation process for the production of soluble glucan using mutant Agrobacterium sp. ATCC31750 has been developed in this research. When the concentration of soluble glucan was higher than 6 g/l, the viscosity of the fermented broth was too high and it needs complex separation process to separate from culture broth. Mathematical models which describe the cell growth and glucan production was developed and they kinetic parameters were estimated with experimental data. They are used for the optimization of continuous fermentation process and calculate optimal dilution rate for easy separation of glucan 4 g/l. With continuous fermentation, glucan production rate was increased 1.8 times more than that with batch fermentation.
