Optimization of Outdoor Cultivation of Spirulina platensis and Control ofContaminant Organisms (Spirulina platensis의 옥외배양 최적화 및 오염생물 구제)

  • Outdoor cultivation of cyanobacterium Spirulina platensis was carried out for 40 days in a batch mode. A half concentration of the SOT based on the underground water was used as culture medium. Working volume was 5.7 tons with 0.2 m depth. During cultivation, mean water temperature, DO and light intensity were all in proper conditions for the S. platensis growth. The adjustment of pH to over 10 with Na2CO3 and addition of the 1.5% natural salt were very effective to delete contaminant organisms, Chlamydomonas moewusii and Chlorella minutissima occurred one after the other in the culture. The mean productivity of the biomass based on the dry cell weight from 14 to 25 days, after the contaminants were deleted, was 7.8 g ·m–2· d–1, which was relatively high productivity in that a half concentration of the SOT was used for the culture. Underground water used in the culture minimized contaminants invasion and addition of the 1.5% natural salt was effective to delete contaminants as well as acted as mineral supplement in outdoor cultivation of S. platensis. Harvesting using the floating activity of S. platensis was effective from mass floating in day time after overnight without agitation and illumination.

  • Strains degrading and decolorizing reactive dyes, Procion blue HEGN and Procion red HE7B were isolated from water system, are named as RBK1 and RRK, the growth characteristics of which were investigated. Decolorization efficiencies after 42 hrs in batch culture were 95% and 77%, respectively. and the optimal culture condition of temperature and pH were $30^{\circ}C$, 7.0. Decolorization efficiencies in condition of aerobic shaking culture by strains RBK1 and RRK conspicuously increased, and culture by strain RBK1 was found as 95% after 42 hrs, while standing culture was 64%, Optimum nitrogen source was peptone, and It was found that decolorization efficiencies by strains RBK1 and RRK increased up to 4,000mg/l of peptone concentration as nitrogen source, but peptone concentration did' nt influence the decolorization efficiency in above 4,000mg/l. When the concentration of dyes were more than 800 mg/l and 400 mg/l respectively, the strains RBK1 and RRK, which degrade Procion bule HEGN and Procion red HE7B, showed a sharply decreased decolorization efficiencies; then the specific growth rate were $0.25hr^{-1}$ and $0.09hr^{-1}$.

  • We attempted to modulate the overall protein expression rate through the addition of a repressor against the araBAD promoter system of Escherichia coli, in which glucose was used as a repressor. Therefore, 0.5% L-arabinose was initially contained as an inducer in culture medium, and either 2% glucose or 2% glycerol was used as a carbon source, and it was found that the expression of recombinant interferon-${\alpha}$ could be observed at the beginning of the batch culture when glycerol was used as a carbon source. However, when glucose was used, the initiation of recombinant interferon-${\alpha}$ expression was delayed compared with that when glycerol was used. Furthermore, when the addition of 0.5% glucose was carried out once or twice after 0.5% L-arabinose induction during DO-stat fed-batch culture, the distributions of soluble and insoluble recombinant interferon-${\alpha}$ were modulated. When glucose was not added after the induction of L-arabinose, all of the expressed recombinant interferon-${\alpha}$ formed an inclusion body during the later half of culturing. However, when glucose was added after induction, the expressed recombinant interferon-${\alpha}$ did not all form an inclusion body, and about half of the total recombinant interferon-${\alpha}$ was expressed in a soluble form. It was deduced that the addition of glucose after the induction of L-arabinose might lower the cAMP level, and thus, CAP (catabolite activator protein) might not be activated. The transcription rate of recombinant interferon-${\alpha}$ in the araBAD promoter system might be delayed by the partial repression. This inhibition of the transcription rate probably resulted in more soluble interferon-${\alpha}$ expression caused by the reduction of the protein synthesis rate.

The Optimum Culture Condition for the Increasement of Biosurfactant Produced by Pseudomonas aeruginosa F722 (생물계면활성제 생산증가를 위한 Pseudomonas aeruginosa F722의 최적배양조건)

  • The hydrogen-producting bacterium was isolated from fresh water and identified as Enterbacter cloacae. The isolated was named Enterobacter cloacae YJ-1. In batch culture, The optimum cultivation temperature and pH of strain YJ-1 was 35$^{\circ}C$ and 7.5, respectively. All of the added glucose was consumed completely during fermentation even though pH was not controlled. Amount of hydrogen produced on each condition of 2% glucose, 4% sucrose and 5% fructose was 950, 1000 and 948 mL/L, respectively and resulted in increasing hydrogen production approximately 2.5-times more than controlled condition. The macimum hydrogen production was obtained when 50mM phosphate was added. was obtained when 50mM phosphate was added. In repeated0batch culture, yeast extract, but the production amount was not changed on the condition of over 0.5%, Most of the organic acides produced during the fermentation were formic and acetic acid, and propionic acid was moiety also generated.

The Production of Sodium Gluconate by Aspergillus niger (Aspergillus niger를 이용한 글루콘산 나트륨의 생산)

  • Sodium gluconate was produced by neutralization of gluconic acid formed during the submerged culture fermentation of glucose with Aspergillus niger ACM 7. The fermentation characteristics of Aspergillus niger ACM 7 were investigated quantitatively according to the change of the initial glucose concentrations and the initial pHs of fermentation broth. The maximum specific growth rate was estimated to be $0.20hr^{-1}$ at 95g/$\ell$ of initial glucose concentration. The maximum fermentability of sodium gluconate was 95% at the initial glucose concentration of 26g/$\ell$. However, the maximum sodium gluconate productivity was 1.18g/$\ell$/hr when the initial glucose concentration was 110g/$\ell$. The optimum pH was found to be 5.5 for both the cell growth and the sodium gluconate production. With optimized culture conditions, the productivity of sodium gluconate in a fed-batch culture(production fermentor, 16,000$\ell$) increased up to 7.1g/$\ell$/hr.

  • For poly- $\beta$ -hydroxybutyrate (PHB) production, a pink-pigmented facultative methylotrophic bacterium (PPFM) P-10 was newly isolated from soils through methanol-enrichment culture. The optimal medium composition for cell growth was 1.0% (vlv) of methanol as carbon source and l.Og/l of ,TEX>$NH_4Cl$, equivalent to C/N ratio of 13.2 at pH 7.0 and $30^{\circ}C$. To investigate the optimal condition for YHB accumulation, two-stage culture technique was adopted; first stage for cell growth and second stage for accumulation of PHB providing unbalanced growth conditions. The optimal PHB accumulation was 1.0% (vIv) of methanol and 0.26gll of $NH_4Cl$, C/N of 50.8 at pH 6.0. To overcome methanol inhibition on cell growth, intermittent feeding fed-batch culture technique was employed, and the cell concentration as high as 14gll with 40% of PHB was achieved. The purified PHB was identified using IR and $1^H NMR$ as homopolymer of 8hydroxybutyric acid. The absorption spectrum of extracted pink colored microbial pigment was alsa investigated.

Medium Composition of Enterobacter cloacae YJ-1 for Maximizing Hydrogen Production (수소생산 증진을 위한 Enterobacter cloacae YJ-1의 배지조성)

  • In order to maximize hydrogen production by Enterobacter cloacae YJ-1, anaerobic hydrogen producing bacteria, the medium composition was optimized. Glucose was better than other carbon sources in hydrogen production and its production was 975.4 mL/L at $2\%$ (w/v) for 48 h. Organic nitrogen sources were more effective than inorganic nitrogen sources and also yeast extract among organic nitrogens was the most effective in hydrogen production. Among metal ions, $Na_2MoO_4$ was most effective, and its production was 1753.3mL/L at $0.04\%$ (w/v). Addition of amino acid was very effective with compare to another components of medium, and cystein was most effective among them. Under the optimum medium obtained in batch culture, semi-batch culture in order to produce continuous hydrogen was run. The highest hydrogen production was earned at $3\%$(w/v) of glucose and the amount was 2215.4 mL/L.

  • A process for the production of bioplastics from wastewater with an open microbial culture was developed and evaluated. The process consists of a selection reactor to select bacteria in feast/famine regime and an accumulation reactor to produce PHA using selected bacteria. Polyhydroxyalkanoate(PHAs) accumulating bacteria could be efficiently grown in a sequencing batch reactor(SBR) without any growth limitation. For the high production of PHA limitation such as oxygen and nutrients seemed to be needed. Accumulation experiments were performed to evaluate the level of accumulation of PHA. Limited aeration had no effect, but nutrients limitation showed high accumulation. Bacteria which were selected in the SBR could accumulate PHA till 60% of cellular dry weight in accumulation experiments under nitrogen limitation. PHA accumulation rate decreased with increasing PHA content in the cells. Clearly, PHA accumulation rate has a strong correlation with the PHA content of the cells.

  • The three speices of miroalgae (Chlorella vulgaris, Dunaliella salina and Porphyridium purpureum) were immobilized in Ca-alginate capsules as a basic study for development of economic cultivation process, and then were cultivated in an air-bubble column bioreactor. Under the batch culture of aerobic conditions, the thickness of the capsule membrane and $CO_2$ supply did not affect the growth of the immobilized microalga, Chlorella vulgaris. Cell concentration of immobilized microalgae in the capsule was higher than those of imobilized microalgae in beads and free cells. The cell concentration of microencapsulated Dunaliella salina was greater about 5 times than that of free cells. Based on these results, it is concluded that the application of microencapsulation technology to the culture of microalgae was an effective method for high-density cultivation.

  • This study investigated on the adsorption of nonpoint pollution source using the Sand, hydroxyapatite(HAP), Zeolite and mixed culture. The adsorption of nonpoint pollution source on Sand, hydroxyapatite(HAP), Zeolite and mixed culture was investigated during a series of batch adsorption experiments. After the batch absorption experiments analysed COD, T-N, T-P on adsorption water. The experimental data was analysed using the pseudo-first-order adsorption kinetic models. Langmuir and Freundlich isotherm models were tested for their applicability. The maximum adsorbed amount $(Q_{max})$ of COD were found to be sand 0.0511mg/g, HAP 0.1905mg/g, Zeolite 1.0366mg/g and Mixed media 0.7444mg/g. The maximum adsorbed amount $(Q_{max})$ of T-N were found to be sand 0.0159mg/g, HAP 0.0537mg/g, Zeolite 0.5496mg/g and Mixed media 0.1374mg/g. The maximum adsorbed amount $(Q_{max})$ of T-P were found to be sand 0.0202mg/g, HAP 0.1342mg/g, Zeolite 0.0462mg/g and Mixed media 0.1180mg/g. As a result, the mixed media was effective to remove nonpoint pollution source.
