Radiation Sensitivity of Basidiospore and Mycelium in Pleurotus ostreatus

  • To assess the effects of gamma-ray (Co-60) on radiation sensitivity and genetic similarity of the basidiospore and mycelium in oyster mushroom, Pleurotus ostreatus, the D$_{10}$ values and RAPD patterns were analysed. Three strains were isolated from basidiospores (PO-Bl, -B2, and - B3 from 2 kGy irradiation group) and five strains from mycelia (PO-Ml, -M2 from 1 key, PO-M3 from 2 kGy, and PO-M4 and -M5 from 2+1 key irradiation group). The D$_{10}$ values of basidiospore and mycelium of P. Preurotus ostreatus were 1,250 Gy and 500 Gy, respectively. The growth rates of the eight strains on the five media were various and the activities of extracellular chitinases of them were generally higher than those of the control. By the gamma-ray radiation, 22-25% of genetic similarities were changed in the basidiospore strains and 23-36% of them in the mycelium strains. From these results, it seems that the basidiospore could be more radio-resistant than the mycelium of P. ostreatus and that the genetic similarity of the mycelium of P ostrentus could be changed easier than that of the basidiospore by the gamma-ray radiation.ion.

  • Apexes of basidia in Rhodophyllus muraii var. albus are divided into four sections or depressed in the center. A spore is formed by inflation of the apex of the sterigma. The apex of the sterigma is swollen and changed from a papilla through a peinsform and a club into a globeform. Six spots of globose spore are regularly or irregularly depressed with hilum axes. Finally the spores come cuboid.

  • Four sterigmata of papillaform E. violaceobrunneum are developed from crator-shaped basidium. The apex of sterigma is swollen to a small globeform. And then it is swollen to a clubform. The clubform is again swollen to a ellipticalform, and then more than six spots of spore surfaces are ramdomly depressed with hilum axes. When the depression of surface of a elliptical spore is over, the spore is a heterodiametrical spore of multi-angular.

  • Germination of the single-basidiospore of Agaricus bisporus, mycelial characteristics of the single spore culture and its fruit body formation were studied. Germination of the single basidiospore was the best on water agar. Of 6372 spores isolated, 1622 spore were germinated and the germination rate was 26 percent. Cultures from single basidiospores showed two distinct mycelial types on the compost extract sucrose agar: one was the strandy type and the other was the fluffy. The strandy type culture produced more sporophores than the fluffy. Of 778 strandy isolates screened, nine cultures yielded 5 percent higher than the origine. No. 1567 and 1708 were selected for the commercial cultivation.

  • Ganoderma lucidum, a divine mushroom, beared the nonbasidiocarpus basidiospore without the normal basidiocarp. The fungus formed the basidial stroma from the aerial mycelium on agar media, and the basidium was developed from apical cells of the mycelium on the surface of the basidial stroma. One to four basidiospores were observed on a basidium. The nonbasidiocarpous basidiospores formed on the basidial stroma were similar to the normal basidiospores in morphology, although the nonbasidiocarpus basidiospores were slightly smaller in size. However, the number of hollow on the spore surface was remarkably smaller than that of the normal spore. The formation of nonbasidiocarpous basidiosproes was influenced by the genetic factor and the environmental factors such as light, temperature and ventilation.

Proton Beam Sensitivity of Basidiospore and Mycelium in Pleurotus ostreatus (느타리버섯 포자와 균사체의 양성자 빔 조사에 대한 민감도)

  • To assess the effects of proton beam on radiation sensitivity of the basidiospore and mycelium of oyster mushroom (Pleurotus ostreatus), the $D_{10}$ values and $L_{50}$ (lethal 50%) values were analysed. By the proton beam radiation, the survival rate and germination rates increased at the dose of $10\;Gy{\sim}100\;Gy$ and then decreased significantly over 500 Gy. $L_{50}$ values of basidiospore and mycelium of Pleurotus ostreatus were over 500 Gy and 400 Gy, respectively. $D_{10}$ values were calculated from linear regression formulae ($D_{10}\;=\;-1/slope(b)$, y = a + bx) as 750 Gy and 1,250 Gy, respectively. Based on our experiment, the optimum dose of proton beam as a mutation source would be between from 500 Gy to 750 Gy for basidiospores and from 400 Gy to 1000 Gy for mycelium of oyster mushroom.

The apex of the sterigma is swollen and changed from a papilla through a penisform and a club into a globeform. Six spots of globose spore are regularly or irregularly depressed with hilum axes. Finally the spores are cuboid. Basidia of Entoloma squamiferrum are developed from hymenium layer of crator-shaped parabasidium. Apex of basidium is flat or depressed in the center. Although four sterigmata are developed, only two sterigmata are symmetrically swollen to two spores in certain basidia. It means that two sterigmata among four sterigmata are infertile. The apex of the sterigma is swollen into a paillaform, and then turned into a penisform. It is swollen from a clubform into a globeform. Six spots of the surfaces of globose spore are deperssed with hilum axes. Finally the spore is cuboid, and then it is released from hilum. Four sterigmata of papillaform of E. violaceobrunneum are developed from cartor-shaped basidium. The apex of sterigma is swollen to a small globeform. And then it is swollen to a clubform. The clubform is again swollen to a ellipticalform, and then more than six spots of spore surfaces are ramomly depressed with hium axes. When the depression of surface of a elliptical spore is over, it is the multi-angular spore of the heterometrical-form.

Isolation of Genes Specifically Expressed in Different Developmental Stages of Pleurotus ostreatus Using Macroarray Analysis

  • The oyster mushroom (Pleurotus ostreatus) is one of the most important edible mushrooms worldwide. The mechanism of P. ostreatus fruiting body development has been of interest both for the basic understanding of the phenotypic change of the mycelium-fruiting body and to improve breeding of the mushrooms. Based on our previous publication of P. ostreatus expressed sequence tag database, 1,528 unigene clones were used in macroarray analysis of mycelium, fruiting body and basidiospore developmental stages of P. ostreatus. Gene expression profile databases generated by evaluating expression levels showed that 33, 10, and 94 genes were abundantly expressed in mycelium, fruiting body and basidiospore developmental stages, respectively. Among them, the genes specifically expressed in the fruiting body stage were further analyzed by reverse transcription-polymerase chain reaction and Northern blot to investigate temporal and spatial expression patterns. These results provide useful information for future studies of edible mushroom development.

Identification of Some Phellinus spp.

  • Four strains of Phellinus spp. was identified based on internal transcribed spacer(ITS) region of rDNA sequence analysis and morphological characteristics. Basidiocarps of all strains were effused-reflexed and hymenial surface was poroid. Hyphal system was dimitic and basidiospore was globose to ellipsoid. The amplification of ITS regions produced a DNA fragment of 500 to 780 by in all strains examined. The determined sequences were analyzed for the reconstruction of phylogenetic tree. From these results, Phellinus sp. KM-1, KM-2, and KM-4 was identified as P. hartigii, P. baumii, and P. linteus, respectively.

Two New Records of Scleroderma Species (Sclerodermataceae, Boletales) in South Korea

  • Two new records of Scleroderma species from South Korea are described here. Comprehensive taxonomic studies of Scleroderma specimens were conducted at the Korea National Arboretum. Based on morphological and molecular data (fungal barcode sequences), two new records (S. laeve and S. nastii) were confirmed. Herein, morphological descriptions, including Scanning Electron Microscope (SEM) images of basidiospore ornamentation, and a taxonomic key of Korean Scleroderma species are provided.
