Ribonucleic acid contents and mononucleotides distribution from the mycelium and fruit bodies of Ganoderma lucidum were studied. P.E.I. cellulose TLC and HPLC were applied in this study. The obtained results are as follows; The levels of ribonucleic acids from the young basidiocarp mycelium were higher than those of mature basidiocarp. Guanosine 5'-monophosphate and xanthosine 5'-monophosphate were found in both young basidiocarp mycelium and mature basidiocarp. The levels of guanosine 5'-monophosphate and xanthosine 5'-monophosphate from the young basidiocarp were higher than those of the mature basidiocarp. However, inosine 5'-monophosphate was not detected.

The context and upper surface of Phellinus basidiocarp become blackened, rimose and woody. The basidiocarp is sessile, dimidiate and elongate. The basidiospores are pigmented and ovoid to globose. Hymenial setae are $17{\sim}35{\times}6{\sim}8{\mu}m$. Nineteen isolates of Phellinus species, including Phellinus linteus, were used for sequencing of the internal transcribed spacer (ITS) region of the nuclear rDNA. Based on these sequence data, specific primers were designed for identification of Phellinus linteus isolates in Korea. The specific primers were within the ITS1 and ITS2 regions and were nested within the universal primers flanking the spacer regions. A total of four primers (the universal primers ITS-1F and ITS-4, and the specific primers PL-F and PL-R) were used for detection of Phellinus linteus collected in Korea. The length of the four amplification products of Phellinus linteus DNA were 800 bp (ITS-1F/ITS-4), two bands of about 720 bp (ITS-1F/PL-R and PL-F/ITS-4), and 610 bp (PL-F/PL-R). Among 23 isolates of Phellinus species collected in Korea, Thirteen isolates were identified as Phellinus linteus based on the presence of the four bands. The other species produced only the single ITS-1F/ITS-4 product.
An unrecorded Ceriporiopsis species was collected at Mt. Gariwang, Gangwon Province, in 2008. Based on morphological characteristics, such as a fully resupinate basidiocarp, a reddish white to pinkish poroid hymenophore and a monomitic hyphal system with clamp connections, the species was identified as Ceriporiopsis resinascens. This is the first report of Ceriporiopsis resinascens in Korea. We confirmed the identity of the species as Ceriporiopsis resinascens based on ITS sequence analysis.
The optimal conditions for mycelial growth of Phellinus linteus ATCC 26710 were determined to be a log length of 20 cm, temperature of $30^{\circ}C$ and pH of 6.0. Mycelial growth was excellent on the mushroom complete medium, and was optimal when sucrose, man nose and glucose were supplied as carbon sources. Potassium nitrate and sodium nitrate as nitrogen sources supported good mycelial growth. To evaluate P. linteus mycelial colonization on logs, sterilized short log inoculation, drilling inoculation and log-end sandwich inoculation techniques were used. Only sterilized short log inoculation produced good mycelial colonization. Initial mycelial growth and full mycelial colonization were best on 20 cm logs having 42% moisture content. The initial mycelial growth of P. linteus was accelerated over 12hr of sterilization. Basidiocarp formation was optimal using a burying method of logs after $5{\sim}6$ months, and fruiting body formation was superior in cultivation house conditions of $31{\sim}35^{\circ}C$ and in excess of 96% relative humidity.
Artificial cultivation of Phellinus linteus was studied. The best yield (B.E. $9.4{\pm}1.6$) of basidiocarps including basidiospore was obtained using Quercus acutissima among the logs of Morus alba, Quercus acutissima, Alnus japonica, Castanea crenata. Optimum temperature and required periods for the basidiocarps formation were $25^{\circ}C$ and about 10 months, respectively.
The Aphyllophorales is a large order containing about 2,000 known species. Many of these are the bracket and coral fungi. The vast majority of these fungi are saprophytic on the plant debris. Many species are significant in decomposing plant remains, as they are able to digest cellulose or lignin that occurs in plant cell walls. Many of these fungi have been involved in everyday human affairs. A few were used medicinally by the Greeks and Romans as a remedy for many complaints, including colic, fractured limbs and bruises. Other bracket fungi have been used as curry combs for horses, as snuff, as razor strops and as a source of dye for clothing. The texture of the basidiocarp may be similar to that of cork, wood, leather, paper, or cartilage. Unlike the basidiocarps of the Order Agaricales, the basidiocarps of the Aphyllophorales are not fleshly and moist. Division of the members of the Aphyllophorales into genera was originally made on the basis of gross morphology of the basidiocarp and hymenium and Donk(1964) recognizes 22 families in this order. The species and genus whose typical in Aphylloporales were listed in Table. with related information. The ITS region sequence of some genus were found by BLAST search. Sequences retrieved from GenBank were visually aligned by the program CLUSTAL G. As a result, the medicinal mushroom was separated in four groups. In this multiple alignment, the sequence analysis among Fomes group, Inonotus group and Phellinus group showed high genetic similarity except Hericium group and Sparassis group.

Interspecific hybrids of Pleurotus ostreatus and Pleurotus florida were formed by using protoplasts of complementing auxotrophs. The genetic markers were shown to segregate and recombine in the first generation of monosporus isolates from basidiocarp of seven fusion products. The analysis provides proof of heterokaryosis and strong evidence for haploidy of vegetative nuclei, a sexual cycle consisting of nuclear fusion and meiosis. In all the crosses there was no evidence of linkage between the genetic markers. Clamp connections were formed in monosporus mycelia from basidiocarp of fusion products.

With universal primer ITS1-F, the specific DHJ2 primer was developed to detect the Ectomycorrhizal(ECM) root tips in soil and to identify the species of ECM fungi, as based on DNA sequences of rDNA stored in GeneBank of NCBI. This primer was designed with the common sites of rDNA of Amanita and Boletus, and was also designed with several DNA programs provided by NCBI. The DNA fragments synthesized by PCR were calculated to be 1,000 to 1,200 bps of DNA located to 18s to 28s rDNA to contain two variable sites of ITS, indicating much diversities for specific species or ecotypes of ECM fungi. The primer DHJ2 reacted with the genomic DNA's extracted from the tissues of basidiocarp at the rate of 73 of 80 fungi collected produced single bands with a 1,100 bps length. The DNA fragment synthesized with the genomic DNA that extracted from eight ECM tips of Pinus densiflora was confirmed and analysized to the rDNAs of ECM in full sequences, and informed to be a ECM fungal species in the forest.

Nine species of genus Ganoderma collected in Korea and abroad including Ganoderma lucidum complex and G. lucidum were compared by investigating growth characteristics. In the bottle culture, the mycelial growth periods of G. lucidum from Taiwan and North America was 26 to 30 days compared to that of Korean G. lucidum, which was 30 to 32 days. Cultivation period of Taiwan and North American isolates was 30 to 32 days which were 11 to 17 days shorter than those of Korean isolates. Biological efficiency of Taiwan and North American isolates were ranged from 3.3 to 5.5%, which were apparently lower than that of Korean isolates which ranged from 6.2 to 9.4%. Korean isolates had longer stipes($15{\sim}40$ mm) and more number of pileus($4{\sim}6$/bottle) than those of Taiwan and North American isolates. The G. lucidum isolates collected from Korea will be regarded as the independent species from the G. lucidum collected from Taiwan and North America since, the G. lucidum from Korea showed much different growth characteristics in various aspects compared to the G. lucidum from Taiwan and North America.
Composition of fatty acids (FAs) in Pleurotus ostreatus were analyzed by gas chromatography and compositional changes according to cultivar, fruitbody section and developmental stage (days after primordia formation) were investigated. Major FAs in oyster mushroom were linoleic acid, oleic acid and palmitic acid. Unsaturated FAs were higher in pileus portion than upper- or lower-stipe portion. Oleic acid content was increased with developmental days but linoleic acid content was highest at $3{\sim}4$ days after primordia formation and then decreased after that. And ratio of unsaturated FAs/saturated FAs was decreased with basidiocarp maturation.
