A Study of Analytical Method for Trace Metal Ions in Whole Blood and Urine by Inductively Coupled Plasma-Mass Spectrometry using Solid-Liquid Extraction Technique (유도결합 플라스마-질량분석법과 고체-액체 추출법을 이용한 혈액 및 소변중 미량금속의 분석에 관한 연구)

  • An analytical method for the simultaneous measurement of trace Cu, Sn, and Bi in blood and urine has been investigated by Inductively Coupled Plasma-Mass Spectrometry (ICP-MS). Microwave oven was used for the pretreatment of blood samples using nitric acid and hydrogen peroxide in a closedvessel digestion system with 1 mL whole blood for 8 minutes. Amberlite IRC-718 resin was used as a solid phase in solid-liquid extraction technique for the removal of matrix interferences such as Na, S, P, and other polyatomic ion species. Detection limits for Cu, Sn, and Bi by this method were 0.000375 ng/mL, 0.000297 ng/mL, and 0.000174 ng/mL, respectively. Recoveries of 99.1% for Cu, 102.5% for Sn, and 98.4% for Bi were obtained for the standard spiked NIST SRM 955a blood sample. The developed method was applied for whole real blood and urine samples.

  • Laser induced breakdown spectroscopy (LIBS) is a recently developed analytical technique that is based upon the measurement of emission lines generated by atomic species close to the surface of the sample, thus allowing their chemical detection, identification and quantification. With powerful advantages of LIBS compared to the conventional analytical methodology, this technique can be applied in the detection of heavy metals in the field. LIBS allows the rapid analysis by avoiding laborious chemical steps. LES have already been applied for the determination of element concentration in a wide range of materials in the solid, liquid and gaseous phase with simplicity of the instrument and diversity of the analytical application. These feasibility of rapid multi elemental analysis are appealing proprieties for the in-situ analytical technique in geochemical investigation, exploration and environmental analysis. There remain still some limitations to be solved for LIBS to be applied in soil environment as an in-situ analytical technology. We would like to provide the basic principle related to the plasma formation and laser-induced breakdown of sample materials. In addition, the matrix effect, laser properties and the various factors affecting on the analytical signal of LIBS was dealt with to enhance understanding of LIBS through literature review. Ultimately, it was investigated the feasibility of LIBS application in soil environment monitoring by considering the basic idea to enhance the data quality of LIBS including the calibration method for the various effects on the analytical signal of LIBS.

  • In this work, we compare the analytical solutions with the numerical solutions for photothermal interactions in semiconductor medium containing cylindrical cavity. This paper is devoted to a study of the photothermal interactions in semiconductor medium in the context of the coupled photo-thermal model. The basic equations are formulated in the domain of Laplace transform and the eigenvalue scheme are used to get the analytical solutions. The numerical solution is obtained by using the implicit finite difference method (IFDM). A comparison between the analytical solution and the numerical solutions are obtained. It is found that the implicit finite difference method (IFDM) is applicable, simple and efficient for such problems.

  • In this study, inter-laboratory comparison was done using elemental analyzer-isotope ratio mass spectrometers (EA-IRMSs) to determine carbon and nitrogen contents as well as stable carbon and nitrogen isotopic compositions (${\delta}^{13}C$ and ${\delta}^{15}N$) of five environmental samples containing lake and marine sediments, higher plant leaves, and fish muscle, and one organic analytical standard (Protein (Casein) Standard OAS). Five national laboratories participated in this comparison study, and each laboratory analyzed all five samples and the analytical standard. Results showed that variations in total organic carbon (TOC) and total nitrogen (TN) contents as well as ${\delta}^{13}C_{TOC}$ and ${\delta}^{15}N_{TN}$ values among the laboratories were large compared to the analytical uncertainties. The results highlighted the inhomogeneity of the test samples and thus, the need to select suitable standard reference materials for future inter-laboratory studies. Further inter-laboratory comparison exercises could promote good measurement practices in the acquisition of stable carbon and nitrogen isotopic composition data.

Determination of Polybrominated Diphenyl Ethers(PBDEs) in Soil using Gas Chromatography/Isotope Dilution Mass Spectrometry (기체크로마토그래피/동위원소 희석 질량분석법을 이용한 토양 중 폴리브롬화 디페닐에테르의 분석법)

  • An analytical method of polybrominated diphenyl ethers in soil samples by isotope dilution method using gas chromatography/mass spectrometry (GC/MS)-selected ion monitoring (SIM) was described. PBDEs in soil were extracted with soxhlet extractor and then silica and florisil solid phase extraction (SPE) methods as purification of extract were compared. After clean-up, the extractions were analyzed by GC/MS with SIM mode. Quantitation was performed isotope dilution method using four $^{13}C$ isotopically labeled PBDEs as internal standards. This developed method was validated for eight congeners of PBDEs in the concentration range 0.04~4 ng/g in soil and the average recovery of the analytes ranged 30.8~110.8% for florisil and 44.4~110.7% for silica, respectively. The method detection limits of PBDEs were 0.04~0.3 ng/g.

PAMAM Dendrimer Conjugated with N-terminal Oligopeptides of Mouse Fibroblast Growth Factor 3 as a Novel Gene Carrier

  • In this study, we introduced the RRLR oligopeptide sequences on the surface of polyamidoamine (PAMAM) dendrimer and characterized the physical properties and gene carrier activity of the novel polymer using HEK 293, NIH3T3, and HeLa cells. The RRLR peptide sequences were derived from a mouse fibroblast growth factor 3 (FGF3) protein containing a bipartite NLS motif. The entire sequence of FGF3 is RLRRDAGGRGGVYEHLGGAPRRRK and it has two functional sequences RLRR and RRRK at N-terminus and C-terminus, respectively. In particular, PAMAM G4-RRLR conferred enhanced transfection efficiency and lower cytotoxicity compared with those of PEI 25 kDa, PAMAM G4-R, and PAMAM G4 in various cell lines. These results suggest that the introduction of N-terminal oligopeptides of FGF3 on the surface of PAMAM holds promise as an effective non-viral gene delivery carrier for gene therapy.

  • Simultaneous determination of mercury and arsenic has been studied by reductive vapor generation-ICP-AES. Samples were digested with a microwave digestion system and extracted with acids. Reductive vapor generation was carried out with 6N HCI and 2% $NaBH_4$. Detection limit of Hg and As are found to be 0.06 ppb and 0.08 ppb, respectively. Measured values of Hg and As in inorganic samples agree well with reference value of SRMs, but the recovery of As from organic samples is obtained approximately 80% of the reference values.

  • Recently there have been increasing consumers' interests in the geographical origin of foods, due to the FTA (Free Trade Agreement) in the global market. Especially, in Korea, in relation to BSE (bovine spongiform encephalopathy), it is considered to be urgent to develop analytical techniques for distinguishing the geographical origin of beef. Korea is facing conclusion of FTA with many countries, and there is a deep national concern about the distinction of the geographical origin of food. Diverse analytical techniques have been used in many of recent researches to obtain data for distinguishing the geographical origin of foods produced in various countries. In this paper, we reviewed national and international researches about tracing of geographical origin and food authentication using stable isotopes. Improvement of the isotopic techniques and their numerous application have been provided useful information of their geographical origin in food products. Furthermore, we expect that this study could be detecting of many frauds and illegal transaction of food products. We look forward to active progressing research of detecting food origin using isotope analysis and numerous application about imported food products.

  • Nuclear materials such as uranium are used as fuel for nuclear power generation, but there is a high possibility that they will be used for non-peaceful purposes, so international inspections and regulations are being conducted. Isotope analysis data of fine particulate obtained from nuclear facilities can provide important information on the origin and concentration method of nuclear material, so it is widely used in the field of nuclear safety and nuclear forensics. In this study we describe the analytical method that can directly identify nuclear particles and measure their isotopic ratios for fine samples using a large-geometry secondary ion mass spectrometer and introduce its preliminary results. Using the U-200 standard material, the location of fine particles was identified and the results consistent with the standard value were obtained through microbeam analysis.

  • Nitrophenols are a group of important priority pollutants classified by the US Environmental Protection Agency. Reverse-phase liquid chromatographic method combined with mass spectromethy employing atmospheric pressure chemical ionization(APCI) interface is utilized to determine a mixture of nitrophenols in water matrix without any pretreatment. The sensitivity and selectivity for the identification of different kind of nitrophenols is enhanced by the use of selected ion monitoring and cone voltage fragmentation. The fragmentation patterns of nitrophenols are compared with those obtained from the collision induced dissociation(CID) MS/MS technique.
