The major cultured marine fishes in sea off the coast Gyeongsangnam-do Province, South Korea, were assessed and included 9.3% rockfish Sebastes schlegelii, 7.8% red seabream Pagrus major, and 2.1% rock bream Oplegnathus fasciatus. The number of insurance payments related to disease mortality in cultured fish in 2017 was fourfold that in 2016. Economic loss in aquaculture due to disease in cultured fish is high and represents an important inhibitory factor affecting marine fishery productivity. In 2018, diseases led to severe production losses in several aquaculture species: 40.0% in rockfish, 11.4% in olive flounder Paralichthys olivaceus, 10.0% in filefish Thamnaconus modestus, and 9.3% in red seabream. Fish-parasitic pathogens such as Microcotyle sebastis, Alella spp., and Dactylogyrus spp. enter mainly via the gills and skin surface. Among bacterial pathogens, Vibrio species were most common, with Vibrio harveyi being the dominant species causing infections in these fishes. The bacterium Lactococcus garvieae is thought to exhibit host specificity in fish. The fish species in the present study exhibited a higher tendency for infection by heterologous pathogens than by a single pathogen; therefore, it is necessary to devise new strategies for treating diseases in cultured fish.

The zebra shark Stegostoma fasciatum which had been reared in the commercial aquaria was found dead and submitted for postmortem examination. A pure bacterial culture was isolated from pale and enlarged liver. The analysis of ureC and 16S rRNA genes confirmed the isolate as Photobacterium (P.) damselae subsp. damselae and this pathogen was sensitive to gentamicin. Although, no mortality in mouse was observed in the experimental infection study, the isolation of this pathogen in aquarium fish is significant because it can act as a reservoir to other aquatic animals and can also be zoonotic potential to human during aquarium management. This paper describes the first isolation of P. damselae subsp. damselae from zebra shark.
Vibrio alginolyticus, a marine fish and shellfish pathogen, has been found at a high frequency around the coastal areas of Korea. Vibrio alginolyticus was purified from various diseased fish, and a V. alginolyticus-specific bacteriophage was isolated from seawater obtained from fish farms located on the west coast of Korea. In a bacterial lysis experiment using the phage and antibiotics, tetracycline, $10^3$ cfu/ml of V. alginolyticus were completely lysed by both the phage and the antibiotic, suggesting that the purified phage in the study could be utilized as an alternative to antibiotics in the control of fish and shellfish diseases caused by V. alginolyticus.
Gradual mortality of look down fish (Selene vomer) was observed in a private aquarium in Seoul, showing abnormal swimming behavior and lethargy. A bacterial pathogen from kidney was cultured, identified, and confirmed as Vibrio harveyi using Vitek System 2 and 16S rRNA gene sequencing. A predominant bacterial strain, SNUVh-LW2 was proved to be most closely related to isolates from China by phylogenetic analysis with minimum evolution method. Also, tetracycline was considered as the most sensitive antibiotic agent via antibiotic susceptibility test. The group of fish was treated according to the diagnostic result and no more mortality was observed.
Fish mycobacteriosis is a common bacterial disease in many species of freshwater and marine fish and has caused severe loss of fish production. Mycobacterium marinum has been the most prevalent pathogen observed in several outbreaks of mycobacteriosis of farmed sturgeons in China. However, the immune responses and pathology of sturgeons in mycobacterial infection are rarely studied. Therefore, we used the Illumina RNA-seq method to analyze the transcriptome profile of Acipenser schrenckii challenged with Mycobacterium marinum. To begin, 168,220 non-redundant contigs were acquired from the infection and control groups, and among these, 33,225 contigs have acquired annotations. A total of 4,043 differently expressed (DE) contigs between the two groups were identified, and among these, 2479 were up-regulated and 1564 were down-regulated in the infected fish. A total of 1,340 DE contigs with acquired annotations in KEGG were enriched for 124 pathways including the TNF signaling pathway, and the Toll-like receptor signaling pathway. The roles of DE genes involved in significant pathways and other processes were discussed. The 2,209 DE contigs that have yet to acquire proper annotation may represent candidate genes associated with infection in sturgeons and are expected to serve as immunogenetic resources for further study. To our best knowledge, this is the first transcriptome study on sturgeons under bacterial infection.
염농도에 따른 E. tarda CK41의 운동성을 알아보기 위하여 1.0%와 3.5%의 염농도를 가지는 운동성 측정 배지에서 집락의 변화를 관찰한 결과, 3.5% 염농도 조건에서 운동성이 감소하는 것을 확인할 수 있었다. 1.0%과 3.5% 염농도 조건에서의 생육도를 측정해본 결과 각 염농도 조건에 따른 생균수의 차이는 매우 적은 것으로 보아, 높은 염농도에서의 운동성의 감소는 생육정체가 아닌 실질적인 운동성의 차이에 의함을 알 수 있었다. 이러한 염농도에 의한 운동성의 차이가 편모에 의한 것인지를 알아보기 위하여 투과 전자 현미경으로 형태학적 관찰을 해본 결과, 3.5% 염농도에서는 편모의 형성이 되지 않음을 확인하였다. E. tarda는 PFAD와 FDP 두개의 편모 유전자를 가지며 이들간의 아미노산 상동률은 93%로 높은 편이다. 편모의 발현양의 확인을 위하여 PFAD 특이적인 다클론성 항체를 제작하기 위하여, PFAD를 과발현시키는 재조합 플라스미드 pBP793을 구축하여 대장균 발현시스템으로 발현시켜 정제한 후, 토끼에서 면역반응을 유도하여 특이 항체를 제작하였다. PFAD 특이적인 다클론성 항체를 이용한 immunoblot assay 결과, 3.5% 염농도 조건에서 배양한 E. tarda CK41의 경우 1.0% 염농도에서 보다 반응하는 면역 활성 단백질 밴드가 낮은 것으로 측정되었다. 이러한 결과를 종합하여 볼 때, 염농도가 높은 해수환경에서의 운동성의 감소는 E. tarda CK41의 편모 단백질이 제대로 발현되지 않아 기능적인 편모의 형성이 이루어지지 않는다는 것을 예증하고 있다. 향후 연구에서 어떠한 메카니즘에 의해 염농도가 flagellin의 발현을 조절하는지를 밝힐 필요가 있다.
A trap identification system for isolating functional sequences to allow the constitutive expression of foreign protein from Edwardsiella tarda was developed. Using the green fluorescent protein (GFP) reporter-based trap system, various functional sequences to drive heterologous expression of the GFP were selectable in Escherichia coli host. However from the bioinformatic sequence analysis, all the segments predicted as regulatory regions were not native promoters actually existing upstream of endogenous E. tarda genes. Instead, a number of non-authentic sequences, possibly resulted from the random shuffling and/or intermolecular ligation were also proven to be able to display a potent GFP expression in the recombinant E. coli. Further analysis with selected clones showed that both authentic and non-authentic sequences could function in as a constitutive promoter, leading quite a consistent and stable GFP expression after repetitive subcultures. Microscopic examination also confirmed the uniform pattern of GFP expression in every host bacterium. Semi-quantitative assay of GFP showed that there was no clear relationship between expression levels and organizational features of the promoters trapped. Functional promoter-like elements achieved in the present study could be a good starting material for multivalent genetic engineering of E. tarda in order to produce recombinant vaccines in a cost-effective fashion.
배양 온도, 배양 배지 중의 pH, NaCl 농도, iron 농도와 같은 배양 조건이 P. damselae의 미생물학적 성상 및 병원성에 미치는 영향을 조사하였다. P. damselae는 배양 온도 15-30${^{\circ}C}$에서 배지의 pH 5-9, NaCl 농도 0-6%와 $FeCl_{3}$ 첨가시 성장이 촉진되었지만, iron-chelator 첨가 시성장이 억제되었다. Iron-limited 조건에서 배양시 ECPs protein 농도, phospholipase, 용혈능, siderophore 및 cytotoxicity 활성 이 높게 나타났고, 넙치 혈청의 살균작용 대한 생존율도 증가 되었으나, iron-added 조건에서는 감소되어, 배양 조건 중 iron 농도에 영향을 가장 많이 받는 것으로 나타났다. 따라서, 본 연구에서 적용한 iron-limited 조건은 철농도가 낮은 숙주 내와 유사한 환경이므로 iron-limited에서 P. damselae 배양 시 나타나는 병원성의 증가는 어체에 감염시 나타나는 병원성 발현 기전과 유사할 것으로 사료된다.

Background: Live fish import may lead to the unintended introduction of pathogens. We examined the monthly distribution of microbial pathogens in ornamental finfish imported into South Korea over a 6-month period. Results: Vibrio alginolyticus was detected in one lemon damsel in June and July; V. vulnificus was detected in one lemon damsel, one caerulean damsel, and one pearl-spot chromis and one ocellaris clownfish in July, April, and May, respectively; Photobacterium damselae was detected in one ocellaris clownfish and one caerulean damsel in June and July, respectively; V. anguillarum was detected in one pearl-spot chromis in February; V. harveyi was detected in one ocellaris clownfish and two mandarin fish in February and April, respectively; Yersinia ruckeri was detected in a pearlscale goldfish group in June and July and in two colored carp groups in July; and Lactococcus garvieae was detected in a lemon damsel group and a sutchi catfish group in July and May, respectively. European catfish virus, the only viral pathogen detected, was found in two sutchi catfish groups in May. Conclusion: This study is the first to identify pathogenic species and the presence or absence of pathogens (non-quarantine diseases) in imported ornamental finfish. These results demonstrate that various pathogens with the potential to harm indigenous fish populations can accompany ornamental finfish imported into South Korea.
본 연구에서는 명태 양식 기술 개발 과정 중 수산생물 병원체의 영향을 알아보고자 사육수 내 총 세균수 변화, 분포와 명태에 대한 영향을 분석하였으며, 성장 단계별 명태의 병원체(세균, 기생충 및 바이러스) 조사 및 비정상 개체 발생에 대한 원인 구명을 수행하였다. 자어 단계에서 사육수의 평균 세균수는 $2.11{\times}10^3cfu/mL$로 나타났으며, 비브리오균이 52%로 가장 높게 분포하였다. 치어 및 미성어 단계에서 사육수의 세균수는 $3.43{\times}10^3cfu/mL$(슈도모나스균 90% 분포) 그리고 성어 단계에서는 $3.2{\times}10^2cfu/mL$(슈도모나스 52%)로 나타났다. 명태 사육수 세균총의 변화는 먹이생물의 종류, 공급 및 잔류량에 의해 영향을 받는 것으로 판단된다. 외관상 건강한 개체에서는 대량 폐사를 일으킬 수 있는 세균, 바이러스가 검출되지 않았으나, 일부 안구돌출, 턱 주변 조직 출혈을 보이는 비정상 개체에서 아가미 새변의 기포 형성, 각막 조직 및 맥락막 주변 조직 괴사가 관찰되었다. 특히, 병변 부위에서 검출된 세균(P. fluorescens, Vibrio sp., V. pelagius, Planococcus maritumus 그리고 Pseudoalteromonas sp.)이 사육수에도 동일하게 분포하였으며, 사육 과정 중 외상 발생 이후 2차 세균 감염이 발생할 수 있음을 시사한다. 명태의 안정적인 양식 산업화를 위한 요소로 기초 질병 데이터 구축과 더불어 지속적인 질병관리가 필요하다.
