Bacteriospermia is a frequent finding in fresh boar semen and can result in detrimental effects on semen quality and longevity. The objectives of this study was to evaluate types of bacterial contaminants in porcine fresh semen and the reducing effect of antibiotic and density gradient with percoll on the bacterial contaminants. Fresh semen was collected by gloved-hand method into a pre-warmed($37^{\circ}C$) thermostable bottle, and was inoculated onto blood agar and MacConkey agar, respectively. After incubated for 48 hour, 7.5% $CO_2$ at $37^{\circ}C$, bacterial colonies were selected and identified by Gram staining, oxidase test, catalase test and finally identified using API kits and Vitek system. Aerobic culture yielded a variety of bacteria from different genera. The most prevalent contaminant of fresh semen were Leclecia adecarboxylata, Acineobacter banmanni, Staphylococcus epidermidis, Staphylococcus cohni spp urealyticus, Proteus mirabilis. Most of identified bacteria were Gram(-) and non-pathogenic bacteria. It seems that bacterial contaminants in fresh semen were seem originated from multiple sources at the stud/farm, and were from animal and non-animal origins. Gentamicin treatment did not eliminate the bacterial contaminants completely but 3 step-density gradient with percoll completely removed the bacterial contaminants in fresh semen. Therefore, future study is necessary to prove that density gradient method with percoll can eliminate bacteria in fresh semen without significantly affecting sperm viability or function.
Bacteroiospermia is a frequently finding in fresh raw and extended porcine semen and can results in detrimental effects on semen quality and longevity. This study aims to evaluate the type of bacterial contaminants in raw and extended porcine semen and the reducing effect of antibiotic test. To investigate bacterial contaminants, out of 387 sample (raw semen 201, extended semen 186) were collected from 6 artifical insemination centers in Gyeongsangnam-do, were inoculated onto blood agar and MacKonkey agar, respectively. Bacterial colonies were selected after culturing for 48 hours, at $37^{\circ}C$, followed by Gram staining, KOH test, oxidase test, catalase test and eventually identified using VITEK System. Total 15 genus and 24 species of bacteria were isolated from these semen samlpes. In raw semen, the most prevalent contaminants were Pseudomonas aeruginosa, Escherichia coli, Proteus mirabilis, Staphylococcus auricularis, Delftia acidovorans, Acinetobacter lowffii, S. aureus and others. And in extended porcine semen, A. lowffii, S. aureus, S. auricularis and other bacteria were identified. Most of them was G(-), which is nonpathogenic bacteria. It seems that bacterial contaminants in fresh raw and extended porcine semen originated from multiple sources at the farms/stud, and were from animal origin and non-animal origins. Whereas, the 7 virus which is known to be detected in porcine semen in 75 cases was not detected. This results showed that removal of bacterial contamination in raw and extended porcine semen is essential and farms were kept for biosecurity and individual hygienes.
Due to the pandemic caused by COVID-19, the demand for face masks is soaring and has often caused a shortage. The aim of this study was to evaluate the effect of ultraviolet (UV) and drying treatments on microbial contaminants in facial masks. To conduct this study, standard procedures were designed to develop samples contaminated by the control bacteria Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa. The contamination level of the standard samples was approximately 6.30 × 106 CFU/ml, and the UV light treatment was performed 1, 3, 5, and 7 times. To evaluate the effect of the UV and drying treatments, the masks were first treated with UV 1, 2, and 3 times, followed by the drying process. As a result, the mask contaminated with E. coli and P. aeruginosa showed a bacterial rate of approximately 99.9% after 1 UV irradiation, and in the case of the S. aureus-contaminated mask, it exhibited a bactericidal rate of approximately 99.9% after 7 UV irradiations. However, when the drying process was included after UV irradiation, all the samples contaminated with E. coli, S. aureus, and P. aeruginosa showed a bactericidal rate of 99.9% or more. The results of this study suggest that UV and drying treatments can effectively reduce the bacterial contaminants in facial masks. In addition, these results provide fundamental data and appropriate sterilization methods for reusing masks.
During transportation and preservation of Takju, alcohol fermentation has continued to produce $C0_2$ from residual sugar and frequently spoiled owing to bacterial contaminants wich produce organic acids. The authors could preserve Takju for more than 50 days at room temperature by pasteurization without any changes of quality. For the optimal condition of pasteurization, fresh Takju was heated at various temperatures and times. D-Value of the Saccharomyces sp. which isolated from Takju collected at seoul area was 19 see at $55^{\circ}C$. Non-spore forming bacterial contaminants, most of which known to cause acid-spoilage, were decreased when heated at $55^{\circ}C$ for 5 min. The optimal pasteurization condition of Takju was at $55^{\circ}C$ for 10 min. Spore forming bacterial contaminants, considered to be EuciiLw sp., were not sterilized after pasteurized at the optimal condition. However, the spore-forming bacteria could not increase any more and also not cause increment of acidity during preservation even at room temperature for 50 days. Reducing sugar was increased during storage of Takju after pasteurization. This suggests that the residual glucoamylase in Takju is still active after pasteurizsation and keep sweet taste.
The bioavailability of sorbed organic contaminants is one of the most important factors used to determine their fate in the environment. This study was conducted to evaluate the bioavailability of slow-desorbable naphthalene in soils. An air sparging system was utilized to remove dissolved (or desorbed) naphthalene continuously and to limit the bacterial utilization of dissolved naphthalene. A biological air sparging system (air sparging system with bacteria) was developed to evaluate the bioavailability of the slow-desorption fraction in soils. Three different strains (Pseudomonas putida G7, Pseudomonas sp. CZ6 and Burkholderia sp. KM1) and two soils were used. Slow-desorbable naphthalene continuously decreased under air sparging; however, a greater decrease was observed in response to the biological air sparging system. Enhanced bioavailability was not observed in the Jangseong soil. Overall, the results of this study suggests that the removal rate of slow-desorbable contaminants may be enhanced by inoculation of degrading bacteria into an air sparging system during the remediation of contaminated soils. However, the enhanced bioavailability was found to depend more on the soil properties than the bacterial characteristics.
정액 내 세균오염 종류와 검출 빈도를 조사한 결과는 Table 1에서 보는 바와 같다. 돼지 정액 내 오염세균 중 검출빈도는 Staphylo- coccus genus, Proteus genus, Bacillus genus, Pasteulla genus, Acinetobacte genus 및 Serratia genus 등의 순으로 나타났다. 동정된 세균 중 33종의 세균에 대하여 디스크법을 이용하여 8종의 항생제에 대한 감수성을 조사 한 결과는 Table 2와 3에서 보는 바와 같다. 검출된 세균의 항생제 감수성을 조사한 결과 항생제 감수성이 높은 항생제는 Amikacin, Polymyxin B, Neomycin, Streptomycin, Kanamycin, Tetracycline, Erythromycin 및 Penicillin순 이었다. Enterococcus faecium와 Streptococcus uberis는 Streptomycin을 제외한 모든 항생제에 대하여 저항성이 있는 것으로 조사되었다.
Bacterial contamination is an unavoidable finding of the semen collection process in boar and can lead in deleterious effects on semen quality and longevity if left uncontrolled. The purpose of this study is to identify the bacteria in extended boar semen and to select the effective antimicrobials to control of the contaminants. Of 116 extended boar semen samples submitted from eight AI centers in Korea, 39 (33.6%) samples were positive for bacterial contamination. Among 39 contaminated semen, most of them (84.6%) were contaminated with one or two bacterial species and there was no significant difference between two age groups $(\leq\;24\;and\;>\;24\;month\;old).$ Stenotrophomonas maltophilia (n = 18) was the most predominant bacterium followed by Elizabethkingia meningoseptica (n = 12), Sphingomonas paucimobilis (n = 12), Myroides spp. (n = 5), Ochrobactrum anthropi (n = 3), and so on. Enrofloxacin (72.9%), florfenicol (72.9%), bacitracin (49.2%) and tylosin (49.2%) showed higher sensitivity compared with penicillin (13.6%) or aminoglycosides (6.8%-18.6%). Brucella spp., Leptospira spp., Mycoplasma hyopneumoniae, Mycoplasma hyorhinis, Mycobacterium tuberculosis complex were not detected in semen by PCR.
The membrane-aerated biofilm reactor (MABR) is a promising municipal wastewater treatment process. In this study, two cross-flow MABRs were constructed to explore the carbon and nitrogen removal performance and bacterial succession, along with changes of influent loading shock comprising flow velocity, COD, and NH4-N concentrations. Redundancy analysis revealed that the function of high flow velocity was mainly embodied in facilitating contaminants diffusion and biosorption rather than the success of overall bacterial populations (p > 0.05). In contrast, the influent NH4-N concentration contributed most to the variance of reactor efficiency and community structure (p < 0.05). Pyrosequencing results showed that Anaerolineae, and Beta- and Alphaproteobacteria were the dominant groups in biofilms for COD and NH4-N removal. Among the identified genera, Nitrosomonas and Nitrospira were the main nitrifiers, and Hyphomicrobium, Hydrogenophaga, and Rhodobacter were the key denitrifiers. Meanwhile, principal component analysis indicated that bacterial shift in MABR was probably the combination of stochastic and deterministic processes.
Bioremediation is the technology to harness nature's biodegradative capabilities to remove or detoxify pollutions that threaten public health as environmental contaminants. Composting may become one of major bioremediation technologies for treating soils contaminated with petroleum if the fate of contaminants during composting is better understood Most composting research of petroleum was primarily focused on removing contaminant by optimizing composting conditions. Accordingly, laboratory feasibility studies may be useful to establish a realistic basis in co-composting complex substrate such as petroleum hydrocarbons. The purpose of this study was to assess the optimal conditions of kerosene biodegradation following supplementation with nutrient amendments under simulated composting conditions. Although it increased the growth of bacterial consortium, addition of co-substrates 0.5%(w/v) such as acetic acid, citric acid, glucose, and malic acid was not beneficial. Combination of nitrogen and phosphorous source enhanced kerosene biodegradation and reduced VOC evolution. These results showed that kerosene was able to utilize in bioremediation technology.
Isolation of pollutant-degrading bacteria is important in bioaugmentation, one of the methods for biological degradation of environmental contaminants. We focused on the magnetic activated sludge (MAS) process as a culture method that efficiently concentrates degrading bacteria, and cultured activated sludge with 1,4-dioxane as a model pollutant. After 860 days of operation, MLVSS, which indicates the amount of sludge, increased from 390 mg/L to 10,000 mg/L, and the removal rate of organic matter including 1,4-dioxane, tetrahydrofuran, and glucose in the artificial wastewater reached up to 97%. Based on these results, the MAS process was successfully used to acclimate activated sludge with 1,4-dioxane. Bacterial flora analysis in the MAS showed that bacteria of the genus Pseudonocardia, already reported as 1,4-dioxane degrading bacteria, play an important role in the degradation of this pollutant. The MAS process is a suitable culture method for acclimation of environmental pollutants, and the findings indicate that it can be used as an enrichment unit for pollutant-degrading bacteria.
