The rhizobacterial composition varies according to the soil properties. To test if the effect of herbicides on the rhizobacterial communities of genetically modified NK603 glyphosate-tolerant maize varies according to different soil locations, a comparison was made between the effects of glyphosate (Roundup Plus), a post-emergence applied herbicide, and a pre-emergence applied herbicide (GTZ) versus untreated soil. The potential effect was monitored by direct amplification, cloning, and sequencing of the soil DNA encoding 16S rRNA, and high-throughput DNA pyrosequencing of the bacterial DNA coding for the 16S rRNA hypervariable V6 region. The results obtained using three different methods to analyze the herbicide effect on the rhizobacterial communities of genetically modified NK603 maize were comparable to those previously obtained when glyphosate-tolerant maize was grown in soil with different characteristics. Both herbicides decreased the bacterial diversity in the rhizosphere, with Actinobacteria being the taxonomic group most affected. The results suggest that both herbicides affected the structure of the maize rhizobacterial community, but glyphosate was environmentally less aggressive.

Soybean is an important source of protein and for a wide range of agricultural, food, and industrial applications. Soybean is being affected by Xanthomonas citri pv. glycines, a causal pathogen of bacterial pustule disease, result in a reduction in yield and quality. Diverse microbial communities of plants are involved in various plant stresses is known. Therefore, we designed to investigate the microbial community differentiation depending on the infection of X. citri pv. glycines. The microbial community's abundance, diversity, and similarity showed a difference between infected and non-infected soybean. Microbiota community analysis, excluding X. citri pv. glycines, revealed that Pseudomonas spp. would increase the population of the infected soybean. Results of DESeq analyses suggested that energy metabolism, secondary metabolite, and TCA cycle metabolism were actively diverse in the non-infected soybeans. Additionally, Streptomyces bacillaris S8, an endophyte microbiota member, was nominated as a key microbe in the healthy soybeans. Genome analysis of S. bacillaris S8 presented that salinomycin may be the critical antibacterial metabolite. Our findings on the composition of soybean microbiota communities and the key strain information will contribute to developing biological control strategies against X. citri pv. glycines.
Understanding soil microbial community structure in the Arctic is essential for predicting the impact of climate change on interactions between organisms living in polar environments. The hypothesis of the present study was that soil microbial communities and soil chemical characteristics would vary depending on their associated plant species and local environments in Arctic mature soils. We analyzed soil bacterial communities and soil chemical characteristics from soil without vegetation (bare soil) and rhizosphere soil of three Arctic plants (Cassiope tetragona [L.] D. Don, Dryas octopetala L. and Silene acaulis [L.] Jacq.) in different local environments (coal-mined site and seashore-adjacent site). We did not observe any clear differences in microbial community structure in samples belonging to different plant rhizospheres; however, samples from different environmental sites had distinct microbial community structure. The samples from coal-mined site had a relatively higher abundance of Bacteroidetes and Firmicutes. On the other hand, Acidobacteria was more prevalent in seashore-adjacent samples. The relative abundance of Proteobacteria and Acidobacteria decreased toward higher soil pH, whereas that of Bacteroidetes and Firmicutes was positively correlated with soil pH. Our results suggest that soil bacterial community dissimilarity can be driven by spatial heterogeneity in deglaciated mature soil. Furthermore, these results indicate that soil microbial composition and relative abundance are more affected by soil pH, an abiotic factor, than plant species, a biotic factor.
To evaluate the effect of reed population on the distribution and activities of microorganisms, vertical distribution of heterotrophic bacteria, degradation rate of cellulose, extracellular aminopeptidase activity (APA) and metabolic diversity based on GN2 Microlog plate were measured at two salt marsh stations in Hogok-ri, Namyang Bay, west coast of Korea. The number of heterotrophic bacteria at station 1 (reed population inhabited area) showed 2 to 6 times higher than that of station 2 (exposed area) with exception in the surface layer. Cellulose degradation rates in station 1 showed more than 50%. month-I and higher than that of station 2 (10.2 to 38.4%. $month^{-1}$). Yet the APA at two stations did not show difference except surface layer and suggested that APA might not be a significant factor in degrading marsh plant debris. Lipid class compounds, cell wall polymers and L-alanine were widely used by microorganisms. The number and activities of bacterial populations especially concerned in plant debris degradation seemed to be stimulated by the reed communities.
To investigate soil bacterial diversity according to vegetation types, directly extracted DNA from 5 different soils were cross-hybridized with each other as a probe and target. Pinus densiflora soil was shown the highest value then agricultured soil>naked soil>grass soil>Quercus mongolicas soil in the order of diversity. Cluster analysis by similarity showed that soil microbial communities were categorized into three groups.
In order to characterize the genetic diversity of bacterial community in groundwater, samples were collected from used for drinking water and polluted with heavy metal wastewater in Seoul city and natural cave of Kangwondo. The DNA was amplified with 165 rDNA-based primers by use of the PCR, and then analysed ARDRA (amplified ribosomal DNA restriction analysis). Restriction endonuclease analysis patterns of amplified 165 rDNA in drinking water and wastewater relatively showed high genetic diversity in situ and drinking groundwater. The number of DNA fragments varied with in situ and drinking water. This method of ARDRA of bacterial communities in groundwater could be used for a quick assessment of genotypic changes between different locations reflecting different environmental conditions and the diversity reflected pollution of groundwater (natural cave water>drinking water>waste water, as in order of grade). Marine beach sands with bacterial pathogens may cause increased outcomes of illness among beachgoers in summer. In this study, pyrosequencing of 16S ribosomal DNAs extracted from 12 beach sands was performed to understand how the environmental factors of wastewaters or human wastes affected the distribution of bacterial communities at the beach of North Gyeongsang province (Yeongdeok and Pohang counties) in the middle of October. It was found that Acidobacteria were dominantly distributed in the sands near the clean seawaters, Proteobacteria in the sands near the river waters, Cyanobacteria in the sands near the wastewaters, and Bacteroidetes in the sands near the beach park. Other phyla groups such as Actinobacteria, Chlorobi, Deferribacteres, Deinococcus-thermus, Firmicutes, Gemmatimonadetes, Nitrospirae, and/or Verrucomicrobia were distributed at low relative abundance (1-5%).
This study was conducted to understand the dynamics of microbial communities of soil microorganisms, and their distribution and abundance in the indigenous microorganisms (IMOs) manipulated from humus collected from the forest near the crop field. The soil microorganisms originated from humus and artificially cultured microbial-based soil amendments were characterized by molecular and biochemical analyses. The bacterial population (2 × 106~13 × 106 CFU/g sample) was approximately 100-fold abundant than the fungal population (2 × 104~8 × 104 CFU/g sample). The 16S rDNA and ITS sequence analyses showed that the bacterial and fungal communities in humus and IMOs were mainly composed of Bacillus and Pseudomonas, and Trichoderma and Aspergillus species, respectively. Some of the bacterial isolates from the humus and IMOs showed strong inhibitory activity against soil-borne pathogenic fungi Fusarium oxysporum and Sclerotinia sclerotiorum. These bacteria also showed the siderophore production activity as well as phosphate solubilizing activity, which are requisite traits for biological control of plant pathogenic fungi. These results suggest that humus and IMOs could be a useful resource for sustainable agriculture.
Dynamics of bacterial communities and its colonization under aerobic gemp retting were observed in air lift fermentor as a closed system, unlike conventional hemp retting as an open system. Dried hemp which was harvested in both 1986 and 1987 was retted at room temperature. Predominant community was facultatively anaerobic Gram-negative rods, and its density was increased from $3.0\times 10^{7}$ cells/ml to $9.0\times 10^{8}$ cells/ml. The density of facultatively nanerobic Gram-positive fods was maintained at the lovel of $5.0\times 10^{6}$ cells/m, and this Gram-positive bacterial community was not participated in retting. In the Gram-negative bacterial community during the retting, five types of colonieswere developed at early stage of pH7.0-8.0, and thereafter, only three types were colonized till later stage, shich were identified as pectolytic strain Erwinia salicis, Erwinia tracheiphila and Enterobacter agglomerans. A community of facultatively Gram-negative rods was mainly proliferated in stems and dispersed into liquor after 6-8 hours. Retting was terminated within 70-80 hours.
To determine the characteristics of heterotrophic bacterial community in estuarine ecosystem, water and sediment samples were taden from Naktong estuary. All isolates were compared with 73 characters and described by cluster analysis. With same characters, 30 reference strains were able to divide into approximate species level at 80% similarity (S value). Diversity indices ($H^{1}$) of sediment column isolates were higher than water column isolates. The bacterial community commonly appeared in water and sediment column was reduced with going to downstream. Tolerance indices for temperature (Pt) and salinity (Ps) were also higher in sediment isolates than in water isolates. The bacterial community in sediment column is believed to be composed with diverse populations compared to water column and maintains its stability against various environmental changes with high physiological tolerances.
