EFFECTS OF AMMONIATED RICE STRAW FEEDING ON MICROBES AND THEIR FERMENTATION END-PRODUCTS IN THE RUMEN AND CAECUM OF SHEEP

  • Three sheep fitted with cannulas in the rumen and the caecum were used in a $3{\times}3$ Latin square design to study the changes in ruminal and caecal microbial populations and their metabolite composition with ammoniated rice straw feeding. The 3 diets contained either 80% untreated rice straw (UTS) or ammoniated rice straw (ATS) and 20% formula feed. These were a control diet (C), a urea supplemented diet (U) containing urea at 1.1% and an ammoniated rice straw diet (AT). Data were analyzed by analysis of variance and means separated by the Student Neumann Kuel's multiple comparison. AT feeding increased ruminal bacterial counts, in particular cellulolytic bacterial counts (p < 0.05) which were 1.8, 2.4 and 7.0 (${\times}10^6/ml$ ruminal fluid) for C, U and AT, respectively. There was an increasing tendency (p < 0.10) in ruminal fungal population with U; values were 2.0, 5.2, 3.1 (${\times}10^3/ml$ ruminal fluid) for C, U and AT, respectively. Ruminal protozoa counts were not significantly (p > 0.05) altered with diets. Caecal total viable bacterial count with AT was about thrice the value with C. Total VFA concentration in the rumen was significantly increased (p < 0.025) (7.7 mmol/dl for C and 8.2 mmol/dl for AT) and correspondingly, pH lowered when AT was fed. Sheep on AT tended to produce less acetate and more butyrate in the rumen without significance (p > 0.05). Similar to the rumen, total VFA concentrations of 4.4, 3.8 and 5.2 mmol/dl were detected, respectively, for C, U and AT. Caecal ammonia-nitrogen concentrations were about six-fold of that in the rumen, though there were no differences (p > 0.05) among treatments.

Effects of Acarbose Addition on Ruminal Bacterial Microbiota, Lipopolysaccharide Levels and Fermentation Characteristics In vitro

  • This study investigated the effects of acarbose addition on changes in ruminal fermentation characteristics and the composition of the ruminal bacterial community in vitro using batch cultures. Rumen fluid was collected from the rumens of three cannulated Holstein cattle fed forage ad libitum that was supplemented with 6 kg of concentrate. The batch cultures consisted of 8 mL of strained rumen fluid in 40 mL of an anaerobic buffer containing 0.49 g of corn grain, 0.21 g of soybean meal, 0.15 g of alfalfa and 0.15g of Leymus chinensis. Acarbose was added to incubation bottles to achieve final concentrations of 0.1, 0.2, and 0.4 mg/mL. After incubation for 24 h, the addition of acarbose linearly decreased (p<0.05) the total gas production and the concentrations of acetate, propionate, butyrate, total volatile fatty acids, lactate and lipopolysaccharide (LPS). It also linearly increased (p<0.05) the ratio of acetate to propionate, the concentrations of isovalerate, valerate and ammonia-nitrogen and the pH value compared with the control. Pyrosequencing of the 16S rRNA gene showed that the addition of acarbose decreased (p<0.05) the proportion of Firmicutes and Proteobacteria and increased (p<0.05) the percentage of Bacteroidetes, Fibrobacteres, and Synergistetes compared with the control. A principal coordinates analysis plot based on unweighted UniFrac values and molecular variance analysis revealed that the structure of the ruminal bacterial communities in the control was different to that of the ruminal microbiota in the acarbose group. In conclusion, acarbose addition can affect the composition of the ruminal microbial community and may be potentially useful for preventing the occurrence of ruminal acidosis and the accumulation of LPS in the rumen.

  • 길항균을 생물 농약으로 개발하기 위해서는 저렴한 산업용 배지를 이용한 대량 생산 체계를 확립하는 것이 중요하다. 본 연구에서는 풋마름병 방제 효과가 뛰어난 Bacillus amyloliquefaciens SKU-78 균주의 배양조건을 확립하였다. 저가의 산업용 기질로는 콩가루와 옥수수 전분 배지가 균 생장에 가장 효과적이었고, 최초 pH 5.5, 배양 온도 $30^{\circ}C$, 교반속도 150-250 rpm의 조건으로 30 L fermenter를 이용한 배양에서 20 시간째에 최대 생균수($1.2{\times}10^{11}$ CFU/ml)에 도달하였다. 저가의 산업용 배지로 배양한 배양액을 관주 처리하였을 때 65%의 발병 억제 효과를 나타냄으로써 SKU-78 균주의 산업용 배지를 이용한 대량배양의 기초가 마련되었다.

토양에서 분리한 Bacilus flexus로부터 Invertase의 생산 (Production of Invertase from Newly Isolated Strain Bacilus flexus)

  • In the present study, we isolated a new bacterial strain producing invertase (EC 3.2.1.26) and determined optimized culture condition in flask culture. The strain was identified as Bacilus flexus determined by the 16S rDNA sequencing method. The invertase was produced only in the sucrose medium as the sole carbon source. Potassium nitrate was an adequate nitrogen source for enzyme production, whereas meat peptone showed the highest bacterial growth. Enzyme production was increased about 2-fold when $MgSO_4\cdot7H_2O$ was supplemented to the growth media. The optimum temperature was found to be $30^{\circ}C$ for both enzyme production and bacterial growth. Invertase exhibited pH optima in the range 5.0-6.0 and have a temperature optimum at $40^{\circ}C$, similarly to other invertases found from different microbial sources. Several mineral ions (K and Fe) stimulated the invertase activity, whereas some bioelements (Ag, Mg, and Mn) inhibited enzyme activity. Under the optimized culture condition, the maximum enzyme production (over 250 units/mL) was achieved at 20 h. To the best of our knowledge, this is the first time to report on invertase production by Bacilus flexus.

지하수 세균 군집에 미치는 물리화학적 환경요인의 영향 (The Effects of Physico-Chemical Factors on the Microbial Population in Groundwater)

  • 지하수 세균 군집에 미치는 환경요인의 영향을 분석하기 위하여, 서울 시내에서 음용수로 사용하는 2개 정점과 음용수외 생활용수로 사용하고 있는 8개 정점을 대상으로 조사하였다. 물리.화학적 환경요인과 중금속의 농도, 및 세균 군집의 분포 등 40개 변수를 분석한 결과, 음용수로 사용하는 정점을 제외한 나머지 정점에서 질산성 질소와 암모니아가 용수목적별 수질기준의 기준치 이상으로 측정되었다. 총세균은 5.1~41.4$\times$$10^{5}$cells/ml 범주로, 종속영양세균과 기능성 세균군집은 0.01~29.6$\times$$10^4$cfu/ml의 범주로 조사되었다. 세포외 효소의 활성도는 0.005~11.3$\mu$M/l/hr 의 범주로 나타났고, lipase, phophatase, $\beta$-glucosidase, cellulase, chitinase, amylase 순으로 활성도가 나타났다. 정점별로 조사된 세균 군집에 미치는 환경요인의 상호관계는 대응분석(correspondence analysis)과 다차원 척도법 분석(multidimensional scaling; MDS)으로 하였으며, 그 결과 4개의 집단으로 구분되었으며, 세균 군집에 미치는 주요한 환경요인은 정점별 잠재오염원과 일치하는 양상을 보였다.다.

황색종 연초에 있어서 변이체의 조합능력 및 Heterosis (Combining-Ability and Heterosis for Mutant Character of Quantitative Characters in Flue-Cured Tobacco Varieties(Nicotiana tabacum L))

  • This experiment were conducted to investigate heterosis and combining ability for several mutant characters by analyzing dialled crosses of flue-cured tobacco. In a dialled cross of 3 flue-cured varieties and the mutant line 83H -5, the heterosis was somewhat higher in Fl than in F2. For growth character, the heterosis was 0.28-6.03% in plant height, leaf number, leaf shape index and yield, and was 43.2% for bacterial wilt disease index. The mutant line 83H-5 showed significantly negative GCA effect for plant height, leaf width and bacterial wilt disease index in Fl and F2, leaf length in F2, and positive GCA effect for total alkaloids, total nitrogen in Fl and days to flower in F2, respectively. Specific combining ability(SCA) in 83H-5 x Hicks was significant in negative effect for leaf length(F2), number of leaves(F2), leaf shape(F1, F2), bacterial wilt(F2) and alkaloids(F1), and in 83H-5 x NC 2326 in positive effect for leaf length(F1, F2) and leaf width(F2), and for 83H-5 x NC 82 in positive effect for plant height(F1, F2) and leaf width(F2), and for 83H-5 x NC 82 in Positive effect for Plant height(F1, F2), leaf length(F2) and yield(F1, F2).

Acetobacter xylinum GS11에 의한 미생물 셀룰로오스의 생산 (Production of Bacterial Cellulose by Acetobacter xylinum GS11.)

  • Acetobacter xylinum GS11 균주를 이용하여 다양한 배양조건에서 미생물 셀룰로오스의 생산성을 검토하였다. 탄소원으로 glucose 이외에 첨가한 기질은 영향이 없었으며, 2% glucose 농도 범위에서 셀룰로오스 생산량이 2.8 g/L으로 가장 양호한 것으로 나타났다. 질소원은 Y.E와 soytone등의 유기질소원 첨가가 효과적이었으나, 무기질소원의 효과는 나타나지 않았다. 무기염류 및 아미노산의 첨가는 대부분 효과적이었으며, 이 중 무기염류는$ MgSO_4$가 1.5배, 아미노산은 phenylalanine이 1.4배의 셀룰로오스 생산성을 나타냈다. 비타민은 $\alpha$-tocopherol의 첨가 시 1.4배의 생산성을 보였다. 또한 기본배지에 Coconut milk와 0.5%의 lignosulfonate의 첨가가 대조구와 비교하여 각각 2.2, 2.1배의 셀룰로오스의 생산성을 나타내어 실험 처리 중 가장 효과적이었다.

황색종 연초 돌연변이 계통 KF8832-85의 특성 (Characteristics of A New Flue-cured Tobacco Mutant Line KF 8832-85)

  • A new flue-cured tobacco mutant line BU 8832-85 was developed at Taegu Experiment Station, Korea Ginseng and Tobacco Research Institute in 1994. KF 8832-85 was resulted from a cross of flue-cured cultivars NC 95$\times$NC 2326, and developed by a pedigree system of breeding ; initial selection was made by plant type and resistance to bacterial wilt(BW) disease(heudomonas solanaceamm) in the F2 generation under the natural field conditions infested with the pathogen. One white flowered plant was occurred by spontaneous mutation in a certain line among the F3 generatioin while the others were pink. Six plants from the seeds by selfing were selected at the field infested with the pathogen among 240 populations with white flowering in the F4, KF 8832-85 was selected based on yield and leaf quality trials among 6 lines in Fs generation. BCF 8832-85 was compared with its Parent for certain agronomic and chemical characteristics at Taegu Experiment Station in 1993 and 1994. The results showed that KF 8832-85 have white flower, the stalk height was approximately that of NC 2326 but averaged about loom taller than NC 95. It produced ground suckers as much as NC 95, and did not breakdown leaf at the same as WC 2326. KF 8832-85 have high resistance to bacterial wilt disease. Yield of KF 8832-85 was 10 and 18% higher then that of NC 2326 and WC 95, respectively. Price per Kg was equal to that of NC 2326. The contents of nicotine and reducing sugar did not differ significantly from NC 95, while total nitrogen was significantly lower than NC 95. Therefore, the new mutant line is genetically stable for agronomic and chemical characteristics and provides a source of bacterial wilt disease resistance for use in breeding resistant flue-cured cultivars. Physicochemical Properties and Bacterial Communities of Meongge (Halocynthia roretzi) Jeotgal Prepared with 3 Different Types of Salts

  • Three types of meongge (Halocynthia roretzi) jeotgal (MJ) were prepared with 3 different types of salts (12%, w/v): purified salt (PS), solar salt aged for 3 years (SS), and bamboo salt that had been recrystalized 3 times (BS). One set of MJ was fermented with starters, Bacillus subtilis JS2 and Tetragenococcus halophilus BS1-37 (each 6 log CFU/g), and another set without starters for 42 days at $10^{\circ}C$. The LAB count of the SSMJ (non-starter) was highest at day 28 (2.30 log CFU/g). The pH of the PSMJ and SSMJ was 5.72-5.77 at day 0, and 5.40-5.50 at day 42. BSMJ showed higher pH and lower titratable acidities than other samples. Amino-type nitrogen (ANN) increased continuously, and SSMJ showed higher values than other samples from day 14. Bacterial species of non-starter MJ were examined by culture independent method. Clone libraries of 16S rRNA genes were constructed in Escherichia coli from total DNA from non-starter MJ samples at day 0, 14, and 28. Thirty clones per each sample were randomly selected and DNA sequences were analyzed. Variovorax sp., uncultured bacterium, and Acidovorax sp. were the most dominant group at day 0, 14, and 28, respectively. Lactobacillus sakei and Streptococcus sp. were the next dominant group in SSMJ at day 28. A Streptococcus sp. was detected from PSMJ at day 28. Sensory evaluation for MJ samples at day 28 showed that SSMJ got higher overall acceptability scores. These results showed that solar salt can cause desirable changes in the microbial community of fermented foods, thereby positively affecting their overall quality.

  • The small and the large intestine of swine represent the organs that extract nutrients from feedstuffs through digestion and fermentation and that allow their absorption and incorporation into the blood circulation. Special attention is directed towards the small intestine of young pigs since the transition to a solid diet at weaning exerts major impacts on the structural and functional integrity of the small intestine. Dietary factors involved in postweaning changes of gut morphology and biochemistry such as removal of bioactive compounds in sows milk at weaning, anti-nutritional factors in weaner diets, dietary fiber and the role of voluntary feed intake will be elucidated. The microbial function of the large intestine which is carried out by a diverse population of microorganisms is dependent on substrate availability. Short chain fatty acids as main fermentation products contribute to the energy supply of the host but they are also important for the maintenance of the morphological and functional integrity of the epithelium in the colon. As a result of bacterial nitrogen assimilation in the large intestine, nitrogen is shifted from the urinary to the fecal excretion route thus saving metabolic energy to the pig because less ammonia would become available for conversion to urea.
