Isolation of Bacteria Associated with Fresh Sponges in Lake Baikal (바이칼 호수에 서식하는 담수 스폰지 내 공생세균의 분리)

  • Sponge in Lake Baikal is an unique organism. Microorganisms in sponges are assumed as precious resources for bioactive materials. For understanding the bacterial community in Baikalian sponges by cultivation, 92 strains of bacteria were isolated from lake water and 2 species of sponges, Baikalospongia sp. and Lubomirskia sp., Thirty five bacterial strains are isolated from ambient water near the sponge, 27 bacterial strains from Baikalospongia sp., 30 bacterial strains from Lubomirskia sp.. As a result, 78.3% and 57.6% of isolated bacterial strains has amylase and protease activity respectively, while strains with cellulose and lipase activities were 38.0% and 34.8%. By 16S rRNA sequence analysis of selected strains, 13 strains which were isolated from Baikalospongia sp. were belong to Pseudomonas spp.. Whereas, 14 strains which were isolated from Lubomirskia sp. were Pseudomonas spp., Buttiauxella agrestis, Pseudomonas fluorescens, Yersinia ruckeri, Bacillus spp., Paenibacillus spp., Bacillus thuringiensis, Bacillus simplex, Brevibacterium spp., Acinetobacter lwoffii. In culture media, Pseudomonas spp. dominance was supposed that according to allelophathy.

A Study on Suppression Components of Spoiling Ginseng (수삼부패억제 활성물질 선발연구)

  • This study carried out to scanning and select the antimicrobial pharmaceuticals which were suppress the rottening of fresh ginseng. We are isolated 20 kinds of microorganisms from rotten ginseng. In these of the microorganisms, five bacteria, fresh ginseng root-rottening, are identified to Pseudomonas putida, Pseudomonas putida biotype A, Bacillus spp, Enterobacter cloacae, and Klebsiella pneumoniae. Particularly. Bacillus spp was rapidly rotted the ginseng root, compared with the others. The antimicrobial activity were tested with 19 kinds of water extracts, and 34 kinds of essential oils. The water extract of Terminaliae Fructus, and Schiandra chinensis are strongly inhibited the growth of bacteria causing the ginseng root-rottening. And 5 kind of essential oils are inhibited bacteria. It was regarded that the water extract of Terminaliae Fructus has weekly water insoluble and polar antimicrobial components.

In Vitro Antagonistic Effects of Bacilli Isolates against Four Soilborne Plant Pathogenic Fungi

  • Twenty isolates of Bacillus spp. obtained from livestock manure composts and cotton-waste composts were tested for in vitro antagonistic effects against soilborne plant pathogenic fungi, Fusarium oxysporum, Phytophthora capsici, Rhizoctonia solani AG-4, and Sclerotinia sclerotiorum. Seven isolates of Bacillus spp. had antagonistic effects on mycelial growth of all the isolates of F. oxysporum tested. The bacterial isolate RM43 was the most effective to inhibit the mycelial growth of the fungal isolates. Twelve isolates of Bacillus spp. had antagonistic effects on mycelial growth of all the isolates of P. capsici tested. The bacterial isolates M34 and M47 were very effective to inhibit the mycelial growth of the fungal isolates. Thirteen isolates of Bacillus spp. had antagonistic effects on mycelial growth of all the isolates of R. solani AG-4 tested. The bacterial isolates M27 and M75 were very effective to inhibit the mycelial growth of the fungal isolates. Fourteen isolates of Bacillus sp. had antagonistic effects on mycelial growth of all the isolates of S. sclerotiorum tested. The bacterial isolates M49 and M75 were very effective to inhibit the mycelial growth of the fungal isolates. The antagonistic effects of most Bacillus spp. isolates against the isolates of the four fungi differed depending on the fungal species and the isolates of each fungus. The bacterial isolates M27 and M75 were the most effective to inhibit the mycelial growth of all four fungi.

Removal of pentachlorophenol by pentachlorophenol resistant strains isolated from activated sludge (활성오니에서 분리한 pentachlorophenol 내성균주의 pentachlorophenol 제거에 관한 연구)

  • Twenty strains of pentachlorophnol (PCP) resistant bacteria were isolated from activated sludge of the sewage treatment plant of Jung Lang Chun, Seoul. The predominant strains were Bacillus spp. including B. sphaericus and E. schlegelii. The other strains were identified as Corynebacterium spp., Staphylococcus aureus, Arthrobacter spp. and Aeromonas spp. The resistant strains could be grouped into two categories; PCP-degrading and PCP-adsorbing/absorbing ones. PCP-degrading strains degraded $75{\sim}90%$ of PCP in the medium containing 100 ppm PCP during the first 24 hours of growth. At the initial period the PCP-adsorbing/absorbing strains removed PCP from the medium but started to release PCP after 24 or 72 hours of growth. PCP degradation products from the culture broth of PCP-degrading strains were identified by comparing their $R_f$ values with those of the reference compounds. 2-chlorophenol and 2.4-dichlorophenol were presumed to be the intermediate products of PCP degradation.

  • 37 Bacillus spp strains(43.0%=37/86), and 18 B. cereus(20.9%=18/86) were isolated on 86 dried marine products. Each isolates were B. cereus 48.6%(18/37), B. mycoides 13.5%(5/37) B. coagulus 5.4%(2/37) B. firmus 5.4%(2/37), B. circulus 2.7%(1/37), B. stearothermophilus 2.7%(1/37), B. pumilus 2.7%(1/37) B. spp. 8.1%(3/37), and Brebacillus brevis 10.8%(4/37) etc.

  • To evaluate antimicrobial activities of chongkukjang slime fermented by different strains, growth characteristics were compared using various standard microorganisms with addition of chongkykjang slime. Chonghkjang slime was prepared by fermenting cooked soybean after inoculating with Bacillus circulans K-1, Baciilus spp N-1 and Bacillus subtilis CH-1, respectively. Significant antimicrobial activity was observed by chongkukjang slime on gram positive bacteria (Staphylococcus aureus, Bacillus cereus, Bacillus subtilis, Micrococcus luteus), gram negative bacteria(Escherichia coli O157:H7, Salmonella Typhimurium, Pseudomonas fluorescens), and yeast (Pichia membranaefaciens, Saccharomyces cerevisiae, Candida albicans). In case of B. cereus growth inhibition of 80% was achieved by the addition of chongkukjang slime; on the contrary, to Escherichia coli O157:H7 only 20% inhibition was observed. Slime from Bacillus subtilis CH-1, in particular, inhibition of 40% toward bacteria and yeast, whereas slime from Bacillus circulans K-1, Bacillus spp N-1 showed only 20% inhibition.

  • In this study, microbes were collected before and after disinfection using ANIOS(ANIOSURF Premium NPC) and compared the areas where the radiological technologist and the patient frequently contacted the chest X-ray. From September 1st to September 7th, 2020. in P Hospital in Deagu, 4 region were collected in a 10×10 size using a sterile cotton swab of the transport medium, and before and after disinfection results were obtained through the colorimetric method. As a result, n the X-ray tube handle Proteus mirabilis, Staphylococcus epidermidis, Bacillus spp., Candida spp., and in the Chin region Proteus mirabilis, Enterococcu faecium, Pseudomonas aeruginosa, NTM, and in the Chest region Proteus mirabilis, Enterococcu faecium, Pseudomonas aeruginosa, and in the Palm region NTM, Candida spp. were detected, and 103 CFU(Colony Forming Unit) or more were measured. After disinfection, only X-ray tube handle was detect Bacillus spp. and more than 102 CFU was measured. Microorganisms found prior to disinfection can cause opportunistic infections, Experimental results showed that Aniosulf(0.25%) is more economical and disinfectant than ethanol(70-90%) and isopropyl alcohol(70-90%). However, further research is needed on the detection of Bacillus spp. resultingly this research is useful basic data of infection control in Radiography room and prevention secondary infections.

A Study on the Relationships Between the Epiphytic Microbes and the Blight of Porphyra Species from the Coastal Waters of the Yellow Sea, Korea -II. Seasonal Variation of Heterotrophic Bacteria in the Surrounding Seawater of Porphyra Farming Area Near Mokp'o- (서해산 김 엽체상의 미소생물과 김의 병해와의 관계에 대한 연구 -II. 목포 인근 김 양식장 주변해수에서의 종속영양세균의 계절적 변화-)

  • To study the seasonal variation of heterotrophic bacteria near the surrounding seawater of Porphyra forming area, samples were collected in the intertidal waters of Mokp'o of the Yellow Sea from February to December, 1990. Annual distribution of heterotrophic bacteria ranged from $7.5\times10^2\;to\;1.1\times10^5\;cfu/ml,$ annual distribution of physiological characteristic bacteria ranged from $5.0\times10\;to\;4.34\times10^4\;cfu/ml$ for proteolytic bacteria, from 0 to $1.35\times10^4\;cfu/ml$ for lipolytic bacteria and from 0 to $1.2\times10^4\;cfu/ml$ for amylolytic bacteria. Sixty-five percent of isolates from seawater were rods, and $76.7\%$ of isolates were Gram-negative. Most isolates were mesophiles and showed utilization of various carbon sources such as glucose, maltose, lactose, sucrose and arabinose. Most isolates also showed tolerance to a broad range of salt concentration. Dominant genus in seawater were Flavobacterium spp., in February, Moraxella spp., Acinetobacter spp. in March, Bacillus spp., Chromobacterium spp., Micrococcus spp., Vibrio spp. in July and Chromobacterium spp., Micrococcus spp., Bacillus spp. in November.

Quantitative Analysis of Bacillus amyloliquefaciens GR4-5 in Soil (Bacillus amyloliquefaciens GR4-5 균주의 토양 내 정량 분석)

  • Bacillus amyloliquefaciens GR4-5 was isolated from the rhizosphere soil of Korean ginseng and displayed broad-spectrum suppression of ginseng root rot pathogens. The survivability of B. amyloliquefaciens GR4-5 in soil was investigated under three different conditions; indoor, outdoor - of which soil was put in 14 mL tube after treatment - and field environments. Soil samples were collected over a four-week period from three experimental designs, and assessed for 16S rRNA gene copy number by quantitative polymerase chain reaction (qPCR). In outdoor condition, the 16S rRNA gene copy number of Bacillus spp. was 8.35 log copies g $soil^{-1}$ immediately after the GR4-5 treatment. Two weeks later, the 16S rRNA gene copy number of Bacillus spp. (6.70 log copies g $soil^{-1}$) was similar to that of the control (6.38 log copies g $soil^{-1}$). In indoor condition, the 16S rRNA gene copy number of Bacillus spp. maintained in a certain level for a longer period than those in outdoor and field. The 16S rRNA gene copy number of Bacillus spp. in field experiment was reduced faster than that of outdoor condition. Our results show that B. amyloliquefaciens GR4-5 can survive in bulk soil for 1 week, indicating its potential use as a biocontrol agent following 7 day application intervals. This study presents that outdoor microcosm system design could be a useful method to assess easily the survivability of beneficial microorganisms.

THE ANTIBIOTIC ACTIVITY OF ACTINOMYCES ISOLATED FROM PLAQUE OF BLACK STAINED PRIMARY TEETH TO STREPTOCOCCUS MUTANS (Black stain을 가진 유치 치태에서 추출한 방선균의 S. mutans에 대한 항생능 평가)

  • The aim of this study is to assess the antibiotic activity of Actinomyces in plaque from black stained primary teeth to Streptococcus mutans. Samples were obtained from four children, 2-6 years of age, who had black stains on all erupted primary teeth. 16 different Actinomyces spp. were isolated, and antibiotic activity test with paper disc method was done. The results were as follows, 1. No.1 and No.5 Actinomyces spp. showed the antibiotic activity to Streptococcus mutans and the activity of No.5 Actinomyces spp. could compete with that of Oxacillin. 2. No.1 and No.5 Actinomyces spp. also exhibited the antibiotic activity to Bacillus cereus, Bacillus subtilis commonly used as experimental bacteria for testing antibiotic activity. 3. For identification of No.1 and No.5 spp., PCR analysis was done. No.5 spp. matched Actinomyces viscosus at 97% level but No.1 spp. didn't match.
