Production of Purplish-red Pigment in Mixed Culture of Streptomyces propurpuratus ATCC 21630 and Bacillus sp. R-89 (Streptomyces propurpuratus ATCC 21630 Bacillus sp. R-89의 혼합배양에 의한 적자색 색소의 생산)

  • The purplish-red pigment was formed on agar plate by superimposed streaking of Streptomyces propurpuratus ATCC 21630 and strain R-89, The strain No.89 was ascribable to the genus Bacillus and designated as Bacillus sp. R-89. Both strain did not produced such pigment when cultivated independently. For hyperpigment production, we selected the mutant S.P-6 from Streptomyces propurpuratus ATCC 21630 by MNNG (N-methyl-N'-nitro-N-nitrosoguanidine) treatment. Maximum purplish-red pigment 1420 mg/$m\ell$ were produced, when the mutant of R-16 and Bacillus sp. R-89 were mixed cultured at 3$0^{\circ}C$ for 72 hr.

Effect of Different Abiotic Factors on Chemotaxis of Bacteria Towards Fungal Propagules (종류가 다른 무생물적 요인이 진균류에 미치는 세균의 주화성)

  • Chemotactic responses of five motile saprophytic and one phytopathogenic bacteria e.g. Agrobacterium radiobacter, Bacillus subtilis, B. polymyxa, Pseudomonas aeruginosa, P. fluorescens and Xanthomonas malvacearum towards exudate of Cochliobolus sativus conidia, Fusarium of oxysporum f. sp. ciceri chlamydospores, Macrophomina phaseolina sclerotia and Phytophthora drechsleri f. sp. cajani oospores were determined in vitro at different abiotic conditions. In general, a positive correlation (r=0.76 to 0.89; P=0.05) was observed between concentration of fungal exudates and attraction of bacterial cells. Similarly, a significant (P=0.05; r=+0.82 to 0.95) positive correlation was noticed between chemotactic response and incubation period. The chemotactic response of bacteria was greatly influenced by temperature and pH of the test fungal exudate. The optimum temperature for maximum chemotaxis was $25^{\circ}C$ for A. radiobacter, $30^{\circ}C$ for B. polymyxa, P. aerugionosa, P. fluorescens and X. malvacearum and $35^{\circ}C$ for B. subtilis. Fungal exudates maintained at pH 7 attracted maximum number of bacteria. The response of bacterial cells to exudates at pH 3 and 11 was not significantly (P=0.05) different than that to the buffer (control). Chemotaxis of bacteria was observed towards attractants (fungal propagules and their exudates) when they were kept apart and bridged with the capillaries filled with non-attractant (buffer) or attractant (exudate).

  • To isolate novel strains that hydrolyzed the rac-ketoprofen ethyl ester to ketoprofen in the stereospecific manner, we screened broad ecological niches and soil samples in which the activity was expected to be found. From thousands of strains, we isolated a Pseudomonas sp. S34 producing a (S)-stereospecific esterase, and a thermostable esterase with (R)-form selectivity was also 。 btained from Bacillus stearothermophilus JYl44. To further analyse the gene structure and to induce a high level expression, two genes from each strain were cloned and sequenced. BLAST search results with the esterase gene from 534 revealed that both of gene (80-84 %) and amino acid sequences (89- 95 %) were highly conserved in the related esterases from Pseudomonas strains (fluorescens and aeruginosa). The thermostable esterase from JY144, however, revealed a relative low homology (45-52 %) to other esterase and/or lipase from related strains. Obviously, a complete conversion with pure enantiomer (R - or S) were readily achieved by recombinant clones expressing either (R)- or (S)- stereospecific esterase.

Biochemical and Molecular Characterization of High Population Density Bacteria Isolated from Sunflower

  • Natural and beneficial associations between plants and bacteria have demonstrated potential commercial application for several agricultural crops. The sunflower has acquired increasing importance in Brazilian agribusiness owing to its agronomic characteristics such as the tolerance to edaphoclimatic variations, resistance to pests and diseases, and adaptation to the implements commonly used for maize and soybean, as well as the versatility of the products and by-products obtained from its cultivation. A study of the cultivable bacteria associated with two sunflower cultivars, using classical microbiological methods, successfully obtained isolates from different plant tissues (roots, stems, florets, and rhizosphere). Out of 57 plant-growth-promoting isolates obtained, 45 were identified at the genus level and phylogenetically positioned based on 16S rRNA gene sequencing: 42 Bacillus (B. subtilis, B. cereus, B. thuringiensis, B. pumilus, B. megaterium, and Bacillus sp.) and 3 Methylobacterium komagatae. Random amplified polymorphic DNA (RAPD) analysis showed a broad diversity among the Bacillus isolates, which clustered into 2 groups with 75% similarity and 13 subgroups with 85% similarity, suggesting that the genetic distance correlated with the source of isolation. The isolates were also analyzed for certain growth-promoting activities. Auxin synthesis was widely distributed among the isolates, with values ranging from 93.34 to 1653.37 ${\mu}M$ auxin per ${\mu}g$ of protein. The phosphate solubilization index ranged from 1.25 to 3.89, and siderophore index varied from 1.15 to 5.25. From a total of 57 isolates, 3 showed an ability to biologically fix atmospheric nitrogen, and 7 showed antagonism against the pathogen Sclerotinia sclerotiorum. The results of biochemical characterization allowed identification of potential candidates for the development of biofertilizers targeted to the sunflower crop.

Diversity and Succession of the Bacterial Community during the Initial Fermentation Period in Modernized Soy Sauce (Ganjang) (개량식 간장의 발효 초기 단계에서의 미생물 다양성 및 천이에 관한 연구)

  • The taste and quality of soy sauce, a fermented liquid condiment, is greatly influenced by microbial metabolism during fermentation. To investigate the microbiological characteristics of ganjang during the initial fermentation process, we prepared meju (fermented soybean) blocks fermented with starter cultures and solar salts and analyzed the microbial community quantitively using 16S rRNA gene profiling from ganjang that had been fermented over a five-week period. The ganjang samples were collected and analyzed after soaking for week one (1W), three (3W), and five (5W) weeks. We found that Halomonadaceae was significantly higher in the 1W group (89.83%) than the 3W and 5W groups (14.46%, and 13.78%, respectively). At a species level, Chromohalobacter beijerinckii and Chromohalobacter canadensis were the dominant species in the 1W group but several taxa such as Bacillus subtilis, Pediococcus acidilactici, and Enterococcus faecalis were more abundant in the 3W and 5W groups. Pearson correlation analysis of the relative abundance of the bacteria showed a negative correlation between Chromohalobacter and two bacterial genera Bacillus and Enterococcus. Beta-diversity showed a statistical distinction between the 1W and the 3W and 5W groups, while no significance was evident between the 3W and 5W groups. Linear discriminant effect size analysis was used to identify biomarkers and significant differences in the relative abundance of several halophilic bacteria, Bacillus sp. and lactic acid bacteria at 1W, 3W, and 5W, recpectively, which indicates the important role of the bacterial community at these time points.
