Metabolism Activity of Bifidobacterium spp. by D.Ps of Konjac Glucomannan Hydrolysates (Konjac Glucomannan 가수분해 올리고당의 중합도별 Bifidobacterium spp.에 대한 대사활성)

  • Bacillus sp. $\beta$-mannanase was purified by DEAE-sephadex ion exchange column chromatography. The partially purified P-mannanase exhibited maximum activity at pH 6.0 and 5$0^{\circ}C$, and was stable at a pH range of 5.5 to 7.0, and at temperature between 30 to 5$0^{\circ}C$. Konjac glucomannan was hydrolyzed by the purified $\beta$-mannanase, and then hydrolysates separated by 1st activated carbon column chromatography and 2nd sephadex G-25 gel filtration. The main hydrolysates were composed of D.P 5 and 7 glucomannooligosaccharides by TLC and FACE method. To investigate the effects of guar gum glucomannooligosaccharides on the in vitro growth of B. longum, B. bifidum, B. infantis, B. adolescentis, B. animalis, and B. breve, Bifidobacterium spp. were cultivated individually on the modified-MRS medium containing carbon SOUTce such as D.P 5, and D.P 7 glucomannooligosaccharides, respectively. B. longum grew up 4.6-fold and 5.3-fold more effectively by the replacement of D.P 5 and 7 glucomannooligosaccharides as the carbon source in a comparasion of standard MRS. Also, B. breve and B. animalis slightly grew up by the treatment of D.P 5 glucomannooligosaccharide.

  • ${\gamma}$-PGA(poly-${\gamma}$-glutamic acid) is an unusual anionic polypeptide that is made of D- and L-glutamic acid units connected by amide linkages between ${\alpha}$-amino and ${\gamma}$-carboxylic acid groups. ${\gamma}$-PGA has been isolated from many kinds of organisms. Many Bacillus strains produce ${\gamma}$-PGA as a capsular material of an extracellular viscous material. It is safe for eating as a viscosity element of fermented soybean products such as Chungkookjang and Natto. It is biodegradable, edible and nontoxic toward humans and the environment and its molecular weight varies from ten thousand to several hundred thousand depending on the kinds of strains used. Therefore, potential applications of ${\gamma}$-PGA and its derivatives have been of interest in the past few years in a broad range of industrial fields such as food, cosmetics, medicine, water-treatment, etc. In this study, a bacterium, Bacillus subtilis GS-2 isolated from the Korean traditional seasoning food, Chungkookjang could produce a large amount of ${\gamma}$-PGA with high productivity and had a simple nutrient requirement. Based on carbon utilization pattern and partial 16S rRNA sequence analysis, the GS-2 strain was identified as B. subtilis. The determination of purified ${\gamma}$-PGA was confirmed with thin layer chromatography (TLC), high performance liquid chromatography (HPLC), fourier transform infrared (FT-IR) spectra, and $^1H$-nuclear magnetic resonance ($^1H$-NMR) spectroscopy.

Antifungfal Activity Against Plant Pathogenic Fungi on Insect Enterobacteriaceae (식물병원성 곰팡이에 대한 곤충장내세균의 항균활성)

  • In order to investigating the effects of antifungal activity of intestinal bacteria obtained from insect, it was identified these bacteria isolated from the gut. In this result, total 49 isolates of intestinal bacteria were identified from 10 kinds of insect species. It was that 4 isolates including Cedecea sp. from Nesidiocoris tenuis, 3 isolates including Enterobacter sp. from Odontotaenius disjunctus, 4 isolates including Acinetobacter sp. from Reticulitermes speratus, 4 isolates including Clavibacter sp. from Riptortus clavatus, 11 isolates including Bacillus sp. from Lema decempunctata, 3 isolates including Enterococcus sp. from Henosepilachna vigintioctopunctata 2 isolates including Staphylococccus sp. from Harmonia axyridis, 5 isolates including Enterobacter asburiae from Popillia mutans, 7 isolates including Aeromonas sp. from Hydrophilus acuminatus, and 7 isolates including Brucella sp. from Anomala octiescostata. In order to investigating antifungal activity against plant-pathogenic fungi, Altanaria solani, Colletotrichum gloeosporioides, Botrytis cinerea, Fusarium oxysporum, Phytophthora capsici, Rhizoctonia solani and Selerotinia sclerotiorum were dual cultured with each 49 gut enterobacteriaceae. As these results showed that many isolates have the antifungal activities including 26 isolates against A. solani, 6 isolates against B. cinerea, 13 isolates against C. gloeosporioides, 11 isolates against F. oxysporum, 17 isolates P. capsici, 2 isolates against R. solani and 2 isolates against S. sclerotiorum. Pseudomonas aeruginosa was showed strong antifungal activity against all of tested plant pathogens. It might be taken a potential for application against plant-pathogenic fungi with useful control agent.

Characterization of Bacteria Isolated from the Traditional Steeping Process and Their Application as Starters for the Production of Yugwa, a Korean Traditional Snack

  • During the traditional production process of yugwa, a Korean traditional oil-puffed snack, three bacterial isolates, 12, 37, and B1 from waxy rice slurry were characterized by using the molecular techniques, and the quality of the yugwa prepared by using three isolates was examined in relation to physical and sensory properties. Isolates 37 and B1 were identified as Lactobacillus sp. by 16S rDNA sequence analysis, while isolate 12 was as Bacillus amyloliquefaciens by gyrA sequence analysis. Waxy rice inoculated with isolate 12 demonstrated the lowest hardness and the highest peak and [mal viscosity. Yugwa prepared from isolate 12-inoculated waxy rice indicated the lowest hardness and the highest volume expansion, the key characteristics of yugwa. This result presents the possibility for the use of a starter in the commercial production of yugwa and other rice-based snacks.

Changes in isoflavone content and quality characteristics of Cheonggukjang prepared with Bacillus subtilis HJ18-3 and KACC 15935 (Bacillus subtilis HJ18-3과 KACC 15935를 이용하여 제조한 청국장의 품질특성과 isoflavone 함량의 변화)

  • This study was conducted in order to investigate the change of isoflavone composition (glycoside and bio-active aglycone), and to evaluate the quality characteristics of Cheonggukjang, which was prepared by different bacillus strains. After the 48-hour fermentation, the contents of daidzein, genistein, and glycitein in the Bacillus subtilis HJ18-3 have significantly increased up to approximately $89.06{\pm}3.59$, $10.36{\pm}0.28$, and $101.37{\pm}3.67ug/g$, respectively. The contents of daidzein, genistein, and glycitein in the Bacillus subtilis KACC 15935 were $38.88{\pm}5.39$, $12.58{\pm}2.14$, and $80.13{\pm}0.71ug/g$, respectively. The original content of daidzein was 3.96 ug/g, while genistein and glycitein were not measured. However, the contents of daidzen and genistein in HJ18-3 and in KACC 15935 were decreased. The ${\alpha}$-Amylase and cellulase activities of Chungkookjang in HJ18-3 were higher than in the KACC 15935. The contents of Chungkookjang in HJ18-3 were $29.70{\pm}11.66$ and $4861.3{\pm}388.07unit/g$, respectively. The amino type nitrogen contents and ammonia type nitrogen contents of Chungkookjang in KACC 15935 were higher than in the HJ18-3. These results suggested that it could be used to increase the bioactivity via fermentation with the Bacillus subtilis possessing a ${\beta}$-glucosidase activity with a view towards the development of functional foods.

Effects of Probiotic Microbes on Growth Performance, Innate Immunity, and Pathogen Sensitivity in Cultured Olive Flounder (Probiotic 기능을 가진 미생물을 함유한 양어용 생균제가 넙치의 성장, 선천성면역 및 항병능에 미치는 영향)

  • Probiotic principles can be applied in aquaculture for the purpose of growth and immunity stimulation, disease prevention and eventually better production performance. This study was to assess effects of combinations of microbes containing two Bacillus sp., plus one Lactobacillus sp. as the basal preparation (BSL-LAB), and additional Nitrosomonas sp. (nitrifying bacteria consortium, NBS) in olive flounder (Paralichthys olivaceus). The effects examined were growth parameters, hematologic parameters, innate immunity and pathogen challenge test. Fish were assigned to 4 treatments as Control (no probiotics), Group A (Bacillius and Lactobacillus to culture water), Group B (Bacillius and Lactobacillus both in water and feed), Group C (same as Group B with additional Nitrosomonas in feed). Fish were allocated to the above 4 groups, each group being composed of triplicate 30 fish, for a 7-week feeding in the laboratory. Positive effects were observed both in growth and pathogen sensitivity with all three probiotic combinations. Such effects were attributed to improved innate immune functions. This result indicates that the tested probiotic microbes are beneficial to olive flounder aquaculture.

Characterization of a Fibrinolytic Enzyme Produced by Bacillus subtilis MJ-226 Isolated from Meju (전통 메주에서 분리한 Bacillus subtilis MJ-226이 생산하는 혈전용해효소의 특성)

  • Among 27 Bacillus sp. isolated from Meju, a traditional Korean soybean fermented food, a strain MJ-226 was selected due to its strong fibrinolytic activity, and it was identified to be Bacillus subtilis MJ-226 according to morphological and biochemical characterization and sugar utilization. The fibrinolytic enzyme of B. subtilis MJ-226 was maximally produced by cultivating in the Tryptic Soy Broth (TSB) for 24~26 h at $37^{\circ}C$, and the enzymes activity was promoted with adding glucose, fructose, peptone or yeast extract to TSB. The fibrinolytic enzyme was stable at the range of pH from 6.0 to 8.0, and between 35 and $40^{\circ}C$. Also, when the crude enzyme was exposed to various metal ions and chemical inhibitors for 12 h, the enzyme stability was maintained by $MnSO_4$, $CaCl_2$, KCl, and NaCl. However, the stability was destroyed by treatment with $CuSO_4$, $MgSO_4$, $ZnSO_4$, $FeSO_4$, and $BaCl_2$, and the enzyme was unstable in the presence of chemical inhibitors such as iodoacetic acid, leupeptin, phenylmethanesulphonyl fluoride (PMSF), sodium dodecyl sulfate (SDS), thiourea, trans-1,2-diaminocyclohexane-N,N,N',N'-tetraacetic acid (CDTA) and ethylenediaminetetraacetic acid (EDTA).

Antagonistic Assay of Bacillus spp. for Eco-friendly Biological Control of Melon Powdery Mildew (멜론 흰가루병 친환경 생물적 방제를 위한 Bacillus속 균의 길항력 평가)

  • Melon powdery mildew, caused by Podosphaera fusca, is one of the serious diseases of melon plant in Korea. In this study, we evaluated the effect of selected antagonistic bacteria on the inhibition of mycelial growth of various plant pathogens, and control of melon powdery mildew. Based on the 16S rDNA and gyrA gene sequences, the selected antagonistic bacteria, M09, M70, and M99-1, were identified as Bacillus velezensis. These bacteria not only inhibited the mycelial growth of 47~69% in various plant pathogens, but also significantly reduced the incidence of powdery mildew. The three strains selected in this study could be used as potential biological control agents for various plant diseases as well as melon powdery mildew.

Characterization of Bacillus thuringiensis subsp. aizawai CAB109 isolate with bioactivities to Spodoptera litura and Spodoptera exigua (Lepidoptera: Noctuidae) (담배거세미나방과 파밤나방에 활성이 있는 Bacillus thuringiensis subsp. aizawai CAB109 균주의 특성)

  • Bacillus thuringiensis subsp. aizawai CAB109 isolated in Korea is known active against Spodoptera sp.. Especially, B. thuringiensis aizawai CAB109 isolates showed 100% mortality against Spodoptera litura and Spodoptera exigua. To screen highly active B. thuringiensis, the pathogenicity of B. thuringiensis CAB109 was compared with that of commercialized B. thuringiensis products. $LC_{50}$ values of CAB109, product TB-WP and product SC strains of B. thuringiensis were $1.3{\times}10^5$, $2.3{\times}10^6$ and $5.2{\times}10^5\;cfu/ml$ against the 2nd larva of S. litura and $1.8{\times}10^4$, $1.3{\times}10^6$ and $1.5{\times}10^6\;cfu/ml$ against the 2nd larva S. exigua, respectively. To determine new gene's existence and absence, the plasmid DNA was extracted, and compared to that of B.t. aizawai HD-133. Both B. thuringiensis were not like plasmid DNA pattern. PCR technique was used to predict both plasmid DNA's cry gene. PCR products analysis showed that B.t. CAB109 harbor Cry1Aa, Cry1Ab, Cry1C and Cry1D and B.t. HD-133 has Cry1Aa and Cry1Ab, respectively.
