Enzymatic activity of Endoplasmic Reticulum Oxidoreductin 1 from Bombyx mori

  • Most proteins produced in the endoplasmic reticulum (ER) of eukaryotic cells fold via disulfide formation (oxidative folding). Oxidative folding is catalyzed by protein disulfide isomerase (PDI) and PDI-related ER protein thiol disulfide oxidoreductases (ER oxidoreductases). In yeast and mammals, ER oxidoreductin-1s (ERO1s) supply oxidizing equivalent to the active centers of PDI. We previously identified and characterized the ERO1 of Bombyx mori (bERO1) as a thioredoxin-like protein that shares primary sequence homology with other ERO1s. Here we compare the reactivation of inactivated rRNase and sRNase by bERO1, and show that bERO1 and bPDI cooperatively refold denatured RNase A. This is the first result suggesting that bERO1 plays an essential role in ER quality control through the combined activities of bERO1 and bPDI as a catalyst of protein folding in the ER and sustaining cellular redox homeostasis.

Analysis of Antioxidant Content and Growth of Agastache rugosa as Affected by LED Light Qualities (LED 광질 조건에 따른 배초향 생장 및 항산화 물질 함량 분석)

  • The aim of this study was to evaluate the effect of light quality using either monochromatic or combined LEDs on the growth and antioxidant accumulation of Agastache rugosa cultivated under hydroponics for 4 weeks. This experiment was performed in a controlled-environment room at $22{\pm}1^{\circ}C$ and $18{\pm}1^{\circ}C$ (day and night temperatures, respectively) and 50-70% relative humidity, with a provided photosynthetic photon flux density (PPFD) of $180{\pm}5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ and irradiated with either monochromatic (W10 and R10) or mixed LEDs (W2B1G1, R3B1, R2B1G1, and W2B1G1) with a differing ratio of each LED's PPFD and fluorescent lighting (FL: control) with a 16/8 h photoperiod. Fresh and dry weights were highest for plants grown under the W2B1G1 treatment. A. rugosa grown with R10 had the greatest plant height but the lowest SPAD among all treatments. The concentration of rosmarinic acid in plants grown under W2B1G1 was significantly higher than that of plants grown under other treatments. Tilianin content was significantly higher in R3B1 than in the other treatments. However, whole-plant rosmarinic acid and tilianin content was the highest under the W2B1G1 condition. To cultivate A. rugosa in a plant factory, mixed-LED light conditions with W2B1G1 is considered to be more advantageous for the growth and antioxidant accumulation of A. rugosa. It is though that the total whole-plant antioxidant content is more crucial for commercial use; the present study demonstrates the potential to achieve higher content of functional materials in plants through the selection of light quality.

The evaluate the usefulness of various CT kernel applications by PET/CT attenuation correction (PET/CT 감쇠보정시 다양한 CT Kernel 적용에 따른 유용성 평가)

  • Purpose Recently PET/CT image's attenuation correction is used CTAC(Computed Tomgraphy Attenuation Correction). it can quantitative evaluation by SUV(Standard Uptake Value). This study's purpose is to evaluate SUV and to find proper CT kernel using CTAC with applied various CT kernel to PET/CT construction. Materials and Methods Biograph mCT 64 was used for the equipment. We were performed on 20 patients who had examed at our hospital from february through March 2017. Using NEMA IEC Body Phantom, The data was reconstructed PET/CT images with CTAC appiled various CT kernel. ANOVA was used to evaluated the significant difference in the result. Results The result of measuring the radioactivity concentration of Phantom was B45F 96% and B80F 6.58% against B08F CT kernel, each respectively. the SUVmax increased to B45F 0.86% and B80F 6.54% against B08F CT kernel, In case of patient's parts data, the Lung SUVmax increased to B45F 1.6% and B80F 6.6%, Liver SUVmax increased to B45F 0.7% and B80F 4.7%, and Bone SUVmax increased to B45F 1.3% and B80F 6.2%, respectively. As for parts of patient's about Standard Deviation(SD), the Lung SD increased to B45F 4.2% and B80F 15.4%, Liver SD increased to B45F 2.1% and B80F 11%, and Bone SD increased to B45F 2.3% and B80F 14.7%, respectively. There was no significant difference discovered in three CT kernel (P >.05). Conclusion When using increased noise CT kernel for PET/CT reconstruction, It tends to change both SUVmax and SD in ROI(region of interest), Due to the increase the CT kernel number, Sharp noise increased in ROI. so SUVmax and SD were highly measured, but there was no statistically significant difference. Therefore Using CT kernel of low variation of SD occur less variation of SUV.

Preferential Flow as Tested by Breakthrough Curves of Cl- and Cu2+ from Saturated Undisturbed Soil Core Samples under Steady Flow Conditions (포화 불교란 토양시료의 Cl- 및 Cu2+ 출현곡선에 의한 preferential flow의 검증)

  • Preferential flow has recently been the subject of increasing interest because these phenomena contribute to solute transport in soils. Commonly, preferential flow paths are associated with macropores or highly structured soils. We presented an analysis of the measured breakthrough curves (BTCs) of $Cl^-$ and $Cu^{2+}$ ions to test the occurrence of preferential flow in soils using miscible displacement technique under steady flow conditions. We also analyzed soil water retention curves and from this curves induced cumulative pore size distribution of undisturbed soils, which sampled from Ap1, B1, and C horizons of Songjeong series soils (the fine loamy, mesic family of Typic Hapludults). In this study, miscible displacement experiment on C horizon was excluded, because it is structureless sandy loam with saturated hydraulic conductivity of $5.2cmhr^{-1}$. The saturated hydraulic conductivity of Ap1 horizon was $2.0cmhr^{-1}$, which was about 7 times higher than that of B1 horizon ($0.27cm hr^{-1}$). Cumulative pore size distribution predicted that Ap1 horizon had more macropores (pore diameter larger than $49{\mu}m$, equivalent to -6 kpa of soil matric potential) than B1 horizon. The hydrodynamic dispersion coefficient from chloride BTCs was estimated as $1.3cm^2hr^{-1}$ for B1 and $34cm^2hr^{-1}$ for Ap1 horizon. However the retardation factors of B1 and Ap1 horizon were significantly different, i.e. 1 and 0.6, respectively, which means that there was distinct partition between mobile water and immobile phase in Ap1 horizon. The copper retardation effect of Ap1 horizon was less than that of B1 horizon, even though cation exchange capacity of Ap1 horizon was higher than that of B1 horizon. Thus, breakthrough curves of $Cl^-$ and $Cu^{2+}$ obviously showed the probability that preferential flow would occur in Ap1 horizon.

  • Supported Pd($Pd/AlF_3$, $Pd/{\gamma}-Al_2O_3$) catalysts and solid-acid catalysts(${\gamma}-Al_2O_3$, ${\alpha}-Al_2O_3$, $AlF_3$) were used to perform dechlorination of HCFC-142b(1-chloro-1,1-difluoroethane) in the presence of excess hydrogen. In the reactions the effects of reaction temperature, the mole ratio(r) of $H_2$ to HCFC-142b and the amount of supported Pd on dechlorination of HCFC-142b into HFC-143a(1,1,1-trifluoroethane) or HFC-152a(1,1-difluoroethane) were investigated. The experimental results showed that the conversion of HCFC-142b to product gases were 60% and 92%, respectively, and the selectivity to HFC-143a in the product gases were 58% and 64% for $Pd/AlF_3$ and $Pd/{\gamma}-Al_2O_3$ catalysts, respectively. On these catalysts an optimum reaction condition was found at $200^{\circ}C$ with the space time of reactant gases as 1.05 second and the mole ratio of $H_2$ to HCFC-142b as 3. Solid-acid catalysts were also tested at the same reaction condition. The results showed that the conversions of HCFC-142b to product gases were 12%, 8% and 7%, and the selectivities to HFC-152a were 94%, 92% and 90% for ${\gamma}-Al_2O_3$, ${\alpha}-Al_2O_3$ and $AlF_3$ catalysts, respectively.

Improvement of a Nitrogen Fertilizer Recommendation Model by introducing a Concept of the Mitscherlich-Baule-Spillman Equation (토양검정에 의한 질소시비량 결정에 Mitscherlich-Baule-Spillman 개념의 응용)

  • In order to improve a soil test-based fertilization model for nitrogen, the Mitscherlich-Baule-Spillman equation modified by Bray and his coworkers was introduced and tested for its possibility of application. The coefficients $C_1$ and $C_2$ in the equation A were calculated by applying 10 pot experimental results done by the National Institute of Agricultural Sciences and Technology. Then, a regression(B) between C_1 and x, the fertilizer level was estimated. log(Ymax-Y) = logYmax-$C_1x-C_2b$ (A) where Ymax : maximum yield. Y : yield without or with a limited amount of fertilizer applied, x : application level of fertilizer, b : content of a nutrient in soil, $C_1$ and $C_2$ : coefficients of x and b, respectively. log(0.05)=$-C_1x-C_2b$ (B) Fertilizer requirement calculated by the equation B was significantly correlated with the real level of fertilizer applied for 95 percentage of the maximum yield. A more extensive and precise study on $C_1$ and $C_2$ in the equation A in expected to improve significantly the fitness of soil analysis to the fertilizer recommendation.

Studies on Bacterial Characteristics of Bacillus cereus Group LS-1 Isolated from Suyeong Bay (수영만에서 분리된 Bacillus cereus Group LS-1 의 세균학적 특성에 관한 연구)

  • These studies were carried out to identify Bacillus cereus group 1..5-] strain isolated from 5uyeong Bay. This strain was differentiated from B. cereus group using conventional, API system and fatty acid composition analysis. Colony characteristics were opague. mucoid, entire margin. convex. circular and non hemolysis on sheep blood agar plates, and were observed with central spore forming positive bacilli in a Gram stained preparation. and had no motility. The carbohydrates tested; glucose.maltose, and sucrose were assimilated but neither trehalose nor salicin were assimilated. This strain ultilized gelatin and was also inhibited by 6.5% NaCI. The results of biochemical examination were differented from B. cereus group LS-1 compared with others B. cereus group. The fatty acid composition contained major amounts of branched chain acids. iso $C_{15}$ and iso $C_{13}$ and the range of chain length was $C_{12}$ to C"$C_{17}$ and n$C_{15}$, acid was not detected. Automated fatty acid computer profile indicated "B. mycoides GC subgroup B of 0.312 similarity index." The results agreed with other research cases. On the other hand. A TB computer prolile index of API system (API 50 CHB & API 20E) identified" Doubtful profile of 99.7% B. firmus" . These results were presented with considerable discrepancies between API system and fatty acid analysis. With 67 biochemical characters. the similarity matrix of B. mycaides (KCTC 1033). B. thuringiensis (KCTC 1033). B. cereus (5-3) and B. mycoides (S-12) showed 42%. 42%. 59%, and 52%. respectively. Through the key tests and fatty acid analyses. we could notice the appearance of B. mycoides of the B. cereus group and this leads us to suspect the existence of a new biotype B. mycoides.

NF-${\kappa}B$ Activation and cIAP Expression in Radiation-induced Cell Death of A549 Lung Cancer Cells (A549 폐암세포주의 방사선-유도성 세포사에서 NF-${\kappa}B$ 활성화 및 cIAP 발현)

  • Background : Activation of the transcription factor NF-${\kappa}B$ has been shown to protect cells from tumor necrosis factor-alpha, chemotherapy, and radiation-induced apoptosis. NF-${\kappa}B$-dependent cIAP expression is a major antiapoptotic mechanism for that. NF-${\kappa}B$ activation and cIAP expression in A549 lung cancer cells which is relatively resistant to radiation-induced cell death were investigated for the mechanism of radioresistance. Materials and methods : We used A549 lung cancer cells and Clinac 1800C linear accelerator for radiation. Cell viability test was done by MTT assay. NF-${\kappa}B$ activation was tested by luciferase reporter gene assay, Western blot for $I{\kappa}B{\alpha}$ degradation, and electromobility shift assay. For blocking ${\kappa}B$, MG132 and transfection of $I{\kappa}B{\alpha}$-superrepressor plasmid construct were used. cIAP expression was analyzed by RT-PCR and cIAP2 promoter activity was performed using luciferase assay system. Results : MTT assay showed that cytotoxicity even 48 hr after radiation in A549 cells were less than 20%. Luciferas assay demonstrated weak NF-${\kappa}B$ activation of $1.6{\pm}0.2$ fold compared to PMA-induced $3.4{\pm}0.9$ fold. Radiation-induced $I{\kappa}B{\alpha}$ degradation was observed in Western blot and NF-${\kappa}B$ DNA binding was confirmed by EMSA. However, blocking NF-${\kappa}B$ using MG132 and $I{\kappa}B{\alpha}$-superrepressor transfection did not show any sensitizing effect for radiation-induced cell death. The result of RT-PCR for cIAP1 & 2 expression was negative induction while TNF-${\alpha}$ showed strong expression for cIAP1 & 2. The cIAP2 promoter activity also did not show any change compared to positive control with TNF-${\alpha}$. Conclusion : We conclude that activation of NF-${\kappa}B$ does not determine the intrinsic radiosensitivity of cancer cells, at least for the cell lines tested in this study.

High Frequencies of the CCR2b-64I and SDF1-3'A Mutations with HIV Infection in Koreans

  • Background: Host genetic polymorphisms in the HIV-1 co-receptor CCR5 and CCR2b and SDF-1, ligand for co-receptor CXCR4, have been known to be associated with the resistance of HIV infection and/or the delayed disease progression in HIV-infected patients. Methods: We examined the frequencies of SDF1-3'A and CCR2b-64I alleles of 354 Koreans including 100 HIV-uninfected persons, 13 discordant spouses of HIV-infected persons, and 241 HIV-infected persons. The genotyping assays of SDF1 and CCR2b genes were carried out by polymerase chain reaction-restriction fragment length polymorphism. Results: The frequencies of CCR2b-64I and SDF1-3'A alleles in Koreans were very high compared with Caucasians and blacks. Observed frequencies of CCR2b-64I and SDF1-3'A allelic variants were 25.1% and 28.7%, respectively. The frequency of the CCR2b-64I allele in Koreans was 2~4 times higher than those of other ethnic groups with the exception of Asian. The frequencies of CCR2b-64I and SDF1-3'A genotypes did not show the significant difference between HIV-infected and uninfected Koreans. However, the prevalence of CCR2b-64I genotype of the LTNP group was about two times higher than that of the remainder group (P< 0.05). Four (45%) out of 9 LTNPs (long-term nonprogressors) showed having the SDF1-3'A allele and 7 (78%) out of 9 LTNPs carried the CCR2b-64I allele. 3 (33%) out of 9 LTNPs had both SDF1-3'A and CCR2b-64I alleles. But none of 5 RPs (rapid progressors) appeared to have both SDF1-3'A and CCR2b-64I alleles. Conclusion: The different genetic backgrounds in study populations may affect the disease progression and the AIDS epidemic in each country. Further studies need to define whether high frequencies of CCR2b-64I and SDF1-3'A allelic variants may affect the HIV disease progression.

Comparison of immunogenecities of three beta-nodavirus proteins, capsid protein, non-structural protein B1 and B2 in olive flounder

  • The genomic and subgenomic RNAs of fish nodavirus encode the four proteins, protein A, capsid protein, non-structural protein B1 and B2. In this study, we describe the immune response of olive flounder Paralichthys olivaceus immunized with live fish nodavirus or recombinant capsid protein, non-structural protein B1 and B2 expressed in E. coli. Nodavirus-infected flounder produced antibodies to capsid protein, B1 and B2 and nodavirus-neutralizing activities were detected in the serum of the nodavirus-infected flounder. The flounder were immunized against the three recombinant proteins of fish nodavirus and the sera from these immunized fishes were assayed for nodavirus-specific antibody by ELISA and a neutralization test. In the immunized flounder, all three recombinant proteins induced the production of similar levels of antibody, but only the antibody to capsid protein significantly neutralized nodavirus. These results indicate that all three nodaviral proteins are immunogenic in flounder, but only the capsid protein can induce neutralizing antibody against nodavirus.
