A Study on the Trend of Changes in the Positive Rate of Hepatitis B Surface Antigens and Antibodies over the Last 9 Years in One Health Care Center

  • The purpose of this study was to provide basic data on hepatitis B infection control in the community through the results of the hepatitis B surface antigen and antibody tests conducted at the Cheongyang-County Health Medical Center. From 2012 to 2020, we retrospectively analyzed the HBsAg, HBsAb, HBeAg, HBeAb, and HBV DNA results of 7,329 hepatitis B-related testers. Among 7,329 subjects, the HBsAg positivity rate was 1.7%, and the positivity rate according to age was the highest at 4.4% in their 30s, 4.2% in their 40s, 4.1% in their 50s, 2.0% in their 60s, 1.9% in their 70s and over, and 10 it was shown in the order of 0.3% from less than large. The HBsAb positivity rate was 43.1% for men, 38.2% for men, and 46.7% for women (P<0.001). To summarize the above results, for infection control of hepatitis B in Cheongyang-County, hepatitis surface antigen proton management is required for those in their 30s or older, and it is thought that efforts to acquire immunity are necessary for those in their 20s or younger.

Antioxidant Effect of Filipendula glaberrima Nakai Extract in HepG2 Cells

  • The imbalance of oxidative stress due to the excessive production of reactive oxygen species (ROS) leads to the pathogenesis of liver disease. To prevent this, the role of antioxidant mechanisms is important. Antioxidant studies have been reported on the Filipendula glaberrima Nakai. However, studies applied to HepG2 cells, which are human liver cells, have not yet been conducted. In this study, 70% ethanol extract of Filipendula glaberrima Nakai (FGE) was prepared and antioxidant activity was investigated. It was confirmed whether FGE pretreatment could reduce hydrogen peroxide-induced oxidative stress in HepG2 cells. The increase in gene expression of antioxidant biomarkers and the scavenging ability of ROS were measured, and Hoechst 33342 staining was used to know the inhibitory effect of the apoptosis. As a result, FGE significantly increased SOD (2.6-fold), CAT (4.4-fold), MT-1A (3.1-fold), GPx (4-fold), and G6PD (2.4)-fold compared to the H2O2-treated group. FGE directly inhibited ROS production from 13.4 to 3.6 (the fluorescence mean of DCF-DA) and also reduced apoptotic cells from 45% to 10% (Hoechst 33342 staining) at 2.5 ㎍/mL. These results demonstrate the excellent antioxidant activity of FGE and show that it can be used as a functional food to prevent liver disease.

The Association of Pulse Pressure and Pre-Metabolic Syndrome in Korean Middle Aged Men

  • Pulse pressure is an independent risk factor for cardiovascular disease. The aim of the present study was to assess the association between pulse pressure and metabolic syndrome in Korean men. The study subjects were 8,439 adults aged 40 to 64 years, who underwent health screening examination from January 2012 to December 2014 at the Health Promotion Center of one hospital in Gyeonggi-do for general health check-up. They include the metabolic syndrome absent group (Absent, n=3,078), the pre-metabolic syndrome group (Pre-MetS, n=4,242) and the metabolic syndrome group (MetS, n=1,119). Progressive increase in pulse pressure was demonstrated for increasing components of the metabolic syndrome (P<0.001). The pulse pressure according to the degree of the metabolic syndrome was higher in the pre-MetS and MetS groups compared to the Absent group (P<0.001). Systolic and diastolic blood pressure, total cholesterol, triglyceride, fasting glucose, and abdominal obesity positively correlated with pulse pressure (P<0.001).

Hypocholesterolemic Effect of Ginkgo Biloba Seeds Extract from High Fat Diet Mice

  • To investigate the efficacy of extract of Ginkgo Biloba seeds in high fat diet (HFD) in mice, the Ginkgo Biloba seeds extract (GSE) was orally administered to mice with a HFD at 300 mg/kg/day for 4 weeks. Our results show that GSE significantly inhibited fat accumulation. Moreover, GSE markedly reduced the final body weight with a decrease in epididymal adipose tissue mass and adipocyte size compared with the untreated HFD-induced group. Additionally, GSE ameliorated serum high-density lipoprotein cholesterol levels. The results show that Ginkgo Biloba seeds possesses hypocholesterolemic effect through down regulating lipid metabolism. Further studies are required in this area to strengthen the anti-obesity effects of GSE with active component, and it can be used a pro-drug instead of whole extract.

  • To investigate the efficacy of extract of Tartary buckwheat (F. tataricum Gaertn.) in high fat diet (HFD) in mice, the F. tataricum Gaertn. extract (FTE) was orally administered to mice with a HFD at 300 mg/kg/day for 4 weeks. 진동하는 익형 주위의 유동장 해석을 위한 SST 난류 모델의 수정 (Modification of SST Turbulence Model for Computation of Oscillating Airfoil Flows)

  • A modified version of SST turbulence model is suggested to simulate unsteady separated flows over oscillating airfoils. The original SST model, which shows good performance in predicting various steady flows, often results in oscillatory behavior of aerodynamic loads in large separated flow regions. It is shown that this oscillatory behavior is due to the adoption of the absolute value of vorticity in generalizing the original model. As a remedy, a modification is made such that the vorticity in the original SST model is replaced by strain rate. The present model is verified for a mild separated airfoil flow at fixed angle of incidence and for unsteady flowfields about oscillating airfoils. The results are compared with BSL model and original SST model. It is illustrated that the present model gives a better agreement with the experimental results than other two models.

The Ameliorative Effect of Adenophorae Radix on Atopic Dermatitis in vivo and in vitro

  • Adenophorae Radix (AR) has been used as a traditional medicine for various diseases. However, the regulatory effects of AR in atopic dermatitis are not yet understood. This study attempted to determine the pharmacological effects of AR and its constituent on both compound 48/80 or histamine-induced scratching behaviors and 2, 4-dinitrochlrobenzene (DNCB)-induced atopic dermatitis in mice. The findings of this study demonstrated that AR reduced compound 48/80 or histamine-induced scratching behaviors in mice. Treatment of AR attenuated the AD symptoms such as eczematous, erythema and dryness and serum IgE and IL-6 levels in AD model. Additionally, AR inhibited the TNF-${\alpha}$-induced the Nuclear factor-${\kappa}B$ activation in HaCaT cells. Collectively, the findings of this study provide us with novel insights into the pharmacological actions of AR as a potential molecule for therapeutic agent against atopic dermatitis.

Inhibitory Effect of S100A8 on Neutrophil Apoptosis by Cytokine Release of Normal and Allergic Monocytes

  • S100A8 functions as an essential factor in inflammatory response. Cytokine release of monocytes and regulation of neutrophil apoptosis are important steps in pathogenesis of allergy. This study aims to examine the relation between cytokine release of monocytes due to S100A8 and neutrophil apoptosis. S100A8 enhanced the release of IL-6 and IL-8 in monocytes of normal and allergic subjects. Treatment of supernatants of normal and allergic monocytes with S100A8 blocked neutrophil apoptosis by inhibition of caspase 9 and caspase 3 activation. The secretion signal induced by S100A8 is involved in TLR4, Src family protein, PKCδ, ERK1/2, p38 MAPK, JNK, and NF-κB. These findings may contribute to understanding the complex pathogenesis of allergic diseases by determining inflammatory responses associated with S100A8, monocytes, and neutrophils.

Inhibition of 5α-reductase of de novo Generation of Short Anti-oxidant Peptides

  • This study aims to investigate the biological activities related to hair loss of short anti-oxidant peptides (DK peptides) $5{\alpha}$-reductase inhibition and anti-oxidation. The series of DK peptides were generated amphipathic helical properties using leucines, lysines and tryptophan residues. Cell viability and free radical scavenging activities were performed using 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl-tetrazolium bromide (MTT) and 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay, respectively. The DK peptides were investigated for inhibitory activity against $5{\alpha}$-reductase. Antioxidant activities were determined by means of, 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assays. All peptides could inhibit $5{\alpha}$-reductase in lipopolysaccharide-stimulated macrophage. In conclusion, DK peptides was suggested as the most attractive ingredients for improving hair loss, because of the high inhibitory against $5{\alpha}$-reductase inhibition.

Noninvasive Detection of Specific Diagnostic Biomarkers for Atopic Dermatitis

  • The diagnosis of atopic dermatitis (AD) includes a test that checks allergen-mediated skin reactions and a method of measuring the total IgE and allergen-specific IgE in blood. However, these test methods are performed directly on the patient, which cause some pain or discomfort. In addition, the skin response test or IgE may result in false negative in about 20% of patients. In the present study, to identify specific biomarkers, HaCaT cells were used as a human keratinocyte that make up the skin, were treated IL-4 and IL-13 for 24 hours to induce a situation similar to keratinocytes in AD patients. In the HaCaT cells, pro-inflammatory cytokine such as IL-5, IL-6, and MCP-1 were increased by IL-4 and IL-13 and skin barrier proteins was reduced by IL-4 and L-13. This results showed that a situation similar to the stratum corneum of an actual patient is induced in HaCaT cells. And then the secretions of Kallikrein (KLK) 5 and KLK7 protease were checked by enzyme-linked immunosorbent assay (ELISA). It was specifically increased by IL-4 and IL-13. This showed that AD-related protease can be detected at the protein level using keratinocytes that can be taken in a non-invasive manner and suggested the possibility of applying it to AD diagnosis.
