Yeast의 FLP/FRT 시스템을 이용한 BmNPV의 유전자 재조합 (Construction of Recombinant Bombyx mori Nuclear Polyhedrosis Virus Using a FLP/FRT System of Yeast, Saccharomyces cerevisiae 2$\mu$m plasmid)

  • For the construction of plasmid and bmNPV sarrying the FRT recognition site for the FLP recombinases, we synthesized the wild type FRT dligonucleotides. The target FRT sequences consist of three 13bp repeated DNA sequences; two repeats in a direct orientation and one inverted relative to the other two. In addition, there is an 8bp spacer region between the repeats which determune the orientation of the FRT recombination site. In order to place the FRT site both in target BmNPV genome and the transfer vector, we constructed a plasmid, FRT site both in the target BmNPv genome and the transfer vector, we constructed a plasmid, pFRT$\beta$-gal, carrying the FRT sites within the cloning sites of pSV vector and a recombinant BmNPV, vFRTPH, carrying the FRT sites at a downstream of polyhedrin promotor, respectively. In order to test the functionality of the FLP/FRT site-specific recombination system, vFRTPH, pFRT$\beta$-gal and pHsFLP DNA were co-transfected into BmN-4 cells. The resulting recombinant virus was designated a vFRT$\beta$2-gal. From construction analysis of the vFRT$\beta$2-gal with PCR technique it was concluded that the entire pFRT$\beta$-gal plasmid with $\beta$-galactosidase gene and origines of replication flanked by two functional hybrid FRT sequences. The efficiency of recombination was 8.7%, which was higher than that(2.2%) of recombination between a conventional transfer vector and the wild type BmNPV.

Optimization of block-matching and 3D filtering (BM3D) algorithm in brain SPECT imaging using fan beam collimator: Phantom study

  • The purpose of this study is to model and optimize the block-matching and 3D filtering (BM3D) algorithm and to evaluate its applicability in brain single-photon emission computed tomography (SPECT) images using a fan beam collimator. For quantitative evaluation of the noise level, the coefficient of variation (COV) and contrast-to-noise ratio (CNR) were used, and finally, a no-reference-based evaluation parameter was used for optimization of the BM3D algorithm in the brain SPECT images. As a result, optimized results were derived when the sigma values of the BM3D algorithm were 0.15, 0.2, and 0.25 in brain SPECT images acquired for 5, 10, and 15 s, respectively. In addition, when the sigma value of the optimized BM3D algorithm was applied, superior results were obtained compared with conventional filtering methods. In particular, we confirmed that the COV and CNR of the images obtained using the BM3D algorithm were improved by 2.40 and 2.33 times, respectively, compared with the original image. In conclusion, the usefulness of the optimized BM3D algorithm in brain SPECT images using a fan beam collimator has been proven, and based on the results, it is expected that its application in various nuclear medicine examinations will be possible.

120 GHz 국부발진기의 설계 및 제작 (A Design and Fabrication of 120 GHz Local Oscillator)

  • 본 논문은 캐리어 주파수가 240 GHz인 THz 대역 송수신기에 있어서, 서브하모닉 믹서의 LO 주파수로 사용되는 120 GHz 국부발진기의 설계 및 제작에 관한 것이다. 120 GHz 국부발진기는 40 GHz PLL(Phase Locked Loop), 40 GHz 대역통과필터(Band Pass Filter), 3 체배기(frequency tripler), 120 GHz 대역통과필터로 구성되어 있으며, 3 체배기는 상용품을 이용하였다. 40 GHz PLL의 위상잡음은 100 kHz offset 주파수에서 -105 dBc/Hz의 성능을 보였고, 120 GHz의 대역통과필터의 중심주파수 119 GHz, 대역폭 5 GHz 일 때 삽입손실은 1.3 dB로 측정되었다. 제작된 120 GHz 국부발진기의 최종 출력은 6.6 dBm이었다.

Gelatinase, a Possible Etiologic Factor of Photoaging, is Present in Healthy Human Facial Skin and is Inhibited by Turmeric Extract

  • Influence of gelatinase on basement membrane (BM) structure was investigated by using a skin equivalent (SE) model. The results showed that (1) gelatinase produced by cells degraded the BM and (2) the addition of matrix metalloproteinase-specific inhibitor to the SE medium accelerated the formation of BM structure, indicating that gelatinase is involved in BM impairment. The activity of gelatinase was also studied in healthy human facial skin tissues. The result of in situ zymography revealed gelatinase activity around the basal layer of the epidermis, where BM integrity was severely compromised. Therefore, this enzyme was suggested to be associated with BM decomposition in human facial skin. To assess the behavior of gelatinase in stratum corneum (SC) non-invasively, an immunological study was performed. Since positive immunostaining of pro-gelatinase B was observed in SC stripped from sun-exposed skin, whereas no positive staining detected in SC of non-irradiated skin, gelatinase in the epidermis could be non-invasively detected by measuring gelatinase in SC. Gelatinase in SC of healthy female volunteers was monitored using a special film that sensitively and conveniently detects gelatinase. Ninetr percent of SC from facial skin (l00 women, 40's-50's) was gelatinase-positive. On the other hand, SC from non-irradiated skin was negative. These results strongly suggest that (1) gelatinase is constantly produced in the facial epidermis of most middle-aged woman during their daily life, and (2) the enzyme might be involved in the aging-related degeneration of both BM and the matrix fibers of the upper layer of the dermis, acting as a very important aging factor. Strong inhibitory activity against gelatinase was found in turmeric extract and identified curcumin as the major ingredient. Topical application of cream containing turmeric extract significantly decreased the number of gelatinase-positive SC clusters in human facial skins. These results indicated that turmeric is an effective ingredient to prevent skin from photo aging by suppressing chlonically upregulated gelatinase activity by UV and to improve skin condition.

BM 미생물제제를 이용한 선박 오·폐수 내 유해물질처리 (Harmful Materials treatment in Shipboard sewage by SBR process with BM)

  • 본 연구에서는 선박에서 발생하는 오 폐수의 처리를 위하여 SBR공정에 유효미생물을 주입하는 변법을 이용하여 Lab scale 실험을 수행하였다. 유해물질 유입에 따른 생물학적 처리 장치의 효율 저하 문제를 해결하기 위하여 SBR공정에 유효미생물을 주입하는 변법은 크루즈선이라는 특수 환경과의 접목성과 생물학적 처리 시 야기될 수 있는 문제를 대비하기 위한 대안으로 선박환경에 매우 적합한 공정으로 평가되었다. 슬러지 관찰 결과 기존의 활성슬러지에 유효미생물의 주입함으로써 슬러지의 안정성을 확보할 수 있었으며, 슬러지의 EPS 함량도 40% 이상 높아진 것을 확인할 수 있었다. 또한 슬러지의 미생물 분석 결과 유효미생물 주입으로 인해 수처리에 유리한 미생물종이 다수 출현하여 휘발성 유기화합물과 같은 유기 유해물질이 생분해되어 안전한 물질로 전환되는 것을 확인할 수 있었으며. 중금속과 같은 무기 유해물질도 중금속의 종류와 유입농도에 영향을 받지 않고 70% 이상의 안정적인 처리 효율도 확보할 수 있는 것으로 확인되었다.

  • 본 논문은 LTE-Advanced 시스템에 적용할 수 있는 2,500 MHz~2,570 MHz 대역 0.13-${\mu}m$ CMOS RF front-end 송신기를 제안하며 I/Q 상향주파수변환기와 구동증폭기로 구성되어있다. 상향주파수변환기는 우수한 선형특성을 얻기 위해 공진회로를 부하로 사용하였으며 국부발진신호의 누설을 줄이기 위해 전류 보상회로를 사용하였다. 또한, 제안하는 구동증폭기는 높은 전류 효율과 우수한 선형특성을 확보하기 위해 Class AB 바이어스 상태로 설계되었다. 측정 결과 제안하는 RF front-end 송신기는 최대 +6 dBm의 출력 파워를 제공하며, +0 dBm 출력 시 이미지 신호 및 국부 발진 누설 신호와 40 dBc의 차이를 보인다. 제작된 칩은 1.2 V의 공급 전압으로부터 36 mA 전류를 소모한다.

Transcription of Some Early and Late Genes of Bombyx mori Nuclear Polyhedrosis Virus in the Cells

  • To understand expression of some early and late genes of Bombyx mori nuclear polyhedrosis virus (BmNPV) in the B. mori-derived BmN cell line, the transcripts were analyzed by polymerase chain reaction with synthetic primers. After infection, the transcript of early genes, which include p35, IE1 and helicase p143, was immediately detected in the infected cells. In addition, the transcript of late genes, which include p10 and polyhedrin, was also detected in just-infected cells. In conclusion, our results revealed that transcripts of early and late genes of BmNPV are immediately expressed from the cells after infection.

  • This paper presents the design and analysis of LNA-Mixer for 2.45GHz RFID reader. The LNA is implemented by PCSNIM method for low power consumption. The Mixer is implemented by using the Gilbert-type configuration, current bleeding technique, and the resonating technique for the tail capacitance. The connection between the two designed circuits is made by active balun. This LNA-Mixer has about 35dB for -40dBm input RF power, LO power is 0dBm and RF frequency is 2.45 GHz and IIP3 is -4dBm. The layout of LNA-Mixer for one-chip design in a $0.18-{\mu}m$ TSMC process has 2.6mm ${\times}$ 1.3mm size.

  • 본 논문에서는 VHF대역 기지국용 전력증폭기에 대해 입력레벨이 $0dBm{\pm}1 dB$, 출력이 $45dBm{\pm}1dB$인 증폭기를 모듈화 하여 설계하고 제작 및 측정하였다. 출력 레벨 제어는 Pin Diode 감쇄기를 이용하여 Dynamic Range 40dB 내에서 5단계로 전력제어가 가능하고 입력레벨이 $-4{\sim}0dBm$로 변동될 때 AGC(Auto Gain Control) 회로가 동작 되도록 설계하였다.

인터넷정보자원을 활용한 부가가치정보서비스 (The Value-Added Information Services Using Internet Resources)

  • 인터넷은 도서관정보서비스에 가장 큰 영향을 미치고 있다. 본고는 인터넷의 개요과 인터넷정보자원을 활용하여 부가가치정보서비스를 개발한 사례를 살펴보았다. 전자메일을 통한 신착 정보서비스의 사례로서 미국워싱턴대학의 Zephyr, 독일 베링거맨하임사의 BM-SwetScan, 한국과학기술원의 Jcontents를, 웹기반 통합시스템으로서 미국의 HarperSource, Ei Village, IPL, 그리고 한국전자통신연구원의 ETLARS를 소개하였다. 인터넷정보자원을 탐색하여 평가하고 조직적으로 체계화하는 것이 큰 과제이다.
