Aronia melanocarpa reduced adiposity via enhanced lipolysis in high-fat diet-induced obese mice

  • Obesity is a critical health issue in Korea, where half of all adults are overweight and a third obese. Aronia melanocarpa -rich in flavonoids and phenolics- with antioxidant and anti-inflammatory activities, could have anti-obesity activity and reduce body fat mass by upregulating lipolysis and β-oxidation in obese mice. Male C57BL/6J mice (n=12) were assigned into four groups: normal chow (18% kcal from fat); high-fat diet control (HFD, 45% kcal from fat); HFD+A. melanocarpa (200 mg/kg diet); HFD+Xenical (500 mg/kg diet, positive control). Antioxidant capacity of A. melanocarpa was established in vitro and in vivo. Weight loss was induced as decreased adiposity and lowered respiratory quotient at rest suggested oxidation of stored fat. Adiposity reduction, accompanied with elevated fat utilization, was owing to enhanced activity of hormone-sensitive lipase. Thus, A. melanocarpa lowered adiposity by enhancing lipolysis and utilization of fatty acids in visceral fat.

  • In this study, we demonstrated the whitening effect of Lagenaria leucantha through the melanin biosynthesis of S bikiniensis and inhibition of melanogenesis in cultured Bl6 melanocytes. And we confirmed the whitening effect of Lagenaria leucantha through the depigmentation of gold fish in vivo. The melanogenesis of B$_{16}$ melanocytes was founded to be activated dose and time dependently by the treatment of u- MSH. When the B$_{16}$ melanocytes was treated with 200nM of $\alpha$-MSH, the morphology of melanocytes was remarkably changed. The melanin content and the synthesis of tyrosinase were strikingly increased. Lagenaria ieucantha inhibited the melanin formation stimulated by $\alpha$-MSH without affection of cell viability. However, Lagenaria leucantha didn't inhibit tyrosinase activity and showed weak suppression on the synthesis of tyrosinase. These results suggest Lagenaria leucantha might inhibit melanin formation with tyrosinase independent manner. Lagenaria ieucantha also inhibition melanin biosynthesis with 18mm inhibition zone in S.bikiniensis. To evaluate the inhibitory activity of melanogenesis of Lagenaria leucantha in vivo, we examined its effect on depigmentation of gold fish. Lagenaria ieucantha remarkably reduced the size and density of melanophores in gold fish. These results suggest that Lagenaria ieucantha can be used as a whitening agent in cosmetics.ics.s.

  • The literatural study on the effect of acupuncture for tinnitus was studied from the viewpoint of acupuncture effect. The conclusions are as follows. 1. The ear manages kidney, it relates with the heart meridian of hand soeum, kidney meridian of foot soeum, lung meridian of hand taeeum, spleen meridian of foot taeeum, stomach meridian of foot yangmyeong, gallbladder meridian of foot soyang, triple energizer meridian of hand soyang, small intestine meridian of hand taeyang. 2. According to classification of meridian in acupuncture treatment of tinnitus triple energizer meridian of hand soyang 18.0%, gallbladder meridian of foot soyang 16.6%, bladder meridian of foot taeyang 16.6%, small intestine meridian of hand taeyang 9.7%, large intestine meridian of hand yangmyeong 8.3%, stomach meridian of foot yangmyeong 5.5%, spleen meridian of foot taeeum 4.1%, pericardium meridian of hand gworeum 4.1%, lung meridian of hand taeeum 2.8%, heart meridian of hand soeum 2.8%, kidney meridian of foot soeum 2.8%, liver meridian of foot gworeum 2.8%, conception channel 2.8%, governor channel 2.8% have been used much in turn. 3. In the general points GB2 24times, TE3 22times, TE17 22times, SI19 20times, TE21 20times, KI 3 19times, BL23 17times, LI4 15times have been used much in turn.

Effect of Cigarette Smoke Exposure on MPTP-Induced Neurotoxicity in Mice (흡연이 MPTP에 의해 유발되는 신경독성에 미치는 영향)

  • Effect of cigarette smoke exposure on 1-methyl-4-phpnyl-1,2,3,6-tetrahydro-pyidine (Mm)-induced neurotoxicity was investigated in C57BL6 mice. Cigarette smoke exposure of mice to the mainstream smoke generated from 15 cigarettes for 10 mins per day, 5 days per week, for fi weeks, effectively attenuated the decline both in the level of striatal dopamine and the number of brrosine hydros:ylase-positive ceils in the brain caused by MPTP treahent. Exposure to cigarette smoke significantly decreased monoamine oxidate B activity in the cerebral cortex and cerebellum. The activity of brain antioxidant enzymes such as catalase, glutathione peroxidase, and Cu, Zn-superoxide dismutase, was not changed by cigarette smoke exposure or MPTP treatment. Sulfhydryl compounds content in all brain regions except for the striatum was uniquely increased by MPTP treatment, however, such an effect of MPTP was not observed in mice exposed to cigarette smoke. These results suggest that cigarette smoke exposure inhibits MPTP-induced neurotoxicity without influencing free radical metabolism in the brain of mice. This protective effect of cigarette smoke seems to be closely related with the decreased activity of brain monoamine oxidase H.   • Objective : The purpose of this study is to review the acupuncture therapy and moxibustion on itching Method : We searched and reviewed chinese journals of and books of the acupuncture therapy and moxibustion on itching. Results & Conclusions : 1. Dermatopathy of itching is described as eczema, neurotic-dermititis, itching in orient medical text. 2. According to text, itching causes were wind symptom produced by deficiency of blood, deficiency syndrom of the spleen, and damp-heat. 3. Frequently used acupoints were LI10(41 frequency), SP10(29), ST36(27), SP6(22), GB20(19), Asi point(18), BL(17), back Su acupoints(13), LI4(13), SP9(10), PC6(7), HT7(7). etc 4. The practice of acupuncture was used by body acupuncture, auriculo-theraphy, loss of bleeding with needle, blum-blosson needle, dermal needle, fire needle. etc. 5. Moxibustions are mild moxibustion, non-scar producing moxibustion, herb- moxibustion. etc.

  • Plasmid Achromobacter secretion (PAS) factor is a putative secretion factor that induces the secretion of periplasmic proteins. PAS factor from Vibrio vulnificus was crystallized at 294 K by the hanging drop vapor-diffusion method. It was isolated as a monomer during the purification procedures. The native crystal belongs to the F222 space group with unit cell parameters a = 56.1, b = 74.4, $c=80.0{\AA}$, ${\alpha}={\beta}={\gamma}=90^{\circ}$. The crystal was soaked in cryoprotectant containing 1 M NaBr for 1 h for MAD phasing. The diffraction limit of the Br-MAD data set was $1.9{\AA}$ using synchrotron X-ray irradiation at beam line BL-18B at the Photon Factory, Japan.

Immunomodulatory Effects of Hominis Placenta Extract Injection into an Acupuncture Point on the Experimental Subcutaneous Tumor Model of Mice

  • Hominis placenta (HP) has been used as an agent for promoting physiological function in traditional asian medicine. The present study was peformed to investigate whether HP acupuncture treatment in an experimental tumor mice model inhibit tumor growth through immunomodulatory effects. Mice were inoculated subcutaneously with colon26-L5 cells on the back. Three days after tumor inoculation, HP herbal acupuncture treatment was conducted on BL18 acupoint every other day for three weeks. HP Herbal acupuncture treatment significantly suppressed the primary tumor growth and prolonged survival rate. To evaluate immunomodulatory effect of HP acupuncture, splenocytes proliferation assay, fluorescence-activated cell sorting (FACS) and ELISA for IFN- ${\gamma}$, and IL-4 cytokine level. HP herbal acupuncture enhanced the mitogenic activity of Balb/c whole splenocytes induced by various mitogenic stimuli and increased immune cell population such as T cell, B cell, Th cell, Tc cell and Macrophages. HP herbal acupuncture caused a marked increase of production of Th1 cytokine (IFN- ${\gamma}$ ,) and decrease of production of Th2 cytokine (IL-4). These results indicated that HP herbal acupuncture suppresses tumor growth through a mechanism leading to a Th1 dominant immune state.

Changes in Contents and Composition of Insoluble Dietary Fiber during Buckwheat Germination (메밀 발아 중 불용성 식이섬유 함량과 조성의 변화)

  • To provide the basic information of buckwheat insoluble dietary fiber (IDF) and efficient material for the buckwheat processed foodstuffs, buckwheat (Fagopyrum egculentum Moench) was germinated at 1$0^{\circ}C$ for 7 days and the contents and composition of the insoluble dietary fiber were examined at 24 hour intervals. The NDF and ADF contents in ungerminated seeds were 20.27%, 18.95% on dry weight basis, respectively. During the germination period, the NDF and ADF contents increased gradually. The cellulose, hemicellulose and lignin contents in ungerminated seeds were 11.10%, 1.32% and 7.85%, respectively. During the germination period, the cellulose and lignin contents increased. However, there was no significant change in the hemicellulose contents. The composition of cellulose and hemicellulose in the NDF decreased with germination time, whereas that of lignin increased. The IDF contents obtained by Prosky method were higher than the NDF contents obtained by Van Soest method. However, the IDF and NDF contents were closely correlated(r=0.9785, p<0.01). The cellulose(Xl), hemicellulose(X2), lignin(X3) and soluble dietary fiber (SDF) (BL) showed the significant regression equation(p<0.001) with the root length(Y). The multiple regression equation was Y = -12.1306 + 0.9213xl - 0.1615$\times$2 + 0.1071$\times$3 + 0.7397$\times$4 and R2(coefficient of determination) was 0.942.

A Review of the Domestic Study Trends on Climacteric Syndrome Treated with Pharmacopuncture (갱년기 증후군의 약침 치료에 대한 국내 연구 동향 고찰)

  • Objectives: The purpose of this review is to analyze the domestic study trends on climacteric syndrome treated with pharmacopuncture. Methods: We investigated the studies on pharmacopuncture for climacteric syndrome via searching 6 online databases. Results: 18 studies were selected. There were 2 case reports, 1 randomised controlled trial, and 15 animal experiments. There were 14 studies about osteoporosis, 1 study about hot flush and sweating, 1 study about abdominal obesity, 1 study about neck pain, and 1 study about learning disability and memory disorder. Nokyong was most frequently used for pharmacopuncture, followed by Honghwa. 陰谷 (KI10) was most frequently used for acupuncture point, followed by 腎兪 (BL23). Conclusions: This study shows that pharmacopuncture could be one of the effective treatments for climacteric syndrome. More well-designed clinical studies using pharmacopuncture for climacteric syndrome will be needed.

Cloning and Characterization of a Methionine Aminopeptidase (MAP) Gene from Tetragenococcus halophilus CY54 Isolated from Myeolchi-Jeotgal

  • A map gene encoding methionyl-specific aminopeptidase (MAP; EC 3.4.11.18) was cloned from Tetragenococcus halophilus CY54. Translated amino acid sequence of CY54 MAP showed high similarities with those from Enterococcus faecalis (83.8%) and Streptococcus salivarius (62.2%) but low similarities with MAPs from Lactobacillus and Lactococcus genera. The map gene was overexpressed in E. coli BL21(DE3) using pET26b(+),pET26b(+), and the recombinant MAP was purified by using an Ni-NTA column. The size of recombinant MAP was 29 kDa as determined by SDS-PAGE. The optimum pH and temperature of CY54 MAP were pH 5.0 and 60℃, respectively. The activity of CY54 MAP was most significantly increased by Co2+ ion (159%), and showed the highest activity at 12% NaCl. Km and Vmax were 0.64 ± 0.006 mM and 10.12 ± 0.014 U/mg protein, respectively when met-pNA was used as the substrate. This is the first report on a MAP from Tetragenococcus species.
