Review on Needling or Moxibustion-prohibited Points in "Emergency Formulas Worth a Thousand in Gold" and "Supplement to the Formulas Worth a Thousand in Gold" ("비급천금요방(備急千金要方)"과 "천금익방(千金翼方)"의 침구금기혈(鍼灸禁忌穴) 연구)

  • Objectives : To classify needling or moxibustion-prohibited acupoints in Emergency Formulas Worth a Thousand in Gold (EFWTG) and Supplement to the Formulas Worth a Thousand in Gold (SFWTG). Methods : We found needling or moxibustion-prohibited acupoints in EFWTG and SFWTG, then investigated the influences of needling or moxibustion-prohibited acupoints on A-B Classic of Acupuncture and Moxibustion. Results : In EFWTG, the needling-prohibited points were LI13, ST17, BL56, TE8, CV8, CV15 and GV24. Acupoints needed careful needling were LU2, ST12, KI2, KI7, TE19, GB3 and jwagak. The moxibustion-prohibited points were LU3, LU8, ST1, ST8, ST9, ST17, ST32, ST33, BL6, BL30, TE18, TE23, GB33, GB42, CV5, CV15, GV6, GV15, GV16 and GV17. Acupoints needed careful moxibustion were ST7, ST30, TE21 and GB22. In SFWTG, the needling-prohibited points were LU2, LI13, ST12, ST17, ST32, BL56, KI2, KI7, TE8, TE19, GB3, CV8, CV15, GV24 and jwagak. The moxibustion-prohibited points were LU3, LU8, ST1, ST7, ST8, ST9, ST17, ST30, ST32, ST33, BL6, BL30, TE18, TE23, GB22, GB33, GB42, CV5, CV15, GV6, GV15, GV16, GV17 and ijung. Conclusions : There were 7 needling-prohibited points, 7 acupoints needed careful needling, 20 moxibustion-prohibited points, and 4 acupoints needed careful needling in EFWTG, and 15 needling-prohibited points and 24 moxibustion-prohibited points in SFWTG. The needling or moxibustion-prohibited acupoints in A-B Classic of Acupuncture and Moxibustion had a strong influence on those in the two literatures.

Effect of Allium cepa (red) and Angelica gigas Nakai on Hair-growth Promotion in C57BL/6 Mice (C57BL/6 마우스에서 모발성장 촉진에 대한 Allium cepa (red)와 Angelica gigas Nakai의 효과)

  • This study was conducted to evaluate the hair-growth promotion effect in C57BL/6 mice of a new substance mixed with Allium cepa (red)-bioconversion extract and Angelica gigas Nakai. The ethanol extract of Allium cepa (red) was bioconverted through the use of the Bacillus subtilis KJ-3 (BS3) strain, which was named Red-BCQ. The quercetin content of Red-BCQ increased by about 7.4-fold after bioconversion. Angelica gigas Nakai extract (Agnex) contains a large amount of coumarins such as decursin (D) and decursinol angelate (DA). A 1 mg portion of Agnex contained 0.4146 mg of D and 0.3659 mg of DA. Minoxidil has been known to promote hair growth. In this study, the hair-growth promotion effects of Red-BCQ, Agnex, and a mixture of both Red-BCQ and Agnex were compared with 5% minoxidil. Twenty-five mice were divided into five experimental groups including saline (CON), 5% minoxidil (PCON), Red-BCQ (RA), Agnex (AG), and a Red-BCQ-Agnex mixture (RAG)-treated group. Samples were administered orally once a day at a fixed time for 4 weeks. Hair growth was monitored by photograph at 7, 14, 21, and 28 days. We also observed 5α-reductase, alkaline phosphatase (ALP), γ-glutamyl transpeptidase (γ-GT), insulin-like growth factor (IGF)-1, transforming growth factor (TGF)-β1, antioxidant enzyme, and the hair follicles of the skin tissue. In all the results, the RAG-administered group showed greater antioxidative and hair-growth promotion effects than the other groups. These data suggest that RAG has potent stimulating activity on hair growth in C57BL/6 mice.

Influence of the Adjuvants and Genetic Background on the Asthma Model Using Recombinant Der f 2 in Mice

  • Der f 2 is the group 2 major allergen of a house dust mite (Dermatophagoides farinae) and its function has been recently suggested. To determine the optimal condition of sensitization to recombinant Der f 2 (rDer f 2) in murine model of asthma, we compared the effectiveness with different adjuvants in BALB/c and C57BL/6 mice. Mice from both strains sensitized with rDer f 2 by intraperitoneal injection or subcutaneous injection on days 1 and 14. The dosage was $20{\mu}g$. Freund's adjuvants with pertussis toxin (FP) or alum alone were used as adjuvants. On days 28, 29, and 30, mice were challenged intranasally with 0.1% rDer f 2. We evaluated airway hyperresponsivenss, eosinophil proportion in lung lavage, airway inflammation, and serum allergen specific antibody responses. Naive mice were used as controls. Airway hyperresponsiveness was increased in C57BL/6 with FP, and BALB/c with alum (PC200: $13.5{\pm}6.3$, $13.2{\pm}6.7$ vs. >50 mg/ml, p<0.05). The eosinophil proportion was increased in all groups; C57BL/6 with FP, BALB/c with FP, C57BL/6 with alum, BALB/c with alum ($24.8{\pm}3.6$, $20.3{\pm}10.3$, $11.0{\pm}6.9$, $5.7{\pm}2.8$, vs. $0.0{\pm}0.0$%, p<0.05). The serum allergen specific IgE levels were increased in C57BL/6 with FP or alum (OD: $0.8{\pm}1.4$, $1.1{\pm}0.8$, vs. $0.0{\pm}0.0$). C57BL/6 mice were better responders to rDer f 2 and as for adjuvants, Freund's adjuvant with pertussis toxin was better.

  • In order to maximize the growth of Bifidobacterium lactis BL 740 and soy isoflavone agycones production, we investigated the optimization of a culture medium containing soy hypocotyls, which are the byproducts of the soy manufacturing process, and soy proteins. The ingredients of the medium containing soy materials (S-medium) were selected by fractional factorial design (FFD) and central composite design (CCD) within a desirable range. The FFD was applied by six factors: glucose, cellobiose, fructooligosaccharide, soy peptone, soy protein, and soy hypocotyl. Soy protein, soy peptone, and soy hypocotyl were found to be significant factors from the result of FFD for both the growth of B. lactis BL 740 and aglycone production. The CCD was then applied with three variables found from FFD at five levels each and the optimum values were determined for the three variables: soy peptone, soy protein, and soy hypocotyl. In the case of the growth of B. lactics BL740, the proposed optimal media contained 12.73 g/L of soy protein, 29.55 g/L of soy peptone, and 130.67 g/L of soy hypocotyl. To produce isoflavone aglycones, optimized media was composed of 2.06 g/L, soy protein, 1.25 g/L of soy peptone, and 60.02 g/L of soy hypocotyl.

  • The study was conducted to find out the gene effects on body weights at weaning and at the age of 60 days in mice, with 343 progenies from full-dialell crosses of four lines of BALB/c, CBA, C3H and C57BL. The progenies were reared at the Experimental Animal Farm, College of Agriculture, Kyungpook National University from November, 1984 to February, 1985, and data collected from the progenies were analyzed into general combining ability, maternal effects, specific combining ability and reciprocal effects with Harvey's model. General combining ability effects estimated in line-crosses were -0.6033~0.5298 for weaning weights and -0.5086~1.0012 for body weights at the age of 60 days. General combining ability for BALB/c and C57BL were significantly better than general combining ability for CBA and C3H for both traits (P<0.05). Maternal effects for C3H were significantly larger than the maternal effects of BALB/c for both traits (P<0.05). The estimates of maternal effects were -0.9678~0.4609 for weaning weights and -1.1886~0.0729 for body weights at the age of 60 days. Specific combining ability effects were estimated to be significant (P<0.05), and the estimates were -0.1999~0.3380 for weaning weights and -0.4056~0.3317 for body weights at the age of 60 days. Reciprocal effects were found to be largest in BALB/c${\times}$C57BL and BALB/c${\times}$C3H. The estimates were -0.5049 from BALB/c${\times}$C57BL and 0.4972 from BALB/c${\times}$C3H form weaning weights, and -1.0336 from BALB/c${\times}$C57BL and 1.2793 from BALB/c${\times}$C3H for body weights at the age of 60 days.

Antidepressant-like Effects of the Gastrodia elata Bl Extract in Mice

  • Objectives : A growing body of evidence has suggested that the dysfunction of glutamatergic systems plays a pivotal role in major depressive disorder (MDD). This study was performed to investigate the antidepressant-like effects of the ethanolic extract of Gastrodia elata Bl (GE) in mouse models and to investigate the role of ${\alpha}$-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid (AMPA) receptors in producing these antidepressant-like effects. Methods : The forced swim test (FST) and tail suspension test (TST) were used to investigate GE's behavioral effects in mice. Additional biochemical and behavioral experiments with NBQX, an AMPA receptor antagonist, were undertaken to determine whether the antidepressant-like properties of GE are involved in AMPA receptor throughput. Results : Oral administration of GE extract (1,600 mg/kg) 1h prior to testing significantly reduced the immobility times in the FST and TST. These antidepressant-like effects of GE extract were increased dose-dependently. Pre-treatment with NBQX significantly attenuated the reduction in immobility time induced by the GE extract in the FST and TST. Conclusions : The ethanolic extract of GE may exert antidepressant-like effects with involvement of AMPA receptor.

  • An agarase gene from Bacillus cereus ASK202 was expressed in high levels by E. coli BL21(DE3)/pEBA1 using pET28a(+) vector system with the inducible T7 promoter in the presence of isopropyl- ${\beta}$ -thiogalactopyranoside. The open reading frame encodes 761 amino acid residues with a calculated molecular weight of 83,300 daltons and a potential signal peptide about 36 amino acid residues at the N-terminus. E. coli BL21(DE3)/pEBA1 produce 1280 unit/ ${\ell}$ of agarase. The optimum physical condition for the agarase activity was pH 5.6, and $40^{\circ}C$, respectively. The agarase activity was stable up pH $4.0{\sim}9.0$ and $4{\sim}40^{\circ}C$. The km and maximum rate of metabolism for agar were 0.068mg/$m{\ell}$ and 0.094mg/$m{\ell}{\cdot}min$, respectively.

  • For antibody development of human serine palmitoyltransferase (SPT, EC 2.3.1.50), SPTLC1 and SPTLC2 genes were subcloned in pRset vector and expressed in E. coli BL21 (DE3)pLys cells. Eucaryotic SPT is a membrane-bound heterodimer enzyme, while all other members are soluble homodimer enzymes. cDNA library were obtained from total RNA from human embryo kidney cell line, HEK293, using RT-PCR and PCR with specific primers was carried out for preparing SPTLC1 and SPTLC2 genes. pRset vector which can express hexahistidine-tag fusion protein was used and the DNA sequences of pRsetB/SPTLC1 and pRsetA/SPTLC2 were confirmed. Recombinant BL21 cells with SPTLC subunits were selected with LB plate containing ampicillin and chroramphenicol. SPTLC1 and SPTLC2 proteins were induced with 1 mM IPTG and seperated on 10% SDS-PAGE gel. Expressed proteins were confirmed by western blotting with His-tag antibody.

Immune response improvement induced by Herbal-acupuncture with Aucklandiae Radix infusion solution into Zusanli(ST36) (족삼리(足三里)에 시술(施術)한 목향(木香) 약침(藥鍼)이 항암(抗癌) 및 면역효과(免疫效果)에 미치는 영향(影響))

  • To study the effects of anti-cancer, anti-metastasis and immune response improvement of herbal-acupuncture with Aucklandiae Radix diffusae herba infusion solution(AKL-HAS), we injected AKL-HAS into Zusanli(St36) of C57BL/6 mice implanted intravenously with B16-F10 melanoma. The results were obtained as follows : 1. In the experiment groups treated with Aucklandiae Radix(AKL) Herbal-Acupuncture, the spleen cell proliferation in Balb/c mouse was significantly increased compared with that of the control group. 2. In the experiment groups treated with Aucklandiae Radix(AKL) Herbal-Acupuncture, the percentage of $CD25^{+}/CD4^{+},\;CD8^{+}/CD3e^{+},\;CD69^{+}/B220^{+},\;NK1.1^{+}/CD3e^{+}$ cells in C57BL/6 mouse PBMCs was increased compared with that of the control group. 3. In the experiment groups treated with Aucklandiae Radix(AKL) Herbal-Acupuncture, the pulmonary colony number of C57BL/6 mice implanted intravenously with B16-F10 melanoma was decreased significantly compared with that of the control group. 4. In the experiment groups treated with Aucklandiae Radix(AKL) Herbal-Acupuncture, MST(Median Survial Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma were increased significantly compared with those of the control group.

  • To evaluate the antitumor activity and antimetastatic effects of HDBT, studies were done experimentally. The results were obtained as follows: 1. HDBT extracts didn't show cytotoxicity against BALB/C mouse lung fibroblast cell. 2. In cytotoxicity against A549, SK-OV-3, B16-BL6 and HT1080 concen- tration inhibiting cell growth up to below 30% of control was recognized at $10^{-3}g/ml$ of HDBT. 3. The concentration inhibiting adhesion of A549 and B16-BL6 to complex extracellular matrix up to below 30% of control was recognized at $10^{-3}g/ml$ of HDBT. 4. In Inhibitory effect on activity of DNA topoisomerase I, the $IC_{50}$ was shown $200-300{\mu}g/m{\ell}$ of HDBT. 5. The T/C% was 137.9% in HDBT-treated group in S-180 bearing ICR mice. 6. In CAM assay, HDBT extracts inhibited angiogenesis significantly at $15{\mu}g/egg$ concentration as compared with control. 7. In pumonary colonization assay, a number of colonies in the lungs were decreased but insignificantly in HDBT-treated group as compared with control group. 8. In hematological changes in B16-BL6 injected C57BL/6, numbers of WBC were decreased significantly in HDBT-treated group but numbers PLT were increased insignificantly as compared with control. From above results it was concluded that HDBT could be usefully applied for the prevention and treatment of cancer.
