  • The effect of bio-logger tagging location on blood characteristics and bio-logger attachment efficiency in spotted sea bass (mean body weight 2356.7 g) was investigated. The fish were tagged at four different tagging locations: no-tag (control), operculum attachment (OA), dorsal muscle attachment (DA), and cauda peduncle muscle attachment (CA). The blood properties and bio-logger attachment efficiencies were examined on days 1, 7, 14, and 35 after tagging the bio-logger at each tagging location. During the experimental periods, the concentrations of hematocrit and hemoglobin in whole blood, and GOT (glutamic oxaloacetic transaminase), GPT (glutamic pyruvic transaminase), total protein (TP), glucose, total cholesterol, cortisol, and superoxide dismutase in plasma were not affected by the attachment location of the bio-logger, however, the TP concentration was significantly lower in OA than in the control group on day 7. After tagging for 35 days, the efficiencies of bio-logger attachment in the OA, DA, and CA after tagging for 35 days were 33.3%, 100.0%, and 33.3%, respectively. These results indicate that, in our experimental condition, the most appropriate bio-logger attachment location is DA, providing basic information on bio-logger utilization methods for ecological and biological biotelemetry surveys of the spotted sea bass.

  • This study aimed to determine the effect of the pop-up satellite archival tag (PSAT) tagging method on the blood indices and PSAT attachment efficiency of yellowtail Seriola quinqueradiata (mean body weight 10.2 kg). Based on tagging method, the fishes were divided in four different groups: untagged (control), single anchor (SA), dual anchor (DA), and silicon tube (ST). The blood indices and PSAT attachment efficiency were investigated on days 1, 14, and 28 after tagging PSAT on the muscle below the dorsal fin for each tagging method in triplicates. After 28 days of tagging with PSAT, a significant increase was observed in plasma glucose level in the ST group and in total protein level in the DA and ST groups. The levels of glucose, total protein, and total cholesterol in the SA group after 28 days of tagging were significantly lower than in the control group. The efficiencies of PSAT attachment were 0% in the SA and DA groups on 14 days post-tagging, and 66.7% in the ST group on 28 days post-tagging. The study results indicate that the proper PSAT tagging method is the ST type. The information obtained in this study presents valuable data that provide the required PSAT operational tool for industrial development and ecological monitoring of yellowtail.

Preparation of Metal-p-aminobenzyl-DOTA Complex Using Magnetic Particles for Bio-tagging in Laser Ablation ICP-MS

  • Metal-p-$NH_2$-Bn-DOTA (paraammionobenzyl-1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid: ABDOTA) complex was synthesized and purified for bio-tagging to quantify biological target materials using laser ablation (LA)-ICP-MS. Since the preparation of a pure and stable tagging complex is the key procedure for quantification, magnetic particles were used to purify the synthesized metal-ABDOTA complex. The magnetic particles immobilized with the complex attracted to a permanent magnet, resulting in fast separation from free un-reacted metal ions in solution. Gd ions formed the metal-complex with a higher yield of 64.3% (${\pm}3.9%$ relative standard deviation (RSD)) than Y ions, 52.3% (${\pm}2.5%$ RSD), in the pH range 4-7. The complex bound to the magnetic particles was released by treatment with a strong base, of which the recovery was 81.7%. As a reference, a solid phase extraction (SPE) column packed with Chelex-100 resin was employed for separation under similar conditions and produced comparable results. The tagging technique complemented polydimethylsiloxane (PDMS) microarray chip sampling in LA-ICP-MS, allowing determination of small sample volumes at high throughputs. For application, immunoglobulin G (IgG) was immobilized on the pillars of PDMS microarray chips and then tagged with the prepared Gd complex. IgG could then be determined through measurement of Gd by LA-ICP-MS. A detection limit of 1.61 ng/mL (${\pm}0.75%$ RSD) for Gd was obtained.

Analytical Techniques Using ICP-MS for Clinical and Biological Analysis

  • This article reviews recent analytical techniques using inductively coupled plasma-mass spectrometry (ICP-MS) immunoassay for clinical and bio analysis. We classified the techniques into two categories, direct and indirect analysis, which depend upon a guideline of whether tagging materials are used or not. Direct analysis is well known, and generally used in conjunction with various other techniques, such as laser ablation, chromatographic separations, etc. Recently, indirect analysis using tagging elements has intensively been discussed because of its importance in future applications to bio and clinical analysis, including environmental and food industries. The method has shown advantages of multiplex detection, excellent sensitivity, and short analysis time owing to signal amplification and magnetic separation. Now, it expands the application field from small biomolecules to large cells.

한국어 생의학 개체명 인식 성능 비교와 오류 분석 (Performance Comparison and Error Analysis of Korean Bio-medical Named Entity Recognition)

  • 딥러닝 분야에서 트랜스포머 아키텍쳐의 출현은 자연어 처리 연구가 획기적인 발전을 가져왔다. 개체명 인식은 자연어 처리의 한 분야로 정보 검색과 같은 태스크에 중요한 연구 분야이다. 생의학 분야에서도 그 중요성이 강조되나 학습용 한국어 생의학 말뭉치의 부족으로 AI를 활용한 한국어 임상 연구 발전에 제약이 되고 있다. 본 연구에서는 한국어 생의학 개체명 인식을 위해 새로운 생의학 말뭉치를 구축하고 대용량 한국어 말뭉치로 사전 학습된 언어 모델들을 선정하여 전이 학습시켰다. F1-score로 선정된 언어 모델의 개체명 인식 성능과 태그별 인식률을 비교하고 오류 분석을 하였다. 인식 성능에서는 KlueRoBERTa가 상대적인 좋은 성능을 보였다. 태깅 과정의 오류 분석 결과 Disease의 인식 성능은 우수하나 상대적으로 Body와 Treatment는 낮았다. 이는 문맥에 기반하여 제대로 개체명을 분류하지 못하는 과분할과 미분할로 인한 것으로, 잘못된 태깅들을 보완하기 위해서는 보다 정밀한 형태소 분석기와 풍부한 어휘사전 구축이 선행되어야 할 것이다.

넙치(Paralichthys olivaceus) 산소 소비율에 미치는 Pop-up Satellite Archival Tag (PSAT) 체외 부착과 수온의 영향 (External Attachment of Pop-up Satellite Archival Tag (PSAT) and Water Temperature Affect Oxygen Consumption Rate of the Olive Flounder Paralichthys olivaceus)

  • This study aimed to examine the effect of external pop-up satellite archival tags (PSATs) attachment and water temperature on the oxygen consumption rate (OCR) of the olive flounder (mean body weight 2281.7 g). The OCRs of fish were measured under conditions of three different water temperature conditions (15, 20, and 25℃) and two different tagging methods [non-tagging, control; bio-logger external attachment with a miniature PSAT (dummy mrPAT), BEA] using a closed flow-through respirometer. The OCRs of fish linearly increased with the increase in water temperature in both the control and BEA (P<0.001); however, the OCRs of BEA were approximately 1.8-1.9 times lower than those of the control at each water temperature (P<0.001). The Q10 values of the control and BEA were the highest in the water temperature range of 15 to 20℃, but sensitivity to water temperature changes was higher in BEA than in the control. The metabolic energy loss rate (MEL) of fish increased with increasing water temperature regardless of external tagging, but the MEL of the control was higher than that of BEA (P<0.001). These results demonstrate that OCR, thermal sensitivity, and energy expenditure are all affected in adult olive flounder with external PSAT attachment.

A Small Epitope Tagging on the C-Terminus of a Target Protein Requires Extra Amino Acids to Enhance the Immune Responses of the Corresponding Antibody

  • Protein-specific antibodies are essential for various aspects of protein research, including detection, purification, and characterization. When specific antibodies are unavailable, protein tagging is a useful alternative. Small epitope tags, typically less than 10 amino acids, are widely used in protein research due to the simple modification through PCR and reduced impact on the target protein's function compared to larger tags. The 2B8 epitope tag (RDPLPFFPP), reported by us in a previous study, has high specificity and sensitivity to the corresponding antibody. However, when attached to the C-terminus of the target protein in immunoprecipitation experiments, we observed a decrease in detection signal with reduced immunity and low protein recovery. This phenomenon was not unique to 2B8 and was also observed with the commercially available Myc tag. Our study revealed that C-terminal tagging of small epitope tags requires the addition of more than one extra amino acid to enhance (restore) antibody immunities. Moreover, among the amino acids we tested, serine was the best for the 2B8 tag. Our findings demonstrated that the interaction between a small epitope and a corresponding paratope of an antibody requires an extra amino acid at the C-terminus of the epitope. This result is important for researchers planning studies on target proteins using small epitope tags.

Response of Saccharomyces cerevisiae to Ethanol Stress Involves Actions of Protein Asr1p

  • During the fermentation process of Saccharomyces cerevisiae, yeast cells must rapidly respond to a wide variety of external stresses in order to survive the constantly changing environment, including ethanol stress. The accumulation of ethanol can severely inhibit cell growth activity and productivity. Thus, the response to changing ethanol concentrations is one of the most important stress reactions in S. cerevisiae and worthy of thorough investigation. Therefore, this study examined the relationship between ethanol tolerance in S. cerevisiae and a unique protein called alcohol sensitive RING/PHD finger 1 protein (Asr1p). A real-time PCR showed that upon exposure to 8% ethanol, the expression of Asr1 was continuously enhanced, reaching a peak 2 h after stimulation. This result was confirmed by monitoring the fluorescence levels using a strain with a green fluorescent protein tagged to the C-terminal of Asr1p. The fluorescent microscopy also revealed a change in the subcellular localization before and after stimulation. Furthermore, the disruption of the Asr1 gene resulted in hypersensitivity on the medium containing ethanol, when compared with the wild-type strain. Thus, when taken together, the present results suggest that Asr1 is involved in the response to ethanol stress in the yeast S. cerevisiae.

모바일 NFC기반의 순번대기 시스템의 설계 (An Implementation of Waiting Queue System based on Mobile NFC)

  • 스마트 디바이스의 보급이 확대되면서 스마트폰은 우리 생활 깊숙이 파고들어 많은 생활방식을 변화시키고 있다. 또한, 스마트폰의 보급률이 높아지면서 스마트폰의 고급화, 고성능화가 진행중이며 이에맞추어 사회전반적인 스마트폰의 활용영역이 확대되고 있다. 최근에는 RFID를 기반으로 하는 근거리 무선통신 기술인 NFC를 탑재한 스마트폰의 보급이 증가하여 전자명함, 태깅 등 여러 영역에서 그 활용도를 높이는 추세다. 본 논문에서는 이를 활용하여 전통적인 번호표 방식의 순번대기 시스템에 NFC를 적용하여 접촉만으로 인체에 해가될 수 있는 전사용지사용의 지양, 스마트폰을 활용한 원거리 예약 등 보다 효율적인 순번대기 시스템을 운영할 수 있는 시스템을 제안한다.
