Antioxidant Activities and Whitening Effects of Extracts from Hippophae rhamnoides L. (비타민 나무(Hippophae rhamnoides L.) 열매 추출물의 항산화 활성 및 미백 효과)

  • This study was performed to investigate the antioxidant activities and the melanin inhibitory effects of Hippophae rhamnoides L. fruit extracts. Two in vitro methods were used; the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method to determine antioxidant activity and measurements of the inhibitory effects of tyrosinase activity to determine melanogenesis in B16/F10 melanoma cells. The radical scavenging activity of the extract was 56.0% at $700{\mu}g/mL$, similar to ascorbic acid (56.9%), in the DPPH assay. The tyrosinase inhibitory activity of the extract was 52.1% and 73.4% at 100 and $500{\mu}g/mL$, which is also similar to ascorbic acid. In B16/F10 mouse melanoma cells, the extract inhibited melanin synthesis by 56% at $500{\mu}g/mL$, a more prominent inhibition of melanin synthesis compared to extracts from arbutin. These results suggest that extracts from H. rhamnoides L. have antioxidant activity and skin-whitening effects; allowing their application in cosmetics as a natural product.

Skin Whitening and Anti-Wrinkle Effects of Extract from Jubak of Oriental Herbal Liquor (한방 발효주 주박 추출물의 미백 및 피부 주름 개선 효과)

  • Oriental herbal liquor (Yakju) is a type of Korean traditional alcoholic beverage that uses Nuruk and oriental herbs for fermentation. The purpose of this study was to develop cosmetic ingredients using Jubak, which is a by-product of alcoholic fermentation of oriental herbal liquor. To investigate antioxidant, whitening, and anti-aging effects of Jubak, we prepared extract of Jubak and its solvent fractions. Ethyl acetate fraction (KSD E4-3) showed the most prominent free radical [1,1-diphenyl-2-picrylhydrazyl (DPPH)] scavenging activity ($SC_{50}$: 0.75 mg/mL). KSD E4-3 significantly inhibited in vitro mushroom tyrosinase activity ($IC_{50}$: 0.82 mg/mL) and reduced the melanin contents in mouse melanoma melanocyte, B16F10 cells. KSD E4-3 down-regulated protein expression of tyrosinase related proteins (TRP)-1, -2, which play key roles in melanogenesis. For anti-aging effects, inhibition of matrix metalloproteinase (MMPs) expression was evaluated using human keratinocyte, HaCaT cells. Treatment of HaCaT cells with KSD E4-3 reduced expression of MMP-1, -2, -9 and inhibited proteolytic activities of MMP-2, -9. These results suggest that KSD E4-3 induces down-regulation of cellular melanogenesis and protects against photoaging induced by UVB-induced damage. Thus KSD E4-3 could potentially be a valuable cosmetic ingredient.

A Study of the Antioxidant Activities and Whitening Activities of Areca semen Extracts as Cosmetic Ingredient (화장품 소재로서 빈랑자의 항산화 및 미백활성에 관한 효과)

  • Herbal plant extracts are good resources to find functional compounds for cosmetic ingredient. In this study, the extract of Areca semen (A. semen) was studied for melanogenesis inhibition and antioxidant activity. The results showed that ethyl acetate fraction of A. semen contained phenolic contents, $301.35{\pm}0.88{\mu}g/mg$, and exhibited potent antioxidant activity with $IC_{50}$ value of $1.02{\pm}0.07{\mu}g/mg$. Further, FRAP value exhibited potent antioxidant activity with $9.07{\pm}0.36mM$. Disk diffusion assay was performed for antibacterial activity. Ethyl acetate fraction of A. semen showed antibacterial activity against Staphylococcus aureus (S. aureus) and Staphylococcus epidermidis (S. epidermidis) at $80{\mu}g/mL$, whereas it showed no significant antibacterial activity against Escherichia coli (E. coli). The results of cell viability indicated that ethyl acetate fraction did not show cytotoxicity to B16/F1 cells at $80{\mu}g/mL$ and showed significant cytotoxicity at $100{\mu}g/mL$ of concentration and showed inhibition of melanin synthesis inhibitory, $29.78{\pm}0.31%$ at $80{\mu}g/mL$. Furthermore, mRNA expressions of tyrosinase and MITF were decreased after treatment with ethyl acetate fraction in a dose-dependent manner. As a result, the ethyl acetate fraction of A. semen could be considered as potential as whitening agents.

Effect of Various Pear Cultivars at Different Fruit Development Stages on Antioxidant and Whitening Activities (배 품종별 생육 단계에 따른 산화방지 및 미백 효과)

  • This study was carried out to investigate the total polyphenols, antioxidant activities, and melanin synthesis inhibition of several pear cultivars (Pyruspyrifolia). The total polypenolic content of five pear cultivars was high in the unripe developmental stage. Total polyphenolic content of the Chuwhangbae cultivar extract was higher than that of other pear cultivars regardless of the developmental stage. However, the total flavonoid content did not differ between cultivars or developmental stages. The phenolic compound, arbutin has an inhibitory effect on melanogenesis. Arbutin levels in pear cultivars declined as the fruit matured. The free radical scavenging activity of the extract also decreased as the fruit ripened. In B16F10 mouse melanoma cells, most of the cultivar extracts inhibited melanin synthesis by about 50% at a $100{\mu}g{\cdot}mL^{-1}$ concentration, except in the Gamcheonbae extract until 90 days after full bloom. We have confirmed that the extract of pear cultivars have antioxidant activity and skin-whitening effects.

Anti-Oxidant, Anti-Melanogenic, and Anti-Inflammatory Activities of Zanthoxylum schinifolium Extract and its Solvent Fractions (산초 추출물 및 분획물의 항산화, 미백 및 항염증 활성)

  • This study was designed to explore new nutraceutical and cosmetic resources possessing biological activities from the plant kingdom. To fulfill this purpose, we analyzed the anti-oxidative, anti-melanogenic, and anti-inflammatory activities of Zanthoxylum schinifolium extract (ZSE) and its solvent fractions using in vitro assays and cell culture model systems. Three kinds of ZSE treated with methanol, ethanol, and water exhibited potent anti-oxidative activities through DPPH radical scavenging capacity, and inhibited in vitro DOPA oxidation. Furthermore, Z. schinifolium methanol extract (ZSME) inhibited the ${\alpha}$-melanocyte stimulating hormone, which induces melanin contents in B16F10 cells. Its anti-melanogenic activity originates from the inhibition of tyrosinase enzyme activity and melanogenesis related protein expression. Moreover, lipopolysaccharide induced nitric oxide production in the RAW 264.7 cell line was also ameliorated by ZSME treatment in a dose dependent manner. Among the four solvent fractions of ZSME treated with dichloromethane, ethyl acetate, n-butanol, and water, three fractions, except water, showed significant anti-melanogenic and anti-inflammatory activities. Taken together, these results provide important new insights into Z. schinifolium, indicating that it possesses numerous biological activities such as anti-oxidative, anti-melanogenic, and anti-inflammatory activities. Therefore, it may well serve as a promising material in the field of nutraceuticals and cosmetics.
