단백질분해효소를 생산하지 않는 균주 B. subtilis MT-2의 염색체 DNA를 추출한 다음, B. subtilis AC819 균주에 상동성 유전자재조합을 이용하여 competent cell 형질전환을 시켰다. 얻어진 형질전환체를 B. subtilis HL-1이라고 명명하였으며, 그 표현형은 histidine 요구성, streptomycin 내성, tetracyclin 내성을 나타내면서 단백질 분해효소를 생산하지 않았다. 플라스미드 pUB110 을 이용한 B. subtilis HL-1의 protoplst 형질전환율은 B. subtilis MT-2의 형질전환율보다 높았다. 따라서 새로운 B. subtilis HL-1균주는 단백질분해효소의 형질전환과 내열성 protease 유전자클로닝에서 숙주로 사용하는데 유용하다.
Two species of antimicrobial agent producing bacteria and one sensitive strain were isolated from soil. Those were identified as B. subtilis, B. licheniformis, and Curtobacterium sp. by morphological, biochemical, physiological and chemotaxonomic characteristics. These were designated as B. subtilis cx1, B. licheniformis cy2 and Curtobacterium sp. cf3, respectively. Antimicrobial agent produced by B. subtilis cx1 showed high antibacterial activity against gram positive bacteria including of B. subtilis, Curtobacterium sp., L. mesenteroids, Staphy. aureus, S. faecalis and even gram negative bacteria, P. aeruginosa. Antimicrobial agent from B. licheniformis cy2 showed slightly lower antimi crobial activity than that from B. subtilis cx1. These two strains showed maximum production of antimicrobial agents at 30oC for 9~21hr cultivation. Curtobacterium sp. cf3 showed more sensitive activity than a sensitive strain of B. subtilis ATCC 6633 which was same genus or species with the B. subtilis cx1 and B. subtilis cy2, when the antimicrobial agent producing strains, B. subtilis cx1 and B. subtilis cy2, were directly applied onto these sensitive lawns.
오이 지상부에 발생하는 주요 곰팡이병인 노균병, 흰가루병 및 잿빛곰팡이병에 대한 생물적 방제를 위하여 오이식물제로부터 Bacillus subtilis B29, B. subtilis M10 and Streptomyces sp. CC19균주를 분리하였다. 오이 노균병에 대한 유묘검정에서 B. subtilis B29, B. subtilis M10과 Streptomyces sp. CC19균주는 0.5%, 20.2% 및 42.0%의 병반면적율을 나타내었지만, 무처리에서는 82.0% 발생하였다. 오이 흰가루병에 대한 비닐하우스 포장실험에서 B. subtilis B29, B. subtilis M10과 Streptomyces sp. CC19균주는 2.8%, 3.6%와 12.3% 발생하였지만 무처리에서 65.6% 발생면적을 나타내었다. 오이 잿빛곰팡이병에 대한 B. subtilis B29, B. subtilis M10과 Streptomyces sp. CC19균주는 8.0%, 30.8% 및 5.2%를 각각 나타내었고 무처리에서는 81.2%의 병반면적율을 나타내었다. 그러므로 B. subtilis B29균주는 오이에 발생하는 흰가루병, 노균병과 잿빛곰팡이병의 생물적 방제에 유망한 균주로 선발하였다.
재래식 메주의 맛을 지닌 코오지의 산업화를 유도하기 위해 재래식 메주에서 분리한 세균과 곰팡이를 이용한 코오지의 특성을 조사하였다. 코오지 제조에 선정된 곰팡이와 세균은 재래식 메주에서 분리한 Bacillus subtilis B-4와 Aspergillus oryzae F-5로, B. subtilis B-4와 A. oryzae F-5는 amylase와 protease 등의 효소분비능이 우수한 것이었다. B. subtilis B-4와 A. oryzae F-5를 각각 코오지 제조에 이용한 결과 색깔은 B. subtilis B-4로 제조한 코오지가 A. oryzae F-5로 제조한 코오지에 비해 약간 어두운 편이었고, 효소분비능은 B. subtilis B-4로 제조한 코오지가 A. oryzae F-5로 제조한 코오지에 비해 우수한 편이었다. 제국이 끝난 코오지에 대한 맛, 향, 색깔 및 전반적인 기호도에 대한 관능검사에서는 B. subtilis B-4로 제조한 코오지가 A. oryzae F-5로 제조한 코오지에 비해 색깔을 제외한 모든 면에서 높은 점수를 얻고 있었다. 수분활성도가 제국에 미치는 영향을 조사한 결과 수분활성도를 조절하여 제조한 코오지가 색도, 효소분비능, 생균수 및 관능검사에서 우수한 것으로 나타났다.
박테리오신 BSCX1은 Bacillus subtilis cx1에 의해 생산되는 항균성 peptide이다. B. subtilis cx1의 박테리오신(BSCX1)은 Bacilius subtilis ATCC6633, Listeria monocytogenes KCTC3569를 포함한 그람양성균과 Salmonella typhi ATCC19430, Escherichia coli ATCC25922와 같은 그람음성균에 대해서도 비교적 넓은 항균활성 범위를 가진다. 박테리오신 생산균주인 B. subtilis cx1과 그것의 감수성 균주인 B. subtilis ATCC6633을 공동 배양한 결과, 박테리오신 BSCX1의 생산이 증가됨을 확인할 수 있다. 이 결과는 박테리오신 생산균주 B. subtilis cx1의 성장 배지내에 박테리오신 감수성 균주가 존재함이 BSCX1 생산을 촉진시키는 것을 의미한다. 감수성 균주의 박테리오신 유도 작용을 확인하였으므로 유도물질이 감수성 균주의 어느 위치에 존재하는지 밝히기 위해 B. subtilis ATCC6633을 분획하여 실험한 결과 세포내 분획과 세포파쇄물에 모두 유도물질이 존재함을 확인하였다. BSCX1 유도물질의 유도활성은 pH 2.5에서 pH 9.5에 걸쳐 전 구간에서 유지되었으며, $50^{\circ}C$이상에서는 3시간 이내에 불활성화 되었다. 유도물질에 단백분해효소인 proteinase K를 처리한 결과 유도활성이 사라져 단백질성 물질임을 알 수 있었다.
본 연구는 E. tarda에 대한 특이박테리오phage와 생균제의 일종인 B. subtilis KM-1의 혼합제가 E. tarda에 대한 항균활성에 미치는 효과를 알아보고자 수행되었다. 가장 효과적인 혼합제의 항균활성을 보인 phage의 적정 수는 B. subtilis 가 $2{\times}10^5$ CFU/ml의 농도로 존재할 때 배양 36시간째 $2{\times}10^5$ PFU/ml로 나타났다. B. subtilis의 경우 $2{\times}10^5$ PFU/ml의 phage가 존재할 때 첨가량에 비례하는 항균활성을 보여주었다. phage와 B. subtilis의 혼합투여는 phage나 B. subtilis 각각을 투여한 그룹보다 훨씬 높은 항균활성을 나타냈다. 본 연구의 결과는 수산양식에서 대두되고 있는 양식어류의 E. tarda 감염성 질병 치료를 위한 항생제 대체제로서의 사료첨가제 개발 가능성을 제시하고 있다.
A gene encoding the mannanase of Bacillus subtilis WL-3, which had been isolated from Korean soybean paste, was cloned into Escherichia coli and the nucleotide sequence of a 2.7-kb DNA fragment containing the mannanase gene was subsequently determined. The mannanase gene, designated manA, consisted of 1,080 nucleotides encoding a polypeptide of 360 amino acid residues. The deduced amino acid sequence was highly homologous to those of mannanases belonging to glycosyl hydrolase family 26. The manA gene was strongly expressed in B. subtilis 168 by cloning the gene downstream of a strong B. subtilis promoter of plasmid $pJ27{\Delta}88U$. In flask cultures, the production of mannanase by recombinant B. subtilis 168 reached maximum levels of 300 units/ml and 450 units/ml in LB medium and LB medium containing 0.3% locust bean gum, respectively. Based on the zymogram ofthe mannanase, it was found that the mannanase produced by recombinant B. subtilis could be maintained stably without proteolytic degradation during the culture time.

Bacillus species were isolated from iru, a traditional fermented condiment in Nigeria. Polyphasic approach was used to evaluate the phylogenetic relationship and strain sub-type of the isolated species. Additionally, the phylogenetic profiles of the species isolated from iru were compared with those of bacilli isolated from different continents. The phylogenetic diversity analysis was performed using the combination of 16S rRNA gene sequencing, ITS-PCR, ITS-PCR-RFLP, and M13 RAPD-PCR. The analysis revealed that Bacillus subtilis U170B and B. subtilis U146A isolated from iru were the closest relatives of strains belonging to the phylogeny of B. subtilis sensu stricto and were related to other bacilli isolated from different continents that had functional benefits. The two isolated species exhibited resistance to acidic pH (pH 2.0). The survival rates of B. subtilis U170B, B. subtilis U146A, and B. clausii UBBC-07 (commercial probiotic strain) cultured at pH 2.0 for 3 h were 33.45, 12.44, and 9.53%, respectively. The strains were highly tolerant to bile salts [0.3% (w/v)]. B. subtilis U170B exhibited the highest cell viability (43.45%) when cultured for 3 h in the presence of bile salts, followed by B. subtilis U146A (25%) and B. clausii UBBC-07 (18.94%). B. subtilis U170B and B. subtilis U146A did not exhibit haemolytic activity and were susceptible to different antibiotics. Additionally, these two strains exhibited weak antagonistic activity against B. cereus. The diverse wild strains of B. subtilis can be used as a safe multifunctional starter culture for the industrial production of condiments with health benefits.

Bacillus subtilis and Bacillus amyloliquefaciens are closely related species that share a similar genomic background, and are both known to secrete large amounts of proteins directly into a medium. The extracellular proteomes of two strains of Bacillus subtilis and two strains of Bacillus amyloliquefaciens were compared by 2-D gel electrophoresis during the late exponential growth phase. The relative abundance of some minor protein spots varied among the four strains of Bacillus. Over 123 spots of extracellular proteins were visualized on the gel for B. subtilis CH 97, 68 spots for B. subtilis 3-5, 230 spots for B. amyloliquefaciens CH 51, and 60 spotsfor B. amyloliquefaciens 86-1. 2D gel electrophoresis images of the four Bacillus strains showed significantly different protein profiles. Consistent with the 2D gel electrophoretic analysis, most of the B. subtilis proteins differed from the proteases secreted by the B. amyloliquefaciensstrains. Among the proteins identified from B. subtilis, approximately 50% were cytoplasmic and 30% were canonically extracellular proteins. The secreted protein profiles for B. subtilis CH 97 and B. subtilis 3-5 were quite different, as were the profiles for B. amyloliquefaciens CH 51 and 86-1. The four proteomes also differed in the major protein composition. The B. subtilis CH 97 and B. amyloliquefaciens CH 51 proteomes both contained large amounts of secreted hydrolytic enzymes. Among the four strains, B. subtilis 3-5 secreted the least number of proteins. Therefore, even closely related bacteria in terms of genomic sequences can still have significant differences in their physiology and proteome layout.

The type strain Bacillus subtilis subsp. subtilis KCTC $3135^T$ was deeply sequenced and annotated, replacing a previous draft genome in this study. The tar and tag genes were involved in synthesizing wall teichoic acids (WTAs), and these genes and their products were previously regarded as the distinguishing difference between B. s. subtilis and B. s. spizizenii. However, a comparative genomic analysis of B. subtilis spp. revealed that both B. s. subtilis and B. s. spizizenii had various types of cell walls. These tar and tag operons were mutually exclusive and the tar genes from B. s. spizizenii were very similar to the genes from non-Bacillus bacteria, unlike the tag genes from B. s. subtilis. The results and previous studies suggest that the tar genes and the tag genes are not inherited after subspecies speciation. The phylogenetic tree based on whole genome sequences showed that each subspecies clearly formed a monophyletic group, while the tree based on tar genes showed that monophyletic groups were formed according to the cell wall type rather than the subspecies. These findings indicate that the tar genes and the presence of ribitol as a cell-wall constituent were not the distinguishing difference between the subspecies of B. subtilis and that the description of subspecies B. s. spizizenii should be updated.
