The hemolmyph and whole body of insect, Bombyx mori, Allomyrina dichotoma and Neotocia brevitarsi, conspicuously inhibited the mycelial growth of several plant pathogenic fungi. The hemolymph of 1087 strain among the 16 strains of B. mori has inhibition activities against the 3 species of fungi, Alternaria panax, Collctotrichum gloeosporioides, and Pyricularia oryzae. The whole body of B. mori was more effective than the hemolymph as a inhibitor on fungi growth. The antifungal activity of B. mori was variable to the fungi species. In addition, A. dichotoma and N. brevitarsi showed antifungal activities against the same fungi as did B. mori. These data showed that the insect has potent antifungal activity. Whereas, the level of activities were differ according to the fungal species. This finding underlines that the possibility of the insect can be use of the agent as a inhibitor against the plant pathogenic fungi.
Objectives : The Skin is composed of multiple layers, including the epidermis, dermis, and hypodermis. It provides a vital barrier structure that protects vertebrates from external environmental antigens, solvents, ultraviolet light, microorganisms, toxins, and weather conditions. Although several biological effects of Mori Follium have been reported, beneficial effects of Mori Follium in skin health remain unclear. In this study, we prepared water extract of Mori Follium (MLE) and evaluated the effects on melanin accumulation and expression levels of skin fibril-related proteins.Methods : The cytotoxicities of MLE in B16F10 melanoma and human skin fibroblasts (HSF) were examined by MTT assay. Inhibitory effect of MLE on the α-MSH- and IBMX-induced melanosis in B16F10 melanoma was examined. The expression levels of fibronectin, collagen 1α2, and CCN2 in MLE-treated HSF were analyzed by reverse transcription-polymer chain reaction (RT-PCR) and western blotting.Results : The MLE treatment for 24 h did not affect to the B16F10 and HSF at concentrations of 1, 10, 50, 100, 200, 400 and 800 ㎍/ml. The MLE treatment for 72 h significantly and dose dependently suppressed melanin accumulation in B16F10 melanoma. In addition, the MLE treatment up-regulated expression levels of skin fibril-related genes such as fibronectin, collagen 1α2, and CCN2 in HSF. Our western blot analysis revealed MLE-induced up-regulation of skin fibril-related genes required the activation of CCN2 protein.Conclusions : In conclusion, these findings suggest that the MLE could be used in development of cosmetic natural material of maintaining healthy skin.

Cathepsins are well-characterized proteases that are ubiquitously expressed in lysosomes. Previous work revealed that $Bombyx$$mori$ cathepsins B and D are expressed in the fat body and undergo decomposition during larval-pupal metamorphosis. Quantitative RT-PCR was performed to detect cathepsin gene expression at the transcription level when challenged by $B.$$mori$ nuclear polyhedrosis virus (BmNPV), temperature and hormones (20-hydroxyecdysone (20E) and juvenile hormone analogue (JHA)). mRNAs encoding cathepsins B and D were significantly enhanced after the larvae were infected with BmNPV, and the peak of the induction appeared at 1 day before spinning. This attenuated the inducing effect on cathepsin expression caused by infection. Temperature shock induced cathepsin expression at the later stage of the $5^{th}$ instar, and transcription levels varied with development stage and temperature. Cathepsin B and D mRNA expression in the fat body were significantly induced by JHA at the day before spinning, and with 20E, the expression reached a peak at the last day of the $5^{th}$ instar. Cathepsin B and D mRNA expression exhibited detectable changes post-treatment, without significant differences between or among the hormone concentrations.

The cytotoxic activity of MeOH extracts of the freeze-dried silkworm (Bombyx mori)-derived materials (4th instar larvae, female and mate pupae, virgin female and male adult), dried Beauveria bassiana-infected silkworm larvae, dried feces from the 4th instar larvae B. mori, and dried mulberry (Morus alba)-derived materials (leaves, fruits, root barks) in vitro was evaluated by sulforhodamine B assay, using the five human solid A 549 lung, SK-OV-2 ovarian, SK-MEL-2 melanoma, XF-498 CNS and HCT-15 colon tumor cell lines. The responses varied with both cell line and material used. The 70% hot MeOH extract of B. mori feces (BFH) revealed potent cytotoxic activity against model tumor cell lines whereas moderate activity was observed from the MeOH extract of B. mori feces. M. alba root barks, and M. alba fruits. The other test materials were ineffective. Because of its potent cytotoxic activity, the activity of each solvent fraction from the BFH was determined. Chloroform and ethyl acetate fractions showed the most potent cytotoxic activity. In conclusion, our results may be an indication of at least one of the pharmacological actions of B. mori feces. M. alba root barks, and M. alba fruits.
This study examined the enzymatic digestion, average molecular weight distribution and structural changes of Bombyx mori silk gland fibroin using gel penneation chromatography and nuclear magnetic resonance methods. The pure-separation of calcium chloride-treated fibroin hydrolysates was carried out by gel filtration chromatography. Also, the effects of fibroin's enzymatic hydrolysis were investigated using an edible enzyme. The average molecular weight of three hydrolysate samples (silk gland fibroin (SF), SF-calcium chloride (SFC), and SFC-enzyme) were measured to compare their characteristics. The molecular weights of SF, SFC, and SFC-enzyme were approximately 108,000, 65,000, and 1,000 Da, respectively. Finally, we determined the structural characteristic changes of the different enzymatically digested samples by $^{13}C$ nuclear magnetic resonance methods. For the enzymatically digested fibroin, the glycine $^{13}C^{\alpha}$ resonance indicated that the amino acid was dramatically changed and/or separated out; however, this was not shown for the normal Bombyx mori silk gland fibroin.
Flacherie symptom was found in the fifth instar larvae of silkworm, Bombyx mori. The bacterial pathogen was isolated from the hemolymph of the infected silkworm and identified. The isolated bacteria caused a significant flacherie pathogenicity to the fifth instar larvae of B. mori when $5{\times}10^{6}$ cfu (colony-forming unit) of the bacteria was injected into each larva. The infected larvae began to die at 6 days after injection and resulted in complete mortality at 10 days. The bacterium was identified as Staphylococcus gallinarum based on the morphological and physiological characteristics described in Bergey's manual. This identification was further supported by the characters of carbohydrate utility analyzed from a bacterial identification system ($MicroLog^{\circledR}$) and also by the molecular structure of 165-23S rDNA internal transcribed spaces. As an insecticidal action, S. gallinarum caused hemolymph septicemia by its cytotoxic effect on the hemocytes of B. mori.

The recombinant plasmids harboring a heterologous gene coding mouse endostatin were transfected and expressed stably in Trichoplusia ni Tn 5B1-4 cells and Bombyx mori BmN cells, respectively. Recombinant endostatin expressed in the stably transformed Tn 5B1-4 and BmN cells was secreted into the medium. BmN cells are relatively lower in maximum cell growth and recombinant endostatin production than Tn 5B 1-4 cells. Recombinant endostatin was also purified to homogeneity using a simple one-step ${Ni^2+}$ affinity fractionation method. Purified recombinant endostatin inhibited endothelial cell proliferation in a dose-dependent manner. The concentration at half-maximum inhibition $({ED_50})$ for recombinant endostatin was approximately 0.35 ${\mu}g$/ml.

The aim of this study was to investigate the potential effects of extracts from silkworm Bombyx mori L. fermented with Bacillus subtilis KACC 91157 at levels of 5%(v/w) and 10%(v/w) in Sprague-Dawley rats intoxicated with 1%(w/w) orotic acid (OA) for 10 days. The rats were divided into a normal group (N), a control group (C: OA), and treatment groups (SP10: OA + 10% extracts from B. mori L.; BSP5: OA + 5% extracts from B. mori L. fermented with B. subtilis KACC 91157; BSP10: OA + 10% extracts from B. mori L. fermented with B. subtilis KACC 91157). Serum activities of aspartate aminotransferase (AST), alanine transferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) increased following OA feeding, but the rise was slightly reduced by administration of BSP10. The total lipid, free fatty acid, phospholipid, total cholesterol, and triglyceride contents in serum were significantly lower in the OA treatment groups than in the N group. However, the contents slightly increased following the administration of BSP10. Glutathione concentrations in liver and serum were reduced in the OA-induced fatty liver, but they increased following the administration of BSP10. Hepatocytes in the OA-induced fatty liver contained numerous large droplets. However, SP10, BSP5, and BSP10 feeding prevented OA-induced lipid droplet accumulation in hepatocytes. Accordingly, extracts from silkworm powder fermented with B. subtilis could be an ideal material as a dietary supplement in healthy functional foods to improve the effects of fatty liver.

Allatotropin is a 13-residue amidated neuropeptide isolated from pharate adult heads of the tobacco hornworm, Manduca serta and strongly stimulates biosynthesis of juvenile hormones in adults, but not larval, lepidopteran corpora allata. From a Bombyx mori midgut cDNA library, a cDNA that encodes a 130-amino-acid polypeptide containing M. sexta allatotropin sequence was isolated. The B. mori allatotropin cDNA consists of 1196 nucleotides. (omitted)
Pulmonary edema is a disease involving the principal symptoms: dyspnea, bloody phlegm, asthma, cough, etc. According to oriental medical references, Jeomglyukdaejosapaetang (J.D.T) was efficacious for dropsy, cough, dysnea, etc, so it was thought to be used for remedy of pulmonary edema. Therefore experimental study was performed to investigate the effects of J.C.T on pulmonary edema of the rats induced by the herbicide, paraquat.Thus the survival rats, respiratory rats, lung weights and histopathological view of the lungs of rats were studied. The results are as follows. 1. The survival rates at 72hours of the rats injected with paraquat and treated with J.D.T increased in B group(J.D.T + Cortex Lycii Radicis + Cortex mori). The others were not different with the control. 2. The respiratory rates of the rats which survived 72hours later significantly decreased in B group(J.D.T + Cortex Lycii Radicis + Cortex mori). 3. The lung weights of the rats which survived for 72hours later significantly decreased in B group(J.B.T + Cortex Lycii Radicis + Cortex mori). 4. The histopathological views of the lungs of rats induced by paraquat were seen severe hemorrhage, edema and some broken alveoli in control group. But B group(J.D.T + Cortex Lycii Radicis + Cortex mori) were seen little hemorrhage and interstitial hyperplasia. According to the above results, J.D.T + Cortex Lycii Radicis + Cortex mort is effective on the remedy for pulmonary edema of rats induced by paraquat.
