Characterization of Rhizobacteria Isolated from Family Solanaceae Plants in Dokdo Island (독도에 서식하는 가지과식물로부터 분리된 근권세균의 특성)

  • To characterize plant root-associated bacteria in wild plant family Solanaceae, Solanum nigrum L. plants were collected in Dokdo island. Forty four strains of nitrogen-fixing or spore-forming bacteria were isolated from rhizosphere of Solanum nigrum L. plants. Among these, 19 strains were able to produce auxin. Thirteen strains of these produced siderophore as determined by color reaction on CAS-blue plate, 8 strains were able to solubilize phosphate. The 16S rDNA genes of the isolated bacteria were amplified and sequenced. Model plants, pepper and tobacco, were established in order to evaluate the bacterial capacities eliciting growth promotion and induced systemic resistance. The plants treated with strain KUDC1009 were more resistant and capable of growth-promotion than control plants when challenged by either Xanthomonas axonopodis pv. vesicatoria or Erwinia carotovora sub. carotovora strain SCC1. Rhizobacteria isolated from Dokdo island can promote growth of wild type Solanum nigrum L. under much environmental stresses.

  • In order to develop a simple and reproducible protocol for red top bentgrass (Agrostis alba L.), effect of different growth regulators was investigated for embrogenic calli induction and subsequent plant regeneration using mature seeds. MS medium containing 2 mg/L 2,4-D was optimal for embryogenic callus induction from mature seeds. The highest plant regeneration frequency (64.4%) was showed when the embryogenic callus tissues were cultured on N6 medium supplemented with 0.5 mg/L 2,4-D and 2 mg/L BA. Regenerated plantlets were grown normally when shoots transplanted to the soil. A high-frequency and efficient regeneration system from mature seeds would be helpful for molecular breeding of new variety of red top bentgrass through Agrobacterium-mediated genetic transformation.

  • Present experiment were attempted to examine in vitro multiplication throughbud culture of Cornus officinalis. Bud derived shoot formation was established successfully on Murashige and Skoog's medium supplemented with $0.5mg\;/\;{\ell}$ BAP(N-benzyl amino purine). The shoot proliferation increased on the Driver Kuniyuki Walnut medium containing $0.5mg\;/\;{\ell}$ NAA(Napthalene acetic acid) and $0.5mg\;/\;{\ell}$ BAP. Addition of 2,4-D(2,4-Dichlorophenoxy acetic acid) to the media produced excessive callus inducton. IAA(Indole-3-acetic acid) and IBA (Indole-3-bu-tyric acid) enhanced multple shooting, and NAA showed callus induction and multiple shooting. Shoot growth was enhanced supplemented with 3% sucrose, $2g\;/\;{\ell}$ activated charcoal, and 1 / 4MS in organic salts. However, root formation of proliferated shoots was low about 5%

Growth Promotion of Tomato Plant under Drought Conditions by Treatment of Rhizobacteria Producing ACC Deaminase and Phytohormones (ACC Deaminase와 식물호르몬 생성 세균 처리에 의한 토마토 식물의 가뭄 조건에서의 생장)

  • Some rhizobacteria producing 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase can make plant to continue growth under the stress conditions through lowering the level of phytohormone, ethylene which inhibits the plant growth and accelerates plant aging. In this study, some rhizobacteria producing ACC deaminase have been isolated from the rhizosphere of plants grown at sand beaches, and identified as Escherichia hermannii m-2, Enterobacter asburiae m-4, Pseudomonas thivervalensis BD2-26 and Pseudomonas brassicacearum subsp. neoaurantiaca BD3-35 through sequencing of 16S rRNA genes. Strain BD3-35 showed the highest activity of ACC deaminase among the isolates, 20.26 ${\alpha}$-ketobutyrate ${\mu}M/mg$ protein/h. Strains BD3-35 and BD2-26 secreted a phytohormone cytokinin, and strains m-4 and m-2 could produce auxin and abscisic acid, respectively. When these bacteria were applied to the 7-day old tomato plant under drought stress for 7 days, strains BD3-35, m-2, and m-4 increased the length of tomato root by 14, 15, and 35%, respectively, and strains m-2, BD2-26 and BD3-35 increased the dry weight of tomato plant by 22, 33, and 68%, respectively compared to the uninoculated control tomatoes. Therefore, these rhizobacteria may be utilized as a microbial fertilizer for the plants under drought stress.

  • Callus of Rhodiola sachalinensis A. Bor were induced from leaf explant on l/2MS solid supplemented with combination of auxin (2.4-D, NAA: 0.1∼2mg/L) and cytokinin (BA: 0.1∼0.2mg/L). The effects of various medium, culture conditions and phytohormones on the growth of callus were investigated. MS, WPM, B$_{5}$ medium and diluted or concentrated media (1/2X, 2X, 3X) were used to investigate the growth of callus on each media. Among these, the highest growth was observed when cultured in in 2B$_{5}$ medium containing 0.5mg/L NAA and 1mg/L BA, 3% sucrose and 49.6 mM KNO$_3$ as nitrogen source, and 2.16mM NaH$_2$PO$_4$ as phoshate source at $25^{\circ}C$ in the dark. The calli cultured with 5% sucrose produced high salidroside content (0.41% on the basis of dry wt) than normal root (0.17%).

Efficient Plant Regeneration from Mature Seed-derived Callus of Italian ryegrass (Lolium multiflorum Lam.) (이탈리안 라이그래스의 성숙종자 유래 캘러스로부터 효율적인 식물체 재분화)

  • As an initial step for future genetic manipulations to improve forage characteristics of Italian ryegrass (Lolium multiflorum Lam.), an efficient tissue culture system was established and the factors affecting plant regeneration were evaluated. MS medium containing 5mg/L 2,4-D was optimal for embryogenic callus induction from mature seed and had a strong effect on successive plant regeneration. The plant regeneration frequency was observed at above 70% when embryogenic calli induced were transferred to N6 medium supplemented with 1mg/L 2,4-D and 5mg/L BA. Among several basic media tested, MS and N6 medium were optimal for callus induction and plant regeneration, respectively. Genotype was an important factor in plant regenerability. 'Jeanne' showed the highest regeneration frequency of 73%. A short tissue culture period and high-frequency regeneration system established in this study will be useful for molecular breeding of Italian ryegrass through genetic transformation.

Auxin and Cytokinin Affect Biomass and Bioactive Compound Production from Adventitious Roots of Eleutherococcus koreanum (섬오갈피 부정근 배양 시 오옥신과 사이토키닌이 생장과 생리활성물질 생산에 미치는 영향)

  • In an attempt to improve biomass and bioactive compound production, we cultured adventitious roots of $Eleutherococcus$ $koreanum$ in 250 mL Erlenmeyer flasks using Murashige and Skoog (MS) medium with different concentrations of auxins (IBA, NAA, IAA) and cytokinins (BA, kinetin, TDZ). Root biomass (fresh and dry weight) was enhanced at $5mg{\cdot}L^{-1}$ indole-3-butyric acid (IBA) after 5 weeks of culture. The content of total phenolics and flavonoids was also increased with $5mg{\cdot}L^{-1}$ IBA compared to ${\alpha}$-naphtalene acetic acid (NAA) or indole-3-acetic acid (IAA) treatments. The combination of $5mg{\cdot}L^{-1}$ IBA with $0.1mg{\cdot}L^{-1}$ thidiazuron (TDZ; N-phenyl-N'-1,2,3,-thidiazol-5-ylurea) enhanced root fresh and dry weight (1.4- and 1.6-fold, respectively) as well as the content of total phenolics and flavonoids compared to the relative control (without cytokinin). On the contrary, $N_6$-benzyladenine (BA) and 6-furfurylaminopurine (kinetin) did not significantly affect root biomass and bioactive compound production in adventitious roots of $E.$ $koreanum$. These results suggested that $5mg{\cdot}L^{-1}$ IBA combination with $0.1mg{\cdot}L^{-1}$ TDZ supplementation was most suitable for both biomass and bioactive compound production from adventitious roots of $E.$ $koreanum$.

  • To optimize tissue culture responses for genetic transformation of Kentucky bluegrass, the effects of culture medium supplements on tissue culture responses were investigated with mature seeds of a cultivar 'Newport' as explant tissues. The optimal concentration of 2,4-D (2.4-dichloro phenoxy acetic acid) for the induction of embryogenic callus from mature seed was 3 mg/L. Plant regeneration frequency was 54% when embryogenic callus was cultured on the regeneration medium supplemented with 1 mg/L 2,4-D and 3 mg/L of BA (6-benzyladenine). Addition of 1 g/L of casein hydrolysate and 500 mg/L of L-proline improved frequencies of embryogenic callus induction and plant regeneration up to 60.8% and 58.3%, respectively. Regenerated plants were grown normally when shoots transplanted to the soil. A rapid and efficient plant regeneration system established in this study. We suggest that the results may be useful for molecular breeding of Kentucky bluegrass through genetic transformation.

In Vitro Selection for Salty Tolerance of Populus nigra ${\times}$ P. maximowiczii (포푸라 내염성(耐塩性) 개체(個體)의 기내선발(器內選拔))

  • The concentration of 50 mM Nacl inhibited the shoot growth of P. nigra ${\times}$ maximowiczii in viro culture. The punctured leaves have produced so many individuals on MS basal medium with cytokinin. Especially MS basal medium with BAP 0.8mg/l showed the best shoot performance in which the average number of shoots were 127.6. For selection of NaCl tolerance shoot of poplar, punctured leaves were inoculated on MS basal medium with BAP 0.8mg/l and various concentrations of NaCl(from 10 mM to 100 mM graded by 10 mM). On the medium with over 50mM of NaCl, 13.7 to 15.7 shoots were obtained. Especially on the medium with go mM and 100mM. 10.7 and 8.3 shoots, respectively. The shoots derived from control medium (non-NaCl) were depressed growth, while the selected shoots from MS with NaCl showed good growth performance on MS basal medium with 50 mM of NaCl. From this results, we suggested that the possibility of in vitro selection to tolerance for inorganic salts in forest tree species.

Effect of Plant Growth Regulators on in vitro Cultured Atractylodes Hybrid 'Dachul' (A. macrocephala x A. japonica) (기내배양 백출 교잡종 '다출'(Dachul, Atractylodes macrocephala x A. japonica)에 미치는 생장조절제처리효과)

  • This study was conducted to establish the tissue culture system for Atractylodes plant which is most frequently used in oriental medicine. Root and auxiliary bud of Dachul cv., which is Atractylodes hybrid (A. macrocephala x A. japonica), were used as target tissues for in vitro culture. In root culture, callus induction rate was higher in the treatment of BAP combined with NAA than others, however, 2-iP was more effective for callus proliferation and root induction. Although calli were effectively induced from the root and proliferated in lower concentration of cytokinin combined with higher auxin, root tissue was inappropriate for shoot regeneration. For plant regeneration with axillary bud, BAP combined with NAA was more effective than 2-iP with NAA or IBA. Number of regenerated plant per bud was 3.8, which was highest, and stem diameters was shown as 5.0mm under the conditions of 1 mg/L BAP combined with 1 mg/L NAA. Although, plant height was tend to be higher in 2-iP than BAP, number of the regenerated plant was lower via versus. Furthermore, root proliferation of regenerated plant was more effective in higher concentration of sucrose (7%) than in lower concentration (3%). In results, auxiliary bud was an efficient target tissue for producing regenerated plant of Atractylodes under the conditions of 1 mg/L BAP combined with 1 mg/L NAA and higher concentration of sucrose was effective for root proliferation of regenerated plants.
